[Effects of small interfering RNA magnetic nanoparticles combination with external magnetic fields on survivin gene expression of bladder cancer cells and apoptosis].

OBJECTIVE: To investigate the effects of the small interfering RNA plasmid-dextran magnetic nanoparticles (siRNA-DMN) combination with external magnetic fields on silencing survivin gene expression of bladder cancer cells and apoptosis when DMN used as gene carrier to transfer siRNA-survivin recombinant plasmid in vivo. METHODS: The siRNA-survivin recombinant plasmid specific targeted survivin was synthesized in previous experiment. DMN were prepared by chemical coprecipitation method and used as gene carrier. The siRNA-DMN were constructed by static electricity of polylysine and transferred into human bladder cancer BIU-87 cells with the help of external magnetic fields. The growth inhibiting rate (IR) of BIU-87 cells was observed by 3-[4, 5-dimethylthiazol-2-yl]-2, 5-diphenyl tetrazolium bromide's test and the apoptosis index (AI) was detected by transferase-mediated dUTP nick end labeling method. The relatively transcription levels of survivin mRNA and protein expression were respectively detected by semi-quantitive reverse transcription polymerase chain reaction and Western Blotting techniques. RESULTS: The diameter, effective diameter and saturation magnetization of DMN-siRNA were about 10 - 12 nm, 94.8 nm and 0.19 emu/g, respectively. The IR (39.60%) and AI (28.72%), the relative expression of survivin mRNA and protein of siRNA-DMN combination with external magnetic fields on BIU-87 cells were significantly higher and lower than those in the control group and single siRNA-DMN group, respectively (P < 0.05). CONCLUSIONS: The siRNA-survivin plasmid-DMN combination with external magnetic fields could effectively inhibit survivin expression and induce BIU-87 cells apoptosis which provided experimental basis for the magnetic targeting gene therapy of bladder tumor.

[Expression of key cell cycle markers in squamous cell carcinoma of the cervix positive in human papillomavirus: a comparative study between Chinese and Australian populations].

OBJECTIVE: To investigate the patterns of expression of key cell cycle proteins targeting on the human papillomavirus (HPV) sites E6 and E7 in cervical carcinoma. METHODS: Semiquantitative immunohistochemistry was used to determine the expression of the cell cycle regulatory factor p53, retinoblastoma gene product pRb, cyclin-dependent kinase inhibitors p21(CIP1/WAF1) (p21), p16(INK4A) (p21), and p27(KIP1) (p21), and cyclin D1 targeting on the HPV sites E6 and E7 in 73 HPV 16-positive specimens of cervical squamous cell carcinoma, 35 from Australia patients and 38 age, lymph node status, and grade of tumor-matched Chinese patients. RESULTS: There were no significant differences in age, lymph node status, and grade of tumor between the Chinese and Australian groups, however, the number of the patients in advanced stage (>Stage IIa) was greater in the Chinese group than in the Australian group (19:7). The positive rate of p53 in the Australian group was 3%, significantly lower than that in the Chinese group (23%, P = 0.028). The upregulation rate of pRb, p21, and p27 in the Australian group were 56%, 63% and 34% respectively, all significantly higher than those in the Chinese group (89%, 97%, and 89% respectively, P = 0.01, P < 0.001, and P < 0.001). There were no significant differences in the upregulation rate of p16 and cyclin D1 expression rate between the Australian group and Chinese group (97% versus 100%, and 3% versus 12%, both P > 0.05). In the combined data of both groups, the positive rate of p53 was 13%; and the upregulation rates of pRb, p16, p21, p27, and cyclin D1 were 74%, 99%, 81%, 63% and 7% respectively. CONCLUSION: Cervical carcinoma overexpresses pRb, p16, p21, and p27. Tumors from Chinese patients are significantly more likely to be positive in p53 and to have upregulated pRb, p21, and p27 than their Australian counterparts. The molecular pathways to human papillomavirus-induced cervical cancer may be influenced by ethnicity and further investigation is necessary.

[The trend of clinical characteristics of colorectal cancer during the past 20 years in Guangdong province].

OBJECTIVE: To study the trend of clinical characteristics of colorectal cancer during the past 20 years in Gaungdong province. METHODS: The clinical data of 3870 cases of colorectal cancers confirmed pathologically who were hospitalized to Nanfang Hospital and Huizhou Municipal Hospital, Guangdong province, 1985 - 2004 were divided into 5-year groups and analyzed. RESULTS: The hospitalization number of colorectal cancer in the 2000 - 20004 5-year period was 102%, higher than that of the 1985 - 1989 period with an annual increase of 5.1%. The median age of incidence 2000 - 2004 was 58.6 years, 8.4 years higher than that in 1985 - 1989. The male to female ratio of the 3870 patients was 1.42:1. The male to female ratio 2000 - 2004 was 1.35:1, lower than that 1985 - 1989 (1.50:1). The proportion of rectal cancer 2000 - 2004 was 49.7%, significantly lower than that in 1985 - 1989 (64.8%), while the proportion of right hemi-colon cancer 2000 - 2004 was 28.7%, significantly higher than that in 1985 - 1989 (18.0%). The proportion of moderately and well differentiated cancer 2000 - 2004 was 80.6%, higher than that in 1985 - 1989 (70.1%), and the proportion of poorly differentiated cancer 2000 - 2004 was 19.4%, lower than that in 1985 - 1989 (29.9%). The proportion of colorectal cancer at Dukes A stage 2000 - 2004 was 9.8%, higher than that in 1985 - 1989 (3.2%). CONCLUSION: In the past 20 years, the incidence of colorectal cancer has increased in Guangdong province with a n increase of median age of incidence, The male to female ratio has decreased, and the incidence of right hemi-colon cancer, the rates of higher differentiated cancer and Dukes A stage cancer have increased.

[Effects of senility on inducible nitric oxide synthase expression and cell apoptosis in rat penis].

OBJECTIVES: To investigate the effect of senility on inducible nitric oxide synthase (iNOS) expression and apoptosis in rat penis. METHODS: The rats were divided into three groups randomly: senescent, aged and young. Their penises were harvested to detect iNOS expression by immunohistochemistry and cell apoptosis rate by TUNEL, respectively. RESULTS: iNOS expression were 0.24 +/- 0.03, 0.17 +/- 0.02, 0.12 +/- 0.03 (relative A) and cell apoptosis rate were (1.41 +/- 0.78)%, (0.94 +/- 0.43)% and (0.50 +/- 0.23 )%, respectively, in the tissues of three groups. The differences among three groups were considerable significance (P <0.05). CONCLUSION: It maybe one of the mechanisms of erectile dysfunction in the aging male that senility causes the increase of iNOS expression and cell apoptosis.

[Clinical epidemiological characteristics of 3870 cases of colorectal cancers in Guangdong region].

OBJECTIVE: To explore the clinical characteristics and pattern of incidence of colorectal cancers in Guangdong region. METHODS: Analysis and summary were made for 3870 colorectal cancer patients pathologically confirmed in the Nanfang Hospital and Huizhou Municipal Central Hospital of Guangdong province. RESULTS: Median age of 3870 cases with colorectal cancer was 55.3. The high-risk age ranged from 41 to 70.204 cases among them were young patients (age < or = 30, 5.3%). With increase of age the number of cases with rectal cancers decreased gradually, while cancers occurring in the right hemi colon increased gradually. The ratio between male and female was 1.42:1. There were altogether 3958 colonic cancer lesions found in all the cases. Among them 3783 (97.8%) cases presented with a single lesson, 87 (2.2%) cases presented with multiple lesions. 2243 (56.7%) lesions located in the rectum, 717 (18.1%) in the left hemi colon, 998 (25.2%) in the right hemi colon. Histological types in all the lesions cases were grouped as follows: tubular adenocarcinoma 2943 (76.0%); papillary adenocarcinoma 256 (6.6%); mucinous carcinoma 425 (11.0%); and miscellaneous types 246 (6.4%). Colorectal cancers with poor differentiation occurring in the young were 38.2% while in the middle age and the elderly were 29.9% and 14.6%, respectively. The difference between two groups showed a statistical significance (P < 0.01). The cases with confirmed stage A, B, C and D were 234 (6.0%), 1936 (50.0%), 1310 (33.9%) and 390 (10.1%), respectively, according to Dukes' staging system. The cases with the progressing stages (B, C, D stages) were 3636 (94.0%) among all the cases. CONCLUSIONS: The number of patients with colorectal cancer admitted in hospital increased gradually in the recent 20 years, and showed a trend with the decrease percentage in rectal cancer and the gradual increasing in right hemi colon cancer with increase of patients age. Half of the colorectal cancer occurred in the rectum, the rest occurred in the left and right hemi colon. The three clinical epidemiological characteristics of colorectal cancer, which once existed in Chinese, has disappeared in Guangdong region.

[Influence of LY294002 on sperm motility in asthenozoospermia patients in vitro].

OBJECTIVE: To investigate the effects of LY294002, a phosphatidylinositol 3-kinase inhibition, on sperm motility in asthenozoospermia patients in vitro, and further analyze the possible molecular mechanism. METHODS: Sperm aseptically obtained by masturbation and prepared by swim-up technique from 10 patients with asthenozoospermia and 10 healthy fertile men were incubated with different concentrations of LY294002. Measurements of motility were carried out at 10, 30 and 60 min in all specimens by CASA. RESULTS: The sperm in asthenozoospermia patients treated with LY294002 showed a significant increase in sperm progressive motility, the percentage of motile cells, VSL and VAP. CONCLUSION: LY294002 can enhance the motility of sperm in asthenozoospermia patients in vitro.

[Association between lower urinary tract symptoms and erectile dysfunction].

Lower urinary tract symptoms (LUTS) and erectile dysfunction (ED) are highly prevalent in aging men and both of the conditions have a significant impact on the quality of life. In the past few years, various epidemiological trials were conducted to assess the association between LUTS and ED. These studies showed that LUTS, particularly the voiding symptoms, nocturia and the others caused by LUTS, independently increased the incidence of ED. There are some factors involved in the link between LUTS and ED: (1) rho-kinase expression/activity increased; (2) nitric oxide release decreased and corpus cavernosum smooth muscle contraction strengthened due to endothelin-1; (3) the composition of myosin isoform altered; (4) sympathetic hyperactivity and innervation of the corpus cavernosum smooth muscles decreased. These findings concerning the relationship between LUTS and ED have offered some new insights into the evaluation and treatment of patients with these conditions. The present paper briefly reviews the recent studies of the association between LUTS and ED.

[Experimental study of augmentation phalloplasty using tunica vaginalis grafts in bilateral albuginea of penile corpus].

OBJECTIVE: To study the effect and complications of augmentation phalloplasty by implanting autogenous tunica vaginalis grafts into the bilateral albuginea of the penile corpus. METHODS: Five mongrel dogs underwent augmentation phalloplasty with bilateral autogenous tunica vaginalis grafts. The increase in the volume of the corpora cavernosa was achieved by applying autogenous tunica vaginalis grafts to longitudinal openings made bilaterally in the albuginea along the whole length of the penile cavernous corpora. The perimeter of the penile cavernous corpora was measured in the flaccid state and during erection and dynamic infusion cavernosometry (DIC) was conducted for each dog before and after the operation. Histology of the penis was examined using hematoxylin and eosin stains. RESULTS: The average increase in the perimeter of the penile cavernous corpora was 21. 1% in the erectile state but unobvious in the flaccid state 3 months after surgery. There were no statistically significant differences in cavernosometric parameters before and after the operation, and neither were there any other severe postoperative complications. The grafts had mild inflammatory reaction and fibrosis with apparent reformation of the tunica albuginea over the patch site. CONCLUSION: The augmentation phalloplasty technique with bilateral autogenous tunica vaginalis grafts was proved to be effective and reliable with few complications, particularly conspicuous in increasing the volume of the erectile tissues during erection.

[Ion channels and asthenospermia].

Asthenospermia is one of the familiar causes of male infertility. Recently, more and more studies have discovered that some diseases result from gene defect and functional variation of ion channels, known as ion channelopathies. Meanwhile, it has been found that even though many diseases do not fall into the category of the ion channelopathies, some links or passages during the disease development are closely related with the malfunction of ion channels, and many drugs can prevent and cure these diseases by acting on ion channels. The relationship of sperm physiology and pathophysiology with ion channels is gradually becoming one of the hot topics in the current researches. Recent progress in the researches on the relationship between sperm motility and ion channels ( including cation channel s and anion channels) is briefly reviewed in this article.

[Identification of stage Ib specific related genes in lung squamous cell cancer by oligonucleotide array].

OBJECTIVE: To establish gene expression profile of stage Ib and IIIa primary lung squamous cell cancer (SCC) within whole genome and identify genes specifically expressed in stage Ib of SCC. METHODS: Total RNA was extracted from the normal, stage Ib and IIIa lung SCC tissue. cRNA probes prepared from total RNA were hybridized with pretreatment oligonucleotide chip containing 22215 genes. The resultant data were treated with MSV 5.0 software and looked up on the affymetrix website. RT-PCR examination were used to validated the results from chip analysis. RESULTS: Comparing with the normal lung, difference genes expressed in Ib SCC are totally 1764, among which 571 were upregulated and 1193 were downregulated, and in stage IIIa, they are 554, 128 and 426 genes respectively. Genes specifically expressed in stage Ib were totally 1329, including 482 upregulation genes and 847 downregulation genes, which were classified into different category which included 480 metabolism related genes, 227 signal transduction genes, 136 cell proliferation genes, 136 immune related genes, 94 cell adhere genes, 88 transcription regulation genes, 86 cell cycle genes, 73 cytoskeleton genes, 45 differentiation genes, 42 apoptosis and 31 extracellular matrix genes. RT-PCR examination of FOXM1 and TNXB genes was consistent with the analysis of gene chip. CONCLUSION: Stage Ib and IIIa Gene expression profile of primary SCC within the whole genome were set up and genes specifically expressed in stage Ib of SCC were identified, which lay a foundation for further research on carcinogenesis mechanism and identifying new markers of early diagnosis.

[Inhibition of binding peptides on replication of duck hepatitis B virus].

OBJECTIVE: To study the inhibitory effect of binding peptides on duck hepatitis B virus (DHBV) replication in duck hepatocytes. METHODS: Specific binding peptides to duck hepatitis B virus polymerase (DHBVP) were screened by phage display technology (PDT), then were sequenced and synthesized. Binding peptides were added into primary culture of duck hepatocytes infected with DHBV in vitro. DHBV-DNA in the cytoplasm, cell nucleus and medium supernatant was assayed over time. RESULTS: Seven binding peptides were obtained after 3-round screening by PDT. Duck primary hepatocytes infected by DHBV were treated with above obtained binding peptides. The DHBV-DNA levels in medium supernatant and cytoplasm of duck hepatocytes treated with synthesized peptides (the 3rd and the 6th peptide) were significantly lower than those of control cells (P<0.05). CONCLUSION: Specific binding peptides to DHBVP could inhibit the replication of DHBV.

[Penile vibratory stimulation and electroejaculation in the treatment of ejaculatory dysfunction].

The fertility potential of infertile men can be enhanced to a great extent by the application of assisted reproduction techniques such as intrauterine insemination or in-vitro fertilization with or without intracytoplasmic sperm injection, but how to obtain semen from men with ejaculatory dysfunction remains a problem. The development and refinement of penile vibratory stimulation (PVS) and electroejaculation (EEJ) have significantly brightened the prospects for the treatment of ejaculatory dysfunction. Because vibratory stimulation is non-invasive and easy to perform, and needs no anaesthesia, it is preferred by most of the patients to EEJ, and recommended to be the first choice of treatment for ejaculatory dysfunction. Approximately 80% of all ejaculatory dysfunction men with an intact ejaculatory reflex arc (above T10 ) can obtain antegrade ejaculation by PVS. Any condition which affects the ejaculatory mechanism of the central and/or peripheral nervous system including surgical nerve injury may be treated successfully by EEJ. The purpose of this review is to present the current understanding of PVS and EEJ procedures and their clinical use in men with ejaculatory dysfunction.

[Immunopharmacological action of sinomenine, an alkaloid isolated from Sinomenium acutum, and its mechanism of action in treating rheumatoid arthritis].

AIM: To observe the effects of sinomenine on the immune functions and apoptosis of murine lymphocyte as well as on human synovial fibroblast proliferation. METHODS: Both in vivo and in vitro tests were adopted. The lymphocyte proliferation induced by mitogens was assayed by MTT method. Spleen T lymphocyte subtypes were tested with flow cytometry. Spleen lymphocyte apoptosis was analyzed by flow cytometry and DNA ladder methods. In vitro test was adopted to observe the effects of sinomenine on the proliferation of human fibroblast of rheumatoid arthritis. RESULTS: Sinomenine can inhibit the proliferation of mouse lymphocytes induced by ConA, LPS and anti-CD3 mAb but not PMA in vitro, and inhibit the proliferation induced by LPS and PMA in vivo. Sinomenine can reduce up-regulated CD4+/CD8+ ratio of T lymphocyte subtype in adjuvant arthritis rat. At the same concentration increased apoptosis ratio. As to human synovial fibroblast, sinomenine can significantly inhibit proliferation of human fibroblast. CONCLUSION: Sinomenine can inhibit the immunological function and correct imbalance of CD4+/CD8+ ratio of T lymphocyte subtype. It can also increase apoptosis ratio of spleen lymphocyte. This may be the mechanism of its immunological inhibitory effect.

[Expression of the kallikreins-kinins system in the corpus cavernosum of rats].

OBJECTIVE: To study the expression of the kallikreins-kinins system in the corpus cavernosum of rats. METHODS: The expression of tissue kallikrein I and kinin B2 receptor gene in the corpus cavernosum and heart of adult rats was detected by reverse transcription polymerase chain reaction (RT-PCR). RESULTS: The tissue kallikrein I and kinin B2 receptor were detected in the corpus cavernosum as well as in the heart of the rats and the contents were similar. CONCLUSION: A kallikreins-kinins system exists in the corpus cavernosum of rats, and the content is rich, almost similar to that in the heart.

[The use of intermaxillary fixation screws in the management of jaw fractures].

48 patients with fractures of the jaw bones were included in this study. Intermaxillary fixation screws were used for tractions between the upper and lower jaw and to regain occlusion relationship, followed by internal rigid fixation. Two days after reduction of the fractures, intermaxillary tractions were carried out and the fixation screws remained for 2 weeks. All the wounds healed primarily with good occlusion. The wounds for screw placement healed well 3 to 5 days after removal of the screws. Excellent occlusion recovered in 45 patients, while in 3 patients, mild open bite developed which returned to normal after 2 weeks of intermaxillary tractions. It is concluded that intermaxillary fixation screws are time saving and efficient in the management of jaw fractures.

[Expression of MUC1 and distribution of tumor-infiltrating dentritic cells in human bladder transitional cell carcinoma].

OBJECTIVE: To study MUC1 expression and distribution of tumor-infiltrating dentritic cells (TIDCs) in human bladder transitional cell carcinoma (BTCC). METHODS: Immunohistochemical staining was employed to detect MUC1 expression and TIDC distribution in 69 surgical specimens of BTCC. MUC1 expression was also detected immunohistochemically in BIU-87, T-24 and drug-resistant BIU87/A cells. Flow cytometry was performed for determining the apoptosis rates of these 3 cells after a 48-hour treatment with adriamycin, vincristine and cisplatin, respectively. RESULTS: MUC1 expression was detected in the BTCC tissues of all stages and the immunohistochemical staining patterns were significantly associated with the pathological grade and clinical stage of the tumors (P<0.001). The number of TIDCs in the tumors was inversely correlated with tumor pathological grades and clinical stages (P<0.005). MUC1 expressed weakly in the cytoplasm and on the membrane of BIU-87 cells and T-24 cells, but strongly in the cytoplasm and membrane of BIU-87/A cells, showing significant differences between the drug-sensitive and -resistant cells (P<0.05). The apoptosis rates of BIU-87 cells and T-24 cells increased obviously after treatment with adriamycin, vincristine and cisplatin, but no significant differences were noted between the two cells or between the 3 drugs. The apoptosis rate of BIU87/A cells, however, exhibited no obvious increase after adriamycin or vincristine treatment, but showed significant increase in response to cisplatin treatment (P<0.05). CONCLUSIONS: The expression pattern of MUC1 and distribution of TIDCs can be useful markers to evaluate the degree of malignancy and prognosis of BTCC. The decrease in the number of TIDCs may have important relation to tumor immune evasion and immune tolerance, and MUC1 over-expression may lead to drug resistance of BTCC, indicating its involvement in tumor infiltration and metastasis.

[A controlled randomized trial of the use of combined L-carnitine and acetyl-L-carnitine treatment in men with oligoasthenozoospermia].

OBJECTIVE: To determine the efficacy and safety of combined L-carnitine and acetyl-L-carnitine therapy in infertile males with oligoasthenozoospermia. METHODS: One hundred fifty patients with oligoasthenozoospermia were randomized selected into treatment and control groups. The treatment group with 90 patients were given L-carnitine (2 g/d) and acetyl-L-carnitine (1 g/d) orally, twice a day. The patients in control group were given Vitamin E 100 mg plus Vitamin C 100 mg, tid. The oral therapy lasted three months and patients accepted sperm analysis every one month. The L-carnitine level in seminal plasma was examined by high performance liquid chromatography (HPC). Side effects as well as pregnant rate were observed. RESULTS: In the treatment group, 85 patients out of 90 finished the three month treatment. Female spouses of 10 patients (11.6%) achieved pregnancy. Moreover, their forward motile sperm per ejaculation, total motile sperm, as well as the concentration of L-carnitine in seminal plasma were increased significantly (P < 0.01). In control group, 53 patients out of 60 completed three months therapy. Two pregnancy (3.7%) was observed. Though some increase was seen in number of forward motile sperm and total motile sperm per ejaculation, the changes were not statistically significant (P > 0.05). The difference of the pregnant rate between two groups was statistically significant. No side effects were found. CONCLUSION: Combined treatment with L-carnitine and acetyl-L-calmitine can be an effective and safe option for treating oligoasthenozoospermia by means of significantly improving forward motile sperm and total motile sperm per ejaculation, as well as increasing pregnant rates.
