[Construct tissue-engineered bone by co-seeding marrow stromal cells and endothelial cells].

To identify the role of co-seeding marrow stromal cells (MSCs) and endothelial cells (ECs) onto biocomposite in constructing tissue-engineered bone for promoting osteogenesis and angiogenesis, we seeded rat marrow stromal cells (rMSCs) and human umbilical vein endothelial cells (hUVECs) onto poly (L-lactic acid)/beta-tricalcium phosphate (PLLA/beta-TCP) macroporous composite (the test group), and seeded rMSCs onto the same kind of composite (the control group). We implanted these biocomposites into the thighs of nude mice intramuscularly and sacrificed these mice at 1, 4, 8, 12, 16 weeks after implantation, respectively. Biocomposites were taken out and histomorphometry was performed. Image manipulation technology was used to calculate the percentages of new bone area and material area in the test group and control group. We found that the percentage of new bone area of test group increased faster than that of control group, the percentage of material of test group decreased faster than that of control group, and the growth of capillary network of test group was much better than that of control group. These results suggest that co-seeding MSCs and ECs onto scaffold for constructing tissue-engineered bone is beneficial to osteogenesis, to angiogenesis in the prosthesis, and to the degradation of scaffold, and it is of significance in the research of bone tissue engineering.

[Analysis on 2071 cases excluded from severe acute respiratory syndrome in Beijing].

OBJECTIVE: To examine the characteristics of cases excluded from severe acute respiratory syndrome (SARS) and the reasons for exclusion. METHODS: 2071 probable or suspected cases excluded from SARS between March and June, 2003 were analyzed. RESULTS: Two-thirds of the excluded cases were males. Construction workers, students and retired people ranked top three in all the occupation categories. Three peaks appeared in the dates of exclusion, and the most obvious one was from June 7 to June 13. There were two peaks in the distribution of time period from onset to exclusion, one was six to ten days and the other was forty-eight to fifty-two days after onset. Patients with history of close contact were more likely to be excluded within fifty days after onset than those without close history of contact. Pneumonia, common cold and lung infection were the leading causes for correction in the 1211 excluded cases. CONCLUSION: Study on the diagnosis and differential diagnosis should be emphasized.

Light-evoked responses of the mouse retinal pigment epithelium.

In response to light, the retinal pigment epithelium (RPE) generates a series of slow potentials that can be recorded as the c-wave, fast oscillation (FO), and light peak (LP) of the electroretinogram (ERG). As these potentials can be related to specific cellular events, they provide information about RPE function and how that may be altered by disease or experimental manipulation. In the present study we describe a noninvasive means for recording the light-evoked responses of the mouse RPE and use this to define the stimulus-response properties of the major components in three inbred strains of mice (BALBc/ByJ, C57BL/6J, and 129/SvJ) and two mouse mutants that reduce activity in the rod pathway. All of the major ERG components generated by the RPE are readily measured in the mouse. In albino strains (BALBc/ByJ and 129/SvJ) the intensity-response functions for the c-wave, FO, and LP are shifted toward lower intensities in comparison to those for C57BL/6J mice. Each of these components was markedly reduced in mice lacking transducin in which rod phototransduction is interrupted, indicating that they reflect primarily rod photoreceptor activity. All components were observed in no b-wave (nob) mutant mice, indicating that inner retinal activity does not make a major contribution to these potentials. Further studies of mutant mice will allow us to define the functional consequences of gene manipulation on RPE function and to evaluate specific hypotheses regarding the generation of ERG components.

Quantitative assessment of pericardial effusion volume by two-dimensional echocardiography.

The clinical applicability of a method for quantifying pericardial effusion (PE) was studied. The pericardial and cardiac volumes were determined from border tracings of 2-dimensional echocardiograms fitted with 3-dimensional disk models. The PE volume was the difference between pericardial and cardiac volumes. A phantom study included 54 cases with different volumes and viewing geometry showed a correlation coefficient r = 0.98 and an accuracy of +/- 6%. A clinical study included 20 cases showing that the estimated PE volume correlated well with that drained surgically (between 100 mL and 1200 mL): y = 0.81 x + 120 mL; r = 0.91, P <.0001. The percent error, determined by the standard error of the estimate (114 mL) over mean (548 mL), was 20%. Intraobserver variability was 4% and interobserver variability 6%. The 3-dimensional disk method provides a quick and convenient way to quantify PE from 2-dimensional echocardiograms with acceptable accuracy and reproducibility.

[Study on the changes of demography and behavioral characteristics of drug users in Beijing].

OBJECTIVE: To understand the demography changes and behaviors in drug users. METHOD: Self-reported questionnaires was used and longitudinal investigation was conducted in one of the detoxication centers in Beijing in 1998 and 2000. Drug users were randomly chosen. RESULTS: Results showed that age of drug users tend to become younger with the numbers of drug users aged below 25, increased from 18.7% in 1998 to 28.2% in 2000. Majority of drug users remained males, but the proportion of females seemed to increase. Distribution of occupation showed that the largest increase fell among individual enterprisers, from 15.2% in 1998 to 25.9% in 2000. With educational back-ground, the proportion of lower than elementary education level, including illiterate, increased. Fifty percent of drug users were unmarried which increased from 40.8% in 1998 to 53.2% in 2000. Needle sharing was quite common, 16.5% in 1998 and 11.9% in 2000, but the decrease was not statistically significant (P > 0.05). Fifty-seven point three percent of the injecting drug users did not have constant partners to share equipments. Proportion of extra-marriage sexual practice increased from 12.5% in 1998 to 27.5% in 2000, and significant difference (Chi-square = 12.50, P < 0.001). Multiple partners in extra-married drug users was also found (mean = 2). Compared to 1998, condom use during every sexual practice increased in 2000, but 47.7% drug users still never used condom. CONCLUSION: In summary, as the quick increase of drug users, sharing of injecting equipment and high-risk sexual behavior, including multiple partners and unprotected sex, were quite common, with the possibility of HIV epidemic in drug users.

[Research in use of vascular endothelial cells to promote osteogenesis of marrow stromal cells].

Osteogenesis deep in the materials is postponed because the blood vessels grow into the materials very slowly. It is one of the key challenges to the bone tissue engineering researchers. However, it was found that marrow stromal cells can produce vascular endothelial growth factor (VEGF) while endothelial cells produce bone morphogenetic protein (BMP). So we presume if we have co-culture of marrow stromal cells and endothelial cells on the biomaterials, osteogenesis and angiogenesis may be promoted simultaneously. In this experiment, we compare the alkaline phosphatase (ALP) activity and osteocalcin (OCN) production of three groups: the marrow stromal cells under activation of endothelial cells culture fluid, the induced marrow stromal cells, and the untreated marrow stromal cells. The results reveal that the ALP activity and OCN production of the marrow stromal cells under activation of endothelial cell culture fluid and the ALP activity and OCN production of the induced marrow stromal cells do not show statistically significant difference, but they are significantly higher than those of the untreated marrow stromal cells (P < 0.01). It is concluded that the VEGF in the endothelial cell culture fluid can promote osteogenesis as effective as osteogenic induction medium can do.

[Research advances in gene-enhanced tissue engineering].

Gene-enhanced engineering deals with the scientific and technologic endeavour to produce cultured cells or polymer matrices transduced with multiple gene vectors encoding cytokine cDNA by means of genetic engineering technique, to make transduced cells or gene activated matrices highly express according cytokine, and then to enhance certain abilities of the artificial tissue. Up to now, various genes encoding modulatory species of ribonucleic of proteins such as growth factors, receptors, and transcription factors have been used in the context of gene-enhanced tissue engineering and expressed within numerous tissues, including artificial blood vessels, bone, cartilage, skin and urinary system, etc. Many experiments in vitro or in vivo have begun to show good prospects and great potential application of the new approach. We believe great changes will take place in the research field of tissue engineering due to the induction the of genetic engineering, and the new approach will become a very promising and valuable tool for therapy.

Complete sequencing of anti-vancomycin fab fragment by liquid chromatography-electrospray ion trap mass spectrometry with a combination of database searching and manual interpretation of the MS/MS spectra.

Sequencing of anti-vancomycin monoclonal antibody (mAb) Fab region (48,000 Da) was carried out using liquid chromatography-electrospray ionization ion trap mass spectrometry (LC/ESI-MS). Comprehensive strategies were employed to ensure complete sequence coverage. The sequence information was obtained from the spectra of collision-induced dissociation (CID) (MS/MS) of the protonated proteolytic peptides resulting from multiple enzymatic digestions of reduced/S-carboxymethylated (RCM) light chain and Fd fragment. Database searching of the spectra against the published immunoglobulin G (IgG) sequences allowed the identification of all the peptides in constant domains as well as partial sequences in variable domains. The rest of the sequences were deduced by manual interpretation of the peptide tandem mass spectrometry (MS/MS) spectra. The analysis showed that the N-terminus of the heavy chain was modified by the conversion of a glutamine residue to pyroglutamic acid.

A role for KH domain proteins (Sam68-like mammalian proteins and quaking proteins) in the post-transcriptional regulation of HIV replication.

Overexpression of Sam68 functionally substitutes for, as well as synergizes with, human immunodeficiency virus type 1 (HIV-1) Rev in RRE (Rev response element)-mediated gene expression and virus replication. In addition, COOH-terminal deletion and/or point mutants of Sam68 exhibit a transdominant negative phenotype for HIV replication. Sam68 is a member of KH domain family that includes SLM-1, SLM-2 (Sam68 like mammalian); and QKI-5, QKI-6, and QKI-7 (mouse quaking) proteins. The objective of this study was to examine the effects of these KH family proteins on RRE- and CTE (constitutive transport element of type-D retrovirus)-mediated transactivation. We now report that SLM-1 and SLM-2 proteins, which are the closest relatives of Sam68, marginally enhanced RRE-mediated transactivation, while QK isoforms that are distant relatives of Sam68 had no effect. Interestingly, these proteins still enhanced the effect of Rev in RRE-mediated gene expression. The increase in chloramphenicol acetyltransferase activity was also reflected at the levels of cytoplasmic RRE-chloramphenicol acetyltransferase mRNAs, indicating that Sam68 and KH proteins may have been involved in the stability or export of unspliced RNA. The increase in Rev activity was sensitive to leptomycin B, but not to olomoucine, indicating that the effect of SLM-1, SLM-2, QKI-5, QKI-6, and QKI-7 is exerted through a CRM-1-dependent mRNA export pathway. Thus, KH family proteins play an important role in the post-transcriptional regulation of HIV.
