[Usefulness of selected circulating biochemical markers in diagnosis and monitoring of endometriosis].

A review of the literature for searching of biochemical marker of endometriosis is presented. The investigations with CA 125, antiendometrial antibodies, placental protein 14 and others have not established a sensitive screening test for predicting endometriosis, especially in early stages of the disease. Most authors confirm usefulness of those markers in monitoring therapy and predicting recurrences. This may allow to avoid commonly performed "second look" laparoscopy.

[The "Bizzare" leiomyoma in histopathological examination of the uterine myoma: dilemma of the range of operative treatment].

The very rare case of leiomyoma sarcomatosum was presented and differentiated to "bizzare" leiomyoma at 39 years old woman, who was conservative preoperation diagnosis of uterine myoma. To get the after-operation prognosis the following elements of tumour have been studied: mitotic index, presence and type of necrosis, separation of the tumour and perivascular infiltration. A decision of conservative treatment was confirmed by shown world wide literature.

[Lymphoma lymphoplasmocytoides: a case report].

Clinical symptoms and haematological disorders in patients suffering from lymphoplasmatic/lymphoplasmocytoidal lymphoma were presented, 82 patients were examined (median age of 63), all of the in clinical stage IV (ANN Arbor scale). In all of the patients bone marrow was invaded. The most common symptom was enlargement of peripheral lymph nodes (86%) less often abdominal ones (21%). Splenomegalia was found in 55% and hepatomegalia in 49% of patients 35% of patients had anaemia and 38% thrombocytopenia. Close to 1/3 of patients dermatoses were found, in most cases allergic ones. In 7% another cancer was found.

[Thrombocytosis in patients with non-Hodgkin's lymphomas and Hodgkin's disease].

Secondary thrombocytosis appears in patients with various neoplastic and autoaggressive diseases and infections. Thrombocytosis occurrence among 358 patients with NHL and 46 with HD treated at the Department of Haematology of CMKP (Postgraduate Training Centre) and Warsaw Medical School in 1986-1993 is here presented. The 11 patients (3%) among the ones with NHL and the 11 (24%) with HD showed thrombocytosis. Much more frequently the increased thrombocyte count could have been found in patients with low grade T-cell lymphomas (14%) and in patients with Hodgkin's disease. The patients with chronic B-cell lymphocytic leukemia (1%) showed thrombocytosis in very few cases. Thrombocyte count in patients with NHL was not high (mean-596.0 x 10(9)/L median-558.0 x 10(9)/L) which was similar as in those with HD (mean-601.0 x 10(9)/L median-558.0 x 10(9)/L).

[Cholesterol and its fractions in women after ovariectomy treated with estraderm TTS].

25 ovariectomized women mean age 47 +/- 5 years were treated with transdermal 17 beta Estradiol in the shape of Estraderm TTS 100 plasters. Drugs were administrated in 28 days cycles during 24 weeks. Chlormadinon in daily dose 2 mg was added from 16 to 28th day of the fourth therapeutic cycle. Total cholesterol, triglycerides, HDL and LDL fractions were checked using enzymatic method. First measurement was done before the treatment, the next in the last day of every cycle. Results were statistically analysed. After treatment there were significant increase in the mean values of HDL, significant decrease of triglycerides. Changes in mean level of LDL and total cholesterol were not statistically significant. Our results seem to confirm the previous observations concerned with transdermal estrogen replacement therapy and its efficacy in atherosclerosis prophylaxis.

[Lymphoplasmacytoid lymphoma and the immune system in patients with this type of lymphoma].

Among 82 patients with L-LP in clinical stage IV, 14% were found to have monoclonal gammapathy and 22% polyclonal gammapathy. The other classes of immunoglobulins were not affected. In 78%, cell immunity was impaired. Decrease in T cell count (CD2+ and CD3+) in blood was found. Particularly T helper cells (CD4+), to a lesser degree T suppressor cells (CD8+) were affected. The life time of patients varied from 3 to 120 months. 60% of deaths were caused by infection.

Morphological picture in paroxysmal nocturnal hemoglobinuria. Case report.

25-year-old woman with paroxysmal nocturnal hemoglobinuria was admitted to the hospital because of headache, progressing right hemiparesis and speech disorders. Several days later patient lost consciousness. Cerebrospinal fluid was xanthochromic with increased pleocytosis and protein level. CT-scan revealed ischemic area with hemorrhagic focus within left cerebral hemisphere. Patient died 3 weeks after the admission. Brain section revealed hemorrhagic infarct in the cortex of the left parietal lobe, thrombosis of the superior sagittal sinus and "respiratory brain" changes. Microscopic examination revealed meningeal venous thrombosis, hemorrhagic infarct, vasculitis, abundant accumulation of bacteria within blood vessels, and other pathological changes such as petechiae, perivascular exudates and small, round areas composed of acellular fibrillary network. There were no macrophages and GFAP-positive astrocytes in any of these areas. Authors suggest that weak cell reactivity may be connected with alterations in cell membranes, mainly low phosphatidylinositol (GPI) content.

[Model studies of milk containing inhibitory substances. I. Sensitivity of microbiological tests for detection of inhibitory substances and studies of turbidity changes in milk].

In the present studies use was made of raw milk derived from a model farm in the Olsztyn voivodeship. Washing and washing-disinfecting agents: Mirax, Bis, Radion, Iosan CP, Rudlak and Ixi, as well as crystalline penicillin were added to milk. Their lowest concentrations inhibiting growth of indicator strain (MIC): Bacillus stearothermophilus var. calidolactis C 953, were determined by two microbiological tests. Moreover, the changes in light diffusion by milk, caused by the applied inhibitory substances, were examined. It was found that the microbiological tests for the determination of the lowest inhibitory concentration of the sanitization agents and of penicillin were much more sensitive in the case of milk with a 0.05% fat content, as compared with whole milk.

Hemagglutinins of Beauveria bassiana strains: the effect of growth conditions on their production.

Forty strains of Beauveria bassiana were screened for their ability to produce hemagglutinins. It has been found that majority of mycelial extracts but not cultural broth display non specific hemagglutinating activity toward animal and human type AB and A, B, O erythrocytes when microorganisms are cultivated on rich in amino acids media supplemented with saccharose. The formation of hemagglutinins in mycelia is strongly dependent on composition of medium and associated with a production of extracellular lipases, chitinases and amylases. No monosaccharide binding specificity of obtained hemagglutinins toward glucose/mannose, N-acetylglucosamine, N-acetylgalactosamine/galactose, L-fucose and sialic acid was observed.

[Microangiopathic hemolytic anemia in patients with diseases of the hematopoietic and lymphatic systems].

Microangiopathic haemolytic anaemia was diagnosed in the course of haematopoietic and lymphatic disorders such as chronic granulocytic leukemia, chronic myelofibrosis, chronic lymphatic leukemia, Osler's disease, chronic monocytic leukemia, and lymphoplasmocytic lymphoma, in 11 patients (6 women and 5 men) aged between 33 and 81 years (mean age 58.8 years) treated at the Haematological Out-Patient Clinic of the Postgraduate Medical Education Centre within 1977-1987. The following laboratory tests were carried out: 1) morphology of the peripheral blood and bone marrow, especially some haematological parameters concerning erythrocytes and blood platelets; 2) biochemical tests reflecting erythrocytes disintegration; 3) haemostasis. All examined patients suffered from haemolytic anaemia of various degree with characteristic changes in erythrocyte shape (helmets, tear-drops etc.). Haemolytic origin of anaemia was confirmed by the increased LDH activity. In the majority of patients no compensative stimulation of haematopoiesis (reticulocytosis, red blood cells hyperproliferation in bone marrow) was seen. Clinical symptoms of haemostatic disorders such as haemorrhagic diathesis and vein thrombosis were diagnosed in 50% of the patients. Blood platelet counts ranged from markedly decreased to significantly increased. Bone marrow smears did not show increased number of megacariocytes. Bleeding time was prolonged in the majority of examined patients while prothrombin index--decreased). Abnormal fibrinogen levels (decreased or increased) were found in the majority of patients with fibrin degradation products. Microangiopathic haemolytic anaemia in these patients differ from the typical Moschowitz's disease clinically probably due to the lack of compensative stimulation of erythropoiesis and lower thrombocytopenia.

Two molecular forms of penicillin amidase synthesized by Escherichia coli.

The Swatek's method was further simplified for the assay of penicillin amidase activity. The absorbance of colour obtained during determination of 6-aminopenicillanic acid was dependent on concentration of 4-dimethylaminobenzaldehyde and on temperature. Antiodies induced in rabbits with one molecular form of penicillin amidase from E. coli PCM 271 (PA-1 or PA-2) did not cross-react with the other amidase form. No differences in substrate specificity on inactivation with SDS and in alkaline medium between the two amidase forms were observed. Concentrated urea inactivated PA-2 irreversibly and PA-1 reversibly. N-Bromosuccinimide inactivated almost completely only PA-1. Two E. coli PCM 271 strain variants were separated by microbial selection. Each of them produced only one amidase form. Also two amidase forms were found in cells of E. coli ATCC 11105, whereas E. coli ATCC 9636 and ATCC 9637 synthesize only PA-1.

Enzyme-linked immunosorbent assay (ELISA) of penicillin amidases.

Non-competitive, sandwich enzyme immunoassay for both penicillin amidases from Escherichia coli is described. The assay involves the use of monospecific antibodies and their conjugates. The amidases inactivated by heating and by acid- or alkali-treatment cannot be assayed. Reproducible results for each amidase were achieved within 5-6 hours in the range of 3-500 ng/ml (0.25-40 mU/ml) and coefficient of variation was 13%.

Antineoplastic activity of new linear hydrazine derivatives.

A series of hydrazine derivatives was tested for antineoplastic activity. Cyanoacetic acid hydrazide (I), cyanoacetic acid methylhydrazide (II) and N-thioamido-N'-cyano-acethylhydrazine (VI) appeared to be the most active agents against sarcoma 180, Ehrlich carcinoma and Nemeth Kellner lymphoma. The maximum tumor weight inhibition ranged from 70 to 90%. N, N'-bis-cyano-acethylhydrazine (IV) and N-isonicotinoyl-hydrazine (VIII) showed significant antitumor activity in Ehrlich carcinoma and Nemeth Kellner lymphoma systems. None of hydrazine derivatives were active against L1210 leukemia. The most active agents, I, II and VI were further evaluated in leukemia P388, melanoma B16, Lewis lung carcinoma and mammary carcinoma 16/C, and the agent VI was additionally tested in plasmacytoma MP26, colon carcinoma 26 abd Walker carcinosarcoma 256 systems. However, there was no effect with any agent or dose tested against any neoplasm.

Kitasatosporia setae, an interesting actinomycete strain isolated from Spitsbergen soil.

The phenotypic and genotypic properties as well as cell chemistry of an interesting actinomycete, which converted certain azacarbazoles into highly cytotoxic derivatives, were established. The strain was also compared with two similar Japanese actinomycetes of Kitasatosporia gen. nov. A great resemblance between the strains was observed. Apart from similar phenotypic properties they were characterized by contents of both L- and meso-DAP, glycine and galactose as the main cell wall components. They proved to have the same phospholipid, glycolipid and fatty acid patterns. The guanine-plus-cytosine contents of the deoxyribonucleic acids of the strains averaged 72.5 and there was a high degree of homology between the DNAs of the strains (approximately 80%). These data provide evidence that the Spitsbergen isolate and the Japanese actinomycetes belong to one genomic species.
