[Study on quality of crude ramulas cinnamomi in different reserve period and their prepared products].

OBJECTIVE: To compare quality of crude Ramulas Cinnamomi in different reserve period and their prepared products. METHOD: Analyzing the content of volatile oil by pharmacopoeia method. RESULTS: The content of volatile oil in prepared Ramulas Cinnamomi by frying decreased, while that in the prepared Ramulas Cinnamomi with honeybee increased a little. The longer the reserve time was, the less the content of volatile oil were. The density becomed smaller in proper order according to crude Ramules Cinnamomi, product processed with honeybee and product processed by frying. But different store time and preparing methods hardly influenced the component of its volatile oil. CONCLUSION: It's useful for Ramulas Cinnamomi to be prepared by frying or with honeybee.

Externalization and internalization of cardiac endothelin receptors during different phases of sepsis in rat.

OBJECTIVE: To study the redistribution of endothelin-1 (ET-1) receptors in two subcellular organelles, the sarcolemmal membrane and the light vesicle, of rat heart during the progression of sepsis. METHODS: Sepsis was induced by cecal ligation and puncture (CLP). ET1 receptor was assayed by using [125I]-ET1 binding. Marker enzyme activities, protein yield, and dry-to-wet weight ratio of cardiac membranes were measured. RESULTS: Septic rat heart exhibited two distinct phases: an initial hyperdynamic phase( 9h after CLP; early stage of sepsis) followed by a hypodynamic (18h after CLP, late stage of sepsis) phase. [125I]-ET1 binding study showed that during early stage of sepsis, the B(max) of ET1 receptors was increased by 30% in sarcolemma but decreased by 19% in light vesicles, while during late stage of sepsis, the B(max) was decreased by 24% in sarcolemma but increased by 38% in light vesicles. The total binding of sarcolemma and light vesicles was increased by 25% during early stage of sepsis but decreased by 17% during late stage of sepsis. CONCLUSIONS: These data indicated that ET1 receptors in the rat heart were externalized from light vesicles to sarcolemmal membranes during early hyperdynamic phase while internalized from surface membranes to intracellular compartment during late hypodynamic phase of sepsis.

Optical patternation: a technique for three-dimensional aerosol diagnostics

A novel technique based on optical patternation is described for three-dimensional diagnostic studies of aerosols used in analytical spectroscopies. The aerosol is illuminated with a thin laser light sheet to capture images of the fluorescence and Lorenz-Mie light-scattering signals from the aerosol field with a charge-coupled detector. These measurements allow for the rapid and nonintrusive elucidation of two-dimensional spray structures, planar mass distributions, and spatial droplet size distributions. The ratio of the fluorescence image to the Lorenz-Mie image is then utilized to construct a spatially resolved map of the volume-to-surface area mean of the aerosol (Sauter mean diameter). Three-dimensional maps of spray structure, mass distribution, and droplet size distribution are obtained for the entire aerosol field by image stacking. The technique is applied to the measurement of the droplet size over the aerosol field at distances of 5-30 mm from the nebulizer tip where droplet sizes ranged from 6 to 12 microm for a direct injection high efficiency nebulizer used in inductively coupled plasma spectrometries.

Reduction of aflatoxin B(1) adduct biomarkers by oltipraz in the tree shrew (Tupaia belangeri chinensis).

The risk of liver cancer is greatest in people both infected with hepatitis B virus (HBV) and highly exposed to aflatoxin B(1) (AFB(1)). The tree shrew (Tupaia belangeri chinensis) is a unique species that can be infected with human HBV, is susceptible to AFB(1)-induced liver cancer, and shows a synergistic interaction between HBV and AFB(1) for liver cancer. In this regard, the tree shrew may be useful for evaluating experimental chemoprevention strategies relevant to high-risk human populations as it mirrors the human epidemiology of liver cancer. To begin developing the model for chemoprevention study, two groups of tree shrews were fed 400 microg AFB(1)/kg b.wt. in milk daily for 4 weeks. One week prior to AFB(1) administration, one group also received oltipraz (0.5 mmol/kg, p.o.) daily for 5 weeks. At weekly intervals, 1 ml of blood and a 24-h urine sample were obtained from each animal. Aflatoxin-albumin adducts in serum were determined by a radioimmunological assay and aflatoxin-N(7)-guanine adducts in urine were measured by HPLC. Aflatoxin-albumin adducts increased rapidly in 2 weeks to plateau at 20 pmol/mg protein, and they diminished after cessation of AFB(1) exposure. Oltipraz significantly attenuated the overall burden of aflatoxin-albumin adducts throughout the exposure period with a median reduction of 80%. In a single cross-sectional analysis at the end of AFB(1) dosing, oltipraz treatment decreased urinary aflatoxin-N(7)-guanine by 93%. Collectively, these results indicate that oltipraz reduces AFB(1) risk biomarkers in the tree shrew in a manner similar to that observed in rodents and humans, and establishes a rationale to evaluate cancer chemoprevention by oltipraz in human HBV-infected, AFB(1) exposed tree shrews.

Development of a human stanniocalcin radioimmunoassay: serum and tissue hormone levels and pharmacokinetics in the rat.

Stanniocalcin (STC) is a polypeptide hormone that was first discovered in fish and recently identified in humans and other mammals. In fish STC is produced by one gland, circulates freely in the blood and plays an integral role in mineral homeostasis. In mammals, STC is produced in a number of different tissues and serves a variety of different functions. In kidney, STC regulates phosphate reabsorption by proximal tubule cells, whereas in ovary it appears to be involved in steroid hormone synthesis. However there is no information on circulating levels of STC in mammals or the regulation of its secretion. In this report we have developed a radioimmunoassay (RIA) for human STC. The RIA was validated for measuring tissue hormone levels. However human and other mammalian sera were completely devoid of immunoreactive STC (irSTC). To explore the possibility that mammalian STC might have a short half-life pharmacokinetic analysis was carried out in rats. STC pharmacokinetics were best described by a two compartment model where the distribution phase (t1/2(alpha)) equaled 1 min and the elimination phase (t1/2(beta)) was 60 min. However the STC in the elimination phase no longer crossreacted in the RIA indicating it had undergone substantial chemical modification, which could explain our inability to detect irSTC in mammalian sera. When we compared the pharmacokinetics of human and fish STC in mammalian and fish models the human hormone was always eliminated faster, indicating that human STC has unique structural properties. There also appears to be a unique clearance mechanism for STC in mammals. Hence there are major differences in the delivery and biology of mammalian STC. Unlike fishes, mammalian STC does not normally circulate in the blood and functions instead as a local mediator of cell function. Future studies will no doubt show that this has had important ramifications on function as well.

LIGHT, a TNF-like molecule, costimulates T cell proliferation and is required for dendritic cell-mediated allogeneic T cell response.

LIGHT is a recently identified member of the TNF superfamily and its receptors, herpesvirus entry mediator and lymphotoxin beta receptor, are found in T cells and stromal cells. In this study, we demonstrate that LIGHT is selectively expressed on immature dendritic cells (DCs) generated from human PBMCs. In contrast, LIGHT is not detectable in DCs either freshly isolated from PBMCs or rendered mature in vitro by LPS treatment. Blockade of LIGHT by its soluble receptors, lymphotoxin beta receptor-Ig or HVEM-Ig, inhibits the induction of DC-mediated primary allogeneic T cell response. Furthermore, engagement of LIGHT costimulates human T cell proliferation, amplifies the NF-kappaB signaling pathway, and preferentially induces the production of IFN-gamma, but not IL-4, in the presence of an antigenic signal. Our results suggest that LIGHT is a costimulatory molecule involved in DC-mediated cellular immune responses.

Deletion of the GPG motif in the HIV type 1 V3 loop does not abrogate infection in all cells.

The three amino acids glycine, proline, and glycine (GPG) constitute a conserved motif at the center of the V3 loop of HIV-1 surface glycoprotein 120. It has been indicated that deletion of this GPG motif is lethal for viral infectivity and abrogates the ability of the virus to form syncytia. In the present work, we studied the effects of GPG deletion on viral infectivity, cell tropism, syncytium formation, and initiation of apoptosis by constructing a mutant provirus based on the infectious clone pBRu-2. Successful infection and replication of GPG-deleted virus were detected in MT-2 cells, although the mutant virus showed lower infectivity. Infection could also be observed in the C8166, C91-PL, Molt-3, and THP-1 cell lines, and in PBMC-derived dendritic cells (DCs), but not in CEM-SS, HUT78, H9, Jurkat, and U937 cell lines or in PBMCs. Mutant virus also induced syncytia and apoptosis in the MT-2 cells. An intact GPG motif is probably necessary for unimpaired induction of fusion in some HIV-1-permissive cells. However, once the virus enters the cells, the GPG sequence does not seem to be indispensable for syncytium formation or apoptosis induction in MT-2 cells. Our data also imply that cell surface molecules other than CD4 and CXCR4 may be involved in entry of the GPG-deleted virus.

Near-tetraploid minimally differentiated acute myeloid leukemia with extensive erythrophagocytosis by leukemic blasts.

Numerical change of chromosomes is common in acute myeloid leukemia (AML). However, a chromosome number as high as near-tetraploidy is very rare, especially in minimally differentiated AML (AML-M0). Erythrophagocytosis by reactive or malignant histiocytes is common in malignant hematological diseases; however, erythrophagocytosis by leukemic blasts is also very rare, especially in AML-M0. We report here the first case of AML-M0 with both of these unique characteristics: a near-tetraploid karyotype and erythrophagocytosis by leukemic blasts.

Expression of inducible nitric oxide synthase (iNOS/NOS II) in the hydropic cochlea of guinea pigs.

Immunohistochemical investigations of the guinea pig cochlea, using a specific antibody to the inducible isoform of NO synthase (iNOS/NOS II), have been performed 3 weeks after closure of the right endolymphatic duct (n=7). Endolymphatic hydrops, the morphological substrate of Meniere's disease, became evident by distension of the Reissner's membrane. iNOS expression could be noted in endothelium, spiral ganglion cells, in nerve fibers, in supporting cells of the organ of Corti and cells of the spiral ligament. Temporal bones of non-operated controls (n=6) as well as of sham-operated animals (n=3) did not show structures positive to iNOS. These findings imply that iNOS-generated NO could be involved in the pathophysiology of cochlear dysfunction in Meniere's disease.

A new cembranolide from the soft coral Sinularia capillosa.

A new cembranolide, capillolide (1), and three known cembranolides were isolated from the soft coral Sinularia capillosa collected from the South China Sea. Their structures and the relative stereochemistry of 1 were deduced on the basis of spectroscopic methods.

The time-effect relationship of central action in acupuncture treatment for weight reduction.

PURPOSE: To study the time-effect relationship of action of acupuncture on the satiety center of ventromedial nucleus of hypothalamus (VMH) in rats with experimental obesity. METHODS: Microelectrode recording method of nerve cells and stereotaxic technique for brain were adopted with discharge frequency (Hz/s) of nerve impulse in VMH as the index, and the time-effect of acupuncture action in different periods between groups were observed. RESULTS: Electric activity in acupuncture group was higher than in the obesity model group (P < 0.001) and the normal group (P < 0.01) respectively, and the curve of acupuncture action within 2 hours showed irregular fluctuation. CONCLUSION: Acupuncture can increase excitability of the satiety center, with a better long-term effect.

[Clinical study of prostate cancer: statistical analysis of 107 cases in the past 12 years].

One hundred and seven patients with prostate cancer were treated at Mie University Hospital during the past 12 years between 1988 and 1999. They were between 53 and 83 years old, with an average age of 70.8 years old. The clinical stage was defined as A, B, C and D in 3 (2.8%), 19 (17.8%), 50 (46.7%) and 35 (32.7%) patients, respectively. At initial diagnosis, the tumor was well, moderately and poorly differentiated adenocarcinoma in 26 (24.3%), 47 (43.9%) and 34 (31.8%) patients, respectively. The median follow-up period was 52.3 months. The overall 1, 3 and 5-year survival rates were 98.0%, 86.8% and 75.2%, respectively. The 5-year survival rates for stage A, B, C and D were 100%, 93.8%, 82.1% and 56.9%, respectively. A significant difference (p = 0.017) in 5-year survival rate was noted between stage C and D. The 5-year survival rate was 100% for well differentiated, 78.0% for moderately differentiated, and 53.2% for poorly differentiated adenocarcinoma. A significant difference (p = 0.0016) in the 5-year survival rate was noted between well differentiated and poorly differentiated adenocarcinoma. According to the therapy, the 5-year survival rate in stage C was 86.2% for the radical prostatectomy group and 84.0% for the endocrine therapy group. There was no significant difference between these 2 treatment groups. Endocrine therapies, classified into maximum androgen blockade (MAB) and endocrine therapy other than MAB were performed for stage D as an initial therapy. Although the prognosis in the patients treated with MAB was better than that with other endocrine therapies, there was no significant difference between these 2 endocrine treatment groups.

[Resistance and efficacy of treatment in relapse pulmonary tuberculosis].

OBJECTIVE: To investigate the resistant status and factors involved and to evaluate the efficacy of treatment in relapse pulmonary tuberculosis. METHODS: The analysis of history of chemotherapy and resistant status in 448 relapse pulmonary tuberculosis cases with culture positive and susceptibility test was conducted. All new registered relapse cases during 1988-1997 in Beijing were treated with regimen of 2RHZS(E)/4RHE or 2RHZS(E)/6RH under fully supervision and prognosis was evaluated with cohort analysis. RESULTS: Of 448 relapse cases 153 (34.2%) were resistant at least to 1 antituberculosis drug, 20.9% resistant to INH, 17.6% to SM, 11.1% to RFP, 7.8% to PAS, 2.9% to EMB and 6.2% resistant to both INH and RFP. The resistance rates in the cases of compliance and default in initial treatment were 22.3% and 55.8%, respectively, and resistance to RFP were 6.3% and 20.8% respectively. The sputum conversion rate in relapse cases during 1988-1997 were 80.5%-89.6%. CONCLUSIONS: The resistant rate in relapse cases in Beijing was 34.1%. The higher compliance in the chemotherapy, the lower resistant rate, conversely the higher default the higher resistance. The majority of resistant cases were resistant to INH, SM and RFP, and it is important in making a rational regimen.

[Role of nitric oxide in heart failure].

The benefit effects of nitric oxide (NO) donors in acute heart failure have led to the development of vasodilators as treatment of chronic heart failure. However, the mechanisms involved in the effects of NO are complex and still discussed. In chronic heart failure, the eNOS downregulation in vascular endothelium explains the alteration of endothelial function. In addition, in the myocardium, cytokines induce the expression of inducible nitric oxide synthase (iNOS) which increase NO production by myocytes and surrounding cells. This excess of NO production, associated with anion superoxide synthesis, limits the inotropic properties of catecholamines and exert proapoptotic effects. The role of NO donors in heart failure treatment is still controversial but by reducing preload they improve patient's symptoms. Beside blockade of the renin-angiotensin system, the angiotensin converting enzyme inhibitors act via the inhibition of bradykinin degradation which increase NO levels. Finally, vascular endothelial NO expression is improved by exercise training and participates in the improvement of exercise capacity in patients with chronic heart failure involved in cardiac readaptation program.

[Optimized models of logging-tending system in cutting areas].

The comprehensive advantages of different logging-tending systems in Pinus massoniana forest cutting area were evaluated by set-pair analysis, based on the comparison of their economic and ecological benefits. The results showed that the optimized model for P. massoniana forests in Northern Fujian comprised 40% selective cutting, manual skidding, clear-cutting in ribbon, and natural regeneration with artificial aids, which could also be used in the nearby forests with conditions similar to the experimental area.

Alternation of Na(+)-Ca2+ exchange in rat cardiac sarcolemmal membranes during different phases of sepsis.

OBJECTIVE: To study the alteration of Na(+)-Ca2+ exchange in rat cardiac sarcolemmal membrane during phases of septic shock. METHODS: Sepsis was induced by cecal ligation and puncture (CLP). Na(+)-Ca2+ exchange was assayed by radioactive analysis. RESULTS: Na(+)-dependent 45Ca2+ uptake was decreased by 62%-69% in late phase of sepsis, whereas it was not affected in early phase of sepsis. Na(+)-Ca2+ exchange stimulated by 5' guanylyl imidodiphosphate [Gpp (NH) p] was decreased by 65.7% in late phase of sepsis but unaltered in early phase of sepsis. Two agonists (angiotensin II and phenylephrine) coupled to Gq and a protein kinase C activator, phorbol 12-myristate 13-acetate (PMA) all inhibited Na(+)-Ca2+ exchange in late phase of sepsis. Na(+)-Ca2+ exchange activities induced by phosphorylation of Na(+)-Ca2+ exchange were decreased in late phase of sepsis, whereas inhibition of Na(+)-Ca2+ exchange by dephosphorylation was increased both in early and late phases of sepsis. CONCLUSION: The alteration of Na(+)-Ca2+ exchange during different phases of sepsis might be related to the activities of Gq, protein kinase C, and phosphorylation/dephosphorylation.

[Evidence for TCR Vbeta clonal expansion T cells in a patient with cGVHD after unrelated donor BMT].

OBJECTIVE: To investigate the distribution and clonal expansion of TCR Vbeta subfamily T cells in patients with cGVHD after unrelated donor bone marrow transplantation (BMT). METHODS: The CDR3 of 24 TCR Vbeta subfamily genes were amplified from peripheral mononuclear cells of a patient with cGVHD after unrelated BMT. RT-PCR was used for detection of the distribution of TCR Vbeta repertoire, and the PCR products were further analyzed by genescan technique for the CDR3 size, to evaluating clonality of the detectable TCR VbetaT cells. RESULTS: Only 8 Vbeta subfamily T cells could be identified in the patient, clonal expansion T cells could be found of Vbeta2, 3, 8 and 13 subfamilies. CONCLUSIONS: The skew distribution and clonal expansion of TCR Vbeta subfamily T cells could be found in the patient. It may relate to the initiation of cGVHD.
