Synthesis of an X-ray opaque biodegradable copolyester by chemical modification of poly (epsilon-caprolactone).

Poly (epsilon-caprolactone-co-alpha-iodo-epsilon-caprolactone) was synthesized by binding iodine to PCL chain bearing carbanionic site on alpha-position of carbonyl groups using lithium diisopropylamide. Copolyesters containing from 10% to 25% of iodo-units were thus obtained. Viscoelastic properties (modulus, loss angle), thermal properties (T(m), T(c), DeltaH(m)), crystallinity and in vitro degradability of this new type of copolymers were measured. Their opacity to X-rays was assessed, and appeared high enough to be of interest for biomedical applications.

Lactic acid-based functionalized polymers via copolymerization and chemical modification.

Poly(lactic acid) polymers (PLA) are presently the most attractive compounds in the field of artificial degradable and biodegradable polymers. In order to enlarge the family, and thus the range of accessible properties, stereocopolymers and copolymers with various co-monomers have been synthesized. However, very few are functionalized, i.e. include functional groups attached to the main chains or as part of the side chains. In the search for degradable PLA-type polymers bearing functional groups to serve as intermediates for further chemical modifications, we are exploring two different routes. The first one is copolymerization with a protected hydroxyl-bearing lactide-type monomer, namely 3-(1,2,3,4-tetraoxobutyldiisopropylidene)dioxane-2,5-dione. The second route consists of the formation of a carbanionic site in the alpha-position to intrachain carbonyl functions by using lithium N,N-diisopropylamide followed by the coupling of electrophiles. Recent advances in this search are presented using several examples. In particular, it is shown that OH-functionalized PLA-type macromolecules can be made fluorescent by chemical coupling. It is also shown that substituents can be attached to PLA-type macromolecules in solution or to the surface of PLA-based devices selectively.

Norfloxacin-poly(L-lysine citramide imide) conjugates and structure-dependence of the drug release.

Norfloxacin (Nflx), an antibiotic which is active against some intracellular bacteria, was coupled to a polymeric carrier, namely poly(L-lysine citramide) via a lysine or an ethylcarbamate spacer to obtain a macromolecular prodrug. The carrier, which derived from the two metabolites citric acid and L-lysine, is known to be biocompatible and slowly degradable under slight acidic conditions. Conjugates were characterised by UV, 1H and 13C NMR and SEC. The presence of Norfloxacin and the lysine type spacer caused chain aggregation, due to a probable physical cure. The release of Norfloxacin from these prodrugs and from a prodrug where Norfloxacin is bound to the carrier backbone without spacer arm was investigated comparatively in vitro. Conjugation via a carbamate-type linkage appeared as a method to achieve the release of Norfloxacin from a PLCA-type conjugate at neutral.

Influence of polymerization conditions on the hydrolytic degradation of poly(DL-lactide) polymerized in the presence of stannous octoate or zinc-metal.

Laboratory- and pilot-scale racemic polylactides (PLA50) were synthesized in the presence of stannous octoate (SnOct2) or zinc-metal as initiators in the absence of alcohol. The resulting polymers were processed by compression molding or injection molding depending on the batch scale. The hydrolytic degradation of compression-molded samples selected to be comparable was investigated first in order to show the influence of the initiator system. Differences in water uptake were found between PLA50-Zn (zinc-metal initiation) and PLA50-Sn (SnOct2 initiation). PLA50-Zn being much more hydrophilic. PLA50-Sn exhibited a slower molecular weight decrease and delayed onsets of weight loss, release of acidity and stereocomplex formation, with respect to PLA-Zn. The concentration in residual tin in PLA50-Sn increased from 306 to 795 ppm during aging. In the case of PLA50-Zn the residual metal remains constant at ca. 40 ppm. In a second series of experiments, high molecular weight PLA50 different in characteristics and in initiator, synthesized under pilot-scale, were compared. The effects of the initiator on the degradation of the polymers well agreed with laboratory-scale findings, differences in hydrophobicity being enlarged by the up scaling. PLA50-Sn polymers appeared much more degradation resistant than PLA50-Zn ones. Contributions of the other characteristics (e.g. molecular weight, purity, stereoregularity, processing) were shown to be important as well.
