Simple sequence repeat-based consensus linkage map of Bombyx mori.

We established a genetic linkage map employing 518 simple sequence repeat (SSR, or microsatellite) markers for Bombyx mori (silkworm), the economically and culturally important lepidopteran insect, as part of an international genomics program. A survey of six representative silkworm strains using 2,500 (CA)n- and (CT)n-based SSR markers revealed 17-24% polymorphism, indicating a high degree of homozygosity resulting from a long history of inbreeding. Twenty-nine SSR linkage groups were established in well characterized Dazao and C108 strains based on genotyping of 189 backcross progeny derived from an F(1) male mated with a C108 female. The clustering was further focused to 28 groups by genotyping 22 backcross progeny derived from an F(1) female mated with a C108 male. This set of SSR linkage groups was further assigned to the 28 chromosomes (established linkage groups) of silkworm aided by visible mutations and cleaved amplified polymorphic sequence markers developed from previously mapped genes, cDNA sequences, and cloned random amplified polymorphic DNAs. By integrating a visible mutation p (plain, larval marking) and 29 well conserved genes of insects onto this SSR-based linkage map, a second generation consensus silkworm genetic map with a range of 7-40 markers per linkage group and a total map length of approximately 3431.9 cM was constructed and its high efficiency for genotyping and potential application for synteny studies of Lepidoptera and other insects was demonstrated.

[Experimental studies on the treatment of colon cancer by cytosine deaminase gene and 5-fluorocytosine].

OBJECTIVE: To investigate the effect of cytosine deaminase (CD) gene plus 5-fluorocytosine (5-Fc) on the growth of human colon cancer xenograftin nude mice. METHODS: Retroviral vector expressing CD gene was transfected into human colon cancer SW1116 cells. Expression of the transfected CD gene in SW1116 (SWCD(2)) was confirmed by RT-PCR. The cytotoxicity of 5-Fc on SW1116 was determined by MTT assay in vitro. In vivo, the CD gene expression vector was injected intratumorally and 5-Fc was given by ip injections. RESULTS: In vitro, SWCD(2) cells were killed by 5-Fc with an IC(50) of 66 micromol/L while the nontrasfected SW1116 cells needed an IC(50) of 16 000 micromol/L to be killed. The growth of SWCD(2) xenografts was significantly inhibited by systemic administration of 5-Fc. CONCLUSION: CD gene/5-Fc system is a potential gene therapy strategy for human colon cancer.

[The acquisition of two silkworm CAPs markers and their use in genetic diversity and phylogenetic relationship].

Cleaved amplified polymorphic sequence (CAPs) markers are based on PCR amplification of known genes, cDNA sequences or RAPD sequences. The PCR products are digested by restriction enzymes, generating the simple type of data as heterozygotes and homozygotes. Here we designed primers based on silkworm attacin and alpha-amylase genes, then digested the PCR products in silkworm strains P50, C108 and their progeny F1 using 4 different restriction enzymes respectively. Furthermore, the genetic diversity and phylogenetic relationship of 12 silkworm strains were investigated using the obtained two CAPs markers.

[Expression of survivin and its correlation to angiogenesis in oral squamous cell carcinoma].

BACKGROUND & OBJECTIVE: Survivin can block the cell apoptosis through inhibiting the functions of Caspase-3 and Caspase-7, and selectively overexpresses in common human cancers. This study was designed to investigate the effects of Survivin on tumorigenesis and development of oral cancer and its correlation to angiogenesis. METHODS: The expression of Survivin in 8 specimens of normal oral mucosa, 14 specimens of dysplastic leukoplakia and 47 specimens of oral squamous cell carcinoma (OSCC), and the expression of CD34 in the 47 specimens of OSCC was detected by SP immunohistochemistry. Microvessel density (MVD) was also assessed. The correlations of Survivin expression to MVD and clinicopathologic features of the OSCC patients were analyzed. RESULT: The positive rates of Survivin were 0% in normal oral mucosa, 14.24% in dysplastic leukoplakia, and 55.32% in OSCC. Survivin expression was significantly stronger in OSCC than in normal oral mucosa and dysplastic leukoplakia (P < 0.05), while there was no difference between the last 2 groups (P > 0.05). Survivin expression was significantly stronger in moderately and poorly differentiated OSCC than in well differentiated OSCC (P < 0.05); the positive rate of Survivin was significantly higher in OSCC with lymph node metastasis than in OSCC without lymph node metastasis(71.43% vs. 38.89%, P < 0.05). In OSCC, MVD was increased (25.87 +/- 12.10, 28.70 +/- 7.69, 35.42 +/- 10.09, 41.13 +/- 9.62, respectively) along with the strengthened Survivin expression (-, +, 2+, 3+) (P < 0.05). CONCLUSION: Survivin expression is up-regulated in OSCC, and closely related with tumor cell differentiation, lymph node metastasis, and MVD; it may play an important role in tumorigenesis and development of OSCC.

[Non-recurrent laryngeal nerve during thyroid surgery: a report of 4 cases and literature review].

OBJECTIVE: To investigate clinical significance of non-recurrent laryngeal nerve. METHODS: Clinical data from 4 cases of non-recurrent laryngeal nerve and related literature review was made to acknowledge its incidence, variant types and matters concerned during thyroid surgery. RESULTS: Seven hundred and nineteen recurrent laryngeal nerves were exposed during 2156 thyroid operations in Xiangya Hospital, from which 4 were confirmed to hold non-recurrent laryngeal nerves (0.56%). Two cases were found on the right side and the other 2 on the left. Among the 4 cases, 3 (patients) were found to have non-recurrent laryngeal nerves during reoperation because of voice horse after the first operation. The other one was recognized during the first operation. CONCLUSIONS: Non-recurrent laryngeal nerve, a rare anomaly, is very vulnerable during thyroid surgery. Knowing the related knowledge of the non-recurrent laryngeal nerve and its types is helpful to avoid its damage during thyroid surgery.

[Experimental study on effect of epimedium flavonoids in protecting telomere length of senescence cells HU].

OBJECTIVE: To investigate the mechanism of senescence delay of human diploid fibroblast (2BS) and protecting telomere length by epimedium flavonoids (EF). METHODS: The drug sera of EF were used to treat the 2BS. The population doublings of 2BS cells were observed, the mRNA expression of p16 gene were determined by fluorescence real-time quantitative RT-PCR, the telomerase activation of 2BS cells were determined by TRAP-Hyb, the total retinoblastoma (Rb) and phosphorated Rb protein content were detected by ELISA, the telomere length of 2BS cells were determined by telomere restriction fragment (TRF) Southern blot assay. RESULTS: EF could significantly extend the population doublings of 2BS cells, the expression of p16 mRNA was decreased and the content of phosphorated Rb protein were increased by EF. The telomere lengthening of 2BS cells were improved by EF, but the telomerase was not activated. CONCLUSION: In senescence human fibroblasts 2BS cells, p16 gene mRNA expression increased, content of phosphorated Rb protein decreased and the telomere length of 2BS shortened, EF might delay the aging of cells through inhibiting the p16 gene expression, promoting the production of phosphorated Rb protein and to protect the length of telomere, but not activating the telomerase.
