Three-drug synergistic inhibition of HIV-1 replication in vitro by 3'-fluoro-3'-deoxythymidine, recombinant soluble CD4, and recombinant interferon-alpha.

3'-Fluoro-3'deoxythymidine (FLT), recombinant soluble CD4 (CD4), and recombinant interferon-alpha (IFN alpha) were evaluated in two- and three-drug regimens against HIV-1 replication in vitro. Peripheral blood mononuclear cells were studied using p24 antigen production as the virologic endpoints. FLT showed 2.5-fold higher efficacy and a similar selectivity index to zidovudine. Drug interactions were evaluated by the median effect principle and the isobologram technique. FLT, CD4, and interferon alpha at noncytotoxic concentrations inhibited HIV-1 synergistically in two- and three-drug combinations with a combination index smaller than one and dose reduction index greater than one. The three-drug regimen provided greater virus suppression than the two-drug regimen. These results suggest that FLT is an alternative agent to AZT for the treatment of HIV infection either as a single agent or in combination with CD4 and/or interferon-alpha.

Candidiasis: detection by gas-liquid chromatography of D-arabinitol, a fungal metabolite, in human serum.

D-Arabinitol was identified as a major metabolite of Candida species in human subjects. Gas-liquid chromatography was used to measure the concentration of D-arabinitol in serum. The study included subjects who were healthy and cancer patients who had proven invasive candidiasis or were colonized with Candida. D-Arabinitol concentrations greater than 1.0 microgram per milliliter were found in serum from patients with invasive infection. This technique may prove valuable in the diagnosis of invasive candidiasis.

Characterization of antibodies to the structural polypeptides of HGSAg: evidence for subtype-specific determinants.

Antisera prepared in guinea pigs to the structural polypeptides of HBAAg/adw and HBSAg/ayw were examined by a modified passive hemagglutination assay for antibodies to the subtype-specific d and y determinants. All of the isolated polypeptide fractions stimulated antibodies to both group specific and subtype-specific antigens of the native HBSAg particle from which they were derived. These data indicate that the polypeptides have similarities in their immunochemical structure.

Hepatitis B core antigen. Detection of antibody by radioimmunoprecipitation.

Radioactive cores were prepared from concentrated Dane particles by DNA polymerase reaction followed by CsCl density gradient centrifugation. Two radioactive peaks were obtained: one peak with an average density of 1.36 g/cm-3 in CsCl contained cores that possessed full serologic reactivity; the other peak, with a density of 1.28 to 1.32 g/cm-3, contained cores associated with globulin. A double antibody immunoprecipitation test was developed, using the radioactive heavy cores as a source of antigen. The test was at least 300 times as sensitive as complement fixation for detecting antibody to core.
