[Structural characteristics of infrared spectra for polyaluminum chloride in enhanced reactions of modification].

As an important preparation method of Inorganic Polymer Flocculants (IPFs) with all-round performance, the use of enhanced reactions of modification for Polyaluminum Chlorides (PAC) is a trend of application. In this paper, auctorial-made multicore oligomers as a complexing agent, polysilicate as a crosslinking agent and polyacrylamide as a compounding agent are respectively used to enhance PAC and prepared three kinds of IPFs, named CO-PAC, CR-PAC and PAM-PAC. The Fourier Transform Infrared spectra (FTIR) of the three flocculants show that there have been found new chemical bonds beyond Al-OH in their structures, and each characteristic band of primary group in PAC has a band shift to a lower frequency due to its structural aberration. These evidences suggested that enhanced reactions of modification for PAC are not at all a simple process of physical mix, but a chemical reaction to form other structural characteristics of inorganic polymers. The excellent coagulation behavior of these new flocculants depends mainly on the chemical actions, but there are a little physical effects between the modifiers and PAC.

[A DRIFTS study of SO2 oxidation on the surface of CaCO3 particles].

CaCO3 is an important component of mineral dust aerosols in the atmosphere, but its role in atmospheric heterogeneous chemistry is still unclear. In this paper, the oxidation of SO2 on CaCO3 particles was studied using diffuse reflectance infrared Fourier transform spectroscopy (DRIFTS). Formation of sulfite and sulfate was identified on the surface, and the role of O3 on the oxidation process is determined by comparing the experiments in different conditions. DRIFTS is proved as a useful tool for studying heterogeneous reactions on CaCO3 particles. The results show that in the presence of O3, SO2 can be oxidized into sulfate on the surface of CaCO3 particles. A two-step mechanism that involves adsorption of SO2 followed by oxidation is presented.

Expression of nephrin, podocin, alpha-actinin, and WT1 in children with nephrotic syndrome.

Recently, nephrin, podocin, alpha-actinin, and WT1, which are located at the slit diaphragm and expressed by the podocyte, were found to be causative in congenital/familial nephrotic syndrome (NS), but their role in acquired NS remains unclear. We studied their expression in NS with the aim of disclosing their possible role in the development of proteinuria. Immunofluorescence, confocal microscopy, and image analysis were used to study the expression and the distribution in 19 children with primary NS, 9 with isolated hematuria, and 9 controls. All the children with NS presented with heavy proteinuria and foot process effacement was identified by electron microscopy. No proteinuria and foot process effacement was seen in the group with hematuria. A dramatic decrease of podocin expression was found in NS (86.66+/-22.74) compared with control groups ( P=0.014). Furthermore, we also found the pattern of distribution of nephrin, podocin, and alpha-actinin changed in children with NS. In conclusion, a dramatic decrease of podocin expression and abnormal distribution of nephrin, podocin, and alpha-actinin were found in children with NS. No differences were found in children with isolated hematuria, suggesting involvement of these molecules in the development of proteinuria in primary NS.

Development of an oral DNA vaccine against MG7-Ag of gastric cancer using attenuated salmonella typhimurium as carrier.

AIM: To develop an oral DNA vaccine against gastric cancer and evaluate its efficacy in mice. METHODS: The genes of the MG7-Ag mimotope and a universal Th epitope (Pan-DR epitope, PADRE) were included in the PCR primers. By PCR, the fusion gene of the two epitopes was amplified. The fusion gene was confirmed by sequencing and was then cloned into pcDNA3.1(+) plasmid. The pcDNA3.1 (+)-MG7/PADRE was used to transfect an attenuated Salmonella typhimurium. C57BL/6 mice were orally immunized with 1X10(8) cfu Salmonella transfectants. Salmonella harboring the empty pcDNA3.1(+) plasmid and phosphate buffer saline (PBS) were used as negative controls. At the 6th week, serum titer of MG7-Ag specific antibody was detected by ELISA. At the 8th week cellular immunity was detected by an unprimed proliferation test of the spleenocytes by using a ((3)H)-thymidine incorporation assay. Ehrlich ascites carcinoma cells expressing MG7-Ag were used as a model in tumor challenge assay to evaluate the protective effect of the vaccine. RESULTS: Serum titer of antibody against MG7-Ag was significantly higher in mice immunized with the vaccine than that in control groups (0.841 vs 0.347, P<0.01; 0.841 vs 0.298, P<0.01), while in vitro unprimed proliferation assay of the spleenocytes showed no statistical difference between those three groups. Two weeks after tumor challenge, 2 in 7 immunized mice were tumor free, while all the mice in the control groups showed tumor formation. CONCLUSION: Oral DNA vaccine against the MG7-Ag momitope of gastric cancer is immunogenic. It can induce significant humoral immunity against tumor in mice, and the vaccine has partially protective effects.

[Analysis of glucocorticoid receptor gene polymorphisms by denaturing high-performance liquid chromatography].

OBJECTIVE: To detect polymorphisms of glucocorticoid receptor gene (NR3C1) in Chinese population with the technique of denaturing high-performance liquid chromatography (DHPLC). METHODS: Genomic DNA was isolated from peripheral blood of 2 healthy volunteer doctors and umbilical cord blood of 62 normal neonates. All the NR3C1-coding exons along with their flanking intron portions were amplified by polymerase chain reaction (PCR). For polymorphism screening, PCR products were analyzed by DHPLC. DNA fragments with aberrant elution profiles revealed by DHPLC were re-amplified and sequenced directly. RESULTS: Eight polymorphisms were identified by DHPLC and sequencing analysis. Among them, 5 (1374A > G, IVSG-68_IVSG-63delAAAAAA, 2193T > G, IVSH-9C > G, 2382C > T) were novel and 3 (1896C > T, 2166C > T, 2430T > C) were reported previously. Two groups of polymorphisms ([1374A > G + IVSG-68_IVSG-63delAAAAAA + IVSH-9C > G + 2382C > T] and [1896C > T + 2166C > T + 2430T > C]) showed to have the complete linkage disequilibrium and were identified as 2 different haplotypes, which was first reported. The frequencies of the 2 novel haplotypes were 3.1% and 14.1%, respectively. Another polymorphism (2193T > G) was relatively rare. CONCLUSION: DHPLC is a rapid and reliable method for polymorphism screening. Eight polymorphisms in the NR3C1 gene are detected in Chinese population with the technique of DHPLC, of which two haplotypes have been identified for the first time.

[Organic acids conversion in methanogenic-phase reactor of the two-phase anaerobic process].

Experiments on organic acids conversion in methanogenic-phase of the two-phase anaerobic process were conducted. The results showed that when the acidogenic-phase was in mixed acids fermentation and its load was 41.5 kg/(m3-d), the load of methanogenic-phase was 6.05 kg/(m3.d), the substrate conversion velocity of bacteria inhabited at the same height of UASB reactor as follows: acetic acid > ethanol > butyric acid > propionic acid; ethanol-utilized microbial had the same pH range as methanogenic bacteria; ethanol-type fermentation was the optimal acidogenci-type fermentation for the two-phase anaerobic process; the conversion of acetic acid was rather high, but the others organic acids will be convert to acetic acid make it the rate-limiting step for anaerobic degradation.

[Analysis on association of glucocorticoid receptor gene polymorphism with steroid-resistance in idiopathic nephrotic syndrome of children].

OBJECTIVE: The nephrotic syndrome is defined by heavy proteinuria, edema, hypoalbuminemia, and hyperlipidemia. Idiopathic nephrotic syndrome (INS) mainly occurs in children, which is generally treated with glucocorticoids. The majority of patients are steroid-sensitive (SSINS) while steroid-resistance occurs in a subset of NS children (SRINS). Although intensive efforts have been undertaken to study the associations between SRINS and renal pathological changes, pharmacokinetics, and the GR density and binding affinity, the mechanisms underlying steroid-resistance are still not elucidated entirely. The authors hypothesized that it might be associated with polymorphisms in the glucocorticoid receptor gene (NR3C1). The study aimed to screen the NR3C1 gene for polymorphisms in genomic DNA samples from SRINS, SSINS children and control group, and to analyze the association of the polymorphisms in the NR3C1 gene and SRINS of children. METHODS: Totally 39 SRINS and 67 SSINS children (81 males and 25 females with the mean age of 7 years) were involved in the study. Umbilical cord blood of 62 normal neonates and peripheral blood of 2 healthy volunteers were selected as controls. Genomic DNA was isolated from peripheral blood lymphocytes of all subjects. All the NR3C1-coding exons and intron-flanking portions were amplified by polymerase chain reaction (PCR). For polymorphism screen, PCR products were analyzed by denaturing high performance liquid chromatography (DHPLC). DNA fragments with aberrant elution profiles were re-amplified and sequenced directly. RESULTS: Twelve aberrant elution profiles were identified with DHPLC in SRINS, SSINS and controls. Among them, 6 previously reported polymorphisms and 6 novel polymorphisms were confirmed by sequencing (198G > A, 200G > A, IVSD-16G > T, 1896C > T, 2166C > T, 2430T > C; novel, 1206C > T, 1374A > G, IVSG-68_IVSG-63delAAAAAA, 2193T > G, IVSH-9C > G, 2382C > T), and 3 groups of SNPs were in complete linkage disequilibrium, which resulted in 3 different haplotypes ([198G > A + 200G > A], [1374A > G + IVSG-68_IVSG-63delAAAAAA + IVSH-9C > G + 2382C > T], [1896C > T + 2166C > T + 2430T > C]). The last two genotypes were first reported. The genotype frequencies of the 2 novel haplotypes were 10.3% vs 1.5% in SRINS and SSINS, and 15.4% vs 7.5% in SRINS and SSINS, respectively. Other polymorphisms were relatively rare detectable both in patients and controls. CONCLUSION: Twelve polymorphisms in the NR3C1 gene were detected with the technique of DHPLC, of which six polymorphisms were identified at the first time. Two types of newly found haplotypes were associated with steroid-resistant idiopathic nephrotic syndrome of children, which might be responsible for steroid-resistance in partial idiopathic nephrotic syndrome of children.

Characterizing ionic liquids on the basis of multiple solvation interactions.

Room-temperature ionic liquids (RTILs) are useful in many chemical applications. Recent publications have attempted to determine the polarity of RTILs using empirical solvent polarity scales. The results have indicated that most RTILs have similar polarities. Nevertheless, RTILs are capable of behaving quite differently when used as solvents in organic synthesis, matrixes in matrix-assisted laser desorption/ionization (MALDI) mass spectrometry, liquid-liquid extraction, and as stationary phases in gas chromatography. The work presented in this study uses a linear free energy approach to characterize 17 RTILs on the basis of their distinct multiple solvation interactions with probe solute molecules. This model provides data that can be used to help identify the interactions and properties that are important for specific chemical applications.

Phenotypic and genotypic features of Alport syndrome in Chinese children.

Chinese Alport syndrome (AS) was analyzed in 44 unrelated patients who were screened for mutations in the COL4A5 gene by polymerase chain reaction (PCR)-single-strand conformation polymorphism analysis or PCR direct sequencing in 30 of the 44 patients. The clinical data showed that all patients had hematuria; 25 of 29 male patients (86%) and 9 of 15 female patients (60%) had proteinuria; 11 of 29 male patients (38%) and 1 of 15 female patients (7%) had nephrotic-level proteinuria; 10 of 21 male patients examined (48%) and 1 of 12 female patients examined (8%) had hearing abnormalities. Renal function remained normal despite hearing abnormalities, and ocular lesions occurred in 10%. Among 30 of 44 patients who had a family history of end-stage renal disease (ESRD), 80% (24/30) belonged to X-linked juvenile kindreds, and 20% (6/30) patients to adult kindreds. Of the 44 patients, 14 did not have a family history of ESRD, while 11 of 14 patients diagnosed with X-linked AS did. DNA analysis revealed four missense mutations, two silent mutations, one substitution, and one in-frame deletion. PCR along with Southern hybridization analysis revealed a large deletion of the paired COL4A5 and COL4A6 genes. Chinese AS patients were characterized clinically with hematuria, heavy proteinuria, and more juvenile forms. Mutations in these patients were usually small mutations, while a large deletion involving the 5' part of both COL4A5 and COL4A6 genes was identified.

Use of ionic liquids for pi-conjugated polymer electrochemical devices.

pi-Conjugated polymers that are electrochemically cycled in ionic liquids have enhanced lifetimes without failure (up to 1 million cycles) and fast cycle switching speeds (100 ms). We report results for electrochemical mechanical actuators, electrochromic windows, and numeric displays made from three types of pi-conjugated polymers: polyaniline, polypyrrole, and polythiophene. Experiments were performed under ambient conditions, yet the polymers showed negligible loss in electroactivity. These performance advantages were obtained by using environmentally stable, room-temperature ionic liquids composed of 1-butyl-3-methyl imidazolium cations together with anions such as tetrafluoroborate or hexafluorophosphate.

Separation of racemic sulfoxides and sulfinate esters on four derivatized cyclodextrin chiral stationary phases using capillary gas chromatography.

The separation of 17 chiral sulfoxides and eight chiral sulfinate esters by gas chromatography (GC) on four derivatized cyclodextrin chiral stationary phases (CSPs) (Chiraldex G-TA, G-BP, G-PN, B-DM) is presented. Many of these compounds are structural isomers or part of a homologous series. Differences in enantioselectivity of the methyl phenyl sulfoxide isomers on the derivatized gamma cyclodextrin and the heptakis 2,6-di-O-methyl-beta-cyclodextrin (i.e. B-DM) CSPs are discussed. Under the conditions of this study, the molecular mass cut-off for the GC separation of these compounds was approximately 230. Compounds of higher molecular mass were not eluted from the CSPs at reasonable times and temperatures, but these higher molecular mass enantiomers can be separated by liquid chromatography and capillary electrophoresis. The enantiomeric separation and elution order of a sulfinate ester containing two stereogenic centers as well as 15 chiral sulfoxides is presented. The G-TA and B-DM CSPs generally gave opposite elution orders for most of the compounds studied.

Direct effect of croton oil on intestinal epithelial cells and colonic smooth muscle cells.

AIM: To investigate the direct effect of croton oil (CO) on human intestinal epithelial cell (HIEC) and guinea pig colonic smooth muscle cells in vitro. METHODS: Growth curves of HIEC were drawn by MTT colorimetry. The dynamics of cell proliferation was analyzed with flow cytometry, and morphological changes were observed under light and electron microscopy after long-term (6 weeks) treatment with CO. Expression of cyclooxygenase-2 (COX-2) mRNA was detected by dot blot in HIEC treated with CO. Genes related to CO were screened by DD-PCR, and the direct effect of CO on the contractility of isolated guinea pig colonic smooth muscle cells was observed. RESULTS: High concentration (20-40 mg x L(-1)) CO inhibited cell growth significantly (1, 3, 5, 7d OD sequence: (20 mg x L(-1)) 0.040+/-0.003, 0.081+/-0.012, 0.147+/-0.022,0.024+/-0.016; (40 mg x L(-1)) 0.033+/-0.044, 0.056+/-0.012, 0.104+/-0.010, 0.189+/-0.006; OD control 0.031+/-0.008, 0.096+/-0.012, 0.173+/-0.009, 0.300+/-0.016, P<0.01), which appeared to be related directly to the dosage. Compared with the control, the fraction number of cells in G1 phase decreased from 0.60 to 0.58, while that in S phase increased from 0.30 to 0.34 and DNA index also increased after 6 weeks of treatment with CO (the dosage was increased gradually from 4 to 40 mg x L(-1)). Light microscopic observation revealed that cells had karyomegaly, less plasma and karyoplasm lopsidedness. Electron microscopy also showed an increase in cell proliferation and in the quantity of abnormal nuclei with pathologic mitosis. Expression of COX-2 mRNA decreased significantly in HIEC treated with CO. Thirteen differential cDNA fragments were cloned from HIEC treated with CO, one of which was 100 percent homologous with human mitochondrial cytochrome C oxidase subunit II. The length of isolated guinea pig colonic smooth muscle cells was significantly shortened after treatment with CO (P<0.05). CONCLUSION: At a high CO concentration (>20 mg x L(-1)), cell growth and proliferation are inhibited in a dosage-dependent manner. Increase in cell proliferation and in malignant conversion of the cellular phenotype is observed in cells cultured chronically with CO. COX-2 mRNA expression decreases significantly, while human mitochondrial cytochrome C oxidase subunit IImRNA expression increases significantly in HIEC treated with CO. CO also has a direct effect on the contractility of Guinea pig colonic smooth muscle cells.

Screening and identification of proteins mediating senna induced gastrointestinal motility enhancement in mouse colon.

AIM: To isolate the proteins involved in pharmacologic action of senna extract (SE) from mouse gastrointestinal tract and to explore the molecular mechanism of gastrointestinal motility change induced by SE. METHODS: SE was administrated to mice by different routes. Gastrointestinal motility of mice was observed using cathartic, gastrointestinal propellant movement experiments and X-ray analysis. Mouse model for gastrointestinal motility enhancement was established through continuous gastric administration of SE at progressively increased dose. At 3 h and week 3, 4, 6 and 10, morphological changes of gastrointestinal tissues were found under light microscope. Ultrastructural changes of intestinal and colonic tissues at week 6 were observed under transmission electron microscope. The colonic proteomic changes in model mice were examined by two-dimension polyacrylamide gel electrophoresis with immobilized pH gradient isoelectric focusing to screen the differentially expressed proteins, and their molecular masses and isoelectric points were determined. Two N-terminal sequences of the samples were also determined by mass spectrometry. RESULTS: SE (0.3g) caused diarrhea after gastric administration in 1-6h and enhanced gastrointestinal propellant (65.1+/-7.5%; 45.8+/-14.6%, P<0.01) in mice, but intramuscular and hypodermic injection had no cathartic effect. X-ray analysis of gastrointestinal motility demonstrated that gastric administration of SE enhanced gastric evacuation and gastrointestinal transferring function. At 3 h and week 3 and 4 after gastric administration of SE, light microscopic examination revealed no apparent change in gastrointestinal mucosal tissues, but transmission electron microscopic examination revealed inflammatory changes in whole layer of intestinal and colonic wall. Twenty differential proteins were detected in the colonic tissues of the model mice by two-dimensional electrophoresis, and the N-terminal amino acid sequences of two proteins were determined. CONCLUSION: SE causes diarrhea and enhances gastrointestinal motility through digestive tract administration. Long-term gastric administration of SE induces inflammatory changes and cell damage in the whole gastrointestinal tract. The differential proteins screened from the colonic tissues of the model mice might mediate the enhancing effect of SE on gastrointestinal motility.

Differential display of vincristine-resistance-related genes in gastric cancer SGC7901 cell.

AIM: To isolate and clone the vincristine-resistance-related genes in gastric cancer SGC7901 cell line and to clarify the multidrug-resistant molecular mechanism of gastric cancer cells. METHODS: The modified differential-display polymerase chain reaction (DD-PCR) was used to examine differences in the mRNA composition of Vincristine-resistant gastric cancer SGC 7901 cells (SGC7901/VCR), induced by vincristine sulfate versus SGC7901cells. The differentially expressed cDNA fragments were confirmed by reverse Northern analysis, sequencing, BLAST analysis and Northern bolt analysis. RESULTS: The DD-PCR identified that 54 cDNA fragments were preferentially expressed in SGC 7901/VCR cells. When these cDNA fragments were analyzed by reverse Northern blot, twenty were reproducibly expressed at high level in SGC7901/VCR. Sequencing and BLAST analysis revealed that seven of the genes were known genes:ADP-ribosylation factor 4, Cytochrome oxidase subunit II, Ss-A/Ro ribonucleoprtein autoantigen 60kd subunit, ribosomal protein S13, galaectin-8 gene, oligophrenin 1 mRNA, ribosomal protein L23 mRNA; thirteen of the genes were unknown genes. The length and abundance of the four unknown genes were further confirmed by Northern blot analysis. CONCLUSION: The twenty differential known and unknown genes may be related to the vincristine-resistant mechanism in human gastric cancer SGC7901 cell line.
