Syntheses, crystal structures, and magneto-structural correlations of novel Cu(II) complexes containing a planar [Cu(mu-L1)]2 (HL1 = 3-(2-pyridyl)pyrazole) unit: from dinuclear to tetranuclear and then to one-dimensional compounds.

Seven new Cu(II) complexes based on a binuclear planar unit [Cu(mu-L(1))](2), [[Cu(mu-L(1))(NO(3))(H(2)O)](2) (1), [Cu(mu-L(1))(HL(1))(ClO(4))](2) (2), [Cu(4)(mu-L(1))(6)(NO(3))(2)] (3), [Cu(4)(mu-L(1))(6)(L(1))(2)] (4), [Cu(4)(mu-L(1))(6)(mu-L(2))](n) (5), [Cu(4)(mu-L(1))(6)(mu-L(3))](n) (6), [[Cu(4)(mu-L(1))(4)(mu-L(4))(2)](H(2)O)(3)](n) (7) (HL(1) = 3-(2-pyridyl)pyrazole, L(2) = 1,8-naphthalenedicarboxylate, L(3) = terephthalate, L(4) = 2,6-pyridinedicarboxylate)}, have been synthesized and characterized by elemental analysis, IR, and X-ray diffraction. In 1 and 2, the Cu(II) centers are linked by deprotonated pyrazolyl groups to form dinuclear structures. 3 and 4 have similar gridlike tetranuclear structures in which two additional deprotonated L(1) ligands bridge two [Cu(mu-L(1))](2) units perpendicularly. 5 and 6 consist of similar one-dimensional (1-D) chains in which gridlike tetranuclear copper(II) units similar to that of 3 are further linked by L(2) or L(3) ligands, respectively. And, in 7, L(4) ligands link [Cu(mu-L(1))](2) binuclear units to form a tetranuclear gridlike structure in chelating/bridging mode and simultaneously bridge the tetranuclear units to form a 1-D chain. The magnetic properties of all complexes were studied by variable-temperature magnetic susceptibility and magnetization measurements. The obtained parameters of J range from -33.1 to -211 cm(-1), indicating very strong antiferromagnetic coupling between Cu(II) ions. The main factor that affects the |J| parameter is the geometry of the Cu(N(2))(2)Cu entity. From the magnetic point of view, 1 and 2 feature "pure" dinuclear, 3 and 5 tetranuclear, and 4, 6, and 7 pseudodinuclear moieties.

Synthesis, structures, and magnetic properties of the copper(II), cobalt(II), and manganese(II) complexes with 9-acridinecarboxylate and 4-quinolinecarboxylate ligands.

To explore the relationships between the structures of ligands and their complexes, we have synthesized and characterized a series of metal complexes with two structurally related ligands, 9-acridinecarboxylic acid (HL(1)) and 4-quinolinecarboxylate acid (HL(2)), [Cu(2)(mu(2)-OMe)(2)(L(1))(2)(H(2)O)(0.69)](n) 1, [Cu(2)(L(1))(4)(CH(3)OH)(2)] 2, [Cu(3)(L(1))(6)(CH(3)OH)(6)].3H(2)O 3, [Mn(3)(L(1))(6)(CH(3)OH)(6)].3H(2)O 4, [Co(3)(L(1))(6)(CH(3)OH)(6)].3H(2)O 5, [Cu(L(2))(2)](n) 6, [Mn(L(2))(2)(H(2)O)](n) 7, and [Co(L(2))(2)(H(2)O)](n) 8. 1 is a three-dimensional (3D) polymer with an interpenetrating NbO type network showing one-dimensional (1D) channels, whereas 2 and 3 take bi- and trinuclear structures, respectively, because of the differences in basicity of the reaction systems in preparing the three complexes. 4 and 5 have trinuclear structures similar to that of 3. In 1-5, ligand L(1) performs different coordination modes with N,O-bridging in 1 and O,O'-bridging in 2-5, and the metal ions also show different coordination geometries: square planar in 1, square pyramidal in 2, and octahedral in 3-5. 6 has a two-dimensional structure containing (4,4) grids in which L(2) adopts the N,O-bridging mode and the Cu(II) center takes square planar geometry. 7 and 8 are isostructural complexes showing 1D chain structures, with L(2) adopting the O,O-bridging mode. In addition, the intermolecular O-H...N hydrogen bonds and pi-pi stacking interactions further extend the complexes (except 1 and 6), forming 3D structures. The magnetic properties of 2-7 have been investigated and discussed in detail.

Novel 3-D framework nickel(II) complex with azide, nicotinic acid, and nicotinate(1-) as coligands: hydrothermal synthesis, structure, and magnetic properties.

The hydrothermal reaction of Ni(NO3)2, NaN3, and nicotinic acid (Hnic) yielded a 3-D supramolecular framework complex, [Ni(1.5)(N3)(nic)2(Hnic)]n (1) with unusual magnetic properties. In 1, Hnic and nic-1 coexist and show different coordination modes (mu2-Hnic-N,O, mu2-nic-N,O, and mu3-nic-N,O,O bridging NiII ions), and azide groups adopt mu-1,1 (EO) bridging mode linking NiII ions with an unusual Ni-N-Ni angle.

Novel dithioether-silver(I) coordination architectures: structural diversities by varying the spacers and terminal groups of ligands.

An investigation into the dependence of the framework formation of coordination architectures on ligand spacers and terminal groups was reported based on the self-assembly of AgClO4 and eight structurally related flexible dithioether ligands, RS(CH2)nSR (Lan, R = ethyl group; Lbn, R = benzyl group, n= 1-4). Eight novel metal-organic architectures, [Ag(La1)3/2ClO4]n (1a), [Ag2(La2)2(ClO4)2]2 (2a), [AgLa3ClO4]n (3a), {[Ag(La4)2]ClO4}n (4a), [AgLb1ClO4]2 (1b), [Ag(Lb2)2]ClO4 (2b), {[Ag(Lb3)3/2(ClO4)1/2](ClO4)1/2}n (3b) and [Ag(Lb4)3/2ClO4]n(4b), were synthesized and structurally characterized by X-ray crystallography. Structure diversities were observed for these complexes: 1a forms a 2-D (6,3) net, while 2a is a discrete tetranuclear complex, in which the AgI ion adopts linear and tetrahedral coordination modes, and the S donors in each ligand show monodentate terminal and mu2-S bridging coordination fashions; 3a has a chiral helical chain structure in which two homo-chiral right-handed single helical chains (Ag-La3-)n are bound together through mu2-S donors, and simultaneously gives rise to left-handed helical entity (Ag-S-)n. In 4a, left- and right-handed helical chains formed by the ligands bridging AgI centers are further linked alternately by single-bridging ligands to form a non-chiral 2-D framework. 1b has a dinuclear structure showing obvious ligand-sustained Ag-Ag interaction, while 2b is a mononuclear complex; 3b is a 3-D framework formed by ClO4- linking the 2-D (6,3) framework, which is similar to that of 1a, and 4b has a single, double-bridging chain structure in which 14-membered dinuclear ring units formed through two ligands bridging two AgI ions are further linked by single-bridging ligands. In addition, a systematic structural comparison of these complexes and other reported AgClO4 complexes of analogous dithioether ligands indicates that the ligand spacers and terminal groups take essential roles on the framework formation of the AgI complexes, and this present feasible ways for adjusting the structures of such complexes by modifying the ligand spacers and terminal groups.

Novel Cd(II) coordination polymers with flexible disulfoxide ligands: effects of ligand spacers, terminal groups and counter anions on the complex framework formations.

Five novel Cd(II) coordination polymers with three structurally related flexible disulfoxide ligands, [[Cd(L1)3](ClO4)2]n (1), [[Cd(L2)3](ClO4)2(CHCl3)]n (2), [Cd(L2)(NO3)2(H2O)]n (3), [Cd2(L3)2(NO3)4]n (4) and [[Cd(L3)3](ClO4)2]n (5), where L1= 1,3-bis(phenylsulfinyl)propane, L2= 1,4-bis(phenylsulfinyl)butane and L3= 1,4-bis(ethylsulfinyl)butane, were synthesized and structurally determined by X-ray diffraction. Complex 1 has a 2D layer structure, in which part of the L1 ligands bridge the Cd(II) ions to form double-bridging chains and the other part of ligands link such chains to form a 2D framework. Complexes 2 and 5 are isomorphous, showing unusual 2D (3,6) network structures containing triangular grids. Complex 3 adopts a 2D (4,4) network formed by L2 linking the NO3- bridged (Cd-O-N-O-)n 1D zigzag chains. By contrast, is a 1D chain, in which two Cd(II) centers are bridged by mu2-O of sulfoxide groups to form a dinuclear unit, and L3 ligands link such dinuclear units to form a 1D double-bridging chain. The structural differences among such complexes show that the ligand nature and counter anions have important influences on the complex structures, which may provide a rational method for controlling the framework formation in metal-organic coordination polymers.

Polymeric silver(I) complexes with pyridyl dithioether ligands: experimental and theoretical investigations on the coordination properties of the ligands.

The reactions of four flexible tetradentate ligands, 1,3-bis(2-pyridylthio)propane (L1), 1,4-bis(2-pyridylthio)butane (L2), 1,5-bis(2-pyridylthio)pentane (L3) and 1,6-bis(2-pyridylthio)hexane (L4) with AgX (X = BF4-, ClO4-, PF6-, or CF3SO3-) lead to the formation of seven new complexes: [AgL1(BF4)]2 (1), [[AgL2](ClO4)]infinity (2), [[AgL2(CH3CN)](PF6)]infinity (3), [[AgL3](BF4)(CHCl3)]2 (4), [[AgL3(CF3SO3)](CH3OH)(0.5)]infinity (5), [[Ag2L4(2)](BF4)2]infinity (6), and [[AgL4](PF6)]infinity (7), which have been characterized by elemental analyses, IR spectroscopy, and X-ray crystallography. Single-crystal X-ray analyses show that complexes 1 and 4 possess dinuclear macrometallacyclic structures, and complexes 2, 3 and 5-7 take chain structures. In all the complexes, the nitrogen atoms of ligands preferentially coordinate to silver atoms to form normal coordination bonds, while the sulfur atoms only show weak interactions with silver atoms and the intermolecular AgS weak contacts connect the low-dimensional complexes into high-dimensional supramolecular networks. Additional weak interactions, such as pi-pi stacking, F...F weak interactions, Ag...O contacts or C-H...O hydrogen bonds, also help to stabilize the crystal structures. It was found that the parity of the -(CH2)n- spacers (n = 3-6) affect the orientation of the two terminal pyridyl rings, thereby significantly influence the framework formations of these complexes. The coordination features of ligands and their conformation changes between free and coordination states have been investigated by DFT calculations.

Novel lanthanide coordination polymers with a flexible disulfoxide ligand, 1,2-bis(ethylsulfinyl)ethane: structures, stereochemistry, and the influences of counteranions on the framework formations.

The reactions of meso-1,2-bis(ethylsulfinyl)ethane (meso-L) with Ln(ClO(4))(3) [Ln(NO(3))(3) or Ln(NCS)(3)] in MeOH and CHCl(3) gave a series of new lanthanide coordination polymers, [[Ln(micro-meso-L)(rac-L)(2)(CH(3)OH)(2)](ClO(4))(3)](n) [Ln: La (1), Nd (2), Eu (3), Gd (4), Tb (5), Dy (6), and Yb (7)], [Yb(micro-meso-L)(1.5)(NO(3))(3)](n) (8), and [La(micro-meso-L)(2.5)(NCS)(3)](n) (9). All the structures were established by single-crystal X-ray diffraction. Complexes 1-7 are isostructural with infinite single micro-chain structure, in which the L ligands take two kinds of coordination modes: bidentate chelating and bis-monodentate bridging. Six sulfur atoms of the sulfoxide groups around each Ln(III) center adopt alternatively the same R or S configuration in the chain. In addition, the configuration change of partial ligands occurred from the meso to the rac form when reacting with Ln(ClO(4))(3). To our knowledge, this is the first example of disulfoxide complexes with two kinds of coordination modes and three kinds of configurations (R,R, S,S, and R,S) occurring simultaneously in the same complex. 8 exhibits single-double bridging chain structure, in which dinuclear macrometallacycles formed through bridging two Yb(III) by two meso-L ligands are further linked by another meso-L ligand. In 9 each La(III) ion is linked to five other La(III) ions by five meso-L ligands to form a 5-connected 2-D (3/4,5) network containing two types of macrometallacyclic arrays: quadrilateral and triangle grids. The structural differences among 1-7, 8, and 9 show that counteranions play important roles in the framework formation of such coordination polymers. In addition, the luminescent properties of 3 and 5 were also investigated.

[Expression of the infectious bursal disease virus polyprotein in Vero cells using attenuated Salmonella typhimurium as transgenic carrier].

To examine if polyprotein gene (VP2/VP4/VP3) of Infectious Bursal Disease Virus (IBDV) could be delivered into mammalian cells and expressed using attenuated Salmonella typhimurium as vector. The IBDV polyprotein gene was amplified by RT-PCR and inserted in to pCI, an eukaryotic expression plasmid. The resulting recombinant pCI-VP2/VP4/VP3 was transformed by electroporation into attenuated Salmonella typhimurium strain ZJ111 (dam- and phoP-), which was then use to transfect the Vero cells. Gene specific RT-PCR revealed that VP2/VP4/VP3 was transcribed into mRNA in the Vero cells. Indirect immunofluorscence assay, SDS-PAGE and Western-blot analysis showed that VP2/VP4/VP3 was expressed and the product was immuno-reactive with anti-IBDV serum. This work provides essential precondition for developing a new oral DNA vaccine against IBDV.

Adjusting the frameworks of silver(I) complexes with new pyridyl thioethers by varying the chain lengths of ligand spacers, solvents, and counteranions.

In our efforts to systematically investigate the effects of the linker units of flexible ligands and other factors on the structures of Ag(I) complexes with thioethers, five new flexible pyridyl thioether ligands, bis(2-pyridylthio)methane (L(1)()), 1,3-bis(2-pyridylthio)propane (L(3)()), 1,4-bis(2-pyridylthio)butane (L(4)), 1,5-bis(2-pyridylthio)pentane (L(5)), and 1,6-bis(2-pyridylthio)hexane (L(6)), have been designed and synthesized, and the reactions of these ligands with Ag(I) salts under varied conditions (varying the solvents and counteranions) lead to the formation of eight novel metal-organic coordination architectures from di- and trinuclear species to two-dimensional networks: [Ag(3)(L(1)())(2)(ClO(4))(2)](ClO(4)) (1), [[AgL(3)](ClO(4))]( infinity ) (2), [[Ag(2)(L(4))(2)](ClO(4))(2)(CHCl(3))]( infinity ) (3), [[AgL(4)](ClO(4))(C(3)H(6)O)]( infinity ) (4), [[Ag(2)L(4)](NO(3))(2)]( infinity ) (5), [Ag(2)L(4)()(CF(3)SO(3))(2)]( infinity ) (6), [[AgL(5)](ClO(4))(CHCl(3))](2) (7), and [[AgL(6)()](ClO(4))]( infinity ) (8). All the structures were established by single-crystal X-ray diffraction analysis. The coordination modes of these ligands were found to vary from N,N-bidentate to N,N,S-tridentate to N,N,S,S-tetradentate modes, while the Ag(I) centers adopt two-, three-, or four-coordination geometries with different coordination environments. The structural differences of 1, 2, 3, 7, and 8 indicate that the subtle variations on the spacer units can greatly affect the coordination modes of the terminal pyridylsulfanyl groups and the coordination geometries of Ag(I) ions. The structural differences of 3 and 4 indicate that solvents also have great influence on the structures of Ag(I) complexes, and the differences between 3, 5, and 6 show counteranion effects in polymerization of Ag(I) complexes. The influences of counterions and solvents on the frameworks of these complexes are probably based upon the flexibility of ligands and the wide coordination geometries of Ag(I) ions. The results of this study indicate that the frameworks of the Ag(I) complexes with pyridyl dithioethers could be adjusted by ligand modifications and variations of the complex formation conditions.

[Rapid construction of infectious clones of infectious bursal disease virus].

A rapid procedure was established for rescuing infectious bursal disease virus (IBDV), an important pathogen in poultry. A full-length cDNA clone of the segment B of a CEF-adapted IBDV strain HZ2 was constructed by long RT-PCR, and the 2827 bp nucleotide sequence, including the 5 - and 3 -noncoding regions (NCR), was established. Then the cDNA clone of segment B was engineered to make it contain three silent nucleotide changes, creating a new EcoRV site that was different from the parent virus sequences, by site-directed silent mutagenesis. Cotransfection of eukaryotic expression recombinants containing modified segment A and segment B with Lipofectamine into Vero cells resulted in the expression of IBDV RNA and proteins, as confirmed by Northern RNA dot hybridization and indirect immunofluorescence assay analysis. The change of cell morphology after cotransfection and passages of cell cultures was similar to that of cells infected by authentic IBDV, causing cellular pathogenic effects (CPE). The virus-like particles at 55-60 nm were observed under electron microscopy, affirming the rescue of IBDV. The genetic markers were retained in the recovered progeny virus.

[Expression of polyprotein of infectious bursal disease virus in Bombyx mori].

Segment A of the genome of infectious bursal disease virus(IBDV) encodes structure protein VP2 and VP3 and protease VP4. In this study a polyprotein gene of IBDV was inserted into a Bombyx mori baculovirus transfer vector pAcHLT--C and contransfected into BmN cells with linear genome DNA of virus Bm-BacPAK6. Dot hybridization suggested that the segment A of the virus genome was inserted in the genome of Bm-BacPAK6. The silkworm of fifth instars were infected by the recombinant virus and the immunogenicity of the infected larvae's blood were examined with ELISA, SDS-PAGE and Western blotting. It appears that the recombinant polyprotein has the property of immunoreactivity and the expression in larvae reached the pick 5-day post infection.

Amplification and Cloning by Long RT-PCR of Full-length Genome of Larger Segment of Chicken Infectious Bursal Disease Virus.

To develop the genetic rescue techniques for infectious bursal disease virus (IBDV), Birnaviridae family, the full-length cDNA of the larger segment of the chicken IBDV was amplified and cloned by long RT-PCR. A comparison of four purification and extraction methods of RNA from IBDV infected chicken embryoid fibroblasts (CEF) showed that the ultracentrifugation followed by proteinase Kdigestion extracted dsRNA more effectively. Then reverse transcription was carried out at 50 degrees using Superscipt II enzyme, followed by RNase H digestion. Amplification of single stranded cDNA in a single step resulted in the synthesis of the full-length segment A of 3 259 bp. The amplified product was cloned and sequenced, identifying that it was an IBDV. This method is superior to other methods based on amplifying different parts of the genome many times, therefore the cloning procedure was simplified.
