DNA point mutation detection based on DNA ligase reaction and nano-Au amplification: a piezoelectric approach.

A novel piezoelectric method for DNA point mutation detection based on DNA ligase reaction and nano-Au-amplified DNA probes is proposed. A capture probe was designed with the potential point mutation site located at the 3' end and a thiol group at the 5' end to be immobilized on the gold electrode surface of quartz crystal microbalance (QCM). Successive hybridization with the target DNA and detection probe of nano-Au-labeled DNA forms a double-strand DNA (dsDNA). After the DNA ligase reaction and denaturing at an elevated temperature, the QCM frequency would revert to the original value for the target with single-base mismatch, whereas a reduced frequency response would be obtained for the case of the perfect match target. In this way, the purpose of point mutation discrimination could be achieved. The current approach is demonstrated with the identification of a single-base mutation in artificial codon CD17 of the beta-thalassemia gene, and the wild type and mutant type were discriminated successfully. The scanning electron microscope (SEM) image showing that plenty of gold nanoparticles remained on the electrode surface demonstrated that the nano-Au label served as an efficient signal amplification agent in QCM assay. A detection limit of 2.6 x 10(-9)mol/L of oligonucleotides was achieved. Owing to its ease of operation and low detection limit, it is expected that the proposed procedure may hold great promise in both research-based and clinical genomic assays.

Use of pseudo-sample extraction and the projection technique to estimate the chemical rank of three-way data arrays.

Determining the rank of a trilinear data array is a first step in subsequent trilinear component decomposition. Different from estimating the rank of bilinear data, it is more difficult to decide the significant number of component to fit the trilinear decompositions exactly. General methods of rank estimation utilize the information contained in the singular values but ignore information from eigenvectors. In this paper, a rank estimating method specifically for trilinear data arrays is proposed. It uses the idea of direct trilinear decomposition (DTLD) to compress the cube matrix into two pseudo sample matrices which are then decomposed by singular value decomposition. Two eigenvectors combined with the projection technique are used to estimate the rank of trilinear data arrays. Simulated trilinear data arrays with homoscedastic and heteroscedastic noise, different noise levels, high collinearity, and real three-way data arrays have been used to illustrate the feasibility of the proposed method. Compared with other factor-determining methods, for example use of the factor indication function (IND), residual percentage variance (RPV), and the two-mode subspace comparison approach (TMSC), the results showed that the new method can give more reliable answers under the different conditions applied.

Carbon nanotube/cobalt hexacyanoferrate nanoparticle-biopolymer system for the fabrication of biosensors.

Cobalt hexacyanoferrate nanoparticles (CoNP) can be easily prepared by mixing hexacyanoferrate and cobalt chloride solution at room temperature. The nanoparticles were solubilized in aqueous solution of a biopolymer chitosan (CHIT). With the introduction of carbon nanotubes (CNT), the CoNP-CNT-CHIT system formed shows synergy between CNT and CoNP with the significant improvement of redox activity of CoNP due to the excellent electron-transfer ability of CNT. The CoNP-CNT-CHIT film modified glassy carbon electrode allows low potential detection of hydrogen peroxide with high sensitivity and fast response time. In particular, with the introduction of CNT, it amplified the H2O2 sensitivity by approximately 70 times compared to film of CoNP-CHIT. With the immobilization of glucose oxidase onto the electrode surface using glutaric dialdehyde, a biosensor that responds sensitively to glucose has been constructed. In pH 6.98 phosphate buffer, interference free determination of glucose has been realized at -0.2V versus saturated calomel electrode (SCE) with a linear range from 0.01 to 10 mM and response time<10s. The detection limit was 5 microM glucose (S/N=3).

Amperometric biosensor for choline based on layer-by-layer assembled functionalized carbon nanotube and polyaniline multilayer film.

Conducting polymer film was prepared by electrochemical polymerization of aniline. Multiwalled carbon nanotubes (MWNTs) were treated with a mixture of concentrated sulfuric and nitric acid to introduce carboxylic acid groups to the nanotubes. By using the layer-by-layer method, homogeneous and stable MWNTs and polyaniline (PANI) multilayer films were alternately assembled on glassy carbon (GC) electrodes. Conducting polymer of PANI had three main functions: (i) excellent antiinterference ability, (ii) protection ability in favor of increasing the amount of the MWNTs immobilized on GC electrodes, and (iii) superior transducing ability. The protection effect of PANI film and the electrostatic interaction between positively charged PANI and negatively charged MWNTs both attributed to immobilizing abundant MWNTs stably, thereby enhancing the catalytic activity. The layer-by-layer assembled MWNTs and PANI-modified GC electrodes offered a significant decrease in the overvoltage for the H2O2 and were shown to be excellent amperometric sensors for H2O2 from +0.2V over a wide range of concentrations. As an application example, by linking choline oxidase (CHOD), an amplified biosensor toward choline was prepared. The choline biosensor exhibited a linear response range of 1x10(-6) to 2x10(-3) M with a correlation coefficient of 0.997, and the response time and detection limit (S/N=3) were determined to be 3 s and 0.3 microM, respectively. The antiinterference biosensor displays a rapid response and an expanded linear response range as well as excellent reproducibility and stability.

Raman mapping and in situ SERS spectroelectrochemical studies of 6-mercaptopurine SAMs on the gold electrode.

The self-assembled monolayers (SAMs) of 6-mercaptopurine (6MP) were formed at the roughened polycrystalline gold surfaces in acid and alkaline media. The time-dependent Raman mapping spectral analysis in conjunction with the quantum calculations for the vibrational modes using ab initio BLYP/6-31G method suggested that both of the resulted 6MP SAMs adopted the same adsorption mode through the S atom of pyrimidine moiety and the N7 atom of the imidazole moiety anchoring the gold surface in a vertical way. The in situ surface-enhanced Raman scattering spectroelectrochemical experiment was conducted to examine the stability of the SAMs at various bias potentials. It was found that the detaching process of the 6MP SAMs from the surface involved one electron reduction as the voltage was applied at ca. 0.7 V vs a standard calomel electrode.

Chemometric resolution of ATR-IR spectra data for polycondensation reaction of bis(hydroxyethylterephthalate) with a combination of self-modeling curve resolution (SMCR) and local rank analysis.

Self-modeling curve resolution (SMCR) methods, simple-to-use interactive self-modeling mixture analysis (SIMPLISMA) and alternating least squares (ALS) were used to calculate pure concentration profiles and pure spectra for the two-way spectral data collected during the on-line polycondensation reaction of bis(hydroxyethylterephthalate) with an ATR-FT-IR spectrometer. In order to improve the resolution results, SIMPLISMA was combined with local rank analysis method, fixed size moving window evolving factor analysis (FSMWEFA) to search for selective regions of various components and then look for the purest wavenumber variables in the selective regions. Such combination allows more accurate determination of the number of chemical components in the reaction system and the calculations of more accurate concentration profiles and spectra.

Dynamics of a ferroelectric liquid crystal with a naphthalene ring during electric-field-induced switching studied by time-resolved infrared spectroscopy combined with two-dimensional correlation spectroscopy.

The dynamics of a ferroelectric liquid crystal with a naphthalene ring (FLC-3) during the electric-field-induced switching between two surface-stabilized ferroelectric liquid crystal states were investigated by time-resolved infrared (IR) spectroscopy combined with two-dimensional (2D) correlation spectroscopy. Time-resolved IR spectra of FLC-3 in a planar-aligned cell were measured as a function of the polarization angle range from 0 degree to 180 degrees under a rectangular electric field of +/- 40 V with a 5 kHz frequency in the smectic-C* (Sm-C*) phase at 137 degrees C. From these spectra we explore details about the reorientation process of the alkyl chains, the core, and the large C=O dipole moments of FLC-3 at all the delay times. The 2D correlation spectroscopy was applied to the polarization-angle-dependent spectra for different delay times and to the time-resolved spectra at certain polarization angles to reveal the relative orientation of the C=O groups and the core moiety during the electric-field-induced switching. It was found from the present study that the relative orientation of the C=O groups and the core remains unchanged during the initial period of the reorientation, while it is reversed at a certain moment and then kept unchanged again. Moreover, the alkyl chains, C=O groups, and core moieties posses different dynamics during the fast course of electric-field-induced switching by analyzing time-resolved spectra.

Using the fluorescence spot test for neonatal screening of G6PD deficiency.

To establish the neonatal screening method of glucose-6 phosphate dehydrogenase (G6PD) deficiency, G6PD activity was measured using the fluorescence spot test (FST) using dried blood samples on filter paper. The G6PD/6PGD rate test of venous blood samples was further performed for confirmation. The positive G6PD deficiency rate was 4.2% and its detection rates were 3.7% for all neonates and 5.2% only for male newborns when FST was used for neonatal screening. Conformation rates by use of G6PD/ 6PGD ratio test for G6PD deficiency were 86.8% and 100% particularly in the severely deficient groups. Both sensitivity and specificity were very high in the severely deficient groups. FST can be used in neonatal screening of G6PD deficiency because of its high accuracy, applicability, and simplicity. Moreover, a high volume of dried blood samples on filter paper can be tested quickly. It is very favorable to diagnose and treat G6PD deficiency early in high incidence districts.

A survey for the incidence of phenylketonuria in Guangdong, China.

A multicenter cooperative investigated the incidence of Phenylketonuria (PKU) in the central, southern and western areas of Guangdong province and its surrounding districts. Tests to measure phenylalanine (Phe) on dried blood spots on filter paper cards used BIA and thefluorescence assay. Four hundred sixty-one thousand eight hundred five (461,805) newborns were screened and 14 cases of persistent hyperphenylalaninemia (PHPA) were detected. The incidence of PHPA was 1/33,000, including 1/77,000 for classical PKU; 1 in 66,000 for hyperphenylalaninemia (HPA) and 1 in 461,805 for BH4 deficiency. A high variation in PKU incidence in the different districts was observed. The differences may be attributed to the variations in gene frequency of PKU in the different districts and to bioethical consideration especially as regards reproduction in different areas.
