Effects of seasonal succession and water pollution on the protozoan community structure in an eutrophic lake.

The purpose of the research is to study the seasonal succession of protozoa community and the effect of water quality on the protozoa community to characterize biochemical processes occurring at a eutrophic Lake Donghu, a large shallow lake in Wuhan City, China. Samples of protozoa communities were obtained monthly at three stations by PFU (polyurethane foam unit) method over a year. Synchronously, water samples also were taken from the stations for the water chemical quality analysis. Six major variables were examined in a principal component analysis (PCA), which indicate the fast changes of water quality in this station I and less within-year variation and a comparatively stable water quality in stations II and III. The community data were analyzed using multivariate techniques, and we show that clusters are rather mixed and poorly separated, suggesting that the community structure is changing gradually, giving a slight merging of clusters form the summer to the autumn and the autumn to the winter. Canonical correspondence analysis (CCA) was used to infer the relationship between water quality variables and phytoplankton community structure, which changed substantially over the survey period. From the analysis of cluster and CCA, coupled by community pollution value (CPV), it is concluded that the key factors driving the change in protozoa community composition in Lake Donghu was water qualities rather than seasons.

[Effects of leachate recirculation volume on its characteristic and landfill stabilization rate].

Impacts of recirculation volume on leachate characteristic and landfill stabilization rate were studied. Four simulated bioreactor landfill columns were operated weekly with different recirculation ratios, respectively 5.3%, 2.7%, 0.67% leachate and 0.33% water, in this comparative research. The results indicate that simulated reactor with 5.3% recirculation ratio has the most rapidly stabilization rate and release the most organic pollutant. The shortest methane generation delay was also observed in this column. While simulated reactor with 2.7% recirculation ratio formed the best microbe environment and kept the highest reactor temperature 35 degrees C. It also had the best impactive load capacity and treating efficiency to leachate, and removal of COD was 77% and BOD5 was 88% respectively. In actual projects, appropriate leachate recirculation volume should be chosen according to design purpose of landfill sites.

[Chemical stabilization technology in treating with flyash using apatite].

Toxicity leaching characteristics and chemical compositions of sample flyash collected from municipal solid waste incinerator was analyzed, it was clear that different heavy metals were contained in flyash and the heavy metal leaching concentration for Pb, Zn, Cd was 11.8, 164.9 and 31.2 mg/L respectively which was much higher than that in Hazardous Waste Toxicity Leaching Standards. A new kind of inorganic chemical agent apatite was chosen to treat with this kind of incineration flyash, the results indicate that the heavy metals (not including Cd in some samples) in the treated flyash using apatite at dosage 7% can reach the landfilling standards for hazardous waste, and the treated flyash can have better stabilization when the size of apatite was 74 microm, and the heavy metal leaching concentration of the treated flyash keep stable at a curing time of 1 day, and it can keep stabilization with a broader pH value of 3 to 11. Thus the risk of secondary pollution for the treated flyash was reduced dramatically when the environment conditions change.

[Immobilization technology and mechanism of fly ash using H3PO4].

Chemical composition and toxicity leaching characteristics of fly ash was analyzed. The experiment results show that many heavy metals were contained; leaching concentration of Pb is 67.03 mg/L, which exceeds the limit of identification standard for hazardous wastes. Effect of input mass of H3PO4 on immobilization of heavy metals and its long-term environmental stability was studied. The results show that when input 8% - 14% (H3PO4 mass/ fly ash mass) of H3PO4 sound immobilization effect can be achieved; 8% and 12% of H3PO4 will bring a satisfactory environmental stability of heavy metals, while more H3PO4 led to less buffer capacity to acid conditions. In fly ash treated by 12% H3PO4, a small quantity of crystal Cr2P2O7, ZnP2, Pb3P4O13, Pb3P2O7, NaZnPO4, NaPbP3O9, Ca2ZnSi2O7 can be detected by XRD; many independent fly ash particles and bar-shaped Pb5 (PO4)3Cl with a diameter of 0.3 - 0.5 microm were observed by SEM; concentrated heavy metal materials were not obtained by CHBr3 floatation. Conclusions can be drawn that, through neutralization reaction of H3PO4 with strongly alkaline fly ash, stabilization reaction conditions were improved, entrapped heavy metals were chemically activated and PO4(3-) needed in stabilization was produced. Activated heavy metals combined with PO4(3-) on surface of fly ash, generated phosphates existing as forms of solid solution in SiO2, CaCO3, CaSO4, KCl, NaCl.

Isolation and characterization of phytoene desaturase cDNA involved in the beta-carotene biosynthetic pathway in Dunaliella salina.

The green alga Dunaliella salina is one of the best and most important biological sources of beta-carotene; however, to date the molecular basis of the beta-carotene biosynthesis process in D. salina is still unresolved. The dehydrogenation of phytoene is the second step in the carotenoids biosynthetic pathway, and the phytoene-related desaturases are the key enzymes in the beta-carotene biosynthetic pathway. A phytoene desaturase (Pds) cDNA with a 1752 bp open reading frame was cloned by RT-PCR and RACE-PCR methods on the basis of a modified switching mechanism at 5' end of the RNA transcript (SMART) technology from D. salina. The predicted protein sequence displays a high identity (up to 65%) with phytoene desaturases of higher plants and cyanobacteria. The highest amino acid sequence identity (91%) is shared with the phytoene desaturase sequence of Dunaliella bardawil, and a dinucleotide-binding motif lies in the N-terminal. The phylogenetic analysis shows that D. salina Pds is closer to higher plants and cyanobacteria than bacterial and fungi. These results together demonstrated the cloned Pds cDNA of D. salina is a Pds-type gene, and it is postulated that in D. salina the first two dehydrogenations, by which phytoene is converted into zeta-carotene, are carried out by this putative phytoene desaturase.

Development of a new biotic index to assess freshwater pollution.

We developed a new biotic index of species pollution value (SPV) and community pollution value (CPV) based on the correlation of protozoan communities with chemical water quality to assess freshwater pollution. Five hundred and twenty-three species of protozoa SPV were established based on the data of River Hangjiang and Lake Donghu. The present research was conducted in order to further consummate the biotic index. Protozoa of the water system in Changde City were collected from 16 stations using the PFU method and the water chemical parameters of the stations were analyzed. The results showed that CPV calculated from SPV had a close correlation with the degree of water pollution (p < 0.00001), which indicated that the method is reliable. By combining the data of River Hangjiang, Lake Donghu and Changde City, the final form of SPV was accomplished and the SPV list increased to 757. The ultimate water standard evaluated by CPV calculated from SPV was proposed.

Cloning and sequence analysis of the phytoene synthase gene from a unicellular chlorophyte, Dunaliella salina.

Phytoene synthase (Psy) catalyzing the dimerization of two molecules of geranylgeranyl pyrophosphate (GGPP) to phytoene may be rate limiting for the synthesis of beta-carotene in Dunaliella salina. To elucidate the carotenogenic pathway in D. salina, the complete Psy gene was first isolated by genome walking technology and suppression PCR. Subsequently, RT-PCR was performed to obtain the Psy cDNA of 1260 bp, which codes 420 amino acids. The Psy gene of total length of 2982 bp was found to consist of five exons and four introns when compared with the cDNA sequence. The D. salina Psy amino acid has a 78-89% similarity with many other higher plants, but a phylogenic analysis shows a closer relationship between the microalgal and cyanobacterial Psy.

Use of the aquatic protozoa to formulate a community biotic index for an urban water system.

Protozoan were collected from 16 stations in water system of Changde City (China) using the PFU method. Sampling programs were conduced on a yearly basis, with seasonal frequency at diverse sites in the water system and 488 species of protozoa was identified. At the same time, Water sampling from these stations was conducted and various water chemical parameters, including DO, COD, BOD(5), NH(3), TP, and Volatile Phenol, were analyzed. The aim of the research was, on one hand, using chemical method to take an investigation to the water pollution status of Changde City; on the other hand, using protozoan to make an evaluation to the water quality. With the chemical water parameters and protozoa data, a biotic index was derived for the investigated region. The species pollution value (SPV) of 469 protozoa species was established, and the community pollution value (CPV) calculated from SPV was used to evaluate water quality. The method of the biotic index was tested and the result showed that CPV calculated from SPV had a close correlation with the degree of water pollution (p<0.00001). This indicated that the method of the biotic index is reliable. The water quality degrees divided by CPV were suggested.

[Heavy metal stabilization in municipal solid waste incineration fly ash using soluble phosphate].

The physico-chemical characteristics of municipal solid waste incineration fly ashes was analyzed. It indicated that the main elements of fly ashes are Ca, Cl, K, S and Si, and many heavy metals such as Pb, Zn, Cu, Mn and Cr can be found in fly ashes, and the heavy metal leaching toxicity such as Pb, Cu, Zn and Cd is much higher than the standards. A new kind of stabilization agent-soluble phosphate is chosen to treat with fly ash, different influence factors including agent dosage, curing time and pH are taken into consideration to test its stabilization for the stabilized fly ash. The experimental results indicate that fly ash treated with soluble phosphate can have excellent stabilization effect, and the leaching toxicity for Pb, Cd and Zn can be reduced by 97.5%, 91.6% and 95.5% at a phosphate dosage of 3%; curing time can not influence its stabilization; and, stabilized fly ash using soluble phosphate can keep long-term stabilization at a broad pH value.

Application and validation of a new biotic index using data from several water systems.

Protozoans of Lake Donghu were collected from five stations using the PFU method. The sampling was conducted for one year and two times a month. The aim of this research was to test the applicability of a new protozoa biotic index, species pollution value (SPV) and community pollution value (CPV), established by the authors using data from the River Hanjiang. Each station's CPV was calculated from the SPV and the correlation analysis between the CPV and the comprehensive chemical index of stations I, II, III showed a significant correlation between them. The pollution status of the five stations was correctly evaluated by the CPV. These results suggested that the biotic index could be applied in water systems other than the River Hanjiang. The SPV of some protozoa species in Lake Donghu, not observed in the River Hanjiang were established. In order to further test the applicability of the biotic index, protozoan and chemistry data from the Rivers Torrente Stirone and Parma of Italy were used. The results showed that the CPV for the two rivers had a close relationship with the chemical water quality, which indicated that the biotic index could be applied in other parts of the world for the monitoring and estimating of water quality. Since the results of testing and verifying the biotic index in some other water systems in China were also satisfactory, this indicated that the biotic index has an extensive suitability for freshwater ecosystems. As long as more than 50% of the species in a sample have a SPV, the CPV calculated from the SPV is reliable for monitoring and evaluating water quality.
