Expression of integrin beta1 and its roles on adhesion between different cell cycle hepatocellular carcinoma cells (SMMC-7721) and human umbilical vein endothelial cells.

To investigate expression of integrin beta1 and its roles on adhesion between different cell cycle hepatocellular carcinoma cell (HCC) and human umbilical vein endothelial cells (HUVEC), the synchronous G1 and S phase HCC were achieved through thymine-2-deoxyriboside and colchicines sequential blockage method and double thymine-2-deoxyriboside blockage method, respectively. Expression of integrin beta1 on hepatocellular carcinoma cells was detected with flow cytometer. Further, the adhesive force of HCC to HUVEC and the role of integrin beta1 in this adhesive course were studied by micropipette aspiration technique. The results showed that percentage of each cyclic phases of the controlled HCC (non-synchronous) are: G2 + M phase, 11%; G1 phase, 54%; S phase, 36%; the synchronous rates of G1 and S phase HCC amount to 74 and 98%, respectively. The expressive fluorescent intensity of integrin beta1 in G1 phase HCC is depressed significantly than the values of S phase and controlled HCC. Accordingly, the adhesive forces of G1 phase HCC to HUVEC was significantly lower than the value of S phase cells (P < 0.01), but it has no remarkable difference when compared the adhesive force values of S phase HCC with control; the contribution of integrin beta1 was about 50% in the adhesion of HCC to HUVEC. It suggested that HCC would be synchronized preferably in G1 and S phase with thymine-2-deoxyriboside and colchicines, the adhesive molecule integrin beta1 expressed in a high lever in HCC and presented differences in vary cell cycle, and integrin beta1 played an important roles in adhesion of HCC to HUVEC. Possibly, S phase HCC take a great action in this adhesive course.

Mouse strains with an active H2-Ea meiotic recombination hot spot exhibit increased levels of H2-Ea-specific DNA breaks in testicular germ cells.

We devised a sensitive method for the site-specific detection of rare meiotic DNA strand breaks in germ cell-enriched testicular cell populations from mice that possess or lack an active recombination hot spot at the H2-Ea gene. Using germ cells from adult animals, we found an excellent correlation between the frequency of DNA breaks in the 418-bp H2-Ea hot spot and crossover activity. The temporal appearance of DNA breaks was also studied in 7- to 18-day-old mice with an active hot spot during the first waves of spermatogenesis. The number of DNA breaks detected rose as leptotene and zygotene spermatocytes populate the testis with a peak at day 14 postpartum, when leptotene, zygotene, and early pachytene spermatocytes are the most common meiotic prophase I cell types. The number of DNA breaks drops precipitously 1 day later, when middle to late pachytene spermatocytes become the dominant subtype. The recombination-related breaks in the hot spot likely reflect SPO11-induced double-strand breaks and/or recombination intermediates containing free 3' hydroxyl groups.

[Integrin beta1 mediates hepatocellular carcinoma cells adhesion & chemotaxis to type IV collagen].

A micropipette technique was adopted to investigate the effect of blockade of integrin betal on adhesion of hepatocellular carcinoma (HCC) cells onto type IV collagen (Col IV) coated surfaces and pseudopod protrusion of HCC cells in response to Col IV stimulation. Adhesion strength was expressed as an adhesion force, which was defined as the product of the cross sectional area and critical negative pressure needed to detach single cell away from the substrate. Chemotactic pseudopod protrusion of an HCC cell was evaluated using a dual-pipette set-up, in which two pipettes filled with Col IV solution were positional in close contact with the same cell and pseudopod protrusion into each pipette was viewed dynamically and recorded with a tape recorder. The lengths of pseudopods were measured and plotted against time to obtain a pseudopod growth curve. The integrin beta1 subunit on the surfaces of HCC cells was analyzed by flow cytometry. The results showed that the adhesion forces for HCC cells adhering on 5 microg/ml Col IV coated surfaces were 932 +/- 134 (x 10(-10) N, n = 60). Upon treatment of HCC cells with Anti-CD29 in a protein concentration of 5 microg/ml and 10 microg/ml, the value decreased significantly to 449 +/- 119 (x 10(-10) N, n = 60) and 220 +/- 78 (x 10(-10) N, n = 55), respectively. In dual pipette chemotaxis experiment, when the two pipettes were filled with Col IV in an identical concentration of 600 microg/ml, pseudopods extended from the HCC cell into each of the pipettes nearly symmetrically, i.e., with nearly identical maximum pseudopod length and similar pseudopod growth curves. Upon addition of Anti-CD29 to one of the pipettes in a protein concentration of 20 microg/ml, pseudopod protrusion was blocked nearly completely while protrusion into the opposite pipette became more evidently, with larger maximum length. Expression of integrin beta1 was up to 95.78% to cells chosen in the experiment. These results suggested that integrin beta1 subunit was important constituent receptor subunit for mediating HCC cell adhesion and chemotactic pseudopod protrusion to Col IV.

[Integrin alpha3beta1 mediates hepatocellular carcinoma cell adhesion and chemotaxis to type IV collagen].

OBJECTIVE: To investigate the effects of Integrin alpha(3)beta(1) on the adhesion and chemotaxis of hepatocellular carcinoma (HCC) cells to type IV collagen (Col IV). METHODS: (1) HCC cells were culture and suspension of HCC cells was made. Anti-alpha(3) and Anti-beta(1) were added into the HCC cell suspension. Flow cytometry was used to determine the expression of integrin alpha(3)beta(1) on the surface of HCC. (2) 5 micro g/ml Col IV was used to coat a cell with the diameter of 25 mm. Digested HCC cells were added. Anti-alpha(3) and Anti-beta(1) of the concentrations of 5 micro g/ml and 10 micro g/ml respectively were added into the cell suspension. Before and after the addition of Anti-alpha(3) and Anti-beta(1), micropipette technique was used to measure the adhesion force of HCC on Col IV-coated surface, as function of the square of internal radius of micropipette and the critical negative pressure needed to detach a single HCC cell away from the substrate. (3) Col IV of the concentration of 600 micro g/ml was added into the dual micropipettes. Then the dual micropipettes were led towards the HCC cells. A HCC cell was made to seal the openings of the 2 micropipettes with different parts of the cell contacting Col IV in different micropipettes. The pseudopod protrusion was observed dynamically and recorded with tape recorder. The length of pseudopod was measured and plotted against the chemotactic time so as to obtain a pseudopod growth curve. RESULTS: (1) The expression rates of integrin subunit alpha(3) and beta(1) on the surface of HCC cells were 95.55% and 95.78% respectively. (2) The adhesion force of HCC cells to the 5 micro g/ml Col IV-coated surface was 932 +/- 134 (x 10(-10) N, n = 60). Upon treatment of the HCC cells with Anti-alpha(3) of the concentrations of 5 micro g/ml and 10 micro g/ml, the adhesion force decreased by 42% and 49%, to 536 +/- 122 (x 10(-10) N, n = 60) and 476 +/- 63 (x 10(-10) N, n = 60) respectively. Upon treatment of the HCC cells with Anti-beta(1) of the concentrations of 5 micro g/ml and 10 micro g/ml, the adhesion force decreased by 52% and 76%, to 449 +/- 119 (x 10(-10) N, n = 60) and 220 +/- 78 (x 10(-10) N, n = 60) respectively. (3) The length of pseudopod increased along with the chemotactic time. The pseudopod length and growth curve were almost identical in the dual micropipettes when they were filled with Col IV. When Anti-alpha(3) or Anti-beta(1) was added into one of the dual micropipettes, the HCC cell pseudopod protrusion was almost blocked completely, while the HCC cell pseudopod in the opposite micropipette became more evident. CONCLUSION: Integrin alpha(3)beta(1) is an important constituent receptor in mediating HCC cell adhesion and chemotactic pseudopod protrusion to Col IV.

Short-term (intestinal) and long-term (postintestinal) conversion of beta-carotene to retinol in adults as assessed by a stable-isotope reference method.

BACKGROUND: Quantitative information on the conversion of beta-carotene to vitamin A in humans is limited. OBJECTIVE: We determined the short- and long-term conversion of labeled beta-carotene to vitamin A by using a stable-isotope reference method. DESIGN: [(2)H(8)]beta-Carotene (11,011 nmol, or 6 mg) in oil was given with a liquid diet (25% of energy from fat) to 22 adult volunteers (10 men, 12 women). Three days after the [(2)H(8)]beta-carotene dose, the volunteers each took a dose of [(2)H(8)]retinyl acetate (8915 nmol, or 3 mg) in oil with the same liquid diet. Blood samples were collected over 56 d. RESULTS: The 53-d area under the serum [(2)H(4)]retinol response curve (from the [(2)H(8)]beta-carotene dose) was 569 +/- 385 nmol. d, and the 53-d area under the serum [(2)H(8)]retinol response curve (from the [(2)H(8)]retinyl acetate dose) was 1798 +/- 1139 nmol. d. With the use of [(2)H(8)]retinyl acetate as the vitamin A reference, the [(2)H(4)]retinol formed from [(2)H(8)]beta-carotene (11,011 nmol) was calculated to be equivalent to 3413.9 +/- 2298.4 nmol retinol. The conversion factor of beta-carotene to retinol varied from 2.4 to 20.2, and the average conversion factor was 9.1 to 1 by wt or 4.8 to 1 by mol. This conversion factor was positively correlated with body mass index (r = 0.57, P = 0.006). The postabsorption conversion of beta-carotene was estimated as 7.8%, 13.6%, 16.4%, and 19.0% of the total converted retinol at 6, 14, 21, or 53 d after the [(2)H(8)]beta-carotene dose, respectively. CONCLUSION: The quantitative determination of the conversion of beta-carotene to vitamin A in humans can be accomplished by using a stable-isotope reference method. This approach provides in vivo metabolic information after a physiologic dose of beta-carotene.

Ste11p, a high-mobility-group box DNA-binding protein, undergoes pheromone- and nutrient-regulated nuclear-cytoplasmic shuttling.

The high-mobility-group (HMG) box is a conserved DNA-binding domain found in a family of transcription factors that regulate growth and development. One family member, Ste11p, directs sexual differentiation of Schizosaccharomyces pombe by binding specific DNA sequences upstream of genes required for mating and meiosis. Here, we show that Ste11p is a shuttling protein. In growing cells, Ste11p is present in low levels and is pancellular. Mating pheromones and nutrient limitation trigger nuclear accumulation and increased expression of the transcription factor. Several mechanisms likely control Ste11p localization. First, the 14-3-3 protein, Rad24p, binds phosphorylated Ste11p and inhibits its nuclear accumulation. Second, the HMG domain of Ste11p contains a basic cluster nuclear localization signal. Finally, treatment of cells with leptomycin B, an exportin inhibitor, results in the nuclear accumulation of Ste11p. A Ste11p deletion mutation, DeltaC54, mimics the effects of leptomycin B. The C54 region contains no identifiable nuclear export signal but instead is required for biological activity and to stimulate Ste11p target gene expression. These results provide evidence that both nuclear import and export mechanisms operate to regulate cellular localization of an HMG box protein. In addition, they establish a paradigm for the potential role of pheromone/hormone-like polypeptides in cellular localization of this important class of developmental regulators.

[Effects of burn serum on the viscoelasticity and the structure of rat intestinal epithelial cells].

OBJECTIVE: To investigate the effects of burn serum on the viscoelasticity and the structure of rat intestinal epithelial cells. METHODS: The rat intestinal epithelial cell strain (IEC-6) was cultured and stimulated by burn serum. The changes of IECs before and after the stimulation were dynamically observed by cytoskeleton immunohistochemistry, ELISA and the measurement of cytomembranous viscoelasticity. RESULTS: During the early stage of burn serum stimulation, the skeleton protein expression in IEC decreased obviously with weakened positive signals of microfilaments and microtubules and with decreased cellular elasticity. CONCLUSION: The cytoskeleton injury could cause the increase of cellular fragility and the decrease of the viscoelasticity, which ultimately lead to the change of cellular biodynamics. These changes might directly participate the development of postburn intestinal epithelial injury.

Inhibition of adhesion of hepatocellular carcinoma cells to basement membrane components by receptor competition with RGD- or YIGSR-containing synthetic peptides.

A micropipette technique was used to investigate the effects of four synthetic peptides, YIGSR, CDPGYIGSR, RGDS and GRGDTP, on the adhesion of hepatocellular carcinoma (HCC) cells onto type IV collagen/laminin/fibronectin coated surfaces. Adhesion of HCC cells to laminin was found to be YIGSR- or CDPGYIGSR-dependent while that to fibronectin and type IV collagen was RGDS- or GRGDTP-dependent. The reduction in adhesion strengths of HCC cells was slight to moderate (up to 55%), and was dependent on the peptide concentration. The decrease in adhesion strengths was reversed by an increase in ligand coating concentration and was compromised by prolonged interaction of the cells with the surfaces. These results suggested that the inhibition was due to competitive retardation rather than to a blockade of adhesion strengthening. A simple asymptotic function was adopted to fit the correlation between the mean of cell adhesion strengths and peptide concentration within defined concentration ranges. Regression analysis showed that cell adhesion strengths appeared to approach a plateau with increasing concentration of the inhibitory peptides, which was not always uniform over the entire concentration range tested. Further reduction in adhesion strengths was observed at higher peptide concentrations. It is suggested that the constants obtained by fitting over a low peptide concentration range might be kinetically representative of the inhibition during early events of adhesion or attachment.

[Clinical study on "sini" decoction in treating stenocardia for coronary heart disease].

OBJECTIVE: To observe clinical effect of "Sini" decoction on stenocardia for coronary heart disease. METHODS: 63 cases of stenocardia for coronary heart disease with deficiency of yang or cold syndrome were randomly divided into "Sini" decoction group (therapeutic group, 35 cases) and isosorbide dinitrate group (contrast group, 28 cases). Drug effects of the two groups were compared by clinical symptoms, electrocardiogram (ECG), myocardial oxygen consumption and cardiac function. RESULTS: Similar to isosorbide dinitrate, "Sini" decoction could improve clinical symptoms and ECG, decrease frequency of stenocardia and dose of nitroglycerin on stenocardia for coronary heart disease. But "Sini" decoction had better effects on reducing myocardial oxygen consumption and improving cardiac funrtion than isosorbide dinitrate. CONCLUSION: "Sini" decoction can treat stenocardia for coronary heart disease with deficiency of yang or cold syndrome.

[Overexpression of TGF beta 1 increases elastin expression and adhesion of smooth muscle cells].

This study was conducted to examine the effectiveness of a gene transfer of human TGF beta 1 gene into smooth muscle cells and whether the TGF beta 1 can increase elastin expression of smooth muscle cells. With the help of DOTAP, smooth muscle cells were transfected with pMAMneoTGF beta 1. The positive cell clones were selected with G418. The stable transfection and expression of TGF beta 1 in the smooth muscle cells were determined by immunofluorescence analysis. The expression of elastin in the transfected and untransfected cells were determined by in situ hybridization. The adhesion force between smooth muscle cells and matrix was detected by micropipette system. The results showed abundant TGF beta 1 stable expression in smooth muscle cells. TGF beta 1 gene can increase two-three times elastin expression and increase the adhesion between smooth muscle cells and matrix. TGF beta 1 can be used in vascular tissue engineering to increase smooth muscle cells adhesion.

[An experimental study of cellular mechanic properties of intestinal epithelial cells by micropipette aspiration].

OBJECTIVE: To further understand the mechanic properties of the Intestinal Epithelial cells (IEC). METHODS: The viscoelastic properties of the IEC-6 were determined by micropipette aspiration. RESULTS: The elastic coefficient K1 was 436.21 +/- 89.64 dyn/cm2 and K2 was 298.37 +/- 75.16 dyn/cm2. The visous coefficient micro was 15.94 +/- 4.93 dyn/(sec x cm2). CONCLUSION: The elastic component reflects the degree of initial deformation of the IEC and makes a notable impact on the deformation of the IEC. The viscous component reflects the time dependency of the deformation of the IEC and influences the velocity of the IEC deformation.

Use of a short-term isotope-dilution method for determining the vitamin A status of children.

BACKGROUND: After a dose of labeled vitamin A is given to humans for estimating body stores of vitamin A, blood is customarily drawn at pseudo-equilibration times ranging from 11 to 26 d. OBJECTIVE: The objective was to determine whether a shorter sample collection interval (6 h or 3 d), which would be more realistic in field settings, can be used. DESIGN: Correlations of enrichment at 6 h or 3 d with enrichment at 21 d were made after an oral dose of deuterium-labeled vitamin A was given to Chinese schoolchildren (aged 10-11 y; n = 58) with marginal-to-normal vitamin A status. A predictive equation was then derived and applied to data obtained from a separate group of children to verify that the calculated enrichment at 21 d (determined by using data obtained at an earlier time point to predict 21-d enrichment) reflected directly measured enrichment at 21 d. RESULTS: Because 3-d isotope enrichment was found to correlate well with 21-d enrichment, a predictive equation was derived whereby 3-d data were used to predict isotope enrichment at pseudo-equilibration (ie, at 21 d). When the 3-d predictive equation was applied to a separate group of Chinese children, the calculated 21-d data (determined by using the 3-d data and the predictive equation) matched the directly measured 21-d data. Body stores of vitamin A determined from either the calculated or directly measured 21-d enrichment data also showed agreement. CONCLUSION: Percentage enrichment at 3 d (but not at 6 h) can be used to evaluate vitamin A body stores in humans.
