New dextran derivatives. I. Nitrogen containing derivatives.

Three new derivatives of dextran were obtained: 3-morpholine-2-hydroxypropyl ether of dextran (I), hydroxamic acid of carboxymethyl dextran (II), and oxime of ketodextran (III). All the three derivatives exhibited low toxicity at parenteral administration to laboratory animals. (I) and (II) showed the capability of weakening the activity of tetanus and malignant edema toxins. Additionally, (I) showed immunotropic activity. Biological activity was observed when the derivative (I) had been obtained from dextran of the molecular weight 4000. At the molecular weight 20,000 the activity did not appear. Similarly, no activity was observed under oral administration.

New dextran derivatives. II. Preparation of compounds of copper (II) with 3-mercapto-2-hydroxypropyl ether of dextran.

The non-ionic compound of 3-mercapto-2-hydroxypropyl ether of dextran with cupric sulphide was obtained. The compound (called C-79) exhibited biological activity at parenteral administration to laboratory animals. The method of obtaining the compound as well as the properties of the latter were described. The results of gel chromatography suggested the existence of certain analogies with the known compounds of iron with dextran.

Immunological properties of 3-morpholine-2-hydroxypropyl ether of dextran (H-2).

The immunobiological investigations of H-2 preparation--a modified derivative of DEAE dextran--revealed that the preparation has a wide immunotropic spectrum with the predominance of the stimulatory effect. It stimulates the I and II humoral immunological responses to SRBC. It stimulates the induction and effector processes of the cellular type reactions. Under the in vivo and in vitro conditions it causes mouse precursor T cells to gain the theta antigen. On the other hand, H-2 preparation suppresses the development of GvH reaction induced either by the administration of cells coming from the donor treated with the preparation (in vivo), or by the incubation of the donor's cells with the compound under examination. H-2 preparation is non-toxic, and does not exhibit any antiviral or interferogenic activity.

Immunobiological properties of the complex of bivalent copper with 3-mercapto-2-hydroxypropyl ether of dextran (C-79).

The immunological investigations were carried out on mice with the aim to determine the immunotropic properties of the complex of bivalent copper with 3-mercapto-2-hydroxypropyl ether of dextran (C-79), a derivative of previously described 3-morpholine-2-hydroxypropyl ether of dextran (H-2). In the examinations of the copper complex also the H-2 preparation was applied, and for comparative purposes--levamisol, a popular stimulator of immunological responses. It was stated that C-79 similarly to H-2 and levamisol, stimulated the humoral and cellular anti-SRBC immunological responses. Moreover, it increased the number of cells producing anti-H-B antibodies, stimulated the appearance of thy-1,2 antigen on the precursor T cells and inhibited the in vivo and in vitro GvH reactions. H-2 and C-79 preparations intensified the effect of the inductor of the Poli I:C.

Anti-infectious properties of the complex of bivalent copper with 3-mercapto-2-hydroxypropyl ether of dextran (C-79).

The anti-infectious activity of the C-79 preparation was examined on mice intravenously infected with Shigella sonnei dysentery of phase I. The activity of the preparation was determined taking into account the number of bacteria cultured from the organs of the examined animals and the number of animals surviving the infection as compared with the control. C-79 preparation administered to the mice before the infection was observed to protect then from death almost in 100%. The activity of the preparation differed according to the strategy of administration.

Oncostatic properties of the complex of bivalent copper with 3-mercapto-2-hydroxypropyl ether of dextran (C-79).

The oncostatic properties of C-79 preparation were examined on the mice with: sarcoma Sa-180, melanoma B-16, Ehrlich carcinoma and leukemia P-388. The preparation was observed to prolong the life of animals with sarcoma Sa-180 and to modulate the morphology of tumors in them. The preparation had no effect on three remaining investigation models. The investigations carried out on mice of AKR strain have revealed that C-79 significantly suppresses the occurrence of spontaneous leukemia, prolonging the mouse survival-time.

Radioprotective activity of the complex of bivalent copper with 3-mercapto-2-hydroxypropyl ether of dextran (C-79).

The radioprotective activity of C-79 preparation was determined in the course of investigations carried out on mice. The activity was measured by the survival time of lethally irradiated animals and by means of the number of colonies (CFUs) on the spleens of irradiated animals. The endogenous and exogenous CFUs were determined. It was stated that the preparation, when introduced into animals several days before the irradiation with an appropriate dose of X rays, protected the homeopoetic cells of the examined animals against the irradiation effects. It was also revealed that the preparation significantly prolonged the survival time of the lethally irradiated mice. The protection activity of the preparation depended on the dose and the strategy of administration.
