Radionuclide imaging of mesenchymal stem cells transplanted into spinal cord.

In vivo tracking of stem cells implanted to spinal cord by radionuclide imaging was investigated. The high expression of transferrin receptor on human mesenchymal stem cells (hMSCs) was verified by flow cytometry, radioligand binding and immunofluorescence. Radiolabelled transferrin was chosen as a tracer for scintigraphic imaging of the hMSCs transplanted into spinal cord of rabbits. Comparative experiments with radiolabelled human serum albumin as tracer and PBS as graft as well as ex vivo autoradiography demonstrated the specific uptake of radiolabelled transferrin of hMSCs. hMSCs could be detected in vivo with radiolabelled transferrin targeting at cellular transferrin receptors at an early stage after transplantation into spinal cord.

In vivo tracking of implanted stem cells using radio-labeled transferrin scintigraphy.

The possibility of monitoring stem cells in vivo with radionuclide imaging after transplantation was investigated. Based on the results of a radioligand receptors assay that human mesenchymal stem cells (hMSCs) express a high level of transferrin receptors, iodinated transferrin (131I-Tf(Fe)2) was chosen as the radiotracer for imaging the cells implanted into the spinal cords of rabbits. Accumulation of radioactivity at the cell transplanted sites was assessed 16 and 24 hours post-intrathecal injection of 131I-Tf(Fe)2. Transferrin receptors expression and Tf binding of the implanted cells were verified by immunofluorescence and ex vivo phosphor imaging. The specificity of Tf uptake of hMSCs was proved through control experiments, i.e., replacing 131I-Tf(Fe)2 with 131I labeled human serum albumin as the tracer or substituting hMSCs with phosphate buffered saline as the grafts. Despite some defects, such as the invasive administration of the tracer and the non-specificity of transferrin receptors as a marker of stem cells in this preliminary study, the technique of nuclear medicine imaging is considered to have great potential in tracking implanted cells in vivo.

The value of vesicant 18F-fluorodeoxyglucose positron emission tomography (18F-FDG PET) in gastric malignancies.

AIM: The value of vesicant 18F-fluorodeoxyglucose positron emission tomography (18F-FDG PET) in the detection and staging of primary gastric malignancies was studied prospectively. METHODS: Thirty-eight patients with suspected gastric tumour were imaged with three-dimensional PET after the injection of 18F-FDG. During the PET study, vesicant was given orally in order to extend the stomach with CO(2) gas. Surgical operation (n=31) or gastric endoscopy with biopsy (n=7) was undertaken 3-26 days after the PET study. The PET results were compared with clinical (computed tomography/magnetic resonance imaging/ultrasound), surgical and histological staging. RESULTS: PET correctly diagnosed 83.3% of primary malignant (25/30) and benign (7/8) lesions, and corrected 11 clinically and five surgically mis-staged cases. Five primaries were false-negatives in quantitative analysis, but qualitative PET readings revealed positive lymph nodes, thereby providing a correct diagnosis in two of the five. PET misdiagnosed seven N1, one N2 and one false-positive case. The focal uptake of 18F-FDG was correlated with the differentiation, size and depth of invasion of the gastric tumours. CONCLUSIONS: 18F-FDG PET is useful for the detection and staging of primary gastric malignancies, and the administration of vesicant may improve the diagnostic confidence.

Noise components on positron emission tomography images.

Noise components on PET images were studied. The results showed that the square of synthetic noise Delta(2) and the square of signal-to-noise ratio (N/Delta)(2) varied with the signal intensity N and the relationships were respectively expressed with: Delta(2)=0.0395N(2)+0.1427N+0.0025 (R(2)=0.9358) and (N/Delta)(2)=-1.13932N(2)+7.0185N-0.0746 (R(2)=0.9377). The synthetic noise on PET image may be categorized into three components. The first one varies directly as signal intensity, which depend on random coincidence; the second one varies directly as the square root of signal intensity, which depend on Poisson fluctuation; and the third one is independent of signal intensity.

PET imaging of brain function while puncturing the acupoint ST36.

OBJECTIVE: To explore the experimental method of obtaining position emission tonogiaphy (PET) imaging evidence of changes in cerebral function by puncturing the Stomach 36 (ST36, Zusanli) acupoint. METHODS: Data on changes of cerebral glycometabolism were obtained from six healthy male volunteers with positron emission tomography. Visual experimental evidence, as well as statistical parametric mapping (SPM), was gathered while puncturing the ST36 (Zusanli, right leg) acupoint. RESULTS: There was increased glycometabolism in the hypothalamus, head of the caudate nucleus, temporal lobe, the sinistral cerebellum, postcentral gyrus, and brain stem while the acupoint ST36 was being punctured. CONCLUSIONS: Acupuncture on ST36 can lead to increase in glycometabolism in the vegetative nerve centers, which is correlated with gastric function. Visual experimental evidence of ST36 acupuncturing on functional gastrointestinal disorder was obtained in our study.

Comparative analysis of patients not responding to a single dose of 153Sm-EDTMP palliative treatment for painful skeletal metastases.

OBJECTIVE: A certain fraction of patients failed palliative treatment of 153Sm- ethylenediaminetetramethy lenephosphate (153Sm-EDTMP) for painful skeletal metastases were reviewd. A comparative analysis was designed to identify the factors related to therapeutic response. METHODS: From a 3-year multi-center clinical trial, 51 cases were collected who did not respond to an intravenous injection of 153Sm-EDTMP at a dosage of 0.5-1.5 mCi/kg. The therapeutic efficacy was evaluated via changes of symptoms, general condition, consumption of analgesics, sum of effect product, and Karnofsky scores. The age, sex, history of treatment, tumor type, location of bony involvement, uptake ratio and number of metastases, and doses used by the patients were compared to those of the responders. RESULTS: In 51 non-responders, 43 were male, 34 suffered from lung cancer, 41 had bone lesions in the vertebrae, 39 in the pelvis, and 24 had metastases in the lower extremities. Sex distribution, tumor type and location of the lesion differed significantly between responders and non-responders. No other factor showed differences between the two groups. Though patients of younger age, and lesions with lower uptake of radiopharmaceutical seemed to fail the treatment more easily as observed clinically, this was not confirmed by statistical analysis. CONCLUSION: The sex of the patients, certain types of primary tumors and metastases to lower parts of the body were found to influence the patients' response to a single dose of 153Sm-EDTMP palliation. Further exploration of a better way to determine dosage and predict response for each individual case is needed.
