Microcytic anemia and changes in ferrokinetics as late after-effects of glucan administration in murine hepatitis virus-infected C57BL/10ScSnPh mice.

Mild microcytic anemia (without changes in mean corpuscular hemoglobin concentration, MCHC) was discovered 6-14 weeks after a single s.c. administration of 4 mg of particulate glucan to C57BL/10ScSnPh mice serologically positive for murine hepatitis (MHV). The anemia was associated with granulocytosis, decreased body weight and spleen hypertrophy. The overall intensity of erythropoiesis was measured by 59Fe-incorporation into the heme of erythropoietic organs. The localization of erythropoiesis became markedly redistributed--heme production was suppressed in the bone marrow while a several-fold increase was recorded for the spleen. A new steady state was also discovered in ferrokinetics: an iron pool localized away from the blood, erythropoietic organs and the liver was significantly elevated, and hypoferremia was detected. Anemia and wasting of mice were not observed in the same mouse strain free of MHV. A single administration of particulate glucan resulted in late impairment of red blood cell formation in the C57BL/10ScSnPh mouse strain infected with the mouse hepatitis virus. The anemia shares a number of features with those observed for the anemia of chronic diseases.

[The first finding of Giardia spp. in roebucks (Capreolus capreolus L.)].

Roe deer (Capreolus capreolus L.) is a representative of the new host of an intestinal flagellate of the genus Giardia. Giardia spp. was found during autopsy in three dead head of roe deer (roe deer calf 7-8 months, doe, buck) sent for examination to the State Veterinary Institute, Praha, from three different localities of the Czech Republik. Localization of the Giardia spp. was studied in the intestinal tract of the roe deer calf in native preparations in the intestinal contents and in the smears of intestinal mucosa stained after Giemsa (at a distance of 30-40 cm along the whole length of the small intestine). Giardia trophozoites were found in the small intestine in the section of 280-770 cm from the caudal part of duodenum. The trophozoites occurred most intensively within the section of 350-420 cm. The trophozoites showed active, motility in this part of the small intestine as late as in more than 48 hours after the death of the row calf. Their size (obtained by measuring 30 trophozoites) was from 12.4-16.2 to 8-11.5 microns with the mean of 14.5 to 9.4 microns. In the contents of the caeca and colons of all three animals after death numerous Giardia cysts were found, in the caecum of the roe calf trophozoites were detected rarely. Most of the cysts were of ovoid shape. Their size, by measuring 50 cysts from each of the animals, was from 11.2-14.4 to 8-10.4 microns with the mean of 13.3 x 9.1 microns. Sporadically, nearly spherical cysts occurred (12.8-13.6 to 9.6-10.4 microns).(ABSTRACT TRUNCATED AT 250 WORDS)

Ferrokinetics and erythropoiesis in mice after long-term inhalation of benzene.

Ferrokinetics and erythropoiesis were examined in mice exposed for 6 or 7 weeks to an airborne concentration of 300 ppm of benzene, for 6 h per day, and 5 days per week. Ferrokinetic indicators showed only a slightly enhanced production of haeme and erythrocytes in the spleen (133% +/- 18% and 122% +/- 17%, respectively). Production did not change in the femoral marrow; a decline of CFU-C, BFU-E and especially CFU-E (34% +/- 8%) took place there and a shift of cellularity into less mature developmental classes in the erythroblast compartment, without this compartment as a whole being damaged. The erythrocytes produced have an enhanced MCV (109% +/- 0%) and MCH (109% +/- 1%) with an unchanged MCHC; their concentration in blood sank to 87% +/- 1%. The absolute reticulocyte count rose to 160% +/- 16%. 59Fe incorporation into the liver declined far below the level attributable to decreased accessibility of the tracer (84% +/- 4%). A shortening of the life span of late erythroblasts and circulating erythrocytes was deduced from these findings and methodological problems related to some of the seemingly controversial findings are discussed.

Hydroxychloroquine sulphate in the treatment of rheumatoid arthritis: a double blind comparison of two dose regimens.

A controlled, double blind, parallel group, long term study of hydroxychloroquine sulphate in the treatment of rheumatoid arthritis, comparing daily doses of 200 mg and 400 mg, is described. The trial involved 54 patients with moderate disease activity who had not previously received antimalarial drugs. Forty three patients completed the one year treatment. The groups receiving different doses were homogeneous and did not differ in any of the 25 monitored indicators. Both dose regimens were effective, and a significant reduction of disease activity was observed after one year's treatment. Of the nine laboratory and 11 clinical indices of efficacy monitored, no statistically significant differences were reported, but in the group of patients treated with the 400 mg daily dose the number of side effects was three times greater. As there have been no reports of retinopathy with hydroxychloroquine at daily doses of 200 mg the effectiveness of this dose is of practical importance.

[The effect of particle size on the pharmacokinetics of piroxicam].

Two preparations of Piroxicam cps. (10 mg) VUFB Prague, prepared from crystalline and micronized substance, were compared. The release of the active ingredient into the solution in the medium of artificial gastric juice in vitro was examined. The results show that Piroxicam prepared from micronized substance corresponds to the preparation Felden cps. On the other hand, the release of the active ingredient from the preparation prepared from crystalline substance is lower. The pharmacokinetic parameters of Piroxicam from micronized substance are practically consonant to similar parameters of the preparation Felden cps. In Piroxicam prepared from crystalline substance there was a decrease in the rapidity of absorption, which, however, was not statistically significant in the group of volunteers under study. Other pharmacokinetic parameters were practically the same as in the other preparations described.

Interstrain differences in the responsiveness of mice to glucan with respect to hematological effects and manifestations of late damage.

A study was made of the effect of a single s.c. injection of 4 mg glucan in the mouse strains C57BL/10, C3H and a hybrid population of mice. In mice of the strain C57BL/10 high responsiveness was demonstrated, manifested in a long-term increase in the granulocyte counts in the peripheral blood. These effects were accompanied by a fall in the mean red cell volume and the mean corpuscular hemoglobin, a fall in body weight, and mortality. 10 weeks after administration of glucan histology revealed necrotic foci in the livers of animals of the C57BL/10 strain, and an increased amount of the fibrous connective component in the pulmonary interstitium. The strain C3H exhibited a low responsiveness in respect of the hematological indices, while the hybrids did not show any significant response.

Stem cell number versus the fraction synthesizing DNA.

It has been widely held that the fraction of spleen colony-forming units (CFU-S) in DNA synthesis is inversely correlated with CFU-S numbers. In 750 measurements the expected negative slope of the linear regression of CFU-S in S-phase on CFU-S was found to be significant only when the measurements from irradiated mice were pooled. In contrast, a significant positive correlation was observed when the measurements from normal controls were pooled. A hypothesis is offered to account for the variable relationship between CFU-S and CFU-S in S. The hypothesis fully recognizes the crucial role of both parameters in normal and regenerative hemopoiesis.
