[Clinical study of the treatment of endometriosis with traditional Chinese medicine].

46 endometriotic patients were treated by the method of removing Blood Stasis and resolving Phlegm, softening and resolving the Lump (RBS-RP-SRL). The total effective rate was 91.3%. Among them, the effective rate of dysmenorrhea was 97.6%, that of anal tenesmic was 94.3%, that of intercourse pain 86.7%, and 86.7% diminished in size of mass or nodule, 6 of 10 infertility patients became pregnant. It was showed by the laboratory assay that the cases were in the abnormal condition of nail bed microcirculation and hemorheology, which could be significantly improved after treatment. The results revealed that it was effective in treating endometriosis with the method of RBS-RP-SRL.

Propafenone and disopyramide enhance post-ischemic contractile and metabolic recovery of perfused hearts.

The effects of sodium channel blockers, propafenone and disopyramide, on post-ischemic contractile dysfunction of perfused rat hearts were examined. Isolated hearts were subjected to 35 min ischemia, followed by 60 min reperfusion with and without administration of either drug during 3 min of pre-ischemia. Ischemia/reperfusion induced complete cardiac dysfunction, rise in left ventricular end-diastolic pressure, increase in perfusion pressure, accumulation of Na+ and Ca2+ and loss of K+ and Mg2+, and release of creatine kinase and purine nucleosides and bases from the heart. These observations suggest that ischemia/reperfusion in the current study induces cardiac cell necrosis or an increase in cell membrane permeability to ions, substrates and macromolecules. Treatment of perfused hearts with either propafenone at concentrations ranging from 5 to 70 microM, or disopyramide at concentrations of 100 microM or higher resulted in a pronounced contractile recovery of the heart, associated with suppression of reperfusion-induced tissue ion alteration and inhibition of reperfusion-induced release of creatine kinase and purine nucleosides and bases. Ischemic insult itself caused tissue Na+ accumulation and K+ loss without any change in tissue Ca2+ and Mg2+. The alterations in the electrolytes were attenuated by treatment with either agent. The results suggest that prevention of ischemia- and reperfusion-induced ionic disturbance of cardiac cells by propafenone and disopyramide plays a role in the improvement of post-ischemic contractile dysfunction.

Beneficial effects of quinidine on post-ischemic contractile failure of isolated rat hearts.

The present study was undertaken to determine whether quinidine may improve ischemia/reperfusion-induced functional and metabolic injury of isolated rat hearts. Thirty-five-min ischemia and the subsequent 60-min reperfusion resulted in no post-ischemic force generation, an increase in left ventricular end-diastolic pressure (about 1500%) and a sustained rise in perfusion pressure (136 +/- 15% of initial). This was associated with an increase in the release of creatine kinase and ATP metabolites from the reperfused heart, a decrease in tissue high-energy phosphates, changes in tissue sodium, calcium, potassium and magnesium contents, and a reduction in the triphenyltetrazolium chloride (TTC)-stained area, an indicator of infarction. Hearts were treated with 3-100 microM quinidine 3 min before ischemia. Quinidine at concentrations of 10 microM or greater resulted in post-ischemic contractile recovery in a concentration-dependent manner (61 +/- 8 to 95 +/- 8% of initial). Ischemia/reperfusion-induced metabolic and histologic alterations were also suppressed by treatment with quinidine in a concentration-dependent manner. The results suggest that quinidine has a cardioprotective effect in ischemic/reperfused hearts. Because transmembrane fluxes of ions, substrates, and enzymes were suppressed by treatment with quinidine, protection of cardiac cell membrane function and/or integrity against ischemia/reperfusion-induced ionic imbalance, presumably sodium imbalance, across the sarcolemma is a possible mechanism by which quinidine may act. Slightly higher levels of ATP were detected in the treated hearts at 10 and 15 min, but not at 35 min of ischemia. Such preservation of high-energy phosphates might also be beneficial for protecting myocardial cells against ischemic damage.

Improvement of ischemia/reperfusion-induced contractile dysfunction of perfused hearts by class Ic antiarrhythmic agents.

The present study was undertaken to determine whether class Ic antiarrhythmic agents, flecainide and pilsicainide, improve ischemia/reperfusion-induced cardiac contractile dysfunction. Isolated rat hearts were subjected to a 35-min global ischemia, followed by a 60-min reperfusion, and the functional and metabolic consequences were examined. Ischemia/reperfusion resulted in no recovery of contractile function and a sustained rise in left ventricular and diastolic pressure of the perfused heart during reperfusion. The ischemia/reperfusion also induced increases in tissue sodium and calcium levels, decreases in potassium and magnesium ion contents and the release of creatine kinase and purine nucleosides and bases (ATP metabolites) from the heart, suggesting a pronounced change in cardiac transmembrane flux of ions, substrates and macromolecules. Treatment of the perfused heart with either 5 to 50 microM flecainide or 10 to 100 microM pilsicainide during preischemia resulted in diminishing the postischemic contractile dysfunction; suppressing the changes in tissue sodium, potassium, calcium and magnesium contents; and attenuating the release of creatine kinase and ATP metabolites in a concentration-dependent manner. Tissue sodium accumulation and potassium loss were also observed at the end of ischemia, which was attenuated by pretreatment with these agents. These results provided evidence that class Ic antiarrhythmic agents are capable of diminishing ischemia/reperfusion-induced contractile dysfunction and tissue ionic disturbance.

[Metrological analysis for efficacy of acupuncture on angina pectoris].

Acupuncture (Acupoints: Neiguan P6, Shenmen H7, Shaohai H3 or auriculo-points: Heart, Shenmen) was administrated once or 7 times in a week on 40 patients with stable type of angina pectoris. The effect was assessed quantitatively or semi-quantitatively according to the extent, area, frequency, duration of attack, the time of attack during exercise, and the vanishing of suffering after exercise. Just after one performance of acupuncture, 15 patients' angina pectoris were significantly alleviated (P < 0.001) both in degree and area. After 7 times of acupuncture 10 patients' angina pectoris were not only significantly alleviated both in extent and area, but also in frequency and duration of attack. 15 patients were randomized to an acupuncture, non-acupuncture or acupuncture at non-acupoints (ANA) in a single blind design. The time from the beginning of exercise to the anginal attack in active acupuncture group was longer than that in non-acupuncture or ANA group (P < 0.01), but they were similar (P > 0.05) in both non-acupuncture group and ANA group. The time from the end of exercise to the disappearance of angina pectoris in acupuncture group was shorter than that in the other two groups (P < 0.05).

[Effect of acupuncture on intestinal motion and sero-enzyme activity in perioperation].

The authors study on abdominal surgical patients (39 cases) who were randomly divided into the acupuncture and control group. In the acupuncture group, the needle inserted into Zusanli (St.36) and Sanyinjiao (Sp. 6) points at 12-24 h after operation and observed the time of the first excretion through anus. Sero-enzyme activity of GPT, GOT and gamma-GT determined before operation and at 1, 3, 5, 7 days after operation. The results showed that the time of excretion in the acupuncture group (57.78 +/- 23.94 h) was obviously faster than the control one (86.14 +/- 20.43 h), P less than 0.001). It suggested that acupuncture has the effect of promoting intestinal motion. After operation sero-enzyme activity was raised 2-3 times than before. It showed that the surgical trauma directly or indirectly impair cells to release enzyme into blood, but the time of reversing to normal level of sero-enzyme activity in the acupuncture group was obviously faster. There is possibility that acupuncture has the effect of regulating reactivity of organism on trauma and promoting the repair of damaged cell.

[Traditional Chinese medicine combined with Western medicine in the treatment of severe renal failure in patients with epidemic hemorrhagic fever].

This article reports the treatment of 23 cases (with the control group 20 cases) of severe renal failure in patients with epidemic hemorrhagic fever (EHF) by integrated traditional Chinese medicine and western medicine (TCM-WM), i.e., the renal protective decoction and modern medical treatment during the period of 1988-1989. The effects were as follows: (1) The curative rate was elevated and the morbidity was dropped (P less than 0.01-0.05). (2) Both the period of oliguria and albuminuria were shortened (P less than 0.01-0.05). (3) The severity of the complication, e.g., massive gastro-intestinal hemorrhage, etc., was much reduced (P less than 0.05). (4) The degree of azotemia was getting milder. And the renal function was much improved (P less than 0.05). The mode of action of the renal protective decoction is based on the point of view of the TCM in the treatment of infectious disease.

Use of antipeptide antibodies to probe the catalytic activity of p37v-mos.

Several site-directed antipeptide antibodies were generated to probe the kinase function of p37v-mos. Anti-mos(37-55) antibodies allowed autophosphorylation of p37v-mos, as well as transphosphorylation of exogenously added purified bovine vimentin. Rabbit antipeptide antibodies against v-mos residues 158-70, 194-206, 260-271, and 363-374 and a mouse monoclonal antibody against residues 344-359 completely inhibited p37v-mos protein kinase activity in vitro. p37v-mos autophosphorylation and vimentin transphosphorylation were affected similarly. These results suggest important roles for insert and the carboxy-terminal domains in the catalytic activity of p37v-mos.

[Early quantitative detection of vascularity of the reduced femoral head of congenital hip dislocation by scintigraphy].

Vascularity of the femoral head of CDH was detected by 99mTc-MDP scintigraphy before and after reduction. The results showed that a decrease of second time-activity, intersection of minute time-activity and remarkable decrease of relative ratio and corrected relative ratio on the part of femoral head which closed to the acetabulum is the diagnosis criterion for the obstruction of blood supply of the reduced femoral head. We confirmed that the mechanical pressure was the factor which caused the obstruction of blood supply of the reduced femoral head.

Cell cycle-mediated structural and functional alteration of P85gag-mos protein kinase activity.

We investigated cell cycle-dependent regulation of protein kinase activity encoded by the viral mos gene by using a normal rat kidney cell line (NRK-6m2) chronically infected with a temperature-sensitive mutant (ts110) of Moloney murine sarcoma virus, which produces the P85gag-mos transforming protein. In elutriation experiments, in which cells in various phases of the cell cycle are separated based upon size, a twofold increase in the specific activity of the P85gag-mos protein kinase was observed as cells progressed from G0/G1 through S and G2/M. A three- to fourfold increase in gas-mos protein kinase specific activity relative to unsynchronized cells was observed in mitotic NRK-6m2 cells synchronized by treatment with thymidine followed by colcemid or with nocodazole alone. Interestingly, the gag-mos protein was structurally altered in mitotic cells generating a protein species moving slower than P85gag-mos in SDS-polyacrylamide gels. Our findings indicate that viral mos protein kinase activity is regulated during the cell cycle via phosphorylation. We propose that the mos transforming protein functions in a pleiotropic manner, affecting both cytoplasmic and nuclear targets.

An experimental animal model of Kashin-Beck disease.

Twelve young macaque monkeys were fed with grain and water from areas actively endemic or non-endemic for Kashin-Beck disease. Both dietary grain and water from geographical areas endemic for Kashin-Beck disease induced a sequence of pathological changes in the growth plates and articular cartilage and biochemical changes in the serum and urine of monkeys. These changes are similar to those in human Kashin-Beck disease. It is considered that this may be a simple and valuable model for the further study of this disease and its management and control. The results suggest that the pathogenetic factors of Kashin-Beck disease relate both to grain and to water in the diet in endemic areas. The experiment also shows that certain serum enzyme concentrations correlate with chondronecrosis.

[CT brain scanning in the diagnosis and localisation of cerebral schistosomiasis].

CT scanning was performed on 25 cases of cerebral schistosomiasis. The images of the granuloma caused by schistosome eggs showed a high density mass shadow while the edema of the white matter showed a low density shadow. Combining CT scanning, with case history, clinical signs and immunologic assay we could make a correct diagnosis for each of the patients about the pathologic site and nature. The correlations between the images of CT scanning and clinical pathology were analysed and a better treatment regimen was discussed based on our experience. Some of the patients treated with praziquantel and checked by CT scanning showed pathologic changes of the granuloma disappeared during a follow-up period of 3-12 months.

[Avidin-biotin-peroxidase complex enzyme-linked immunosorbent assay in the diagnosis of human multilocular echinococcosis].

The Avidin-Biotin-Peroxidase Complex enzyme-linked immunosorbent assay (ABC-ELISA) with antigen from gerbils infected with alveolar hydatid (GAH) was performed on the sera of 54 patients with multilocular echinococcosis, 107 patients with other diseases and 102 healthy adults. All the sera were diluted 1:200 and an S/N value greater than 2.2 was taken as positive. Both the sensitivity and specificity were 98.1%. The S/N value (mean +/- SD) of sera of the patients with multilocular echinococcosis, other parasitic diseases, tuberculosis, other diseases and healthy adults were: 5.9 +/- 3.0, 1.1 +/- 0.4, 1.4 +/- 0.8, 1.1 +/- 0.5 and 1.0 +/- 0.4. The geometric mean titre (GMT) of antibody measured by ABC-ELISA was 13.7 times that of ELISA. The quantity of serum used in ELISA was 8 times that of ABC-ELISA. ABC-ELISA might be particularly helpful to identify cases with low immune responsiveness. In the detection of alveolar hydatidosis and cystic disease with GAH antigens, the positivity was 98.1 and 67.1% with 87 and 25.6% having S/N greater than 5, respectively. It is suggested that in the diagnosis of hydatid diseases by ABC-ELISA, homologous antigen should be used. The temperature and the incubation time needed for the present assay are practical, for the total time requires only 55 minutes.

The effect of radix Salviae miltiorrhizae on vasoactive intestinal peptide in cerebral ischemia: an animal experiment.

The levels of vasoactive intestinal peptide (VIP) in three regions of rat brain were assayed in 62 rats. Bilateral common carotid artery ligation was done in 50 rats. Half an hour before ligation 26 rats were given 10 g/kg of Radix Salviae Miltiorrhizae (RSM); 24 rats were given same volume of normal saline as controls. A sham operation was done in 12 rats. Half an hour (n = 30) and 3 hours (n = 32) after operation, the rats were quickly decapitated. VIP levels were assayed in cerebral cortex, hippocampus and caudate nucleus. In salin-treated animals, VIP levels of cerebral cortex and caudate nucleus at 3 hour group were significantly decreased compared with the sham-operated group. No significant difference was found between RSM-treated and sham-operated groups. The preliminary results suggest that VIP may be involved in the pathophysiological procedures of cerebral ischemia and RSM may attenuate the dysfunction of VIP during cerebral ischemia.
