Life on the edge: African malaria mosquito (Anopheles gambiae s. l.) larvae are amphibious.

Anopheles gambiae s.l. is the main vector of malaria in Sub-Saharan Africa. Here, an estimated 1 million people die every year from this disease. Despite considerable research on An. gambiae that increasingly explores sub-organismal phenomena, important facets of the field biology of this deadly insect are yet being discovered. In the current study, we used simple observational tools to reveal that the habitat of larval An. gambiae is not limited within the boundaries of temporary mud puddles, as has been the accepted generalization. Thus, control tactics aimed at immatures must consider zones larger than puddles per se. In fact, eggs are more likely to be found outside than inside puddles. Eggs can develop and larvae can emerge on mud. Larvae are then capable of three distinct modes of terrestrial displacement (two active and one passive), whereby, they can reach standing water. On mud bearing a film of water, larvae actively displace backwards by sinusoidal undulations shown to be only a slight variation of the swimming motor program. On drying mud, larvae switch to a slower and forward form of active locomotion resembling that of a crawling caterpillar. During rains, small larvae may be passively displaced by flowing rainwater so as to be deposited into puddles. These capabilities for being amphibious, along with very rapid growth and development, help explain how An. gambiae thrives in a highly uncertain and often hostile larval environment.

Daily temperature profiles in and around Western Kenyan larval habitats of Anopheles gambiae as related to egg mortality.

BACKGROUND: Anopheles gambiae eggs are more frequently found on soil around puddle habitats of the larvae, than on the water surface itself in Western Kenya. Thus, eggs can experience temperatures more wide-ranging and lethal than those experienced by larvae or pupae confined within puddles. METHODS: Small batches of eggs from house-collected An. gambiae as well as from the Kisumu laboratory strain were bathed for defined times in water whose temperature was precisely controlled. Daily temperature profiles were recorded by an infrared thermometer on seven different days in and around three types of typical An. gambiae larval habitats at Kisian. RESULTS: For wild eggs, significant mortality occurred upon brief heating between 42-44 degrees C. Few eggs hatched after 10 min at 45 degrees C and none hatched above this temperature. A similar pattern occurred for eggs of the Kisumu strain, except it was shifted downwards by 1 degrees C. Egg mortality was time-dependent above 40 degrees C. Temperatures of water in the three types of larval habitats never exceeded 35 degrees C. Wet or damp mud rarely and only briefly exceeded 40 degrees C; thus, water and mud would be highly conducive to egg survival and development. However, dry soils frequently reached 40-50 degrees C for several h. Eggs stranded on dry surfaces would experience substantial mortality on hot, sunny days. CONCLUSION: Moist mud around puddles constitutes suitable habitat for An. gambiae eggs; however, eggs on the surface of dry soil under direct sunlight are unlikely to survive for more than a few hours.

Laboratory tests of oviposition by the African malaria mosquito, Anopheles gambiae, on dark soil as influenced by presence or absence of vegetation.

BACKGROUND: Physical objects like vegetation can influence oviposition by mosquitoes on soil or water substrates. Anopheles gambiae s. l. is generally thought to utilize puddles over bare soil as its prime larval habitat and to avoid standing water populated with vegetation. In Kisian, Kenya near Kisumu, water often pools in grassy drainage areas both during and after periods of infrequent rains, when typical puddle habitats become scarce because of drying. This raised the question of whether An. gambiae has the behavioural flexibility to switch ovipositional sites when puddles over bare soil are unavailable. METHODS: To test whether presence and height of grasses influenced oviposition, wild-caught gravid An. gambiae s. l. were offered paired choices between wet, bare soil and wet soil populated with mixed grasses or grasses of differing height. No-choice tests were also conducted by giving females either grassy soil or bare soil. RESULTS: In choice tests, females laid four times more eggs on bare, wet soil than soil populated with grasses. However in no-choice tests, egg output was not significantly different whether grasses were present or not. Females laid significantly more eggs on soil populated with short grass than with medium, or tall grass. CONCLUSION: This work shows An. gambiae s. l. has the capacity to oviposit into grassy aquatic habitats when typical puddles over bare soil are unavailable. This knowledge will need to be considered in the design and implementation of programmes aimed at reducing malaria transmission by suppression of An. gambiae s. l. immatures.

Prognostic significance of nuclear receptor coactivator-3 overexpression in primary cutaneous melanoma.

PURPOSE: To assess the prognostic significance of nuclear receptor coactivator-3 (NCOA3) overexpression in primary cutaneous melanoma. PATIENTS AND METHODS: NCOA3 expression was assessed using immunohistochemical analysis of a melanoma tissue microarray (TMA) containing primary melanomas from 343 patients with defined histology and follow-up. The impact of the presence or absence of various prognostic factors on relapse-free survival (RFS) and disease-specific survival (DSS) of melanoma patients was assessed using Cox regression and Kaplan-Meier analysis. The impact of presence or absence of various factors on sentinel lymph node (SLN) metastasis was assessed using logistic regression analysis. RESULTS: Increasing degree of NCOA3 expression was significantly predictive of SLN metastasis (P = .013) and the mean number of SLN metastases (P = .031). Kaplan-Meier analysis demonstrated a significant association between NCOA3 overexpression and reduced RFS (P = .021) and DSS (P = .030). Logistic regression analysis revealed increasing degree of NCOA3 expression to be an independent predictor of SLN status (P = .017). Multivariate Cox regression analysis showed the independent impact of NCOA3 expression on RFS (P = .0095) and DSS (P = .021). NCOA3 was the most powerful factor predicting DSS, outperforming tumor thickness and ulceration. CONCLUSION: These results identify NCOA3 as a novel, independent marker of melanoma outcome, with a significant impact on SLN metastasis, RFS, and DSS.

Efficacy of severe acute respiratory syndrome vaccine based on a nonhuman primate adenovirus in the presence of immunity against human adenovirus.

Replication-deficient human adenovirus type 5 (AdH5) vectors can induce strong transgene product-specific cellular and humoral responses. However, many adult humans have neutralizing antibodies (NAbs) against AdH5 as a result of natural infection with this virus. Therefore, a chimpanzee adenovirus C7 (AdC7) vector was developed to circumvent interference by preexisting immunity to AdH5. This study evaluated the impact of preexisting immunity to human adenovirus on the efficacy of adenovirus-based vaccines against the coronavirus that causes severe acute respiratory syndrome (SARS-CoV). Efficacy was assessed after intramuscular injection of the vector into mice and was measured as the frequency of SARS-CoV-specific T cells and NAbs against SARS-CoV. Immunogenicity of the AdH5-based vaccine was significantly attenuated or completely abolished when the preexisting anti-AdH5 NAb titer was higher than 40. Because 27% of human serum samples from the United States tested so far have an anti-AdH5 NAb titer higher than 40, our results suggested that a significant percentage of humans with preexisting anti-AdH5 immunity would not be candidates for vaccination with an AdH5-based genetic vaccine. In contrast, preexisting anti-AdH5 NAbs have a minimal effect on the potency of the AdC7-based genetic vaccine. Taken together, our studies warrant the further development of AdC7 as a vaccine carrier for human trials.

Primary lung abscess caused by multidrug-nonsusceptible Streptococcus pneumoniae in a child.

We report a 21-month-old boy with primary lung abscess caused by multidrug-nonsusceptible Streptococcus pneumoniae. A computed tomography-guided aspirate obtained on admission grew penicillin-, ceftriaxone-, erythromycin- and clindamycin-nonsusceptible S. pneumoniae. The patient was treated for 10 days with parental rifampin and cefotaxime and then for 4 weeks with oral cefpodoxime and recovered without complications.

Prognostic significance of extent of ulceration in primary cutaneous melanoma.

Ulceration has been shown to be an adverse prognostic factor in primary cutaneous melanoma. However, the extent of ulceration required for histologic identification and biologic significance is unclear. We examined the impact of extent of ulceration on melanoma outcome in a cohort of 235 melanoma patients by evaluating the relationship between percentage of ulceration in the vertical growth phase of the primary tumor and 2 outcome parameters: sentinel lymph node status and overall survival. We measured the diameter of the ulcerated area in millimeters over the diameter of the entire vertical growth phase. There was a statistically significant relationship between increasing percentage of tumor ulceration and both sentinel lymph node status as well as overall survival, with a binary cut-off point of 2% for sentinel lymph node status and 5% for overall survival. The percentage of ulceration provides additional prognostic information in predicting sentinel lymph node status and in determining survival in melanoma patients. These results suggest that no more than minimal ulceration is required to have a prognostic impact on melanoma survival.

Generation of an adenoviral vaccine vector based on simian adenovirus 21.

Adenoviral vectors can be used to generate potent humoral and cellular immune responses to transgene products. Use of adenoviral vectors based on non-human isolates may allow for their utilization in populations harbouring neutralizing antibodies to common human serotypes. A vector chimera was constructed using simian adenovirus 22 (a serotype belonging to the species Human adenovirus E) and simian adenovirus 21 (a serotype belonging to the species Human adenovirus B) expressing the Ebola (Zaire) virus glycoprotein (Ad C5/C1-ZGP). This chimeric adenovirus vector was used as a model to test its efficacy as a genetic vaccine and comparisons were made to a vector based on the commonly used human adenovirus C serotype 5 (Adhu5-ZGP). Ebola glycoprotein-specific T- and B-cell responses were measured in B10BR mice vaccinated with either Adhu5-ZGP or Ad C5/C1-ZGP vectors. Both vectors resulted in Ebola glycoprotein-specific gamma interferon-expressing T cells, although the Ad C5/C1-ZGP vector appeared to induce lower frequencies with kinetics slower than those elicited by the Adhu5-ZGP vector. The total immunoglobulin G response to Ebola glycoprotein was similar in sera from mice vaccinated with either vector. Two rhesus macaques vaccinated with the Ad C5/C1-ZGP vector were found to mount T-cell and antibody responses to the Ebola glycoprotein. It was found that a single administration of the chimeric Ad C5/C1-ZGP vector protected mice against a lethal challenge with a mouse-adapted strain of the Ebola (Zaire) virus.

Anopheles gambiae (Diptera: Culicidae) oviposition in response to agarose media and cultured bacterial volatiles.

Anopheles gambiae s.s. Giles accepted a range of agarose gels, varying from 0.5 to 8% (wt:vol), for oviposition; laid more eggs on 0.5% agarose gels than on moist filter paper or on drier agarose; and laid equal numbers of eggs on 0.5% agarose gels and distilled water. Larvae hatched on agarose gel substrates and crawled onto the surface of moist agarose gels, but they tended only to burst the egg cap and remain within the egg case on drier gels. A mixture of cultured bacteria, originating from a natural larval habitat of An. gambaiae s.s. in western Kenya, was classified by 16S rDNA sequence analysis, and then this mixture or individual colonies from it was used as odor sources in ovipositional experiments with agarose substrates. Of 61 sequences from a mixed clone sequence library, most (78%) were Pseudomonas strains, whereas the remainder were Stenotrophomonas, Enterobacter, Pantoea, Klebsiella, Acinetobacter, Aeromonas, and Bacillus. Oviposition was significantly reduced when mixtures of bacterial colonies of these strains, or a lawn of colonies of a field isolate of S. maltophilia, was presented. Oviposition was neither reduced nor enhanced when field isolates of Pseudomonas putida or Pseudomonas alcaligenes colonies were presented. These results suggest that gravid An. gambiae females are sensitive to bacterial-derived odors emanating from cultured bacteria from natural larval habitats and that some bacterial odors may be repellent.

Biodiversity conservation and the extinction of experience.

Biodiversity loss is a matter of great concern among conservation scientists, but the wherewithal to reverse this trend is generally lacking. One reason is that nearly half of the world's people live in urban areas and are increasingly disconnected from nature. If there is to be broad-based public support for biodiversity conservation, the places where people live and work should be designed so as to provide opportunities for meaningful interactions with the natural world. Doing so has the potential not only to engender support for protecting native species, but also to enhance human well-being. Accomplishing these goals will necessitate conservation scientists forging new collaborations with design professionals, health practitioners and social scientists, as well as encouraging the participation of the general public.

The gene expression signatures of melanoma progression.

Because of the paucity of available tissue, little information has previously been available regarding the gene expression profiles of primary melanomas. To understand the molecular basis of melanoma progression, we compared the gene expression profiles of a series of nevi, primary melanomas, and melanoma metastases. We found that metastatic melanomas exhibit two dichotomous patterns of gene expression, which unexpectedly reflect gene expression differences already apparent in comparing laser-capture microdissected radial and vertical phases of a large primary melanoma. Unsupervised hierarchical clustering accurately separated nevi and primary melanomas. Multiclass significance analysis of microarrays comparing normal skin, nevi, primary melanomas, and the two types of metastatic melanoma identified 2,602 transcripts that significantly correlated with sample class. These results suggest that melanoma pathogenesis can be understood as a series of distinct molecular events. The gene expression signatures identified here provide the basis for developing new diagnostics and targeting therapies for patients with malignant melanoma.

The role of microsatellites as a prognostic factor in primary malignant melanoma.

OBJECTIVE: To determine the impact of microsatellites as a prognostic factor in primary cutaneous melanoma. DESIGN: Retrospective cohort study. SETTING: Tertiary referral center. Patients A total of 504 patients with a history of primary melanoma observed for 2 years or having experienced a first relapse. MAIN OUTCOME MEASURES: Overall survival (OS) and relapse-free survival (RFS). RESULTS: Forty-five patients had evidence of microsatellites in their primary melanoma. Presence of microsatellites significantly correlated with the presence of several other histologic high-risk factors such as tumor thickness, ulceration, Clark level, vascular factors, and mitotic rate. Univariate analysis demonstrated decreased RFS and OS in patients with microsatellites. Presence of microsatellites was associated with increased locoregional metastasis but not distant metastasis. In multivariate analysis, with the inclusion of 6 other clinical and histologic factors, presence of microsatellites was a significant predictor of RFS but not OS. Patients with clinical macrosatellites had a trend toward worsening OS compared with those with microsatellites. CONCLUSIONS: The presence of microsatellites is intimately tied to other markers of melanoma aggressiveness. Microsatellites appear to predict locoregional relapse and RFS but neither distant metastasis nor OS. These results may have implications for patient care as well as the inclusion of microsatellites in stage III of the current classification.

Method for the determination of perfluorooctanoic acid in air samples using liquid chromatography with mass spectrometry.

Perfluorooctanoic acid is a completely fluorinated carboxylic acid that is usually used in the ammonium salt form as a processing aid in the production of many fluoropolymers and fluoroelastomers. Ammonium perfluorooctanoate readily dissociates in water to give the ammonium and perfluorooctanoate ions. Perfluorooctanoate has been reported to be present in low levels in human serum in the United States and Europe. This study reports on the development and validation of a method for the determination of perfluorooctanoic acid in air samples. This method uses the Occupational Safety and Health Administration (OSHA) Versatile Sampler (OVS) with a nominal 0.3 micro m filter and polystyrene resin sorbent (XAD-2 or XAD-4) followed by determination of the perfluorooctanoate anion by liquid chromatography mass spectrometry. The method was validated in the range of 0.474 to 47.4 microg/m3 for a 480-L sample. Breakthrough studies showed samples could be collected at 1 L/min for 24 hours or at 15 L/min up to 8 hours without breakthrough. Extract storage stability tests showed that sample extracts in methanol remain stable in glass autosampler vials for up to 13 days following initial injection. Perfluorooctanoic acid stability on OVS tubes was unaffected at both refrigerated and ambient temperatures. The overall average retention efficiency was 92.1% with a pooled RSD95 of 5.8% at five concentration levels (0.474 microg/m3 to 47.4 microg/m3).

Severe retrognathia as a risk factor for recent onset painful TMJ disorders among adult females.

OBJECTIVE: To evaluate the importance of severe retrognathia as a risk factor for the development of recent onset painful TMJ disorders among adult females. DESIGN: Case-control study. SETTING: This study was conducted in a large health maintenance organization between 1998 and 1999 [Kaiser Permanente Northwest (KPNW), Portland, OR, USA]. PARTICIPANTS: Adult females with recent onset painful TMJ disorders (n=29) and normal controls (n=104). METHODS: Cases were recruited from the TMD clinic at Kaiser Permanente Northwest (KPNW). Controls were recruited from a dental clinic at KPNW. Case status was determined using a questionnaire; mandibular sagittal position was determined by measuring a research angle on facial photographs. The mean research angle for cases was compared to the mean for controls. Multivariable exact conditional logistic regression analysis was used to examine the demographic characteristics of cases and controls, and to determine the strength of association between recent onset painful TMJ disorders and severe retrognathia. The population attributable risk percentage (PAR%) and the attributable risk percentage (AR%) were calculated to further evaluate severe retrognathia as a risk factor. RESULTS: The mean research angle among cases (67.7 degrees; 95% CI=66.0-69.4) was smaller than among controls (71.6 degrees; 95% CI=70.7-72.5, P<0.001). The odds ratio for the association between case status and the presence or absence of severe retrognathia was elevated (OR=6.3; 95% CI=1.1-47.5, P=0.039). The PAR% and AR%, associated with severe retrognathia, were 13.3 and 84.1%, respectively. CONCLUSIONS: Severe retrognathia is strongly associated with recent onset painful TMJ disorders (OR=6.3). Only a small proportion of these disorders are attributable to severe retrognathia among the total population of adult females (PAR%=13.3%). However, a large proportion of these TMJ disorders are potentially attributable to severe retrognathia among adult females with severe retrognathia (AR%=84.1%).

The use of magnetic resonance imaging in the diagnosis and long-term management of multiple sclerosis.

Magnetic resonance imaging (MRI) continues to evolve, providing almost boundless information about tissue structure and function that can be obtained noninvasively in vivo. MRI is a key tool in the diagnosis and longitudinal monitoring of patients with multiple sclerosis (MS). The technique is highly sensitive for the definition of brain and spinal cord involvement in MS, including the ability to detect multifocal lesions, diffuse (occult) disease, and macroscopic atrophy. Conventional MRI techniques, which include T2-weighted, T1-weighted, and gadolinium-enhanced imaging, are used primarily to detect overt lesions but can also be used to quantify tissue atrophy. Variations of these techniques also are important, and refinements are under way. The ability to demonstrate lesion dissemination in space and time has led to the common use of MRI as a paraclinical measure to support a diagnosis of MS, including in patients with clinically isolated demyelinating syndromes. In addition, MRI is used to monitor the progress of disease in patients with clinically definite MS, including assessment of lesions and atrophy. The use of conventional MRI in the diagnosis and longitudinal management of patients with MS is the focus of this review.

NF-kappa B in the vascular progression of melanoma.

PURPOSE: To examine a model of melanoma progression based on vascular factors and the role of NF-kappa B in the vascular progression of melanoma. PATIENTS AND METHODS: A data set of 526 patients from the University of California San Francisco Melanoma Center with 2 years of follow-up or first relapse was studied. The impact of the presence or absence of various prognostic factors on overall survival of melanoma patients was assessed using Cox regression and Kaplan-Meier analysis. A matched-pair analysis of NF-kappa B expression was performed in cases with vascular involvement and increased tumor vascularity versus matched controls lacking these factors. RESULTS: Cox regression analysis of factors evaluated by the American Joint Committee on Cancer Melanoma Staging Committee reproduced the powerful impact of tumor thickness and ulceration in this data set. With the inclusion of vascular factors such as tumor vascularity and vascular involvement, ulceration was no longer significant in predicting overall survival. By multivariate analysis, vascular involvement and tumor vascularity were the strongest predictors of melanoma outcome. Tumor vascularity seems to be a precursor of both vascular involvement and ulceration. A matched-pair tissue array analysis demonstrated the significant correlation between overexpression of NF-kappa B-p65 and the development of vascular factors. CONCLUSION: Vascular factors play an important role in the progression of malignant melanoma. Ulceration may be a surrogate marker for the interactions between melanoma and the tumor vasculature. NF-kappa B seems to play an important role in the development of these factors.
