Determination of hexamethylene-based isocyanates in spray-painting operations. Part 1. Evaluation of a polyurethane foam sponge sampler.

A polyurethane foam (PUF) sponge was mounted in a cassette sampler and evaluated as a sorbent for the collection of hexamethylene diisocyanate (HDI) monomer and HDI-based oligomers. Recovery studies indicated 112 +/- 34% average recovery of HDI monomer and 92 +/- 9% and 97 +/- 25% average recovery of HDI-based oligomers when using impregnated PUF sponges. The PUF sponge was also evaluated during actual spray-painting operations. In a series of side-by-side sampling events, an impinger filled with 1-(2-methoxyphenyl)piperazine (MOP) in toluene was compared directly with a cassette sampler containing a PUF sponge impregnated with MOP or 1-(9-anthracenylmethyl)piperazine (MAP) in dimethyl sulfoxide (DMSO). For the analysis of HDI-based oligomer, there is no significant difference (p < 0.05, n = 7) in the air concentration when sampling with either the PUF sponge cassette or the impinger. The results are significant because they indicate that a PUF sponge, which is more convenient than an impinger, may be used for the collection of HDI-based oligomer generated during spray-painting operations.

Effects of the estrous cycle and early pregnancy on uterine expression of Mx protein in sheep (Ovis aries).

Conceptuses of ruminant ungulates produce large amounts of a type I interferon, interferon-tau (IFNtau), which is the signal for maternal recognition of pregnancy. Induction of cellular Mx proteins is an important component of the response to type I interferon in the immune system, but Mx regulation and function have not been studied in the uterus. This study examined temporal and spatial alterations in ovine uterine Mx expression during the cycle and early pregnancy using immunohistochemistry, in situ hybridization, and Northern and slot-blot analysis. Sheep uterine endometrium expressed a single approximately 2.5-kilobase Mx mRNA transcript that was detectable at all stages of the estrous cycle and early pregnancy examined. In cyclic ewes, mRNA abundance in endometrium increased from Day 1 to peak levels at Day 13 and then declined to Day 15. In pregnant ewes, steady-state levels of Mx mRNA were first detected above the level in cyclic ewes at Day 13 postmating, were greater than 10-fold higher at Day 15, and remained elevated at Day 19. Expression of Mx mRNA in the myometrium did not change during the estrous cycle but increased approximately 23-fold between Days 11 and 15 of pregnancy. Immunohistochemical and in situ hybridization analysis revealed a similar temporal pattern of Mx expression. In cyclic ewes, Mx protein and mRNA were initially localized to the luminal epithelium at Days 1 and 3, increased from Days 5 to 13, especially in the shallow uterine glands, and then declined at Day 15. Pregnancy resulted in up-regulation of Mx expression in the luminal and glandular epithelium, stroma, and myometrium. Punctate Mx immunostaining and Mx mRNA concentrations were greatest when progesterone production was maximal during the estrous cycle and were strongly up-regulated by the conceptus across the entire uterine wall. It is suggested that a cascade of induction of Mx gene expression proceeds from the luminal epithelium to the outer longitudinal myometrium and that transcriptional activation of the promoter may involve both soluble cytokines (i.e., IFNtau) and steroid hormones (i.e., progesterone).

[Investigation on the epidemiology of Histoplasma capsulatum infection in Nanjing district].

Two hundred and ninety-two permanent or temporary residents in Nanjing district were tested with histoplasmin (Histolyn-CYL, ALK/Berkeley Laboratories, USA) and PPD. Forty-nine (16.78%) subjects reacted to histoplasmin with 5.0-23.0 (9.5 +/- 4.2) mm induration. Positive reaction rate among people without pulmonary diseases (normal group) was 15.10% comparing to 17.74% to patients with pulmonary diseases. Positive reaction to PPD with 5-50 (14.2 +/- 4.7) mm induration was 56.16%, with 59.43% in normal group and 54.30% in patients with pulmonary diseases while 8.90% subjects reacted to both histoplasmin and PPD, 7.88% of the research subjects reacted to histoplasmin but not to PPD. Result suggested that there was herd infection of Histoplasma capsulatum in Nanjing district.

Hyperoxia-induced lung injury in premature rat: description of a suitable model for the study of lung diseases in newborns.

OBJECTIVE: To provide suitable animal model (hyperoxia-induced premature rat lung damage) for research of bronchopulmonary dysplasia (BPD) and to better understand pathogenesis of BPD and look for effective drugs to prevent and treat BPD. METHODS: Rat litters delivered prematurely at 21-day gestation by hysterotomy. Vigorous resuscitation at birth resulted in a high survival rate. Surfactant and antioxidant enzyme (AOE) system were measured. The model was tested in an experiment of hyperoxia-induced lung injury. RESULTS: Compared to litters delivered spontaneously at term (gestation 22 days), these preterm rats had immature pulmonary surfactant composition with low total phospholipid (x +/- s: 10.09 +/- 1.49 micrograms/mg wet weight vs 12.04 +/- 1.31 micrograms/mg wet weight; P = 0.0367) and phostidylcholine (5.06 +/- 1.82 micrograms/mg wet weight vs 8.28 +/- 2.35 micrograms/mg wet weight; P = 0.0238) levels. The concentrations of AOE enzymes, superoxide dismutase (11.40 +/- 2.04 mu/mg DNA vs 15.78 +/- 1.84 mu/mg DNA; P < 0.01) and catalase (92.81 +/- 62.25 mu/mg DNA vs 412.24 +/- 117.50 mu/mg DNA; P < 0.01) were also significantly lower. Animals exposed to hyperoxia had a significantly higher mortality. Pulmonary edema and histological features of lung damage were observed in the pups exposed to hyperoxia. CONCLUSIONS: The premature rat model is relatively cheap, readily available and has a high survival rate. Pulmonary surfactant and AOE systems are immature. These properties make them a suitable model for the study of acute and chronic lung damage related to prematurity and O2 toxicity.

[An ELISA assay for humulus scandens specific IgE antibodies].

OBJECTIVE: Humulus scandens pollen (HSP) has been proved to be the one of the most important causative factors of autumnal hay fever in China. We established an enzyme immunoassay for specific IgE against HSP by using monoclonal anti-IgE antibody to detect sIgE in 143 patients with autumnal hay fever and 30 healthy volunteers. METHODS: An indirect method of ELISA was developed. The optimal conditions for the test were: coating the allergen at 4 degrees C overnight with a concentration of 12 micrograms/ml; the test serum was diluted 1:16 and incubated with allergen overnight at 37 degrees C; the most suitable dilution and incubative time of the peroxidase-conjugated mouse anti-human IgE antibody were 1:3,000 and 2 hours respectively. RESULTS: The patients' serum level of sIgE was significantly higher than controls (P < 0.001) and correlated significantly with the degree of skin test reaction (P < 0.01). The concordance between the results of skin test and ELISA was 74%-83%. The specificity of the assay was confirmed by heat inactivation and multiple absorption experiments. The coefficients of variation for the intraassay and interassay reproducibility ranged from 1.20%-8.30% and 8.63%-9.22% respectively. CONCLUSIONS: Determination of specific HSP IgE with ELISA assay shows good specificity, sensitivity and reproducibility. It favours clinical practice due to its lower cost.

[Identification of gravity-related esterases (grEST1 and grEST2) in carrot callus cells].

A horizontal clinostat which mimics the microgravity in space was used to study its effects on carrot cells. After using boric acid buffer (pH 8.8) as extraction medium, PAGE pattern of esterase isozymes of carrot callus cells displayed 8 bands of which the activities of only 2 were affected by microgravity. They were named grEST1 and grEST2, where grEST stands for gravity-related esterase. The rates of increase in activity of them in carrot cells when cultured on a rotating horizontal clinostat were lower than that cultured in normal gravitational environments, and the difference increased with the culture time. The activities of grEST1 and grEST2 in carrot callus cells subjected to horizontal rotation were found to return to their original levels after being placed under normal gravity (1 x g) for 7 days. We suggest that the effect of simulated microgravity conditions on grEST1 and grEST2 activities in carrot callus cells is through affecting their synthesis. In addition, the activities of grEST1 and grEST2 were not inhibited by eserine, acetylcholine iodide, diisopropyl fluorophosphate and p-chloromercuribenzoate, which indicates that they are acetylesterases.

[Studies on extensive application of bromoacetamide in various types of schistosomiasis endemic area].

AIM: To provide scientific basis for the popularization and application of molluscide bromoacetamide. METHODS: Large scale application of bromoacetamide in twenty eight counties of four provinces was investigated and its molluscicidal effect in marshland and influence on marshland grass (Cyperus) and fishes were observed. RESULTS: High molluscicidal effect of bromoacetamide was found against both snails and snail eggs in various environments. According to the results observed in different fields, the applicable dosage of bromoacetamide is 1.5-2 g/m2 for spraying method, 1.5-2 g/m3 for immersing method and 1.5-2 g/m for immersing in combination with shovelling the turf along the bank. When it was applied in fish-raising ponds or rivers at an applicable dosage, no toxic effect on fishes was found. The spraying dosage of bromoacetamide did not affect the rice seedling but it did some damage to the leaf-tips of marshland grass, however, the grass did turn green and grow. CONCLUSION: Bromoacetamide has proved to be a safe and effective mulluscicde against Oncomelania under various field conditions.

FHIT gene alterations in esophageal cancer and ulcerative colitis (UC).

FHIT (fragile histidine triad gene), a candidate tumor suppressor gene, was recently identified and cloned at chromosome 3p14.2. Alterations of this gene have been reported in a number of primary human tumors, including colorectal, esophageal, gastric and lung carcinomas. However, some reports have found no abnormalities in this gene. We investigated a total of 63 primary esophageal tumors, nine esophageal cancer cell lines and 17 ulcerative colitis-associated neoplasms (UCANs) for alterations of FHIT. In 13 esophageal tumors, we employed overlapping reverse transcriptase-PCRs (RT-PCRs) to amplify and sequence the complete open reading frame of FHIT. One of 13 primary esophageal tumors analysed by RT-PCR expressed no detectable FHIT transcript; the remaining 12 expressed normal-sized transcripts with wild-type open reading frame sequences. In an additional 50 esophageal tumors, the polymorphic microsatellite loci D3S1300 and D3S1313 were used to evaluate loss of heterozygosity (LOH) at 3p14.2. Eleven of these 50 tumors showed LOH at one or both loci. In all these 11 tumors, genomic PCR and direct sequencing of FHIT exons 5-9 was performed. This analysis revealed that none of these 11 primary esophageal tumors contained any alterations in the FHIT open reading frame or adjacent intron sequences. Finally, among 17 UCANs, the in vitro synthesized protein (IVSP) assay detected no truncated protein products, nor were there any abnormalities in size or DNA sequence of FHIT RT-PCR products. However, in six of nine esophageal carcinoma cell lines, no FHIT RT-PCR product was detectable using either of the overlapping primer sets. Genomic PCR and direct sequencing of exons 5-9, also performed in these nine cell lines, revealed wild-type sequence in eight cell lines; however, one cell line contained no exon 5 PCR product. This cell line also lacked detectable FHIT transcript. These data suggest that the open reading frame of FHIT is not important in the development or progression of most primary esophageal carcinomas or UCANs, although lack of expression of the FHIT transcript may be common in esophageal cancer-derived cell lines. The possibility of an additional tumor suppressor gene at chromosome 3p14.2 remains to be evaluated.

Deficient transforming growth factor-beta1 activation and excessive insulin-like growth factor II (IGFII) expression in IGFII receptor-mutant tumors.

The insulin-like growth factor II receptor (IGFIIR) gene has been identified as a coding region target of microsatellite instability in human gastrointestinal (GI) tumors. IGFIIR normally has two growth-suppressive functions: it binds and stimulates the plasmin-mediated cleavage and activation of the latent transforming growth factor-beta1 (LTGF-beta1) complex, and it mediates the internalization and degradation of IGFII ligand, a mitogen. We used an immunohistochemical approach to determine whether IGFIIR mutation affected expression of these proteins in GI tumors. Four highly specific antibodies were used: LC(1-30), which recognizes the active form of TGF-beta1; anti-LTGF-beta1, which detects the LTGF-beta1 precursor protein; anti-IGFIIR; and anti-IGFII ligand. Twenty GI tumors either with (6 of 20) or without (14 of 20) known IGFIIR mutation were examined, along with matching normal tissues. Results were statistically significant in the following categories: (a) decreased active TGF-beta1 protein expression in IGFIIR-mutant tumor tissues versus matching normal tissues or IGFIIR-wild-type tumor tissues; (b) increased LTGF-beta1 protein expression in IGFIIR-mutant tumor tissues versus matching normal tissues or IGFIIR-wild-type tumor tissues; and (c) increased IGFII ligand protein expression in IGFIIR-mutant tumor tissues versus matching normal tissues or IGFIIR-wild-type tumor tissues. These data suggest that in genetically unstable GI tumors, mutation of a microsatellite within the coding region of IGFIIR functionally inactivates this gene, causing both diminished growth suppression (via decreased activation of TGF-beta1) and augmented growth stimulation (via decreased degradation of the IGFII ligand).

Frequent mutation of the E2F-4 cell cycle gene in primary human gastrointestinal tumors.

The E2F group of transcription factors transactivates genes that promote progression through the G1-S transition of the cell cycle. Members of the retinoblastoma (Rb) family of proteins bind to E2Fs and inhibit this function. E2F-4, one example of the E2F group, functions as an oncogene when transfected into nontransformed cells in vitro. On the other hand, mice that are homozygously lacking a normal E2F-1 gene develop cancers, consistent with a tumor-suppressive role for this gene. The exact function of E2Fs has thus been unclear; moreover, direct involvement of this gene in primary human tumorigenesis has not been shown. We, therefore, investigated mutation within the E2F-4 coding region in 16 primary gastric adenocarcinomas, 12 ulcerative colitis-associated neoplasms, 46 sporadic colorectal carcinomas, 9 endometrial cancers, and 3 prostatic carcinomas. We limited our investigation to the serine repeat within E2F-4, reasoning that this tract might be altered in genetically unstable tumors (replication error-positive, or RER+). All tumors were RER+, with the exception of a control group of 15 RER- sporadic colorectal carcinomas. PCR with incorporation of [32P]dCTP was performed using primers flanking the serine trinucleotide (AGC) repeat. Twenty-two of 59 gastrointestinal tumors (37%) contained E2F-4 mutations; these comprised 5 of 16 gastric tumors (31%), 4 of 12 ulcerative colitis-associated neoplasms (33%, including 1 dysplastic lesion), and 13 of 31 sporadic colorectal cancers (42%). No mutation was present in any of the endometrial, prostate, or RER- colorectal tumors. Of note, homozygous mutations occurred in three cases, and two of seven informative patients showed loss of one E2F-4 allele in their tumors. Furthermore, the RER+ sporadic colorectal tumors were evaluated at trinucleotide repeats within the genes for N-cadherin and B-catenin; no tumors demonstrated mutation of these genes. These data suggest that E2F-4 is a target of defective DNA repair in these tumors.

Mutation of hMSH3 and hMSH6 mismatch repair genes in genetically unstable human colorectal and gastric carcinomas.

Mutations within microsatellite sequences, consisting of additions or deletions of repeat units, are known as the replication/repair error positive (RER+) phenotype or micorsatellite instability (MI). Microsatellite instability has been demonstrated in hereditary and sporadic colorectal carcinomas and is usually observed in noncoding regions of genomic DNA. However, relatively few coding region targets of MI have been identified thus far. Using PCR, we amplified regions encompassing (A)8 and (C)8 microsatellite tracts within hMSH3 and hMSH6 from 31 RER+ sporadic colorectal tumors, 8 hereditary colon cancers, 23 RER+ gastric carcinomas, and 32 RER- gastric tumors. Mutations were found in 11 (36%) of 31 sporadic colon carcinomas, 4 (50%) of 8 hereditary colorectal cancers, and 5 (22%) of 23 RER+ gastric carcinomas, but in only 2 (6%) of 32 RER- gastric carcinomas. These frameshift mutations cause premature stop codons downstream that are predicted to abolish normal protein function. Our results and those of others suggest that DNA mismatch repair genes, such as hMSH3 and hMSH6, are targets for the mutagenic activity of upstream mismatch repair gene mutations and that this enhanced genomic instability may accelerate the accumulation of mutations in RER+ tumors.

A transforming growth factor beta 1 receptor type II mutation in ulcerative colitis-associated neoplasms.

BACKGROUND & AIMS: Numerous gastrointestinal tumors, notably sporadic and ulcerative colitis (UC)-associated colorectal carcinomas and dysplasias, gastric cancers, and esophageal carcinomas, manifest microsatellite instability. Recently, a transforming growth factor beta 1 type II receptor (TGF-beta 1RII) mutation in a coding microsatellite was described in colorectal carcinomas showing instability. One hundred thirty-eight human neoplasms (61 UC-associated, 35 gastric, 26 esophageal, and 16 sporadic colorectal) were evaluated for this TGF-beta 1RII mutation. METHODS: Whether instability was present at other chromosomal loci in these lesions was determined. In lesions manifesting or lacking instability, the TGF-beta 1RII coding region polydeoxyadenine (poly A) microsatellite tract was polymerase chain reaction amplified with 32P-labeled deoxycytidine triphosphate. Polymerase chain reaction products were electrophoresed on denaturing gels and exposed to radiographic film. RESULTS: Three of 18 UC specimens with instability at other chromosomal loci (17%) showed TGF-beta 1RII poly A tract mutation, including 2 cancers and 1 dysplasia; moreover, 2% of UC specimens without instability (1 of 43) (1 cancer), 81% of unstable sporadic colorectal cancers (13 of 16), and none of the 61 stable or unstable gastric or esophageal cancers contained TGF-beta 1RII mutations. CONCLUSIONS: Mutational inactivation of the poly A microsatellite tract within TGF-beta 1RII occurs early and in a subset of unstable UC neoplasms and commonly in sporadic colorectal cancers but may be rare in unstable gastric and esophageal tumors.

Irreversible binding of N-methyl-N-[(1S)-1-(4-isothiocyanatophenyl)-2-(1-pyrrolidinyl)ethyl-3,4 -dichlorophenylacetamide to the cloned rat kappa opioid receptor.

N-Methyl-N-[(1S)-1-(4-isothiocyanatophenyl)-2-(1-pyrrolidinyl)ethyl-3,4- dichlorophenylacetamide (MITPD), is an isothiocyanate derivative of the kappa agonist ICI-199,441. In this study, interaction of MITPD with cloned mu, delta, and kappa opioid receptors was characterized. MITPD inhibited [3H]diprenorphine binding to kappa receptors with high affinity and with approximately 700- and approximately 870-fold selectivity over mu and delta receptors. Pretreatment with MITPD followed by extensive washing reduced kappa receptor binding with an IC50 value of 3.7 nM, but did not affect mu or delta binding at < or = 0.1 microM. Preincubation with 1 microM MITPD abolished [3H]diprenorphine binding, while pretreatment with 1 microM ICI-199,441 increased Kd of [3H]diprenorphine binding with no change in Bmax. Thus, MITPD is a selective kappa irreversible ligand. The region of the kappa receptor that conferred selectivity for MITPD was determined by examining its binding to four mu/kappa chimeras. IC50 values of MITPD for inhibition of [3H]diprenorphine binding were determined to be 430 nM for Chimera III (kappa1-141/mu151-398), 1.8 nM for Chimera IV (mu1-150/kappa142-380), 40 nM for Chimera XI (mu1-268/kappa263-380) and 14 nM for Chimera XII (kappa1-262/mu269-398). Pretreatment with MITPD followed by extensive washing reduced binding to chimera IV with an IC50 value of 75 nM, but did not affect III, XI or XII binding (IC50 >1 microM). Thus, the region from the third transmembrane helix to the C-terminus of the kappa receptor is important for the binding of MITPD.

Stable transfection of Acanthamoeba.

The promoter activity of an Acanthamoeba polyubiquitin gene was analyzed in its homologous system. A modified calcium phosphate transfection method using a neomycin marker vector was developed to achieve highly efficient transfection of the Acanthamoeba polyubiquitin gene into Acanthamoeba cells. In this transfection procedure, the calcium phosphate-DNA complex was formed gradually in the medium during incubation with cells and precipitated on the cells. The crucial factors for obtaining efficient transfection were the pH (6.95) of the transfection buffer used for the calcium phosphate precipitation and the amount (25 micrograms/96-well tissue culture plate) and form (circular) of transfecting DNA. Under these conditions, Acanthamoeba isolate 1B6 was transfected at an efficiency of about 40% with the constructed vector pOPSBU, a pOP13CAT-based polyubiquitin gene incorporated neomycin resistance vector. Acanthamoeba polyphaga was transfected at an efficiency of about 10% with this vector. Transfection of both Acanthamoeba strains appeared to result in low copy plasmid integration (about two copies per cell are suggested). The chloramphenicol acetyltransferase (CAT) assays showed that the promoter of the Acanthamoeba polyubiquitin gene in the constructed vector was especially strong in A. polyphaga, thus the pOPSBU-Acanthamoeba system may be useful for the construction of cDNA expression libraries, as well as for the expression of cloned genes.

[Effect of phentolamine on portal pressure in cirrhotic patients with portal hypertension].

Changes of hepatic venous wedge pressure (WHVP) and portal venous pressure (PVP) in fourteen patients with postnecrotic cirrhosis proved pathologically and seven patients with colon carcinoma without hepatopathy were observed before and after the infusion of alpha-adrenergic receptor antagonist phentolamine from peripheral venous. The results indicate that phentolamine can reduce WHVP and PVP of cirrhotic patients with portal hypertension efficiently (0.56-0.63 kPa, 0.42-0.50 kPa, respectively, P < 0.01), but it effects arterial diastolic pressure and heart rate simultaneously (arterial diastolic pressure reduced by 0.94-10.7 kPa, while heart rate increased by 8.67-10.67 per minute). Phentolamine doec not reduce portal pressure on patients without portal hypertension.

[Significance of endocrine cells and their hormones in colorectal cancer].

OBJECTIVE: To analyse the expression and clinical pathologic significance of endocrine cell (EC) and their hormones in colorectal cancer (CC). METHODS: Chromograin A (CGA) and five kinds of hormones in 117 cases of CC were detected by immunohistochemistry with 13 observed by electron microscopy. RESULTS: CGA positive cells (EC) occurred in 46 cases (39.3%). 69.2%(27/39) expressed more than one hormone. The lymph node metastasis rate (68.0%) in the EC-positive CC was higher than EC-negative CC (43.7%) (P < 0.05). The incidence of EC(+2) in CC with morderative and poor differentiated (25.5%) was higher than well differentiated (P < 0.05). EC-positive CC gave shorter survival. It's an independent factor for evaluating prognosis in CC. The expression of 5-HT, GLU, GAST and beta-HCG occurred more offen in tumor with moderate and poor differentiation giving poor prognosis (P < 0.05). Cancer cells containing endocrine granules were found in 8 cases of CC by electron microscopy. The two methods were comformable in 92.3%. CONCLUSION: EC-positive CC has poor histological differemtiation, giving rise to more lymph mode metastasis and poor prognosis. Some hormones secreted by EC could partly account for the growth and metastasis of CC.

[Determination of minor flavor components in Chinese spirits by direct-injection technique with capillary columns].

Chinese spirits are among the best-known distilled alcoholic beverages in the world. Because of the longer multiple-strain fermentation, Chinese spirits are richer than yeast fermented western liquors in fragrance and mellow. The complex nature of Chinese spirits often makes it difficult to separate, identify and quantify major and minor flavor components. Among the several analytical methods for flavor components in Chinese spirits developed in recent years, the direct-injection technique using capillary columns is by far the best. In this study of direct injection with crosslinked PEG 20M, HP-INNOWax and bonded SGE BP21 columns, up to 68 flavor components including alcohols, organic acids, esters, acetals, carbonyl and heterocyclic compounds were identified with fine-tuning separation and followed by GC/MS to identify. The gas chromatographic separation of flavour components of Maotai Liquor by the method on PEG 20M, SGE BP21 and HP-INNOWax are shown. The compounds of the numbered peaks in the chromatograms are listed. Three compounds of, ie., amyl acetate, t-pentanol and 2-ethyl butyric acid, were used as internal standards to minimize discrimination in quantitative determination. The operation procedures for these tests were provided. The application identities of three capillary columns for the analysis of minor flavor components with direct injection were summarized respectively.
