Studies on cellulosic ethanol production for sustainable supply of liquid fuel in China.

As the biggest developing country, China faces a serious challenge in satisfying its need for huge amounts of energy resources, especially for liquid fuel. The Chinese government has recently started a bioethanol project, and has produced about 1 million tons of ethanol fuel from corn and wheat in 2005. As it has the largest population in the world and limited lands for food production, cellulosic ethanol would be a more suitable choice for China. Many research projects in China on biodegradation and biotransformation of lignocellulosics have been carried out. Furthermore, understanding the biodegradation mechanism of lignocellulosics and developing practical processes for ethanol production have been ongoing. After more than 30 years of research, several pilot scale facilities have been set up, and lots of experience has been acquired. However, the calculated production cost of cellulosic ethanol is still higher than that of corn ethanol. To overcome this problem, the biorefinery conception has been introduced into research on lignocellulosics transformation. A corncob biorefinery process has been developed in Shandong University. By combining the cellulase and ethanol production with a xylose-related products production, the total production cost can be reduced. A scale of 50,000-ton/year cellulosic ethanol biorefinery is being planned to be built at Yucheng.

Production of lactic acid from paper sludge by simultaneous saccharification and fermentation.

Production of lactic acid from paper sludge has been performed by simultaneous saccharification and fermentation (SSF). The SSF process design was based on experimental data obtained from cellulose hydrolysis and fermentation. The SSF process was employed to avoid an excessively dense solution when the sludge content of the feed was higher than 15%; this is one of several benefits of SSF. The enzyme system used for hydrolysis of paper sludge for production of glucose was optimized. CMCase and beta-glucosidase with activities of 2.5 and 10 U mL(-1), respectively, were found to be optimum for hydrolyzing 5% sludge. In batch SSF 16 g L(-1) lactic acid was produced from 5% paper sludge with an yield of 80%. Paper sludge which served as a feed seemed to have a buffering effect during SSF, probably because of the inorganic ash component in the sludge. The final product concentration by SSF was observed to be limited by the cellulose content of the system, which can probably be resolved by intermittent feeding of the paper sludge. SSF of paper sludge fed in batch mode, with intermittent feeding, produced lactic acid at 162 g L(-1), with a yield of 74% and a productivity of 1.4 g L(-1) h(-1). The lactic acid production performance of the modified bioreactor improved after removal of indigestible solid materials from the upper compartment, which enabled the feed of paper sludge to be increased. A mathematical model is described which predicts glucose and subsequent lactic acid production on the basis of the rate expressions of each step of the SSF process. Saccharification kinetics were determined by experiments on enzymatic cellulose hydrolysis, by use of a Michaelis-Menten equation; growth kinetics of L. rhamnosus were determined by use of a Monod expression which incorporated lactic acid inhibition. The kinetic model is expected to predict the performance of the SSF process. For further use of the lactic acid, i.e. polylactic acid, it must be recovered and purified. Results from application of the simulated moving-bed (SMB) process for separation of lactic acid and acetic acid are given, as are several methods of lactic acid purification.

[RAPD analysis of four species of Cuscuta in Shandong Province].

OBJECTIVE: To explore the genome difference of four species of Cuscuta in different hosts. METHOD: RAPD was used by 50 primers. RESULT: Four species of genus Cuscuta can be identified by 8 primers. Both Cuscuta chinensis and C. australis from Subg. Grammica had 3 bands whose molecular weights were 1.3 kb, 1.45 kb and 1.53 kb respectively. C. japonica and C. lupuliformis from Subg. Monogyna had a 1.48 kb specific band. Cuscuta of same subgenus had similar RAPD result and close genetic relationship. Same species of Cuscuta in different hosts showed DNA polymorphism. It indicated that hosts can affect genome of Cuscuta to some extent. CONCLUSION: RAPD can be used to identify the species of Cuscuta or same Cuscuta in different hosts.

[Study on xylose fermentation by Neurospora crassa].

The influence of oxygen limitation and medium initial pH on xylose fermentation by Neurospora crassa AS 3.1602 was investigated. N. crassa AS 3.1602 has high ability of xylose fermentation producing ethanol and xylitol. Oxygen limitation has big influence on ethanol and xylitol production. The maximum conversion rate of ethanol was 63.2% obtained under semi-aerobic conditions. The maximum conversion rate of xylitol was 31.8% obtained under micro-aerobic conditions. Xylitol accumulation is behind the accumulation of ethanol. The optimal pH is 5.0 for ethanol fermentation, and 4.0 for xylitol fermentation, respectively. Xylose fermentation is greatly inhibited at high pH. The yields of ethanol and xylitol are greatly influenced by the initial concentration of xylose. The addition of glucose inhibits both the xylose utilization and the ethanol and xylitol production.
