Food Contamination

Porcine cytomegalovirus (PCMV) in early gestation.

Following intranasal exposure to PCMV at or within 48 h of coitus transplacental infection occurred in two groups of gilts. Five out of 22 embryos were infected in the first group but only 2 out of 63 in the second. A more rapid immune response as measured by circulating antibody was probably instrumental in abrogating infection in the second group. In the infected embryos the virus localized in leptomeningeal cells, hepatic sinusoidal cells, peritoneal macrophages, periosteal cells and occasional alveolar cells, but the placenta did not appear to be a primary site of viral replication.

Superinfection with porcine cytomegalovirus initiating transplacental infection.

Four sows with circulating antibody were exposed to porcine cytomegalovirus. Virus was detected in 8 of 24 foetuses by immunofluorescence and/or virus isolation from 2 sows with low levels of antibody. In 6 of the infected foetuses, the virus was in capillary endothelium and macrophages of the lung, and was associated with interlobular oedema in 2 of these. Virus was also detected in the nasal mucosa, spleen and brain. The majority of the virus-positive foetuses were grossly normal.

Border disease of sheep: spinal cord morphometry.

Semi-automated planimetry was used to determine cross-sectional areas of spinal grey and white matter by direct microscopy of paraffin sections of spinal cord from Border disease (BD) and normal lambs at segments C4, C8, T6 and L4. Spinal cord cross-sectional area was significantly reduced in cases of BD produced by either intramuscular or intranasal inoculation of ewes in the first half of pregnancy with Weybridge strains of BD virus. The reduction was apparent at all 4 levels and in both grey and white matter, though the white matter was the more severely affected. Spinal cord areas tended to be smaller in the progeny of ewes exposed earlier in gestation or to larger doses of virus, but these differences were not statistically significant.

Rhinitis of pigs: A gross staining method for macroscopic examination of the nasal mucosa.

A phloxine-alcian blue (PhAB) method has been used to stain the porcine nasal mucosa for macroscopic examination. Applied to either fresh or fixed specimens it stains respiratory epithelium dark blue and non-cilated epithelium, including the metaplastic epithelia of rhinitis, bright pink. The PhAB technique is simple, inexpensive and dependable, and is suitable for routine application in the post mortem room. Stained and fixed specimens may be preserved virtually indefinitely.

Alcian Blue

Porcine atrophic rhinitis: snout radiography as an aid to diagnosis and detection of the disease.

Snout radiography is a useful and practicable aid to the diagnosis of atrophic rhinitis. It is also a fairly reliable means of detecting and measuring turbinate atrophy and distortion of the septum nasi in the live pig. Suitable physical and pharmacological methods of restraining pigs for snout radiography are described and guide lines for producing satisfactory, standard snout radiographs are given. Parameters and criteria are recommended for qualitative and quantitative methods of interpreting snout radiographs. The quantitative scoring system approximates the macroscopic post mortem snout scoring system generally employed in Britain and provides data which might be used in genetic or other control programmes. Radiographic standards are tentatively suggested for certifying pigs free from gross lesions of atrophic rhinitis for sale or export purposes.
