Spectroscopic and computational studies on the coordination-driven self-assembly complexes (ZnL)2 and (NiL)2 [L= bis(2,4-dimethyldipyrrin-3-yl)methane].

FT-IR, FT-Raman and electronic absorption spectroscopies were utilized in conjunction with density functional theory (DFT) calculations to investigate the ground and excited states of self-assembled dinuclear dimeric helicates (ZnL)2 and (NiL)2 [L = bis(2,4-dimethyldipyrrin-3-yl)methane]. These studies afford a detailed description of the ground-state geometric and electronic structures of (ZnL)2 and (NiL)2 and provide a comparison with similar geometrical metal-porphyrins. The results demonstrate that enlarging the basis set used in the DFT calculations results in an obvious alteration of the calculated bond lengths but negligible alteration of the calculated bond angles. The predicted spectra are in good agreement with the experimental ones with the deviations generally less than 30 cm(-1). In comparison with vibrational spectra of metal-porphyrins, the breathing vibration of the pyrrole ring is shifted by over 100 cm(-1) toward higher wavenumber due to local conjugation of molecular geometry. Time-dependent density functional theory (TD-DFT) provides a good description of the excitation energy. Because of the break in symmetry, the absorption band (corresponding to the Q-band of porphyrin) of (ZnL)2 and (NiL)2 is no longer weak. Local conjugation makes the absorption wavelength of (NiL)2 and (ZnL)2 shift to the blue compared with those of NiP and ZnP.

Studies on induction of L-aspartic acid modified chitosan to crystal growth of the calcium phosphate in supersaturated calcification solution by quartz crystal microbalance.

Acidic amino acids, such as aspartic acid (L-Asp) and glutamic acid, are the primary bioactive molecules of the glycoprotein on the organic/inorganic interface of biomineralized tissues. In this study, the induction of chitosan film modified with L-Asp on the crystal growth of hydroxyapatite (HAP) was investigated by a novel in situ analysis approach, quartz crystal microbalance (QCM), associated with the dynamically structural and morphological characterization of precipitation products on various phases by X-ray diffraction (XRD), Fourier-transformed infrared (FT-IR) spectroscopy and scanning electron microscopy (SEM). The natural chitosan exhibited no inducing ability on the crystal growth of HAP. However, the growth rate of induced HAP was dramatically accelerated by the L-Asp modification of chitosan film and increased with the increase of the concentration of L-Asp in the chitosan substrate. It was shown that the chelation of calcium ion with L-Asp provided a nucleation centre and the cluster nuclei was formed by adsorbing further PO(4)(3-), Ca(2+), and then HAP deposited on the original HAP coating in the supersaturated calcification solution (SCS). The developed method allows a kinetic evaluation of the induction of organic film on crystal nucleation and the growth of HAP in vitro.

[Effects of three different etching methods on surface morphology and element of pure titanium].

The pure titanium disks were divided into three groups and etched for 30 minutes with HNO3, hot H2SO4/H2O2 or hot H2SO4/HCl respectively. The treated disks were studied and analyzed with scanning electron microscope (SEM) and X-ray photoelectron spectroscopy (XPS). The disks etched with HNO3 had a smooth surface, while those etched with hot H2SO4/H2O2 or hot H2SO4/HCl had rough surfaces, and the surface etched with hot H2SO4/HCl had larger micropores. The XPS analysis demonstrated that the main elements of the surface in three groups were titanium, oxygen and carbon. The carbon concentration was the lowest on the surface etched with hot H2SO4/H2O2 and the highest on that etched with hot H2SO4/HCl. The substances were TiO2, Ti2O3, TiO and metal Ti on the surface etched with HNO3 or hot H2SO4/H2O2. Only TiO2 was detected on the surface etched with hot H2SO4/HCl.

Meretrix lusoria--a natural biocomposite material: in situ analysis of hierarchical fabrication and micro-hardness.

The ultrastructure of clam (Meretrix lusoria) was investigated by means of scanning electron microscope (SEM), transmission electron microscope (TEM) and X-ray diffraction analyzer (XRD) combining with in situ texture decalcified technique and the micro-hardness of clam was determined, in order to understand the spatial relationship between the mineral phase and organic matrix and further explain the correlation between the property and structure. The results showed that hierarchical fabrication is the major structure character of this mollusc shell. There is specific braided structure forming from domains composed of needle-like structure made up of the single crystal of aragonite. High magnification TEM image of clam indicates the intracrystal region of the aragonite single crystal is made up of subgrain phase and some amorphous substance. There are various crystal grain growth preferential orientations in the different growth direction of the shell. An amount of organic microtubule distribute evenly in the base of calcium carbonate as reinforcement phase. The mechanical property of this natural biological composite is better than other aragonite layer of mollusc shells and pearls according to the data of micro-hardness testing. The braided structure and organic microtubule reinforcement phase are responsible for its high mechanical performance. The stereo hierarchical fabrication of clam was elucidated for the first time.

Electrophoretic separation of DNA using a new matrix in uncoated capillaries.

A new separation matrix, consisting of polymer poly(N-isopropylacrylamide) (PNIPAM) and small molecule additive mannitol, was used for double-stranded (ds) DNA and plasmid DNA separation by capillary electrophoresis. The matrix had a low viscosity, which made it very easy to handle. The additive mannitol dramatically enhanced the sieving performance of PNIPAM in TBE buffer. The optimal mannitol concentration 6% in polymer solution, was determined with the consideration of both speed and resolution. A resolution of 0.95 was achieved on the separation of 271/281 bp in the phiX174/HaeIII digest by using 1.5% PNIPAM + 6% mannitol, while the supercoiled, linear and nicked conformers of lambda plasmid were separated in 1% PNIPAM + 6% mannitol, demonstrating the potential use of this new matrix for effective DNA separations. The dramatic impact of mannitol on sieving performance of PNIPAM solution was investigated. pH dependent self-coating ability of PNIPAM was revealed. The presence of mannitol in TBE buffer decreased the pH of the buffer, which led to more efficient self-coating ability of PNIPAM probable due to the formation of hydrogen bonds between PNIPAM molecules and silanol groups at the silica wall.

The fabrication and characterization of a formaldehyde odor sensor using molecularly imprinted polymers.

The fabrication and characterization of odor sensors based on molecularly imprinted polymers is reported as the first case of imprinting formaldehyde. A quartz crystal microbalance is employed as a sensitive apparatus of a sensor for the determination of odor formaldehyde. An equation is deduced to characterize the interaction between molecularly imprinted films and the template. A linear relationship between the frequency shifts and the concentration of analyte in the range of 1.25 to 14.25 microM is found. The detection limit is about 20.5 microM. The sensor can selectively distinguish gaseous formaldehyde. It is envisaged that this novel and handy method could be employed to determine formaldehyde gas in the atmosphere.

Biosensor for the determination of sorbitol based on molecularly imprinted electrosynthesized polymers.

Despite the increasing number of applications of biosensors in many fields, the construction of a steady biosensor remains still challenging. The high selectivity and stability of molecularly imprinted polymers for the template molecule make them ideal alternatives as recognition elements for sensors. In this work, the fabrication and characterization of biosensor based on molecularly imprinted electrosynthesized polymers is reported as the first case of imprinting sorbitol. A relevant molecularly imprinted film is prepared by o-phenylenediamine (o-PD) using the electrochemical method. Quartz crystal microbalance is employed as a sensitive apparatus of biosensor for the determination of sorbitol. An equation is deduced to characterize the interaction between molecularly imprinted films and the template. A linear relationship between the frequency shift and the concentration of analyte in the range of 1-15 mM was found. The detection limit is about 1mM.

Molecularly imprinted TiO2 thin film by liquid phase deposition for the determination of L-glutamic acid.

For the first time, the feasibility of a molecularly imprinted liquid phase deposition (LPD) thin film has been demonstrated. Thin films of titanium oxide imprinted with L-glutamic acid were prepared by the LPD method on a gold-coated quartz crystal microbalance. The imprinted molecule could be removed upon treatment with immersion in deionized water. A sensor was developed on the basis of this method and showed good sensitivity, selectivity, and reproducibility to the template molecule. An equation was deduced to characterize the interaction between molecularly imprinted films and the template by virtue of Scatchard analysis. X-ray photoelectron spectroscopy was introduced to show the evidence for the molecular imprinting phenomenon. The linear relationship between the frequency shifts and the concentration of analyte in the range of 10-200 microM was obtained. LPD proves to be a powerful method for imprinting titanium oxide thin films.

Control over the crystal phase, shape, size and aggregation of calcium carbonate via a L-aspartic acid inducing process.

The acidic amino acid, such as aspartic acid (l-Asp), and glutamic acid are the primary active molecules of the glycoprotein on the organic/inorganic interface of biomineralized tissue. In this study, aspartic acid was used as the organic template in inducing the nucleation and growth of calcium carbonate. With the analysis of X-ray diffraction we investigated the relationship between the l-Asp concentration and the precipitation phase crystal structure of calcium carbonate. SEM and TEM were employed in the analysis of the morphological characteristic of the precipitation and the aggregation of the nanoscale porous phase. In order to get the direct evidence of the interaction between Ca2+ and l-Asp, the technique of QCM was used in the investigation of the coordinate interaction of Ca2+/l-Asp. As the results have shown, l-Asp alone is adequate to switch the transformation between calcite and vaterite, and neither soluble organic additions nor metal ions are needed. Meanwhile, the morphology, size and aggregative way of the deposition are also mediated with change of l-Asp concentration. To interpret the cause of the hierarchic structure range from nanoscale to micron-scale and the formation of the porous spheres of vaterite, an assumption of limited-fusion was proposed from the view of the small biomolecules polarity that can control over the growth of the crystals and the aggregation of the micro crystals. The conclusion also provide a new material synthesize strategy.

Density functional theory and ab initio studies of geometry, electronic structure and vibrational spectra of novel benzothiazole and benzotriazole herbicides.

The ground-state geometries, electronic structures and vibrational wavenumbers of S-1,3-benzothiazolyl-4-bromobenzenecarbothioate and S-(5,7-dimethyl-3H-4lambda-5-[1,2,4]triazolo[1,5-a]pyridinyl)4-chlorobenzenecarbothioate were studied by DFT-B3LYP, BLYP and ab initio RHF method with different basis sets. The comparison was performed for optimized geometries, thermodynamic parameters and electronic structures at different levels of theory. Because of the larger repulsion effect in triazole ring, the vibrational wavenumbers of skeleton vibration of triazole ring are significantly lower than that of thiazole ring.

[Study of normal mucosa and gastric carcinoma by confocal Raman microspectroscopy].

The high SNR (Signal-to-Noise) spectra from 20 cases of gastric carcinoma and normal mucosa were obtained by confocal Raman microspectroscopy. Consistent spectral features were observed at different sites of one tissue slice for both typical tissues. Raman spectra of normal gastric mucosa shows two peaks which are assigned to amide I vibration of proteins in 1640-1670(cm(-1)) spectral region. However, in gastric carcinoma, only one peak of amino I vibrational mode of proteins can be observed. The obviously different spectral characters of the two types of tissues are in accordance to clinicopathologic diagnosis. The result shows Raman spectroscopy might be a potential method in the diagnosis of gastric carcinoma.

Perivascular space: possible anatomical substrate for the meridian.

BACKGROUND: Despite the meridian system being an important concept in Traditional Chinese Medicine (TCM), modern biology and Western medical systems have failed to find an anatomic substrate. Since the 1960s, a variety of phenomena along meridians have been reported, among which quite a few suggest that along meridians there is a fluid pathway (but not blood vessels or lymphatics). On the other hand, perivascular space (PVS) has been demonstrated to be a body fluid pathway in addition to blood vessels and lymphatics in some mammalian tissues, such as brain, thymus, and lung. OBJECTIVES: The present study was designed to examine the relationship between PVS and the meridian. We studied characteristics of the tissues around the blood vessels along the Stomach Meridian of Foot-Yangming and the Gallbladder Meridian of Foot-shaoyang, with the goal of identifying anatomical structure corresponding to the meridian described in TCM. DESIGN AND RESULTS: Through perivascular dye injection and frozen section histology, we found that there is PVS around the blood vessels along the meridians, and it is a fluid pathway. Subsequent physiologic studies revealed that the PVS shows significantly greater electrical conductivity and significantly higher partial oxygen pressure (pO(2)) compared to medial and lateral tissues. CONCLUSIONS: The PVS along the meridians has properties offering good explanation for the meridian phenomena. The work sheds new light on the studies of meridians and may contribute to research on the mechanism of Chinese acupuncture.

1H-NMR study of the effect of acetonitrile on the interaction of ibuprofen with human serum albumin.

The effect of acetonitrile (ACN) on the low-affinity interaction between human serum albumin (HSA) and ibuprofen (IBP) was studied using 1H-NMR techniques. Both chemical shift and relaxation measurements showed the addition of ACN to the solutions decreased the binding affinity of IBP to HSA and reduced the hydrophobic interaction between them. The self-diffusion coefficients of IBP were measured as a function of the drug concentration at different ACN concentrations. The association constant, K(a), for ligand-HSA complexes and the number of binding sites, n, are evaluated by the application of Langmuir isotherm. The results indicated that the value of n was about 38 without ACN, and about 26 with ACN concentration 12% (v/v%). The decreased binding capacity of IBP to HSA in the presence of ACN was mainly attributed to the competition of ACN with IBP to the low-affinity binding sites of HSA molecule.

Study of electron transfer across the liquid/ice-like matrix interface by scanning electrochemical microscopy.

In this work, we report the findings of a study on scanning electrochemical microscopy (SECM) to investigate the interfacial electron-transfer (ET) reaction between the 7,7,8,8-tetracyanoquinodimethane radical anion (TCNQ*-) in 1,2-dichloroethane and ferricyanide in an ice-like matrix (a mixture of insulting ice and conductive liquid) under low temperatures. Experimental results indicate that the formed liquid/ice-like matrix interface is superficially similar in electrochemical characteristics to a liquid/liquid interface at temperatures above -20 degrees C. Furthermore, imaging data show that the surface of the ice-like matrix is microscopically flat and physically stable and can be applied as either a conductive or an insulting substrate for SECM studies. Perchlorate ion was selected as the common ion in both phases, the concentrations of which controlled the interfacial potential difference. The effect of perchlorate concentration in the DCE phase on interfacial reactions has been studied in detail. The apparent heterogeneous rate constants for TCNQ*- oxidation by Fe(CN)6(3-) in another phase under different temperatures have been calculated by a best-fit analysis, where the experimental approach curves are compared with the theoretically derived relationships. Reaction rate data obey Butler-Volmer formulation before and after the freezing point, which is similar to most other known cases of ET reactions at liquid/liquid interfaces. However, there is a sharp change observed for heterogeneous rate constants around the freezing point of the aqueous phase, which reflects the phase transition. At temperatures below -20 degrees C, surface-confined voltammograms for the reduction of ferricyanide were obtained, and the ice-like matrix became an insulating one, which indicates that the aqueous phase is really a frozen phase.

Detection of hepatitis B virus by piezoelectric biosensor.

A highly sensitive piezoelectric HBV DNA biosensor has been developed based on the sensitive mass-transducing function of the quartz crystal microbalance and the speciality of nucleic acid hybridization reaction. HBV nucleic acid probe was immobilized onto the gold electrodes of a 9 MHz AT-cut piezoelectric quartz crystal with the polyethyleneimine adhesion, glutaraldehyde cross-linking (PEI-Glu) method or the physical adsorption method. The coated crystal with the PEI-Glu method to immobilized HBV nucleic acid probe showed the better results than the physical adsorption method with respect to sensitivity reproducibility and stability. The frequency shifts of hybridization have better linear relationship with the amount of HBV DNA, when the amount was in range 0.02-0.14 microg/ml. The crystal could be regenerated nearly five times without perceptible decrease of sensitivity.

In situ analysis of the organic framework in the prismatic layer of mollusc shell.

A novel in situ analytic approach was constructed by means of ion sputtering, decalcification and deprotein techniques combining with scanning electron microscopy (SEM) and transmission electron microscope (TEM) ultrastructural analysis. The method was employed to determine the spatial distribution of the organic framework outside and the inner crystal and organic/inorganic interface spatial geometrical relationship in the prismatic layer of cristaris plicate (leach). The results show that there is a substructure of organic matrix in the intracrystalline region. The prismatic layer forms according to strict hierarchical configuration of regular pattern. Each unit of organic template of prismatic layer can uniquely determine the column crystal growth direction, spatial orientation and size. Cavity templates are responsible for supporting. limiting size and shape and determining the crystal growth spatial orientation, while the intracrystal organic matrix is responsible for providing nucleation point and inducing the nucleation process of calcite. The stereo hierarchical fabrication of prismatic layer was elucidated for the first time.

Raman scattering properties of human pterygium tissue.

Pterygia, caused by fibrovascular growth of conjunctiva, are a common ophthalmic disease. However, the molecular composition of pterygium tissue has not been completely understood, and therefore the aim of this study is to investigate the spectroscopic differences between normal human bulbar conjunctiva and human pterygium tissue using a confocal Raman system. The high signal-to-noise ratio spectra from pterygium and conjunctiva tissue were obtained by this technique without any sample preparation and the time of detection required less than 3 min. Comparing Raman spectra of two types of tissue, there are obvious changes, including intensity decrease at approximately 1585 cm(-1) and intensity increase at approximately 1748, 1156, and 1521 cm(-1) with the lesion of conjunctiva. Additionally, the amide I vibrational mode of proteins in conjunctiva is significantly different than that in pterygium. The use of pathology, immunology, and the peroxidation of the lipids in conjunction with the Raman results indicate that the presence of additional elastic fibers, mast cells, and lymphocytes in pterygium, as compared with normal bulbar conjunctiva, have fewer unsaturated fat acids. The present study demonstrates that Raman spectroscopy can be potentially applied to diagnose pterygia clinically.
