Examining the relationship between obsessive-compulsive disorder and attention-deficit/hyperactivity disorder in children and adolescents: a familial risk analysis.

BACKGROUND: To use family study methodology to examine the relationship between obsessive-compulsive disorder (OCD) and attention-deficit/hyperactivity disorder (ADHD) in children and adolescents. METHODS: We assessed for ADHD and OCD in the 1533 first-degree relatives of three groups of index children: those with ADHD and OCD, those with ADHD but no OCD, and matched controls with neither disorder. RESULTS: The risk for ADHD was similarly elevated in families of ADHD youth with (18.9%) and without OCD (20.1%; p = .91), and both groups had significantly higher rates of ADHD compared with controls (4.6%; p < or = .001), which was consistent with previous research showing a strong familial risk for ADHD. The risk for OCD was significantly elevated only among relatives of youth with ADHD plus comorbid OCD (13.0%) compared with controls (.5%; p < or = .001) and was consistent with previous research showing a strong familial risk for OCD. Relatives affected with ADHD had a significantly elevated risk for OCD compared with relatives unaffected by ADHD (7.4% vs. 1.3%; p < .001), suggestive of co-segregation between these disorders. There was no evidence of nonrandom mating between ADHD- and OCD-affected spouses. CONCLUSIONS: These results extend previously reported findings regarding the heritability of both ADHD and OCD and provide new evidence of a familial relationship between ADHD and pediatric OCD that best fits the hypothesis of a unique familial subtype.

High-throughput TILLING for Arabidopsis.

Targeting induced local lesions in genomes (TILLING) is a general strategy for identifying induced point mutations that can be applied to almost any organism. In this chapter, we describe the basic methodology for high-throughput TILLING. Gene segments are amplified using fluorescently tagged primers, and products are denatured and reannealed to form heteroduplexes between the mutated sequence and its wild-type counterpart. These heteroduplexes are substrates for cleavage by the endonuclease CEL I. Following cleavage, products are analyzed on denaturing polyacrylamide gels using the LI-COR DNA analyzer system. High-throughput TILLING has been adopted by the Arabidopsis TILLING Project (ATP) to provide allelic series of point mutations for the general Arabidopsis community.

The effects of a typing task on the hand volume of the older worker.

UNLABELLED: Few studies have examined the relationship between changes in hand volume and typing. Volumetric evaluation documents the presence of an inflammatory response to activity. METHOD: To study this relationship, researchers used a convenience sample of 31 participants who were aged 45 and older and worked full time where they typed at least one hour per day. Volumetric measurements were taken before a 15-minute typing activity, immediately after and 10 minutes after typing. Before typing, ergonomic adjustments were made to computer workstations, when necessary. Participants typed using an advanced speed test of the Mavis Beacon Teaches Typing program, which calculated adjusted words per minute. RESULTS: The data showed no significant (F = 0.321, p > 0.05) change in hand volume resulting from the typing activity. CONCLUSION: The amount of exercise provided in this study, a fifteen minute typing activity, did not result in changed hand volume.

Porcelain gallbladder.

OBJECTIVE: To discuss the case of a porcelain gallbladder found incidentally in a patient with low back and heel pain. CLINICAL FEATURES: A 70-year-old woman had low back pain, numbness in the left lower leg, and sharp pain in her left heel. Plain films of the lumbar spine necessitated diagnostic abdominal ultrasound, the findings of which were consistent with porcelain gallbladder. INTERVENTION AND OUTCOME: The patient has been recommended for prophylactic cholecystectomy and is concurrently being treated for mechanical low back and heel pain. CONCLUSIONS: Porcelain gallbladder is an uncommon finding; however, due to the greatly increased chance of malignancy, it must be considered in patients who have cystic type calcification in the right upper abdominal quadrant.
