HCV NS2 protein inhibits cell proliferation and induces cell cycle arrest in the S-phase in mammalian cells through down-regulation of cyclin A expression.

Chronic hepatitis C virus (HCV) infection often leads to liver cancer. NS2 protein is a HCV hydrophobic transmembrane protein that associates with several cellular proteins in mammalian cells. In this report, we investigated the functions of NS2 protein by examining its effects on cell growth and cell cycle progression. Stable NS2-expressing HeLa and Vero cell lines were established by transfection of the cells with pcDNA3.1(-)-NS2 followed by selection of the transfected cells in the presence of G418. We found that the proliferation rates of both NS2-expressing cell lines were inhibited by 40-50% compared with the control cells that were transfected with pcDNA3.1(-) control vector. Cell cycle analysis of these NS2-expressing cell lines shows that the proportion of cells in the S-phase increased significantly compared to that of control cells that do not express NS2 protein, suggesting NS2 protein induces cell cycle arrest in the S-phase. Further studies showed that the induction of cell cycle arrest in the S-phase by NS2 protein is associated with the decrease of cyclin A level. In contrast, the expression of NS2 protein does not affect the levels of cyclin-dependent kinase CDK2, CDK4, cyclin D1, or cyclin E. Our results suggest that HCV NS2 protein inhibits cell growth and induces the cell cycle arrest in the S-phase through down-regulation of cyclin A expression, which may be beneficial to HCV viral replication. Our findings not only provide information in the understanding mechanism of HCV infection, but also provide guidance for the future development of potential therapeutics for the prevention and treatment of the viral infection.

Human endogenous retroviral pol RNA and protein detected and identified in the blood of individuals with schizophrenia.

Retrovirus has been speculated as one of the potential infectious agents involved in the development of schizophrenia. Here we used nested RT-PCR to detect the RNA of HERV pol gene in blood from schizophrenic patients and normal human. We found retroviral pol genes expressed in blood from 20 of 58 (34.5%) individuals with recent-onset schizophrenia, but not from 38 normal persons (p<0.01). Sequence analysis revealed that the expressed gene was homologous to those of the human endogenous retroviral (HERV) family. The ERV9 family was the closest, with 90% homology in the gene sequence. In addition, Western blots showed that antibody against ERV9 pol protein in serum from the HERV+ schizophrenia patients, but not from control (p<0.01). Our data suggested that the transcriptional activation of certain retroviral elements might be associated with the development of schizophrenia in some patients. Further characterization of retroviral elements in subjects with schizophrenia may aid in better diagnosis and treatment of this disorder.

[Study on developing calibration models of fatty acid composition in intact rapeseed by near infrared reflectance spectroscopy].

Four hundred ninety seven rapeseed samples, which feature multi-year, multi-loci and highly variant characteristics, were collected as a raw set. The NIR spectra were pretreated by scatter correction and mathematics treatments, and calibration models of fat acid composition of intact rapeseed were developed by using the algorithm method of modified partial least square (MPLS). Meanwhile, three types of sample cups with different capacity were used to screen the suitable calibration model for rapeseed quality breeding. The results showed that the calibration model of 8-gram-sample was the best, and the calibration determination coefficient was in the range of 0.74-0.98. The calibration effects of 3-gram-sample, which were similar to those of 0.6-gram-sample, were good with high determination coefficient (RSQ1, 1-VR) and low error (SEC, SECV). Therefore, the calibration set with multi-year and multi-loci samples can improve the accuracy and repeatability of NIRS models. The fat acids NIRS models of intact rapeseed developed could be introduced into breeding lines' selection, mutant screening and germplasm evaluation.

[Creating a software package and improving the multimedia teaching of genetics course].

Multimedia as a pedagogical tool has been widely adopted for improving the quality of teaching. This paper describes the construction of a teaching software package based on the characteristics of the "Genetics" course. It also discusses some particulars of multimedia teaching, the collection of teaching materials, the improvement of teaching effects, and the compilation of a multimedia textbook. How to enhance the quality of the teachers as well as the motivation of the students are also discussed.

[Effects of N, N'-Di-(m-methylphenyi)-3, 6-dimethyl-1, 4-dihydro-1, 2, 4, 5-tetrazine-1, 4-dicarboamide on proliferation, apoptosis and differentiation of NB4 leukemia cells in vitro].

The purpose of this study was to explore the effect of N, N'-di-(m-methylphenyi)-3, 6-dimethyl-1, 4-dihydro-1, 2, 4, 5-tetrazine-1, 4-dicarboamide (ZGDHu-1) on proliferation, differentiation and apoptosis in NB4 human leukemia cell line and its possible mechanism. Different concentrations of ZGDHu-1 and the different time of cultivation were used to treat NB4 cells. The proliferation inhibition of NB4 cells was analysed by cell counting, alive cell count, MTT assay. Cell apoptosis was determined by cell morphology, DNA agarose gel electrophoresis, DNA content, Annexin-V/PI and Hoechst 33258 labeling method. The analysis of cell morphological change, expression of CD11b, CD13 and NBT reduction were performed to evaluate the differentiation of NB4 cells. The expressions of bcl-2, bax and phosphorylated p38MAPK or STAT3 were detected by flow cytometry. While the expression of hTERT mRNA in transcriptional level was measured by fluorescence quantitative RT-PCR. The results showed that ZGDHu-1 could inhibit NB4 cell proliferation viability within a certain range of treating time and does, IC(50) values at 48 and 72 hours were 450 ng/ml and 200 ng/ml respectively. A majority of NB4 cells were arrested in G(2/M) phase and a progressive decline of cells was seen in G(0/1). The NB4 cells apoptosis was confirmed by cell typical cell morphology, DNA fragments and sub-G(1) phase peak as well as Hoechst33258 and Annexin-V/PI labeling method with a time-dose-related manner. The morphology of NB4 cells cultured in the presence of 2 - 100 ng/ml ZGDHu-1 for three days was more mature with higher NBT positivity and expressions of CD11b and CD13 than those in control. The expression of phosphor-p38MAPK and bax was increased while phosphor-STAT3 and bcl-2 were unchanged by the treatment of ZGDHu-1. ZGDHu-1 could decrease the expression of hTERT-mRNA in a dose-dependent manner. It is concluded that ZGDHu-1 can inhibit proliferation, induce differentiation and apoptosis of NB4 cells. The mechanism may be associated with up-regulation of bax expression, enhancement of phosphor-p38MAPK activation and inhibition of hTERT-mRNA.

Morphogenesis and molecular basis on naked seed rice, a novel homeotic mutation of OsMADS1 regulating transcript level of AP3 homologue in rice.

The floral organs are formed from floral meristem with a regular initiation pattern in angiosperm species. Flowers of naked seed rice (nsr) were characterized by the overdeveloped lemma and palea, the transformation of lodicules to palea-/lemma-like organs, the decreased number of stamens and occasionally extra pistils. Some nsr spikelets contained additional floral organs of four whorls and/or abnormal internal florets. The floral primordium of nsr spikelet is differentiated under an irregular pattern and an incomplete determination. And molecular analysis indicated that nsr was a novel homeotic mutation in OsMADS1, suggesting that OsMADS1 played a distinct role in regulating the differentiation pattern of floral primordium and in conferring the determination of flower meristem. The gain-of-function of OsMADS1 transgenic lines presented the transformation of outer glumes to lemma-/palea-like organs and no changes in length of lemma and palea, but loss-of-function of OsMADS1 transgenic lines displayed the overdeveloped lemma and palea. Both findings revealed that OsMADS1 played a role in specifying lemma and palea and acted as a repressor of overdevelopment of lemma and palea. Moreover, it was indicated that OsMADS1 upregulated the transcript level of AP3 homologue OsMADS16, using real-time PCR analysis on gain- and loss-of-function of OsMADS1 transgenic lines.

Selective cytolysis of tumor cells by mumps virus S79.

We studied the sensitivity of several human cancer cell strains (HeLa, HT1080, SPC-A1, and ACHN) and normal cell strains (MRC-5 and Wish) to mumps virus (MuV) S79, a live attenuated vaccine strain. These cells exhibited a differential sensitivity to infection with MuV, and the susceptible sequences were ACHN > HeLa > HT1080 > SPC-A1 > Wish > MRC-5. In experiments in vivo, nude mice with HT1080 fibrosarcoma xenografts were randomly divided into three groups for intratumoral treatment with MuVS79, UV-inactivated MuVS79, and PBS. At 10(7) PFU, live MuVS79 injection caused in 7 of 9 mice complete regression by day 15 while rapid tumor growth occurred in all 9 mice treated with PBS. Rapid tumor progression also occurred in all 8 mice treated with UV-inactivated virus; however, tumor growth was delayed in the logarithmic phase relative to the PBS-treated tumors.

[Establishment of Genic Male Sterile Lines of Brassica napus by Wide Cross and Their Cytology and Morphology.].

One and three male sterile plants were found in the progenies of the intergeneric hybrids by crossing Brassica napus and Brassica juncea to Orychophragmus violaceus, respectively. After interspecific cross and full-sib crosses, four genic male sterile lines were obtained. The results of cytological observations indicated that somatic chromosome number was 38 in all types of sterile lines, which were considered as the type of B. napus. Most of pollen mother cells (PMC) in all sterile plants were observed normally for chromosome behaviour. But abnormal phenomena of chromosome, which present for chromosome lagging and bridge, were still found at each stage of meiosis in these sterile lines. These lines belonged to complete sterile type. There were distinct differences between sterile plants and normal plants in the flower morphology. Bud death at different degrees was found on anthotaxies in sterile plants. Studies on the flower growth showed that the growths of the pistils in sterile plants were gradually accelerated along with pollen sterility. At the same time there were two slowly growth stages of the pistils growth in normal plants. The prospects of these sterile lines were discussed in this paper.

[Improvement and optimization for "genetics" course teaching].

"Genetics" is one of the main courses for the study scheme of undergraduate students in university. This paper pointed out some problems found in the teaching process according to the characteristics of "Genetics" course. At the same time, the thought on the construction of "Genetics" course were discussed concerning the reform of course contents, the improvement of teaching method, the creation of teaching CAI (computer assisted instruction), the compile of electronic edition for teaching materials, the evaluation of teaching level, the innovation of experimental contents and the methods for improving the teaching effect.

[Change of Bcl-2, Bax proteins and mitochondrial membrane protein on nitric oxide induced apoptosis in HL-60 cells].

To investigate the possible mechanisms of nitric oxide (NO)-induced apoptosis in leukemia cell line HL-60, HL-60 cells in vitro were incubated with sodium nitroprusside (SNP), the in situ cell apoptosis quantitatively was assayed by TdT-mediated dUTP nick end labeling (TUNEL), the cell cycle DNA and proteins expression of Bcl-2, Bax, mitochondrial membrane protein (APO2.7) were analyzed by flow cytometry. The results showed that SNP induced HL-60 cell apoptosis in a dosage- and time-dependent manner. After exposure to SNP at the concentration of 1.0 mmol/L for 48 hours, the percentage of apoptosis HL-60 was (42.2 +/- 3.5)% for subG1 and (52.5 +/- 7.6)% for TUNEL respectively, and they are significantly higher than those in control and potassium ferricyanide (PFC) groups as same concentration. During the apoptosis process, it showed a decrease of Bcl-2 protein and an increase of Bax protein and mitochondrial membrane protein in HL-60 cell, proteins of Bcl-2, Bax and mitochondrial membrane were expressed in a dosage- and time-dependent manner too. In conclusion, during the process of SNP induced apoptosis in HL-60 cell, the expression of mitochondrial membrane protein was increased, Bcl-2 and Bax proteins may be important regulators.

[Hypoploid groups and their cytogenetic analysis in the progenies between Brassica napus and Orychophragmus violaceus].

Several groups of hypoploids were characterized in pentaploid (p) progenies of B. napus and O. violaceus. Cytological characterization of the P3 population revealed allosubstitution of one pair of B. napus chromosomes with O. violaceus chromosomes. This population had normal fertility and had three kinds of somatic cells with 36-38 chromosomes. Its somatic cells and pollen mother cells (PMC) mainly had 38 chromosomes. From a P4 population, three monosomic plants with 37 chromosomes were identified with differing morphology and fertility. One plant which grew vigorously was composed of various types of somatic cell and PMC, in which the cells with 37 chromosomes were observed mainly. The unpaired chromosome was small and with no negative effects on plant development. In the offsprings of the aneuploids (P3) with 41-44 chromosomes, there are four types of plants with 29-32 chromosomes. Except the plant with 29 chromosomes, others which appeared abnormal meiosis behavior and poor pollen fertility are mixoploid. The origin of these hypoploids and their prospects of application have been discussed.

Effects of litter removal on plant species diversity: a case study in tropical Eucalyptus forest ecosystems in South China.

The removal of litterfall in the Eucalyptus plantations in South China affected the plant biodiversity in these ecosystems was found based on the field observation and lab analysis. The protection times of species diversity of three Eucalyptus communities were different (Community I with no protection, Community II with 7-year-protection, Community III with 35-year-protection). The total numbers of species in these communities (from Community I to II to III) are 1, 6, and 17 respectively. The results showed that the protection of litterfall from being taken out of the ecosystem is important and can increase plant species diversity. This study combined biomass data, the chemical and physical properties of the soil, and the diversity of microbes in the communities. It is concluded that the mechanism of the effects of litter removal on biodiversity includes three factors: removing the suitable habitat of microbe and animal, decreasing the soil nutrient, and changing the special habitat for the germination and growth of invading plants. These results should have important implications for managing these Eucalyptus forest ecosystems in South China.

[Developmental genetic analysis for transparency of rice (Oryza sativa L.) at different environments].

The developmental behavior of gene expression for transparency of rice (TR) was analyzed from two-year experimental data at different filling stages by using both conditional and unconditional analysis methods for quantitative traits of seeds in cereal crops. It was revealed that the gene expression was most active for the performance of TR at the middle and late filling stages (8-21 d after flowering). The differences of expression at different environments for triploid endosperm nuclear genes, cytoplasmic genes and diploid maternal plant nuclear genes were found for the performance of TR. The phenomena of which some genetic effects were spasmodically expressible were detected among filling stages of rice. Because of the higher additive and cytoplasmic effects along with their interaction effects for TR at the filling stages, the better improving effects for this trait could be expected by selection in rice breeding. Some parents such as Zhenan 3 and 1391 were better than others for improving TR.
