Genome-wide Parallelism Underlies Rapid Freshwater Adaptation Fueled by Standing Genetic Variation in a Wild Fish.

A fundamental focus of ecological and evolutionary biology is determining how natural populations adapt to environmental changes. Rapid parallel phenotypic evolution can be leveraged to uncover the genetics of adaptation. Using population genomic approaches, we investigated the genetic architecture underlying rapid parallel freshwater adaptation of Neosalanx brevirostris by comparing four freshwater-resident populations with their common ancestral anadromous population. We demonstrated that the rapid parallel adaptation to freshwater followed a complex polygenic architecture and was characterized by genomic-level parallelism, which proceeded predominantly through repeated selection on the preexisting standing genetic variations. Frequencies of the genome-wide adaptive standing variations were moderate in the ancestral anadromous population, which had pre-adapted to fluctuating salinities. Relatively large allele frequency shifts were observed at some adaptive single-nucleotide polymorphisms (SNPs) during parallel adaptation to freshwater environments, with a large fraction of freshwater-favored alleles being fixed or nearly fixed. These adaptive SNPs were involved in multiple biological functions associated with osmoregulation, immunoregulation, locomotion, metabolism, etc., which were highly consistent with the polygenic architecture of adaptive divergence between the two ecotypes involving multiple complex physiological and behavioral traits. This work provides insight into the mechanisms by which natural populations rapidly evolve to changes in the environment and highlights the importance of standing genetic variation for the evolutionary potential of populations facing global environmental changes.

Investigation of destruxin A and B from 80 Metarhizium strains in China, and the optimization of cultural conditions for the strain MaQ10.

The production levels of destruxin A and B (DA and DB) of 80 Metarhizium strains from China were investigated. The average yields of DA and DB were 14.87+/-2.30 and 3.65+/-0.58 microg/mL, respectively. The strains isolated from soil had significantly (p<0.05) lower production of DA and DB than the strains isolated from insect cadavers. Also, there was a positive correlation in the yields of DA and DB, and the regressive equations were established. Of the 80 strains, MaQ10 had the highest yields of DA and DB, amounted to 106.78+/-9.41 and 29.52+/-2.63 microg/mL, respectively, at 10 days fermentation period. Followed by the strains MaQ05, MaQ07 and MaQ12, their yields of DA were 79.72+/-5.36, 77.69+/-8.54 and 70.04+/-10.1 microg/mL, respectively, and DB were 16.81+/-1.51, 13.38+/-0.41 and 16.88+/-1.15 microg/mL, respectively. Furthermore, the cultural conditions of MaQ10 were optimized to produce DA and DB. The optimal inoculum, initial pH, temperature and rotary speed were 8%, pH 9.0, 27 degrees C and 240 r/min for DA, and 8%, pH 9.0, 25 degrees C and 220 r/min for DB. Under these optimal culture conditions, the predicted production of DA and DB was 193.87 and 39.85 microg/mL, and the actual production was 189.13 and 43.35 microg/mL, respectively.

"Aha!" effects in a guessing riddle task: an event-related potential study.

Insight problem solving has been the topic of much investigation. It is believed widely that insight critically contains the process of breaking one's mental set. Recent functional magnetic resonance imaging (fMRI) research on puzzle solving showed that insight was associated with activities in anterior cingulate cortex (ACC) and other areas (Luo and Niki [2003]: Hippocampus 13:274-281). We proposed ACC might mediate processes of breaking one's mental set, given its well-known role in cognitive conflict. In the present research, high-density event-related potentials (ERPs) were recorded to examine the electrophysiologic correlates of insight problem solving. One hundred twenty interesting Chinese riddles (half difficult and half easy) were adopted as materials. For each trial, subjects were either given an easy puzzle followed by a keyword that was consistent with the subject's initial thinking ("No-aha answer"), or a difficult puzzle followed by a keyword that was consistent with an unusual interpretation, so that it broke the subject's initial mental set ("Aha answer"). Results from 14 subjects showed that Aha answers elicited a more negative ERP deflection than did No-aha answers in the time window from 250-500 msec after onset of the answer. The ERP difference wave (Aha minus No-aha answer) showed the maximum amplitude over the central site (Cz) with a peak latency of 380 msec (N380). Voltage and current density maps of the difference wave showed strong activity and current density in the frontocentral region. Dipole analysis localized the generator of the N380 in the ACC. N380 therefore probably reflects an "Aha!" effect, and the ACC generator may be involved in the breaking of mental set.

[Prognostic analysis of 151 patients with maxillary sinus malignant neoplasms].

BACKGROUND & OBJECTIVE: Five-year survival rate of patients with maxillary malignant neoplasms is low, the prognostic factors of these neoplasms were unclear. This study was to investigate prognostic factors of maxillary sinus malignant neoplasms. METHODS: Records of 151 inpatients with malignant neoplasms of maxillary sinus initially treated at Cancer Center of Sun Yat-sen University from Sep. 1983 to Mar. 1999 were reviewed. Of 151 cases, 72 were squamous cell carcinoma (SCC), 44 were adenocarcinoma, 16 were sarcoma, and 19 were other histological types; according to 1997 UICC classification, 7 were stage II, 55 were stage III, and 89 were stage IV; 66 patients received combined therapy of surgery and radiotherapy, 14 received surgery alone, 25 received radiotherapy alone, 39 received other treatments, and 7 gave up treatment. All patients were followed up for more than 5 years. Influences of clinicopathologic factors on prognosis of patients with maxillary sinus malignant neoplasms were analyzed by Kaplan-Meier method, and Cox regression model with SPSS10.0 software. RESULTS: Five-year overall survival rate of patients of </=40 years old was 55.7%, that of patients of >40 years old was 33.3%(P=0.030); that of patients with SCC was 30.2%, of patients with adenocarcinoma was 57.5%, of patients with sarcoma was 24.3%, of patients with tumor of other histological types was 50.7% (P=0.011); that of patients with tumor of stage II, III, and IV were 85.7%, 45.8%, and 32.7%, respectively (P=0.029); that of patients with cervical metastases was 14.4%, of patients without cervical metastases was 44.1% (P=0.005); that of patients with distant metastases was 14.3%, of patients without distant metastases was 41.1% (P=0.011); that of patients without treatment was 14.3%, of patients treated with surgery alone was 42.9%, of patients treated with radiotherapy alone was 32.3%, of patients treated with combined therapy of surgery and radiotherapy was 50.8%, of patients treated with other treatments was 29.1% (P=0.004). Univariate survival analysis showed that the above 6 factors were prognostic factors of patients with maxillary sinus malignant neoplasms. Multivariate analysis showed that combination of surgery and radiotherapy (P=0.004, OR< 1), clinical stage (P=0.025, OR >1), SCC (P=0.016, OR >1), and sarcoma (P=0.003, OR >1) were independent prognostic factors of patients with maxillary sinus malignant neoplasms. CONCLUSION: For maxillary sinus malignant neoplasms, patients with SCC or sarcoma had poorer survival than patients with adenocarcinoma or other histological types of tumor; patients with sarcoma had poorer survival than patients with SCC. The higher the patient's clinical stage was, the worse his prognosis was. Combination of surgery and radiotherapy may be the best treatment for patients with maxillary sinus malignant neoplasms.

[Establishment of prostatic hyperplasia model with castration beagle canines].

OBJECTIVE: To establish a prostatic hyperplasia model with Beagle canines. METHODS: Twenty-four two-year-old male Beagle canines were divided into treatment and control groups at random and were administrated testosterone propionate (TP) through intramuscular injection two months after castration. Three treatment groups were given 0.8, 2.5 and 7.5 mg/kg TP respectively, and the control was given the same volume of vehicle. Two months later, half of the animals were killed and the serum and prostate were prepared. After the wet weight and volume of prostate were measured, the dihydrotestosterone (DHT) level of serum and prostate were detected with DHT radioimmunoassay (RIA) kit, and paraffine section from canine prostate was stained by the HE methods. Pictures were taken by digital camera under microscope, and all the pictures were analyzed by computer for epithelial cell height and acinar luminal area of prostate with micro image analysis software. The canine prostate volume was measured with ultrasonic diagnosis instrument before castration, at two months after castration and at two months after being given TP. RESULTS: The ultrasonic results showed that the prostate volumes of all the canines were smaller at two months after castration than before castration (P < 0.05), and after having been administrated TP for two months, and the prostate volumes of all treatment groups were larger than those of the control group (P < 0.01). The wet weight of the prostate of the treatment group was higher than that of the control group (P < 0.05), and both had dose-dependent relationship. The DHT level of serum and prostate of the canines became higher with the increase of TP dose. The results of micro image analysis showed that the acinar luminal area of prostate was enlarged, and the epithelial cell height increased with larger dose of TP. CONCLUSIONS: It is practicable to establish prostatic hyperplasia model in Beagle canines after two months of TP administration.
