OBJECTIVE: To establish morphine withdrawal tachycardia model of rat and to observe the effect of electroacupuncture (EA) with different frequency on its heart rate. METHODS: Morphine dependence was induced in Wistar rats by injection of morphine for 8 days successively, and EA 1 mA in strength was given after withdrawal of morphine with different frequencies of 2 Hz, 15 Hz and 100 Hz, the heart rate and blood pressure were recorded in the awaked rats. RESULTS: Morphine abstinent rats showed a 24% increase of heart rate as compared with that of the normal control group, but no significant change was observed in blood pressure. The 100 Hz and 15 Hz EA produced a 13.4% and 13.1% decrease of heart rate respectively, whereas 2 Hz EA produced insignificant change. CONCLUSION: The reducing effect of percutaneous electric stimulation on tachycardia in heroin addicts revealed clinically can be replicated in the rat model.
OBJECTIVE: To explore the effect of Tiangui Recipe (TGR) on obesity and anovulation in androgen-sterilized rats (ASR) from the gene transcription level. METHODS: In situs hybridization and autoradiography were adopted, using alpha-32 P-radiolabelled leptin receptor (OB-R) and neuropeptide Y (NPY) oligonucleotides probes and integrating with image pattern analysis, to observe the effect of TGR on changes of OB-R, NPY mRNA expression in the hypothalamic arcuate nucleus (ARC). RESULTS: Compared to the normal rats, the levels of hypothalamic OB-R mRNA expression decreased, NPY mRNA expression elevated significantly in ASR (P < 0.01). ASR characterized by obesity and anovulation. There was no significant difference in hypothalamic OB-R, NPY mRNA expressions between normal rats and ASR with TGR administration. CONCLUSION: The increased hypothalamic NPY and decreased OB-R mRNA expression may be important contributing factors to the development of obesity and anovulation in ASR. TGR may play a role in reducing weight and inducing ovulation by regulating NPY/OB-R mRNA expression.
OBJECTIVES: To evaluate the optimal position of bony tunnel, the firmness of tendon graft fixation and the effectiveness of postoperative function recovery in an arthroscopic approach for the minimally invasive reconstruction of anterior cruciate ligament (ACL). METHODS: In a single arthroscopic approach, the ACL was reconstructed by the bone-tendon-bone compound patellar tendon autograft with the fixation of an interference screw. RESULTS: ACL reconstruction was performed in 49 patients, of whom 20 were followed up for over one year (average 1 year and 5 months). Among the 20 patients, 13 were rated as excellent, 5 good, and 2 fair. The excellent and good rate went up to 90%. Reconstructed ACLs were reevaluated arthroscopically in 9 patients, in whom, 6 had good remodeling of ACLs. CONCLUSIONS: Our results indicated that this method has the advantages of less injury, optimal positioning of bone tunnel, firm graft fixation, and early rehabilitation. Compound bone-tendon-bone autograft allows firm fixation and biologic healing with good shape and tension remodeling.
OBJECTIVE: To early reconstruct acute and complete rupture of anterior cruciate ligament (ACL) and treat combined injuries for the recovery of knee joint stability. METHODS: Ten cases of acute complete rupture of anterior cruciate ligament and medial collateral ligament were treated arthroscopically by anterior cruciate ligament reconstruction using patellar tendon autograft fixed with interference screw from February 1998 to March 1999. RESULTS: Follow-up ranged from 5 months to 1 year and 3 months (average 10 months). Clinical results showed that the stability of knee joint was satisfactory in the early stage. CONCLUSIONS: Acute the ACL rupture can be reconstructed arthroscopically in the early stage and the injuries were moderate, combined injuries could be treated at the same time, and the stability of knee joint could recover in the early stage after operation.
OBJECTIVE: To evaluate the effect of seawater immersion on multiple organ dysfunction syndrome (MODS) after chest trauma. METHODS: Twenty health dogs were divided into two groups. Right open pneumothorax was induced in both control group (n = 10) and experimental group (n = 10). After induction of chest trauma, animals in the experimental group were immersed in artificial seawater. Blood samples were taken at seven different intervals for assessing blood gas, plasma level of TNFalpha, and IL-1 beta. Changes of serum alanine aminotransferase (ALT), aspartate amino-transferase (AST), creatine kinase (CK) lactate dehydrogenase (LDH) and creatinine (Cr) were measured. At the end of study, lung was harvested for assessing lung water content and ratio of wet weight and dry weight. RESULTS: A significant elevation of ALT, AST, CK and LDH was observed in both groups. Organ functional parameters in the experimental group were consistent with the failure standard at 30 minutes after seawater immersion, and those in the control group at 4 hours. Post-trauma mortality and incidence of MODS were much higher in the experimental group than those in the control group (P < 0.01). The plasma levels of TNFalpha and IL-1 beta significantly increased at 30 minutes and reached the highest level at 60 minutes after seawater immersion. The time of peak level appeared earlier in the experimental group than that in the control group. CONCLUSION: Seawater immersion after open chest trauma results in high incidence of MODS and high mortality rate due to progressive dysfunction of multiple organs.
OBJECTIVE: To analyses the clinical results of transmyocardial laser revascularization (TMLR) for 77 patients with coronary artery diseases (CAD). METHODS: The mean age of the patients was (65 +/- 7) years. Previous medical record included CABG (6 patients), PTCA (9), AMI (66.2%), hypertension (70.1%), and diabetes mellitus (45.5%). TMLR was performed on the beating heart via a left anterolateral thoracotomy at the fifth intercostal space. Transesophageal echocardiography showed transmyocardial penetration of 23 +/- 6 channels. RESULTS: The hospital mortality was 3.8%, and postoperative complications were AMI (3.8%), left ventricular failure (2.6%), PVC (5.2%). After operation, the mean CCS angina class was improved from the baseline 3.5 +/- 0.7 to 2.1 +/- 0.3 at 3 months, 1.7 +/- 0.3 at 6 months, 1.7 +/- 0.3 at 12 months and 1.8 +/- 0.4 at 24 months. One patient died of AMI and two died not due to cardiac events during a follow-up of 3 to approximately 24 months. Echocardiography showed that the average of left ventricular ejection fractions was improved significantly at 6 months after operation compared with the preoperative value (P = 0.0457). (201)TI-SPECT showed a remarkable improvement in reversible ischemia in 70% patients followed up. Metabolic stress test for 20 patients followed up patients demonstrated an average increase in exercise tolerance from 7.1 +/- 3.2 min at the baseline to 9.6 +/- 1.3 min at 12 months (P = 0.021). Similarly the METs increased from 4.3 +/- 2.1 at the baseline to 5.4 +/- 2.0 at 12 months. CONCLUSIONS: TMLR is a safe and effective procedure for the treatment of end stage coronary artery diseases not amenable to PTCA or CABG. The effect of TMLR is associated with indication, correct evaluation of myocardial ischemia, and management for postoperative complications.
OBJECTIVE: To study the experimental basis for the treatment to the sick and wounded with abdominal open wound in land warfare and naval battle. METHODS: Animal model was made with adult dogs to observe the changes of mean artery pressure (MAP), pulmonary capillary wedge pressure (PCWP), central vein pressure (CVP), cardiac output (CO), cardiac index (CI) and pathologic changes of heart, pulmonary, hepatic, nephritic, intestinal, and gastric tissues under the optical and electrical microscope. RESULTS: Comparing with simple open abdominal, the hemodynamic disorder in open wound after seawater immersion was more serious; with a higher death rate. CONCLUSIONS: There was differences in hemodynamic and pathologic changes in seawater immersion wound and general wound. The body damaged is seriously when open abdominal wound immersed in seawater. Special measure must be taken in treating open abdominal wound after seawater immersion.
The inhibitory effects of atrial natriuretic peptide (ANP), C-type natriuretic peptide (CNP) and vasonatrin peptide (VNP) on the proliferation of pulmonary artery smooth muscle cells (PASMCs) were compared in rats. Total protein and OD value of MTT were detected in cultured rat PASMCs to examine the influence of the three peptides on PASMC proliforation induced by phorbol 12-myristate 13-acetate (PMA). The results show that PMA (10( 9) 10( 7) mol/L) increased and VNP (10( 8) 10( 6) mol/L), ANP and CNP (10( 7) 10( 6) mol/L) decreased the total protein and OD value in a dose-dependent manner (P<0.05). The data above suggest that PMA stimulates the proliferation of PASMCs while the three peptides inhibit the proliferation induced by PMA. Of the three peptides VNP has the strongest inhibitory effect.
In growth chambers, a system was developed for infection of Polymyxa graminis to wheat(Triticum aestivum L.), which is an obligate parasite. By the system, the fungus could be propagated in large quantity rapidly and the life cycle of P. graminis in wheat was observed in a period of 13 to 15 days. The methods for separating P. graminis as single cystosori from cultured wheat roots or dried roots were simplified. By tests for the factors affecting on P. graminis infection, including inocula types, seedling age of host plants, temperatures, pH value and nutrient contents etc., the system was modified to more perfection. With an integration of optimal temperature and other elements in the system, which affect on infection and development of P. graminis, wheat yellow mosaic virus (WYMV) was transmitted constantly and efficiently by the fungal vector. In average, 70% of wheat plants inoculated by the viruliferous fungi could infected by WYMV and the typical symptom of wheat mosaic disease would appear at 30 days post-inoculation mechanically.

In order to obtain a real-time and correct test for body water equilibrium in hemodialysis patients, an instrument based on bioimpedance measurement has been developed. The hardware and software design of the instrument was introduced in this paper. The instrument is of safty and good duplicity. Preliminary clinical experiments shows that it can reflect the state of body water with relative high sensitivity.
This report is the summary of our research on the chemical constitutents and their antitumor activities of Annonaceae plants. Annonaceous acetogenins, polyoxy-cyclohexenoids, and styryl-lactones from Annonaceae are the main constituents of antitumor activities. They are expected to develop a kind of new antitumor medicines.
A rapid graphite furnace atomic absorption spectrometric method for the determination of trace copper and lead in surface water is investigated in this paper. The influences of the program for graphite furnace and matrix modifier are systematically researched. A rapid method without ashing step and chemical matrix modifier is established. The determination limits of copper and lead are 0.9 microgram.L and 1.2 micrograms.L, respectively. The relatively standard deviations (RSD) (n = 1) are 2.5% and 4.1%, respectively. The recoveries are 96.0%-97.2% and 90.0%-92.0%, respectively.
The latest development of the molecular vibrational spectroscopy (Fourier Transform Raman Spectroscopy and Fourier Transform Infraed Spectroscopy) and the latest application of the non-destructive identification, quantitative analysis, thermal-stability monitoring and Chinese medicine optimizing were described in this paper. Some Chinese crude herbs can be directly and quickly identified by Fourier Transform Raman spectroscopy (FT - Raman) and Diffused Reflection Fourier Transform Infrared Spectroscopy (DR-FTIR). The content of Chinese herbs, medicine and medicament can be quantified without destruction by Diffused Reflection Fourier Transform and Near Infrared Spectroscopy (DR-NIR-FTIR). The mechanism of interaction between the Chinese medicine and some cells or bacteria can be in situ monitored by FT-Raman and HATR-FTIR. Best medicine could be selected and medicament could be optimized according to the injury degree of the cell or bacteria. The decomposing and degenerating process of medicines can also be timely monitored by FT-Raman and the temperature controlled FTIR. These are the latest use of molecular spectroscopy in Chinese medicine. The fast indentification, quantitative analysis monitoring, thermal stability predicting and the optimizing of Chinese herbs, medicine and medicament can be guided by the above mentioned methods. It is proved that these methods are nondestructive to samples, fast accurate, and easy to use.
The microcavity is sandwiched between a quarterwavelength distributed Bragg reflector(DBR) and a metal Ag reflective mirror. A single layer of a Tris(8-quinolinolato) aluminum (Alq) film was used as the light-emitting layer. The photoluminescent properties of the optical microcavity and that of the Alq film were studied at the same excitation condition. Compared with the Alq film, the significantly narrowed spectral emission linewidth from 90 nm to 10 nm was observed, the PL emission intensity of the microcavity at the resonant mode is enhanced by the order of 1. The spectral narrowing and intensity enhancement of the microcavity is attributed to the microcavity effect.
Sequence-specific transactivation by p53 is essential to its role as a tumor suppressor. A modified tetracycline-inducible system was established to search for transcripts that were activated soon after p53 induction. Among 9,954 unique transcripts identified by serial analysis of gene expression, 34 were increased more than 10-fold; 31 of these had not previously been known to be regulated by p53. The transcription patterns of these genes, as well as previously described p53-regulated genes, were evaluated and classified in a panel of widely studied colorectal cancer cell lines. "Class I" genes were uniformly induced by p53 in all cell lines; "class II" genes were induced in a subset of the lines; and "class III" genes were not induced in any of the lines. These genes were also distinguished by the timing of their induction, their induction by clinically relevant chemotherapeutic agents, the absolute requirement for p53 in this induction, and their inducibility by p73, a p53 homolog. The results revealed substantial heterogeneity in the transcriptional responses to p53, even in cells derived from a single epithelial cell type, and pave the way to a deeper understanding of p53 tumor suppressor action.
We have identified a novel Src homology 2 domain-containing leukocyte protein of 76 kD (SLP-76)-related molecule which we have termed Clnk (for cytokine-dependent hemopoietic cell linker). Unlike its relatives SLP-76 and B cell linker protein (Blnk), Clnk is not expressed uniformly within a given hemopoietic cell lineage. Even though it can be detected in several cell types, including T cells, natural killer cells, and mast cells, its expression seems to be strictly dependent on sustained exposure to cytokines such as interleukin (IL)-2 and IL-3. Strong support for the notion that Clnk is involved in immunoreceptor signaling was provided by the observation that it inducibly associated with at least one tyrosine-phosphorylated polypeptide (p92) in response to immunoreceptor stimulation. Moreover, transient expression of Clnk caused an increase in immunoreceptor-mediated signaling events in a T cell line. Taken together, these results show that Clnk is a novel member of the SLP-76 family selectively expressed in cytokine-stimulated hemopoietic cells. Furthermore, they suggest that Clnk may be involved in a cross-talk mechanism between cytokine receptor and immunoreceptor signaling.
