The effect of verapamil on intestinal calcium transport.

Verapamil (1.5 mM) completely inhibits active calcium transport by everted rat duodenum sacs and depresses oxygen consumption by duodenal rings. Increasing the Ca2+ concentration in the incubation medium from 0.4 mM 5.0 mM reverses the inhibition of oxygen consumption by verapamil but is without effect on the extent of inhibition of calcium active transport.

The effect of calcium restriction in the diet of calcium transport in rat small intestine.

1. The everted-sac technique has been used to study the time-dependent effect of low-calcium diet on calcium active transport along rat small intestine. 2. In animals maintained on standard diet active translocation of calcium was limited to proximal 10 cm of the intestine. 3. In response to calcium restriction, calcium transport in duodenum was highly stimulated after 3 days, then gradually declined and after 28 days almost disappeared. In proximal jejunum it was the highest between 7 and 21 days. In distal ileum, the transport appeared after 3 days and increased progressively until 21 days, but markedly decreased at the 28-th day. The normal pattern of calcium transport was reestablished on refeeding the animals with standard diet.

Diurnal variations of active calcium transport in the intestine of pregnant and lactating rat.

Everted gut sac technique was used to study diurnal fluctuations of active calcium transport in the intestine of pregnant and lactating rats maintained under 12 hours light-dark cycle. At the second and third week of gestation an increase of calcium transport occurred in duodenum and adjacent part of jejunum at light phase, but the transport remained unchanged at darkness. Simultaneously the length of jejunal region able for active transport increased. In pregnant females around the term and in lactating mothers no diurnal differences in calcium transport were seen and at the 21st day of lactation the transport were seen and at the 21st day of lactation the transport was nearly equal in both duodenum and proximal jejunum. Restricted to darkness feeding of lactating rat evoked diurnal differences in jejunal but not in the duodenal transport.

Development of circadian variations in intestinal calcium transport during maturation of the rat.

Everted gut sac technique was used to study diurnal pattern of active calcium transport along the small intestine of developing and adult rats kept under 12:12 h light-dark cycle. In animals aged 6 weeks to 5 months calcium transport displayed essential diurnal fluctuations with maximum transport at darkness whereas in 23 days old rats diurnal fluctuations were negligible. During maturation of the rat a gradual increase in the amplitude of calcium transport rhythm was observed. Since the 10th week of life diurnal variations of the calcium transport involved both the transporting efficiency and the intestinal area participating in the transport. Nevertheless an age-dependent decrease occurred in the intestinal region capable of transporting actively calcium. Diurnal pattern of calcium transport was almost the same in 6-week-old rats either fed ad libitum or adapted from weaning to feeding restricted to darkness. Feeding restricted to light period resulted in arrhythmic transport fluctuations. A shift in the phase of the diurnal rhythm of calcium transport was observed in 6-week-old rats adapted from the 14th day of life on to reversed light dark cycle..
