SRXRF microprobe as a technique for studying elements distribution in Elsholtzia splendens.

Elsholtzia splendens is a copper tolerant plant growing in copper mine areas in south of China and accumulates considerable heavy metals in plant tissue. In this study, synchrotron radiation X-ray fluorescence spectroscopy (SRXRF) microprobe was used to study the Cu and other elements distribution in E. splendens. The element (P, S, Cl, K, Ca, Mn, Fe, Cu, Zn) in the leaf epidermis and cross-sections of the stem and leaf could be checked by SRXRF which was considered a sensitive technique for trace element analysis. The highest Cu levels were measured in the vascular tissues of stem and petiole, while Cu levels in mesophyll were higher than in leaf epidermis. The levels of most elements were not higher in trichomes than in other tissues. It seems that the celluar compartmentation of heavy metals in epidermis and epidermal trichomes was not the general feature of all plants. There was a significant correlation between Cu and P, S, Ca in distribution, which suggested P, S, and Ca played an important role in Cu accumulation of E. splendens. Based on the significant correlation between Cu and elements Mn, Fe, and Zn in distribution, it seemed that Cu, Mn, Fe, and Zn could be transported by the same transporters with a broad substrate range.

Comprehensive study on binding capacity of human immunoglobulin G to Avid AL affinity gel.

Avid AL is an affinity chromatographic gel based on a synthetic ligand which has been designed for the purification of immunoglobulins. The binding capacities of the gel for human IgG were investigated under different conditions; various salts; salt concentrations; different buffer systems with pH ranging from 3.8 to 10.0; and, tissue culture supernatant supplemented with 20% serum. At a similar ionic strength, the binding capacity changed according to the salting out effect of the ions used. Kinetic parameters of the affinity adsorption showed that a strong salting out effect significantly decreased the dissociation constant. Avid AL gel purified IgG from human serum with a capacity of 26.2 mg/ml gel using binding buffer containing sodium sulfate. Recombinant Protein A gel was used as a control. In this study it also exhibited enhanced binding capacity with a high salt concentration binding buffer.

[Identification of fruits of Xanthium sibiricum Patr. and X. mongolicum Kitag].

The protein electrophorogram, tissue separation picture and complete powder characteristic picture of these two kinds of fruit of siberian cocklebur are made for the first time, and TLC chromatograms of different solvent extracts are also presented. All these present a scientific basis for crude drugs selling, breed identification and composition analysis of the fruit of siberian cocklebur.

Treatment of experimental Coxsackie B-3 viral myocarditis with Astragalus membranaceus in mice.

A murine model system for observing the effect of Astragalus Membranaceus (AM) on experimental myocarditis caused by Coxsackie B-3 virus (CB3V) was developed in 4-week-old male BALB/C mice. Gross, histopathologic and ultrastructural examinations of the infected-AM treated group showed that the severity and involved area of the myocardial lesions became milder and smaller than those in the infected-NS treated mice. The total lesion area, and the total lesion area/total myocardial area examined (%) and virus titer in the former group were also smaller and lower than those in the latter group. The results suggest that AM is effective in the inhibition of Coxsackie B virus propagation and protection of myocardium in mouse myocarditis.
