p53 Transgenic mice are highly susceptible to 4-nitroquinoline-1-oxide-induced oral cancer.

In this study, we did a bioassay employing mice with a dominant-negative p53 mutation (p53(Val135/WT)) to assess whether a germ-line p53 mutation predisposed mice toward the development of squamous cell carcinomas (SCC) in the oral cavity. Treatment of the mouse oral cavity with 4-nitroquinoline-1-oxide produced a 66%, 91%, and 20% tumor incidence in the oral cavity, esophagus, and forestomach/stomach, respectively, in p53(Val135/WT) mice. In contrast, only a 25%, 58%, and 4% tumor incidence was observed in oral cavity, esophagus, and forestomach/stomach, respectively, in wild-type littermates (p53(WT/WT)). The most striking difference between p53(Val135/WT) and p53(WT/WT) mice following the carcinogen treatment was the higher prevalence and more rapid development of SSC in p53(Val135/WT) mice than in wild-type mice. To identify the precise genes or pathways involved in these differences during tumor development, we examined gene expression profiles of 4-nitroquinoline-1-oxide-treated normal tongues as well as tongue SCC in p53(Val135/WT) and p53(WT/WT) mice. Microarray and GenMAPP analysis revealed that dominant-negative p53 ((135)Valp53) affects several cellular processes involved in SCC development. Affected processes included apoptosis and cell cycle arrest pathways, which were modulated in both tumor and normal epithelium. These results showed that reduction of p53-dependent apoptosis and increases in cell proliferation might contribute to the observed increase in oral cavity and gastroesophageal malignancies in p53(Val135/WT) mice as well as to the more rapid growth and progression of tumors.

Cell proliferation in ependymal/subventricular zone and nNOS expression following focal cerebral ischemia in adult rats.

Neuronal nitric oxide synthase (nNOS) regulates neurogenesis in normal developing brain, but the role of nNOS in neurogenesis in the ischemic brain remains unclear. To investigate the temporal and spatial relationship between cell proliferation of the ependymal/subventricular zone (SVZ), a principal neuroproliferative region in the adult brain, and nNOS expression, the male Sprague-Dawley rats weighing 250-350 g were used. The focal cerebral ischemia was induced by middle cerebral artery occlusion (MCAO). 10 microl of 0.2% fluorescence dye DiI was injected into the right lateral ventricle to prelabel ependymal/subventricular zone cells before ischemia. The rats were killed immediately after ischemia and days 1, 3, 7, 11, 14, 21 and 28 after ischemia. DiI-labeled cell counting was employed to assess cell proliferation. Immunohistochemistry and grayscale analysis were performed to determine nNOS localization and its quantity in the specific regions. Compared with control, the density of DiI-labeled cells in the ipsilateral ependyma/SVZ was significantly higher at days 1, 3, 7 and 11 after ischemia, whereas the quantity of nNOS expression in the ependyma/SVZ adjacent regions was significantly lower at the above time points. Additionally, nNOS positive cells were largely excluded from SVZ, and their long processes did not enter the ependyma/SVZ. Our results indicate that after focal cerebral ischemia, decreased nNOS expression in the ipsilateral ependymal/SVZ adjacent regions might be related to cell proliferation in the ependymal/SVZ.

Exercise pre-conditioning strengthens brain microvascular integrity in a rat stroke model.

Increasing evidence indicates that physical activity reduces brain damage after stroke. The purpose of this study was to determine whether exercise-induced neuroprotection is associated with improved brain integrity in stroke. Adult male Sprague-Dawley rats (3 months old, n=38) exercised on a treadmill, which required repetitive locomotor movement, for 30 minutes each day for 3 weeks. Then, using an intraluminal filament, stroke was induced by either 2 hours middle cerebral artery (MCA) occlusion followed by 24 or 48 hours of reperfusion. Brain damage was determined by evaluating brain infarction and brain edema, as well as ultrastructural alteration in endothelial-matrix-astrocyte interfaces.Pre-ischemic motor exercise significantly (p<0.01) reduced infarct volume in the frontoparietal cortex and the dorsolateral striatum by 79%. By comparing the percentage difference in brain volume between the right (stroke site) and left hemispheres, we demonstrated a significant (p<0.01) reduction in brain edema associated with reduced infarct volume in a 3 week exercise group (Group 1, n=10) and a 3 week exercise plus 3 week rest group (Group 2, n=10). Edema in cortex and striatum was 19 +/- 4% without exercise pre-conditioning (n=10), in contrast to 5 +/- 3% (Group 1) or 6 +/- 4% (Group 2). The thickness of the basal lamina was enhanced by exercise. In ischemic rats without pre-exercise, alterations in microvessel ultrastructure with decreased luminal area, parenchymal edema and swollen astrocyte end-feet, as well as an abnormally thin basal lamina were observed. In contrast, exercise pre-conditioning significantly reduced the ischemic alterations, decreasing brain edema and increasing basal lamina thickness. This study suggests that exercise pre-conditioning reduces brain injury by decreasing cerebral permeability and enhancing brain integrity after stroke. This exercise-induced endogenous neuroprotection could be an effective strategy to ameliorate ischemic brain injury from stroke.

MUC1 cytoplasmic tail: a potential therapeutic target for ovarian carcinoma.

Ovarian cancer is often a lethal disease, since the occult progression of the tumor within the peritoneal cavity results in late diagnosis and treatment failure. The identification of molecular events specific to metastasis is critical for the development of effective therapies. MUC1 is aberrantly overexpressed by most ovarian cancer and regarded as a molecular target for ovarian cancer. This review focuses on the latest advances regarding a signaling region in the MUC1 C-terminal subunit-mediated c-Src signaling pathways in malignant transformation, invasion and metastasis. Disruption of MUC1-C-terminal subunit-associated c-Src signaling by targeting the specific sites might represent a novel immunotherapeutic approach for the treatment of ovarian cancer.

Cerebral angiogenesis and expression of angiogenic factors in aging rats after exercise.

The effect that exercise has on angiogenesis in the aging rat is unknown. We initiated this study with the intent to determine if exercise could induce angiogenesis in aging rats, as well as in adult rats reported previously. The markers we used to determine our endpoint were vascular endothelial growth factor (VEGF) and angiopoietin 1 and 2, as well as vascular density. Aged (22 month old) female Fisher 344 rats (n=16) were exercised on a treadmill 30 minutes each day for 3 weeks, or housed as non-exercised controls for the same duration. At the end of the exercise protocol, a significant (p<0.01) increase in the density of microvessels was found within the cerebral vasculature of the rats. Exercise was also associated with a significantly (p<0.01) increased mRNA expression of angiopoietin 1 and 2 in the aged cohort of rats. A mild but significant (p<0.01) increase in the four isoforms of VEGF mRNA (120, 144, 164, 188) were observed, with VEGF120 and VEGF144 being more markedly up-regulated than the other two. VEGF protein expression was also significantly (p<0.01) increased. This study demonstrates that angiogenesis can be induced in aging rats via exercise. The induced angiogenesis was associated with overexpression of angiogenic factors. These results support the hypothesis that an angiogenic response to chronic physical exercise is maintained with aging.

Exercise preconditioning reduces brain damage and inhibits TNF-alpha receptor expression after hypoxia/reoxygenation: an in vivo and in vitro study.

Exercise reduces ischemia and reperfusion injury in rat stroke models. We investigated whether gradual increases in tumor necrosis factor-alpha (TNF-alpha) reported during exercise down-regulates expression of TNF-alpha receptors I and II (TNFRI and II) in stroke, leading to reduced brain damage. Adult male Sprague Dawley rats were subjected to 30 minutes of exercise on a treadmill each day for 3 weeks. Then, stroke was induced by a 2-hour middle cerebral artery (MCA) occlusion using an intra-luminal filament. Expressions of TNFRI and II mRNA in the brain were detected using a real-time reverse transcriptase-polymerase chain reaction (RT-PCR). Protein expressions of TNFRI and II were determined by enzyme-linked immunoabsorbant assay (ELISA) in serum and brain homogenates. Spatial distribution of TNF-alpha receptors in brain regions was determined with immunocytochemistry. In human umbilical vein endothelial cells (HUVEC), we addressed the causal effect of TNF-alpha pretreatment on TNF I and II expression using ELISA and real-time PCR. In exercised rats after stroke, brain infarct was significantly (p<0.01) reduced in the entire MCA supplied regions, associated with a mild expression of TNFRI and II mRNA and protein. The TNF-alpha receptors were restricted to the ischemic core. In contrast, a robust expression of TNFRI and II molecules was found in non-exercised rats subjected to similar ischemia/reperfusion insults. An in vitro study revealed a causal link between TNF-alpha pretreatment and reduced cellular expression of TNF-alpha receptors under hypoxic/reoxygenated conditions. Our results suggest that reduced-brain damage in ischemic rats after exercise preconditioning may be attributable to the reduced expression of TNF-alpha receptors. Chronically increased TNF-alpha expression was also found to reduce TNFI and II responding to acute ischemia/reperfusion insult.

Gray scale contrast enhancement and quantification in different positions of rabbit liver.

OBJECTIVE: The purpose of this study was to evaluate the difference between central and ventral peripheral positions and the difference between left and right lobes in rabbit liver with gray scale contrast enhancement. METHODS: An in vivo model of perfusion was studied with a sulfur hexafluoride contrast agent and low-mechanical-index, real-time, gray scale harmonic imaging. The contrast agent (0.1 mL/kg body weight) was applied respectively in 10 rabbits by intravenous bolus injection. The time-intensity curve was used to obtain flow-related parameters such as time to enhancement (ET), time to peak intensity (PIT), peak signal intensity (PSI), enhancement duration (ED), and area under the curve (AUC). RESULTS: There was a significant difference in parameters of the time-intensity curve between central and peripheral ventral positions of liver parenchyma (P < .05), except for the ED in the left liver. The ET and PIT were earlier, the PSI higher, the ED longer, and the AUC larger in the central position of parenchyma than in the peripheral position. In addition, the ET and PIT were earlier, the PSI higher, the ED longer, and the AUC larger in the right lobe of liver parenchyma than in the left lobe. There was a significant difference in parameters of the time-intensity curve between the left and right lobes of liver parenchyma (P < .05), except for the ET of the peripheral position. CONCLUSIONS: Flow parameters are different between central and ventral peripheral positions and between left and right lobes of hepatic parenchyma.

[Establishment of an undifferential orchitis model in rats and changes of spermatogenic epithelium].

OBJECTIVE: To explore the changes of rat testicular spermatogenic epithelium stimulated by bacterial lipopolysaccharide (LPS) in vivo. METHODS: Twenty Wistar rats were divided into two groups: control group and experimental group. The control group was treated with pyrogen-free saline (1 ml/kg) and the experimental group was injected ip with saline containing LPS (1 mg/kg) once every two days. Two groups were operated after ten days in order to investigate the testicular pathological changes by HE staining and the expression of proliferating cell nuclear antigen( PCNA), alpha-catenin in spermatogenic epithelium by immunohistochemistry assay. RESULTS: The testes of the experimental group showed inflammatory changes. The positive expression of PCNA in seminiferous epithelium was significantly lower than that of control group. The number of positive cells in every seminiferous, in which only spermatogonia were stained in experimental group were 59 +/- 5 and it showed significant decrease compared with the control (P < 0.01). Furthermore, the percentage of such seminiferous tubules was 0.673 +/- 0.054 and increased apparently (P < 0.01). The expression of alpha-catenin in testicular tissue of the experimental group declined (P < 0.01), and cellular positive granular light density was 0.150 +/- 0.014. CONCLUSION: The ability of spermatogonium proliferation and the function of conglutination of cells under inflammatory condition of the testes declined, which may be one of the etiologies of male infertility.

[Effect of electroacupuncture on myoelectric activity of Jejunal limb After Roux-en-Y esophagojejunostomy].

OBJECTIVE: To explore the effect of electroacupuncture on myoelectric activity of Jejunal limb after Roux-en-Y esophagojejunostomy. METHODS: Fourteen health young pigs were randomly divided into 2 groups, an experimental group (total gastrectomy and Roux-en-Y esophagojejunostomy was carried out) and a control group (the abdominal cavity was closed after the electrode was placed), 7 pigs in each group. Electroacupunture was given at "Zusanli" (ST 36) in the experimental group. The changes of myoelectrogram of the jejunal limb was investigated. RESULTS: Compared with the control group, the amplitude and the frequency of the slow wave, and the amplitude and incidence rate of the spike potential in the experimental group were changed significantly; the duration of migrating motor complex (MMC) phase III was (2.6 +/- 0.7) minutes in the experimental group, which was significantly shorter than (7.1 +/- 1.1) minutes in the control group. Electroacupuncture did not significantly influence the amplitude and the frequency of the slow wave, but could increased significantly the incidence rate and the amplitude of the spike potential; after electroacupuncture, the duration of MMC phase III was (5.7 +/- 0.9) minutes, which was significantly longer than (2.6 +/- 0.7) minutes before electroacupuncture. CONCLUSION: Electroacupuncture at "Zusanli" (ST 36) can relieve the Roux-en-Y stasis syndrome through influencing myoelectric activity of the jejunum.

[Diagnosis and treatment of deltoid contracture in children].

OBJECTIVE: To reinforce the recognization of deltoid contracture in children and probe its causes, pathomechanism, diagnosis and treatments. METHODS: The Clinical data were summarized and analysed, including etiological factors, clinical situations, examinations and treatments from 9 child patients with deltoid contracture from 1992 to 2004. The 9 child patients were followed up to evaluate therapeutic efficacy. RESULTS: Nine child patients with deltoid contracture included 4 boys and 5 girls, who aged from 5 to 14 years. The main clinical features were presented as abductive deformity, limitation of adduction of the shoulder, winging of the scapula, and dimpling of the skin with a palpable fibrous hard band in deltoid muscle. Four cases had a history of intramuscular injections in deltoid and 1 case was accompanied with bilateral gluteul contracture. Among the 9 patients, 8 cases were misdiagnosed before correct diagnosis and treatment. The misdiagnosis rate was up to 90%. All the cases received the operative treatments to release fibrous bands and were followed up for 8 months to 12 years. Abductive deformity disappeared and functions of the shoulders recovered in all patients, except 1 patient who had a lightly winged scapula. CONCLUSION: Deltoid contracture may be caused by multiple factors, but its clinical features are conductive to correct diagnosis and excellent therapeutic efficacy can be acquired by operation early.

[Effects of nitrite on phosphorus uptake in anaerobic/oxic process].

Abstract:Intermediate products of biological nitrogen removal process, nitrate and nitrite, could be used as electron acceptors for phosphorus removal. This study investigated the effects of nitrite on phosphorus uptake in anaerobic/oxic (A/O) biological phosphorus removal process. The results indicated that in addition to oxygen and nitrate (DPB(Na), Denitrifying Phosphorus removal Bacteria with nitrate as electron acceptor), to some extent, nitrite can also serve as electron acceptor to achieve the nitrite denitrifying phosphorus removal (DPB(Ni)). The quantity and rate of phosphorus uptake of DPBNi, however, were evidently lower than that of DPBN,. The nitrite existed in anoxic reactor made no difference to the quantity and rate of denitrifying phosphorus removal. But it would reduce the consumption of nitrate. Moreover, the data showed that the aerobic phosphate uptake of DPBNi was lower than that of anaerobic phosphorus-released sludge in traditional A/O process. However, there was no much difference between these two kinds of sludge in terms of the total phosphorus uptake quantity and the effluent quality.

[Spectral analysis and photoluminescence properties of 1,5-naphthalene diamine derivative].

In the present paper, 1,5-bis[N-(1-naphthyl)-N-phenyl] naphthalene diamine (NPN) with high purity was synthesized by liquid-phase Ullmann reaction, and high quality film of NPN was formed on cleaned glass substrates by vacuum evaporation. Photoluminescence properties were studied by solvent effects on UV-Vis absorption spectra and fluorescence spectra. In addition the structure of energy band of NPN was studied by cyclic voltammetry, UV-Vis absorption spectra, and fluorescence spectra. The results indicate that the fluorescence emitting is attributed to the charge transfer transition from nitrogen to aromatic ring. The molecules of NPN in its film form "J-aggregate". The HOMO energy and optical gap of NPN are -5.74 and 2.79 eV respectively. NPN film could emit intense blue fluorescence with a peak wavelength at 448.6 nm and a bandwidth of 72.6 nm under UV-Vis excitation at 365 nm.

[Involvement of nitric oxide in regulation of salt stress-induced ABA accumulation in maize seedling].

Possible regulation of salt stress-induced ABA accumulation by nitric oxide (NO) in maize seedling was investigated. Both NO and ABA contents of maize leaves and root tips were increased in response to salt stress (Fig.1,2). Similar to the effects of salt stress, ABA contents of maize leaves and root tips were increased after the treatment of maize leaves with sodium nitroprusside (SNP, a nitric oxide donor) alone (Fig.3). Compared to the salt stress-induced ABA accumulation, this SNP-induced ABA increase was much faster, suggesting that NO may be an intermediate signal from salt stress to ABA accumulation. When NO production inhibitors L-NAME and NaN(3) treatments were applied, salt stress-induced ABA accumulation was lowered (Fig.4). Treatment with NO scavenger cPTIO also inhibited the salt stress-induced ABA increase (Fig.5). From these results it is deduced that NO is involved in regulation of ABA accumulation under salt stress.

[Effects of grape procyanidins on the concentration of intracellular calcium and the proliferation activity of the hepatoma cells].

OBJECTIVE: To investigate the effects of grape procyanidins (GPC) on concentration of intracellular calcium and the proliferation activity of normal hepatic cells and the hepatic cell injuried by alcohol. METHODS: Rat hepatic cells and the cell injuried by alcohol were cultured with different concentration of GPC. The proliferation activity and concentration of intracellular calcium of the hepatic cells were measured by MTT assay and Fura-2 fluorescence methods. RESULTS: (1) The concentration of intracellular calcium of the normal control group and alcohol injury group were (108.26 +/- 14.17) and (651.24 +/- 47.95) nmol/L respectively, and that of both the high and the medium dose GPC groups were lower than the alcohol injury group, all differences are significant (P < 0.05). (2) The concentration of intracellular calcium in the normal hepatic cells treated with GPC is the group with calcium of extracellular fluid > the group free from calcium of extracellular fluid > the normal control group. (3) The proliferation activity of the high and the medium dose GPC group of normal hepatic cell and the cell injuried by alcohol were higher than the normal control group and alcohol injury group, all differences are significant (P < 0.05). CONCLUSION: GPC can raise the intracellular calcium concentration of normal hepatic cell by increasing extracellular fluidca introaffluxion and release from calcium pool, and enhance the proliferation activity of hepatic cells. It also can inhibit the abnormal rise (overload) of intracellular calcium concentration and the proliferation activity injury of hepatic cell induced by alcohol.

[Subtype and sequence analysis of human immunodeficiency virus type-1 strains in Guangdong province].

OBJECTIVE: To investigate subtype and genetic analysis of human immunodeficiency virus type-1 (HIV-1). METHODS: DNA sequences were amplified by nested-PCR from uncultured peripheral blood mononuclear cells (PBMC) obtained from 100 HIV-1 patients from Guangdong Province. The C2 to V3 region of the envelope glycoprotein gp120 of HIV-1 was sequenced directly. The analysis of the gene-based phylogenetic tree and variation of amino acid were carried out by using Wisconsin software package or genetics computer group (GCG). RESULTS: DNA fragments were amplified from 75 PBMC samples by using nested polymerase chain reaction (PCR). Sequence analysis showed that there were 3 HIV-1 subtypes or circulating recombinant forms (CRF): CRF01-AE (n = 44), CRF-BC (n = 27) and B' (n = 4). CONCLUSIONS: Three HIV-1 subtypes or circulating recombinant forms: CRF01-AE, CRF-BC and B' might be circulating in Guangdong Province. Findings from this study suggested that several subtypes might exist in Guangdong Province and the epidemic situation of AIDS be serious. It should be a challenge for Guangdong Province in treating patients, preventing and controlling AIDS in the future.
