Synthesis and activity of NAcSerAspLysPro analogues on cellular interactions between T-cell and erythrocytes in rosette formation.

Analogues of NAcSerAspLysPro (AcSDKP), a natural regulator of hematopoiesis isolated from fetal calf bone marrow, were synthesized. The biological activity of these molecules were evaluated in vitro in the rosette assay, which measures the interaction between human Jurkat T-cells and sheep red blood cells. In this test, the tripeptide SerAspLys was the most efficient. Inhibitory activity was detected at the concentration 10(-14) M for the analogue and at 10(-9) M for the parent tetrapeptide. The dipeptide NAcSerAsp still showed activity but at much higher doses (10(-6) M). Substitution of polar amino acids led mostly to inactive molecules. Thus, replacement of Ser by Ala, or Lys by Orn yielded completely inactive compounds and replacement of Asp by Glu decreased the activity (10(-6) M). The present study gives an insight into the role of individual amino acids of AcSDKP in the inhibition of the rosette formation which implicates interactions with T-cell CD2 glycoprotein.

[Drug resistance of Plasmodium falciparum malaria in Equatorial Africa. Apropos of cases observed at a military unit stationed in Gabon].

The existence of Plasmodium falciparum chloroquine-resisting clones in equatorial Africa is quite a recent event. Among a French military unit of 138 people who had lived in Gabon from April to August 1986, 24 of them have caught malaria due to Plasmodium falciparum. This allows us to perceive the impact of this event because of the perfect control of chloroquine prophylaxis and the good knowledge of all the attacks of fever which occurred in the field. Admitting 10 of those people into the department of internal medicine of the Military Hospital Desgenettes located in Lyons, we could have seen that trophozoites of Plasmodium falciparum were still persisting 2 months after their return for 8 of them in spite of carrying on chemoprophylaxis. Observing that in the field, sets the problem of the chemoprophylaxis of malaria for the troops maneuvering in chloroquine-resisting areas and warrants, if justified, a work protocol aimed at solving it.

[Cutaneous South-American Leishmaniasis. Parasitological and serological diagnosis by indirect immunofluorescence and enzyme linked immunoassay (ELISA). 94 cases].

A parasitological (direct test and culture) and serological (IIF and ELISA) survey was carried out in 94 soldiers infested by a cutaneous South American leishmaniasis during a training in the guyanese forest. 52 p.c of the parasitological tests were positive, direct test being much more sensitive than cultures. By this technique, Leishmania Viannia guyanensis was detected 8 times, and Leishmania Viannia braziliensis twice. I.I.F. was negative in all cases because the utilization of a non-homologous antigen in the promastigote form and of the condition of its preparation. ELISA test utilizing the same antigen in a soluble form was specific and its sensitivity was 52 p.c. This technique was positive in 52 p.c of the cases, corroborating some diagnosis for which parasitological test was negative. 66 p.c of the total cases were confirmed.

[Focus on Schistosoma intercalatum].

Area of frequent occurrence of Schistosoma intercalatum is limited to few countries along the Guinea Gulf although the main intervening agents, Bulinus globosus, Bulinus forskalii are dispersed all over Africa. Reservoir of Schistosoma intercalatum is only human and its cycle is difficult to keep up in test animals. Clinical manifestations are, the most often, a simultaneous inflammation of the rectum and sigmoid. Diagnosis is based on the presence of ova with terminal spine in feces or in biopsies of rectal mucosae. Serology, not specific of this species, eases finding of low infestations. Treatment is based on Praziquantel.
