Transcription in nematodes: early Ascaris embryos are transcriptionally active.

In this report we analyze early zygotic gene expression in the parasitic nematode Ascaris lumbricoides var. suum. Using synchronous populations of early embryonic stages, nuclei were isolated, and in vitro run-off transcription assays were performed. We find transcriptional activity as early as the 4- to 8-cell stage. The percentage of RNA polymerase II activity, as measured in these assays, is greater than 80% of the total transcription at the 60-cell stage. Furthermore, we show that a specific transcript (actin) can be identified in all early stages tested.

Usefulness of the transcutaneous Po2 monitor during exercise testing in adults.

The usefulness of the transcutaneous Po2 monitor for monitoring during stress testing was evaluated. Seventy-five simultaneous recordings of arterial and transcutaneous Po2 were made on 11 patients with chronic obstructive pulmonary disease while they were undergoing treadmill exercise testing. All arterial blood samples were drawn from an indwelling brachial arterial line and analyzed in duplicate. The range of arterial Po2 measured was 50 to 145 mm Hg. The correlation coefficient (relationship between arterial and transcutaneous Po2) was .96 and the slope of the best fit line was 1.047 (Y = 1.047 x -7.2). The transcutaneous Po2 was an average of 5.99 +/- 4.76 mm Hg below the arterial Po2 in the entire group. The mean difference in transcutaneous and arterial Po2 was greater at low Po2 level, than at high levels. We found the electrode easy to maintain and calibrate, with a low frequency of malfunction. This instrument provides a safe, simple, and reliable noninvasive method for recording serial changes in Po2 during exercise testing.

Vasculitis and Sjögren's syndrome with IgA-IgG cryoglobulinemia terminating in immunoblastic sarcoma.

Generalized lymphadenopathy and other manifestations of Sjögren's syndrome developed in a 68 year old woman with a long history of systemic vasculitis and arthralgia. An unusual immunologic feature was hypogammaglobulinemia and immunoglobulin A (IgA) monoclonal immunoglobulinemia with mixed IgA-IgG cryoglobulin. At autopsy, the histopathologic findings were compatible with immunoblastic sarcoma. The monoclonal IgA protein, found in serum, pleural and pericardial fluids, showed rheumatoid factor activity. Immunocytes from the immunoblastic sarcoma were found to be the source of the monoclonal IgA protein.

Antimicrobial and other properties of a new stabilized alkaline glutaraldehyde disinfectant/sterilizer.

The properties of stabilized alkaline 2% glutaraldehyde solution (SGS) are discussed. SGS is discussed with regard to its chemistry, antimicrobial properties, organic soil resistance, toxicity, corrosivity and chemical stability. SGS retains the maximum antimicrobial activity of alkaline glutaraldehyde solutions and the chemical stability heretofore observed only with acidic glutaraldehyde solutions. These improvements, along with the inherent resistance of glutaraldehyde to neutralization by organic soil, allow SGS to be continuously used for 14 days in situations of high dilution, or 28 days in situations of low dilution.
