Adolescent depression as a systemic multimorbidity catalyst: integrated genetic and metabolic pathway analysis.

BACKGROUND: Although adolescent depression has been linked to individual chronic conditions, its broader role in shaping multimorbidity risk remains understudied. METHODS: A total of 87,562 UK Biobank participants were included, of whom 18,851 had documented adolescent depression. Cox proportional hazards models were applied to evaluate associations between adolescent depression and 24 chronic diseases, followed by stratified analyses by sex and age. Two-sample Mendelian randomization (MR) was then conducted to infer causality for diseases showing significant associations. Genomic colocalization analyses were performed using relevant GWAS data to identify shared causal variants. Mediation analyses were performed to detect possible mediating factors, including the frailty index, KDM biological age acceleration, allostatic load and 30 circulating biomarkers. RESULTS: Adolescent depression was associated with elevated risk for 12 chronic diseases, with strongest associations for hypothyroidism (HR = 1.29 [1.18-1.42]), diabetes (HR = 1.25 [1.13-1.38]) and chronic obstructive pulmonary disease (COPD) (HR = 1.74 [1.50-2.01]). Risks were notably higher among females and younger adults. MR confirmed likely causal relationships for hypothyroidism (OR = 1.45 [1.03-2.05]), diabetes (OR = 1.01 [1.01-1.02]) and COPD (OR = 1.04 [1.02-1.06]). Genomic colocalization revealed a shared genetic signal at the CDSN/PSORS1C1 locus between adolescent depression and hypothyroidism. Mediation analyses revealed disease-specific pathways: creatinine for hypothyroidism, testosterone for diabetes, KDM biological ageing for COPD and frailty index across all three conditions. CONCLUSIONS: Adolescent depression confers systemic vulnerability through genetic and metabolic mechanisms, with amplified risks in females and individuals aged ≤55 years. These findings support early, integrated interventions to mitigate long-term multimorbidity.

Genetic heterogeneity affects the risk of incident depression, comorbidity, and response to environment: A prospective trajectory study.

BACKGROUND: Depression exhibits significant heterogeneity in its genetic underpinnings. The role of genetic components in the development of depression and its comorbidities remains insufficiently explored. METHODS: First, depression risk loci from a large-scale genome-wide meta-analysis were annotated to Gene Ontology (GO) terms by functional enrichment. GO-based polygenic risk scores (GO-PRS) were then calculated for individuals in the UK Biobank. Principal component analysis (PCA) was applied for dimensionality reduction, followed by cluster analysis to identify genetic subtypes of depression. Multistate models were applied to assess the impact of genetic patterns on the trajectory from healthy status to incident depression, and depression to 26 subsequent diseases, as well as the associations between environmental factors and disease trajectories across genetic subtypes. RESULTS: Participants were categorized into three genetic subtypes: immune-dominant, neuro-dominant, and comprehensive-risk. Significant differences in risk of depression and subsequent diseases, and susceptibility to environmental factors were observed across subtypes. Comprehensive-risk subtype showed higher risks of depression compared to immune-dominant (HR: 1.10, 95% CI: 1.05-1.15) and neuro-dominant subtype (HR: 1.12, 95% CI: 1.08-1.16). Comprehensive-risk subtype exhibited higher risks of transition from depression to subsequent diseases, such as anemia compared to immune-dominant subtype, and diseases of the digestive system compared to neuro-dominant subtype. Environmental factors were more strongly associated with the transition from depression to subsequent diseases in immune-dominant and comprehensive-risk subtypes, including cardiovascular, respiratory, and metabolic diseases. CONCLUSIONS: Our findings highlight the genetic heterogeneity of depression and comorbidities, and shed light on how genetic components modulate responses to environmental factors.

[Functional groups of natural enemies and their temporal-spatial dynamics in jujube orchard intercropped with herbage].

In this paper, an investigation was made on the temporal-spatial dynamics of the functional groups of natural enemies in the jujube orchard 2.5 km west of Taigu (112 degrees 8'E, 38 degrees 9'N, 780 m elevation), Shanxi Province in 2002, aimed to work out an effective strategy to control the pests there. In this orchard, four management modes were applied, i. e., intercropping herbage combined with integrated pest management (IPM), intercropping combined with conventional pest management (CM), intercropping with no pest management (NM), and non- intercropping (CK). The results indicated that the total number of natural enemies in IPM, CM and NM was 187.99%, 151.82% and 210.03% higher than CK, respectively, and the recovery and reconstruction rates as well as the average breadth and overlap indices of two dimensional temporal-spatial niches of the functional groups were higher in intercropping modes than in CK. The average diversity index of the functional groups in intercropping modes was significantly bigger than that in CK, but the fluctuation of the diversity standard error of the functional groups was significantly smaller (P < 0.05). The fluctuation trends of the dominance, diversity, and evenness of functional groups had a significant correlation (P< 0.01) with those of species. These results showed that intercropping herbage in jujube orchard was favorable to the conservation, proliferation, and making use of natural enemies. The natural enemy's community in jujube orchard intercropped with herbage was more stable than that without intercropping, and the numbers and functions of the functional groups were differed with different management modes and different stages. Among the functional groups of natural enemies, ladybirds had a stronger ability of recovery, while spiders and parasitoids were weaker. It was feasible to use functional groups instead of species to study the natural enemy's community in jujube orchard.

The influence of surfactant mixing ratio on nano-emulsion formation by the pit method.

The formation of O/W nano-emulsions by the PIT emulsification method in water/mixed nonionic surfactant/oil systems has been studied. The hydrophilic-lipophilic properties of the surfactant were varied by mixing polyoxyethylene 4-lauryl ether (C12E4) and polyoxyethylene 6-lauryl ether (C12E6). Emulsification was performed in samples with constant oil concentration (20 wt%) by fast cooling from the corresponding HLB temperature to 25 degrees C. Nano-emulsions with droplet radius 60-70 nm and 25-30 nm were obtained at total surfactant concentrations of 4 and 8 wt%, respectively. Moreover, droplet size remained practically unchanged, independent of the surfactant mixing ratio, X(C12E6). At 4 wt% surfactant concentration, the polydispersity and instability of nano-emulsions increased with the increase in X(C12E6). However, at 8 wt% surfactant concentration, nano-emulsions with low polydispersity and high stability were obtained in a wide range of surfactant mixing ratios. Phase behavior studies showed that at 4 wt% surfactant concentration, three-liquid phases (W+D+O) coexist at the starting emulsification temperature. Furthermore, the excess oil phase with respect to the microemulsion D-phase increases with the increase in X(C12E6), which could explain the increase in instability. At 8 wt% surfactant concentration, a microemulsion D-phase is present when emulsification starts. The low droplet size and polydispersity and higher stability of these nano-emulsions have been attributed, in addition to the increase in the surface or interfacial activity, to the spontaneous emulsification produced in the microemulsion D-phase.

A nonparametric comparison of conditional distributions with nonnegligible cure fractions.

Survival data with nonnegligible cure fractions are commonly encountered in clinical cancer clinical research. Recently, several authors (e.g. Kuk and Chen, Biometrika 79 (1992) 531; Maller and Zhou, Journal of Applied Probability, 30 (1993) 602; Peng and Dear, Biometrics, 56 (2000) 237; Sy and Taylor, Biometrics 56 (2000) 227) have proposed to use semiparametric cure models to analyze such data. Much of the existing work has been emphasized on cure detections and regression techniques. In contrast, this project focuses on the hypothesis testing in the presence of a cure fraction. Specifically, our interest lies in detecting whether there exists survival differences among noncured patients between treatment arms. For this purpose, we investigate the use of a modified Cramér-von Mises statistic for two-sample survival comparisons within the framework of cure models. Such a test has been studied by Tamura et al., (Statistics in Medicine 19, 2000, 2169) using bootstrap procedure. We will focus on developing asymptotic theory and convergent algorithms in this paper. We show that the limiting distributions of the Cramér-von Mises statistic under the null hypothesis can be represented by stochastic integrals and a weighted noncentral chi-squares. Both representations lead to concrete numerical schemes for computing the limiting distributions. The algorithms can be easily implemented for data analysis and significantly reduce computing time compared to the bootstrap approach. For illustrative purposes, we apply the proposed test to a published clinical trial.

[Community succession patterns of pests and natural enemies in jujube orchard ecosystem].

Through investigation and by the methods of optimal partitioning, principal component analysis, and one-or two-dimensional ordination, this paper analyzed the main groups and succession trends of the pest and natural enemy communities at a jujube orchard of Taigu area. The results showed that the communities had more obvious dominant factors and temporal succession pattern. The first three principal components of the communities had a bigger variation, with an obvious follow effects. The community succession was temporally divided into four stages, i. e., in the periods of pre-budding, leaf expanding and blooming, fruiting, and maturing. The dominant species of pests and their natural enemies were demonstrated.

Phase behavior and nano-emulsion formation by the phase inversion temperature method.

Formation of oil-in-water nano-emulsions has been studied in the water/C12E4/isohexadecane system by the phase inversion temperature emulsification method. Emulsification started at the corresponding hydrophilic-lipophilic balance temperature, and then the samples were quickly cooled to 25 degrees C. The influence of phase behavior on nano-emulsion droplet size and stability has been studied. Droplet size was determined by dynamic light scattering, and nano-emulsion stability was assessed, measuring the variation of droplet size as a function of time. The results obtained showed that the smallest droplet sizes were produced in samples where the emulsification started in a bicontinuous microemulsion (D) phase region or in a two-phase region consisting of a microemulsion (D) and a liquid crystalline phase (L(alpha)). Although the breakdown process of nano-emulsions could be attributed to the oil transference from the smaller to the bigger droplets, the increase in instability found with the increase in surfactant concentration may be related to the higher surfactant excess, favoring the oil micellar transport between the emulsion droplets.

[An experiment study and clinical observation of the testicle spermatogenesis after scrotum reconstruction].

OBJECTIVE: To explore the effect of scrotum reconstruction with a skin flap on spermatogenesis. METHODS: Two patients who underwent scrotum reconstruction with the skin flap were followed up for four years. Their sperm quality, sex function, sexual hormone, and testis biopsy were examined. To exclude the influential factors of testis and spermatic cord contusing, an experiment study was designed and performed in rabbits. The scrotal skin of the rabbits was stripped off and the scrotum was reconstructed with a hypogastric skin flap. RESULTS: The clinical follow-up indicated that in the early postoperative period, the reconstruction did not impede spermatogenesis, but the arrest of spermatogenesis happened with time. The experimental results showed that the sperm count of the rabbits decreased obviously and the rabbits became sterile two months after scrotum reconstruction. CONCLUSION: The thick skin flap is not recommended for scrotum reconstruction.

Cloning of the correct full length cDNA of NF-kappaB-repressing factor.

NF-kappaB-repression factor (NRF) is a nuclear inhibitor of NF-kappaB proteins that can silence the IFNbeta promoter. Since NRF was cloned in 1999, in-depth studies have been conducted on the biological functions of this constitutive repressor of NF-kappaB proteins. During large-scale sequencing of a human fetal brain cDNA library we isolated a novel human cDNA that proved to be a correct full-length NRF cDNA. The deduced protein contains 690 aa, and has a G-patch and an R3H domain at its C-terminus. The size of the protein is consistent with its counterparts in mouse and rat. There is considerable evidence that there are some mistakes in the NRF cDNA sequence reported by Nourbakhsh. Here we report the correct, full-length cDNA and protein sequences of NRF. Full-length NRF cDNA is 3247 bp long, contains three exons and maps to human chromosome Xq24. RT-PCR shows that NRF is widely expressed in human tissues.

Phase behavior of polyglycerol didodecanoates in water.

A phase diagram of a water-polyglyceryl didodecanoate ((C11)2Gn) system was constructed as a function of polyglycerol chain length (n) at 25 degrees C. The average number of dodecanoic acid residues attached to polyglycerol is in the range of 1.6-2.3, and unlike commercial long-chain polyglycerol surfactants, unreacted polyglycerols were removed in the surfactants used. With an increase in the polyglycerol chain, the surfactant changes from lipophilic to hydrophilic, and the type of self-organized structure also changes from lamellar liquid crystals to the aqueous micellar solution phase via hexagonal liquid crystals. However, a discontinuous micellar cubic phase does not appear in the phase diagram, while it is formed in a long poly(oxyethylene)-chain nonionic surfactant system. In a dilute region, a cloud point is observed at a moderate polyglycerol chain length, n approximate to 7. The cloud temperature is dramatically increased with a slight increase in hydrophilic chain because the dehydration of the hydrophilic chain length at high temperature is low compared with that of the poly(oxyethylene) chain. In other words, the phase behavior of (C11)2Gn is not very temperature sensitive. Three-phase microemulsion is formed in a water/(C11)2.3G7.3/m-xylene system. The three-phase temperature or HLB temperature is highly dependent on the polyglycerol chain length.

[The experimental study of bioabsorbable mini-plate for rigid fixation on rabbits].

OBJECTIVE: To study the biodegradation process of the bioabsorbable mini-plate (LactoSorb) and its influence on bone healing. METHODS: 15 mature New Zealand White Rabbits were selected as the experimental animals. The bilateral zygomatic arch were fractured using a fissure bur. One side was fixed with the bioabsorbable mini-plate and the other side was fixed with Titanium mini-plate as a control. 5 animals were killed after 4, 12, and 24 weeks post operation respectively for radiographic and histologic study. RESULTS: At 4 weeks, the absorbable plate changed from transparent to opaque and its dimensions was enlarged. The bone gaps of both groups were filled with new bone. No inflammatory cells were found in both groups. At 12 weeks, the absorbable plate broke to small pieces or particles, its dimensions reduced greatly but the outline could still been recognized. The bone continuity had been reestablished in both groups. At 24 weeks, the absorbable plate was resorbed completely. Only the residual screw could be seen in the screw hole. Complete bone healing was seen in both groups. No histological differences of bone healing were found at this stage. CONCLUSION: The absorbable plate demonstrates excellent biocompatibility. No adverse local inflammatory reactions were found during the plate was absorbed gradually. In the non-weight baring area, the absorbable plate provides the same fixation stability as that of the metal plate.
