Effects of cysteamine on growth performance, digestive enzyme activities, and metabolic hormones in broilers.

A total of 600 avian male broilers at the age of 1 d were used to investigate the effects of cysteamine (CSH) on growth performance, digestive enzyme activities, and concentrations of serum hormones. The broilers received the same basal diets, with CSH added at 0 (control), 60, 90, 120, or 150 mg/kg. The feeding program consisted of a starter diet until 21 d and a grower diet until 42 d. The broilers with addition of CSH at 60 or 90 mg/kg had significantly higher growth rates during d 1 to 21 or d 21 to 42 compared with the control, respectively. However, adding 150 mg of CSH/kg significantly suppressed the growth of broilers. Adding 60 mg of CSH/kg significantly increased the activities of protease, amylase, and lipase in the pancreas and small intestinal contents during d 1 to 21, and the activities of protease and amylase in the small intestinal contents during d 21 to 42. Adding 90 mg of CSH/kg significantly increased the activities of lipase during d 1 to 21 and protease, amylase, and lipase during d 21 to 42 in small intestines. The activities of digestive enzymes during the whole period were suppressed by adding 150 mg of CSH/kg. The concentration of serum thyroxine was higher in the CSH-added birds during the whole period, whereas serum triiodothyronine was higher only during d 1 to 21 compared with the control. These findings indicate that low doses of dietary CSH may improve the growth performance and the activities of the digestive enzyme, but high doses of CSH appear to be detrimental to growth and digestion.

Effects of dietary sources of vegetable oils on performance of high-yielding lactating cows and conjugated linoleic acids in milk.

This study was conducted to examine the effects of dietary supplementation with vegetable oils on performance of high-yielding lactating cows and conjugated linoleic acid (CLA) content in milk fat. Twelve lactating Holstein cows in early lactation (30 to 45 d postpartum) were used in a triple 4 x 4 Latin square design. In each period, the cows in each group were fed the same basal diet and received one of the following treatments: 1) control (without oil), 2) 500 g of cottonseed oil, 3) 500 g of soybean oil, and 4) 500 g of corn oil. Each experimental period lasted for 3 wk, with the first 2 wk used for adaptation to the diet. Supplementation with vegetable oils tended to increase milk yield, with the highest milk yield in the cottonseed oil group (35.0 kg/d), compared with the control (34.4 kg/d). Milk fat percentage was decreased, but there were few effects on percentage and yield of milk protein as well as milk fat yield. The cows fed added soybean oil produced milk with the highest content of trans-11 C(18:1) (23.8 mg/g of fat), which was twice that of the control (12.6 mg/g of fat). Content of cis-9, trans-11 CLA in milk fat increased from 3.5 mg/g in the control to 6.0, 7.1, and 10.3 mg/g for the cows fed oils from cottonseed, corn, and soybean, respectively. A significant linear relationship existed between trans-11 C(18:1) and cis-9, trans-11 CLA. Supplementation with oils doubled the content of total fatty acids in blood plasma, with little difference between different vegetable oil sources. Octadecenoic acid content was significantly higher in blood plasma of animals fed added oils from cottonseed and soybean than those fed with corn oil and control. The plasma trans-11 C(18:1) content was significantly higher in the oil-added animals than in control. Supplementation of vegetable oils tended to improve milk production of lactating cows, and the CLA content in milk fat was significantly increased. Soybean oil seemed to be the optimal source to increase CLA production.

Optimization of operational factors of a membrane bioreactor with gravity drain.

A new membrane bioreactor with gravity drain for municipal wastewater treatment was tested and its operational factors were investigated in this study. These factors include pressure head, MLSS, aeration intensity (an air flow rate per unit floor area) and temperature. Results of batch experiments showed that a critical pressure head of the MBR was 0.85-1.5 m-H2O. At a pressure head of 0.85 m-H2O, statistical analysis of batch experiments showed that aeration intensity significantly affected membrane flux, and the MLSS had no impact on membrane flux under a temperature of 22.0 +/- 1.0 degrees C. Results of the long-term continuous experiment showed that temperature significantly affected membrane flux. The impact of temperature on membrane flux in this case was about 4-10 times of that analyzed by using a classical cake layer model. During this experiment, the average removal efficiencies of COD, BOD5 and NH4+-N were over 85%, 97% and 94%, respectively.

Membrane-based gas transfer: an environmental engineering laboratory.

We propose an educational experience in which students design a membrane gas transfer reactor, construct a bench-scale version in the laboratory, and employ the reactor to measure mass transfer coefficients. The membrane reactor is useful for teaching mass transfer principles because the mass transfer interface is well defined and easily observed. The system can be modeled successfully using straightforward mathematics. The reactor can be designed and constructed by students, using the mathematical model as a basis, providing insight into the physical meaning of model parameters. The proposed membrane system can be readily operated to obtain data that can be employed to develop or modify existing mass transfer correlations. This can provide students with significant insight into the development of mass transfer correlations and how the constants in such correlations are typically determined. These features help promote a deeper understanding of mass transfer principles.

In vitro gas production measurements to evaluate interactions between untreated and chemically treated rice straws, grass hay, and mulberry leaves.

In vitro gas production was measured to investigate associative effects of untreated and chemically treated rice straw (RS) and of RS and grass hay or mulberry leaves (ML). The RS was treated with NaOH, urea, or (NH4)HCO3. Cumulative gas production was recorded at 2, 4, 6, 8, 12, 16, 24, 36, 48, 60, 72, and 96 h of incubation, and the Gompertz function was used to describe the kinetics of gas production. Treatment with NaOH, urea, and NH4HCO3 increased (P < 0.05) gas production at 48 h by 55, 52, and 37% and the maximum rate of gas production of RS from 0.64 to 1.51, 1.27, and 1.13 mL/h, respectively. The inclusion of treated straws, hay, and ML in a mixture with RS at increasing proportions (25, 50, 75, 100%) elevated cumulative gas production and its rate. Maximum gas production was not different between the RS and its mixtures with the treated straws, but inclusion of hay into RS or NH4HCO3-treated RS increased the maximum gas production. Associative effects were defined as the difference between the observed gas production for the mixtures and the sum of the individual component feeds. The NaOH-treated RS, included at higher proportions, had positive associative effects at all times of incubation < 96 h; effects were negative or absent at 96 h. No effects were observed with mixtures of RS with urea-treated RS. The NH4HCO3-treated RS mixture tended to have negative associative effects at all three levels. Positive associative effects (P < 0.05) were observed for both RS and NH4HCO3-treated RS at almost all inclusion levels of hay or ML after 12 to 96 h of incubation. The response was more pronounced with ML than with hay. Associative effects generally declined with duration of incubation. We conclude that positive associative effects on in vitro gas production occurred more consistently when RS was incubated in mixtures with hay or ML than when incubated in mixtures with chemically treated RS.

A direct reduction of aliphatic aldehyde, acyl chloride, ester, and carboxylic functions into a methyl group.

The aliphatic carboxylic group was efficiently reduced to the methyl group by HSiEt(3) in the presence of catalytic amounts of B(C(6)F(5))(3). To the best of our knowledge, this is the first example of a direct exhaustive reduction of aliphatic carboxylic function. Aliphatic aldehydes, acyl chlorides, anhydrides, and esters also underwent complete reduction under similar reaction conditions. Aromatic carboxylic acids, as well as other carbonyl functional equivalents, underwent smooth partial reduction to the corresponding TES-protected benzylic alcohols. It was shown that, unlike the reduction of aliphatic substrates, the exhaustive reduction of aromatic substrates was not straightforward: a concurrent Friedel-Crafts-like alkylation process competed with the reduction yielding trace to notable amounts of dimeric products, thus decreasing the overall selectivity of the reduction process.

[High expression of rabbit sperm membrane protein rSP10 in Escherichia coli and preparation of its specific antisera].

To study the position and immunogenicity of rSP10, the rabbit rsp10 gene which did not include sequences coding for signal peptides was inserted into expression vector pET30a. An in-frame fusion protein was made such that a His6 stretch was produced at the N terminus of re-rSP10. High expression was obtained, the amount of re-rSP10 up to 67% in the total bacterial protein. The re-rSP10 was purified by DEAE chromatography and the yield of purified re-rSP10 was approximately 50 micrograms/mL of culture. Western blotting analysis of re-rSP10 with rabbit polyclonal sera raised against rabbit sperm membrane protein showed that the synthesized antigen possessed immunogenicity of rSP10. Specific antisera against re-rSP10 was induced using purified re-rSP10 as an antigen. The motility of capacitated sperms were affected but no aggregation was observed.

G1 arrest and relative protein expressions in mouse thymocytes induced by whole body X-ray irradiation.

OBJECTIVE: To investigate the molecular regulation of G1 arrest of mouse thymocytes induced by ionizing radiation. METHODS: Cell cycle was analyzed by flow cytometry (FCM) following staining of cells with propidium iodide. Fluorescent staining and flow cytometry analysis were employed for measurement of protein expression. RESULTS: It was demonstrated that G1 phase of mouse thymocytes increased significantly at 12 h after whole body irradiation (WBI) with the doses of 0.5, 1.0 and 2.0 Gy, and at 24 h following 2.0 Gy exposure, measured by FCM. In the time course experiment, it was found that G1 phase of thymocytes increased significantly at 4 h, reached a peak level at 24 h and came down toward 48 h after WBI with 2.0 Gy X-rays. The results also showed that after 2.0 Gy exposure, the expression of proteins in mouse thymocytes increased significantly from 1 h to 8 h for p53, for p21 from 4 h to 48 h, and for MDM2 at 4 h and 8 h, measured by FCM. But no change was found for GADD45 protein expression. CONCLUSION: These results suggest that G1 arrest could be induced by a single dose of 0.5 Gy, 1.0 Gy or 2.0 Gy, and its molecular control might be established through the p53-p21 pathway.

[Clinical and experimental study on the ovulation promoting effect of yangjing decoction].

OBJECTIVE: To observe the effect of Yangjing Decoction (YJD) to promote ovulation and to explore its mechanism. METHODS: The clinical effect of YJD in treating patients of functional non-ovulation and changes of gonadotropic hormone, sex hormone and diameter of main follicle before and after treatment were observed. In experimental study, the influence of YJD on monoamine neurotransmitter of hypothalamus, endorphin (beta-EOP), serum sex hormone, trace element, endometrium and histomorphology of ovary were observed. RESULTS: In the 36 patients observed clinically, the ovulation rate was 61.1%, the serum contents of follicular stimulation hormone (FSH), luteinizing hormone (LH) and estradiol (E2) increased and the diameter of main follicle enlarged significantly after treatment (P < 0.01, P < 0.001). The experimental study showed that the totals of follicle at different levels, the corpus luteum and the proportion of superior follicle in ovary were improved obviously in the YJD group as compared with those in the control group. After YJD treatment the hypothalamus contents of monoamine neurotransmitter, including norepinephrine (NE) and dopamine (DA) increased, the 5-hydroxytryptamine (5-HT) 5-hydroxyindoleaceticacid (5-HIAA) and beta-EOP decreased significantly, and the NE/DA ratio and NE/5-HT ratio elevated obviously (P < 0.05, P < 0.001). The serum levels of E2, P and trace elements, including Zn, Cu and Fe increased significantly (P < 0.05, P < 0.01). CONCLUSION: YJD is an effective recipe in promoting ovulation, the effect may be achieved through the multi-linked, multi-systemetic synthetic effect of hypothalamus, hypophysis and ovary, etc.

Resistance of helicobacter pylori to metronidazole, tetracycline and amoxycillin.

Resistance to metronidazole, tetracycline and amoxycillin, and beta-lactamase production were determined for 153 clinical isolates of Helicobacter pylori. Of these isolates, 77.8% were resistant to metronidazole (MIC > 8 mg/L), 58.8% to tetracycline (MIC > 16 mg/L) and 71.9% to amoxycillin (MIC > 0.5 mg/L); 39.2% were multiresistant. Resistance to metronidazole was more common in isolates from females than in those from males (P < 0.05). None of the isolates produced beta-lactamase, so the mechanism of amoxycillin resistance was not linked to production of beta-lactamase.

Adaptive response of thymocyte apoptosis and cell cycle progression induced by low dose X-ray irradiation in mice.

The dose-effect of adaptive response of thymocyte apoptosis and cell cycle progression induced by whole-body X-ray irradiation (WBI) was studied in male Kunming mice. The inductive doses (D1) were 25, 50, 75, 100 or 200 mGy 6 h before the challenging doses (D2) of 1.0, 1.5 or 2.0 Gy. The changes in the percentages of the thymocyte apoptotic bodies (TAB) and the cells in different phases of cell cycle were measured with flow cytometry. The percentages of TAB decreased, the arrests of G1 and G2 + M phases diminished, and the cells of DNA synthesis of S phase increased when the D1 + D2 groups was compared with the D2 groups. When D1 was 200 mGy, the adaptive response of thymocyte apoptosis and cell cycle progression were no longer induced by low dose radiation (LDR). In addition, the extracellular fluid from the splenocytes were cultured with Con A for 48 h in vitro 24 h after 75 mGy WBI was placed in the murine thymocyte suspension from mice irradiated with 2.0 Gy WBI and co-incubated. The thymocyte apoptosis decreased. Especially, noteworthy was that the percentages of TAB after the incubation for 72 h were significantly lower than those in 2.0 Gy irradiated thymocytes (P < 0.05). These results indicate that when the mice were irradiated with 25-100 mGy (D1, 12.5 mGy/min) 6 h before 1.0-2.0 Gy (D2, 0.287 Gy/min) exposure, an adaptive response of thymocyte apoptosis and cell cycle progression may be induced under the condition of WBI, and LDR (75 mGy) may change the microenvironment of immune cells and decrease the thymocyte apoptosis.

[Study of helical CT low-contrast resolution and its influencing factors].

This paper studies the helical CT low-contrast resolution and its influencing factors by experimental analysis. The CT low-contrast resolution increased with mA, kV, slice thickness, scan time and FOV, and decreased with matrix, pitch and thickness of the scanned objective. The reconstruction algorithm of high resolution results lower low-contrast resolution. Mostly helical image low-contrast resolution is slightly lower than axial image. It's very important for us to understand CT low-contrast resolution and its influencing factors for the correct evaluation of the performance of CT scanner.

Studies on treatment of acute promyelocytic leukemia with arsenic trioxide: remission induction, follow-up, and molecular monitoring in 11 newly diagnosed and 47 relapsed acute promyelocytic leukemia patients.

Fifty-eight acute promyelocytic leukemia (APL) patients (11 newly diagnosed and 47 relapsed) were studied for arsenic trioxide (As2O3) treatment. Clinical complete remission (CR) was obtained in 8 of 11 (72.7%) newly diagnosed cases. However, As2O3 treatment resulted in hepatic toxicity in 7 cases including 2 deaths, in contrast to the mild liver dysfunction in one third of the relapsed patients. Forty of forty-seven (85.1%) relapsed patients achieved CR. Two of three nonresponders showed clonal evolution at relapse, with disappearance of t(15;17) and PML-RARalpha fusion gene in 1 and shift to a dominant AML-1-ETO population in another, suggesting a correlation between PML-RARalpha expression and therapeutic response. In a follow-up of 33 relapsed cases over 7 to 48 months, the estimated disease-free survival (DFS) rates for 1 and 2 years were 63.6% and 41.6%, respectively, and the actual median DFS was 17 months. Patients with white blood cell (WBC) count below 10 x 10(9)/L at relapse had better survival than those with WBC count over 10 x 10(9)/L (P =.038). The duration of As2O3-induced CR was related to postremission therapy, because there was only 2 of 11 relapses in patients treated with As2O3 combined with chemotherapy, compared with 12 of 18 relapses with As2O3 alone (P =.01). Reverse transcription polymerase chain reaction (RT-PCR) analysis in both newly diagnosed and relapsed groups showed long-term use of As2O3 could lead to a molecular remission in some patients. We thus recommend that ATRA be used as first choice for remission induction in newly diagnosed APL cases, whereas As2O3 can be either used as a rescue for relapsed cases or included into multidrug consolidation/maintenance clinical trials.

Genomic sequence, structural organization, molecular evolution, and aberrant rearrangement of promyelocytic leukemia zinc finger gene.

The promyelocytic leukemia zinc finger gene (PLZF) is involved in chromosomal translocation t(11;17) associated with acute promyelocytic leukemia. In this work, a 201-kilobase genomic DNA region containing the entire PLZF gene was sequenced. Repeated elements account for 19.83%, and no obvious coding information other than PLZF is present over this region. PLZF contains six exons and five introns, and the exon organization corresponds well with protein domains. There are at least four alternative splicings (AS-I, -II, -III, and -IV) within exon 1. AS-I could be detected in most tissues tested whereas AS-II, -III, and -IV were present in the stomach, testis, and heart, respectively. Although splicing donor and acceptor signals at exon-intron boundaries for AS-I and exons 1-6 were classical (gt-ag), AS-II, -III, and -IV had atypical splicing sites. These alternative splicings, nevertheless, maintained the ORF and may encode isoforms with absence of important functional domains. In mRNA species without AS-I, there is a relatively long 5' UTR of 6.0 kilobases. A TATA box and several transcription factor binding sites were found in the putative promoter region upstream of the transcription start site. PLZF is a well conserved gene from Caenorhabditis elegans to human. PLZF paralogous sequences are found in human genome. The presence of two MLL/PLZF-like alignments on human chromosome 11q23 and 19 suggests a syntenic replication during evolution. The chromosomal breakpoints and joining sites in the index acute promyelocytic leukemia case with t(11;17) also were characterized, which suggests the involvement of DNA damage-repair mechanism.

Distinct leukemia phenotypes in transgenic mice and different corepressor interactions generated by promyelocytic leukemia variant fusion genes PLZF-RARalpha and NPM-RARalpha.

Acute promyelocytic leukemia (APL) is characterized by a specific chromosome translocation involving RARalpha and one of four fusion partners: PML, PLZF, NPM, and NuMA genes. To study the leukemogenic potential of the fusion genes in vivo, we generated transgenic mice with PLZF-RARalpha and NPM-RARalpha. PLZF-RARalpha transgenic animals developed chronic myeloid leukemia-like phenotypes at an early stage of life (within 3 months in five of six mice), whereas three NPM-RARalpha transgenic mice showed a spectrum of phenotypes from typical APL to chronic myeloid leukemia relatively late in life (from 12 to 15 months). In contrast to bone marrow cells from PLZF-RARalpha transgenic mice, those from NPM-RARalpha transgenic mice could be induced to differentiate by all-trans-retinoic acid (ATRA). We also studied RARE binding properties and interactions between nuclear corepressor SMRT and various fusion proteins in response to ATRA. Dissociation of SMRT from different receptors was observed at ATRA concentrations of 0.01 microM, 0.1 microM, and 1.0 microM for RARalpha-RXRalpha, NPM-RARalpha, and PML-RARalpha, respectively, but not observed for PLZF-RARalpha even in the presence of 10 microM ATRA. We also determined the expression of the tissue factor gene in transgenic mice, which was detected only in bone marrow cells of mice expressing the fusion genes. These data clearly establish the leukemogenic role of PLZF-RARalpha and NPM-RARalpha and the importance of fusion receptor/corepressor interactions in the pathogenesis as well as in determining different clinical phenotypes of APL.

Adverse changes in fibre type and myosin heavy chain compositions of human jaw muscle vs. limb muscle during ageing.

This review shows that human jaw muscles not only have unique fibre type and myosin heavy chain (MyHC) compositions but also undergo muscle and region-specific changes in fibre composition during ageing. Alterations in the masseter and the lateral pterygoid muscles in the elderly are opposite to those reported for limb and trunk muscles, whereas changes in the anterior and posterior bellies of the digastric muscle resemble those of limb and trunk muscles. We conclude that age-related alterations in fibre type composition and MyHC expression are muscle and region specific, probably reflecting muscular differences in genetic programs and epigenetic influences.
