N-acetylglucosaminyltranferase VB expression enhances beta1 integrin- dependent PC12 neurite outgrowth on laminin and collagen.

N-acetylglucosaminyltransferase VB (GnT-VB, -IX) is a newly discovered glycosyltransferase expressed exclusively in high levels in neuronal tissue during early development. Its homolog, GnT-V, is expressed in many tissues and modulates cell-cell and cell-matrix adhesion. The ability of GnT-VB to regulate cell-matrix interactions was initially investigated using the rat pheochromocytoma PC12 neurite outgrowth model. PC12 cells stably transfected with GnT-VB consistently showed an enhanced rate of nerve growth factor (NGF)-induced neurite outgrowth on collagen and laminin substrates. Levels of TrkA receptor phosphorylation and downstream ERK activation induced by NGF were not influenced by GnT-VB expression. No significant difference was observed in the rate of neurite outgrowth when cells were cultured on non-coated culture dishes, indicating that integrin-ECM interaction is required for the stimulatory effects. Neurite outgrowth induced by manganese-dependent activation of beta1 integrin on collagen and laminin substrates, however, showed a significant increase in neurite length for the PC12/GnT-VB cells, compared with control cells, suggesting that the enhancement is most likely mediated by alteration of beta1 integrin-ECM interaction by GnT-VB. These results demonstrate that GnT-VB expression can modulate the rate of neurite outgrowth by affecting beta1 integrin-ECM interaction.

Cellular uptake of magnetic nanoparticle is mediated through energy-dependent endocytosis in A549 cells.

Biocompatible silica-overcoated magnetic nanoparticles containing an organic fluorescence dye, rhodamine B isothiocyanate (RITC), within a silica shell [50 nm size, MNP@SiO2(RITC)s] were synthesized. For future application of the MNP@SiO2(RITC)s into diverse areas of research such as drug or gene delivery, bioimaging, and biosensors, detailed information of the cellular uptake process of the nanoparticles is essential. Thus, this study was performed to elucidate the precise mechanism by which the lung cancer cells uptake the magnetic nanoparticles. Lung cells were chosen for this study because inhalation is the most likely route of exposure and lung cancer cells were also found to uptake magnetic nanoparticles rapidly in preliminary experiments. The lung cells were pretreated with different metabolic inhibitors. Our results revealed that low temperature disturbed the uptake of magnetic nanoparticles into the cells. Metabolic inhibitors also prevented the delivery of the materials into cells. Use of TEM clearly demonstrated that uptake of the nanoparticles was mediated through endosomes. Taken together, our results demonstrate that magnetic nanoparticles can be internalized into the cells through an energy-dependent endosomal-lysosomal mechanism.

Toxicity and tissue distribution of magnetic nanoparticles in mice.

The development of technology enables the reduction of material size in science. The use of particle reduction in size from micro to nanoscale not only provides benefits to diverse scientific fields but also poses potential risks to humans and the environment. For the successful application of nanomaterials in bioscience, it is essential to understand the biological fate and potential toxicity of nanoparticles. The aim of this study was to evaluate the biological distribution as well as the potential toxicity of magnetic nanoparticles to enable their diverse applications in life science, such as drug development, protein detection, and gene delivery. We recently synthesized biocompatible silica-overcoated magnetic nanoparticles containing rhodamine B isothiocyanate (RITC) within a silica shell of controllable thickness [MNPs@SiO2(RITC)]. In this study, the MNPs@SiO2(RITC) with 50-nm thickness were used as a model nanomaterial. After intraperitoneal administration of MNPs@SiO2(RITC) for 4 weeks into mice, the nanoparticles were detected in the brain, indicating that such nanosized materials can penetrate blood-brain barrier (BBB) without disturbing its function or producing apparent toxicity. After a 4-week observation, MNPs@SiO2(RITC) was still present in various organs without causing apparent toxicity. Taken together, our results demonstrated that magnetic nanoparticles of 50-nm size did not cause apparent toxicity under the experimental conditions of this study.

Tunable magnetic arrangement of iron oxide nanoparticles in situ synthesized on the solid substrate from diblock copolymer micelles.

Hexagonal arrangement of iron oxide nanoparticles was fabricated by utilizing a single-layered film of diblock copolymer micelles. The synthesis was directly performed on the solid substrate by oxygen plasma with preserving the dimensional order of micelles so that separate procedures for synthesis and deposition of nanoparticles were not necessary. Since the oxygen plasma treatment also eliminated polymers, pure patterns of iron oxide nanoparticles were obtained. Moreover, easy control over the size of nanoparticles enabled us to selectively create a ferrimagnetic or a superparamagnetic pattern of iron oxide nanoparticles without altering the fabrication process.

Efficient electrogenerated chemiluminescence from cyclometalated iridium(III) complexes.

Very efficient electrogenerated chemiluminescence (ECL) phenomena were realized by deliberately tuning electron-transfer reactions from electrochemically generated electron donor to metal complex radical cations. By controlling the relative positions of HOMO and LUMO levels (oxidation potential and reduction potential) of Ir(III) complexes, we could obtain 77 times higher ECL from iridium(III) complexes in the presence of TPA than that of the Ru(bpy)32+/TPA system. This high ECL efficiency of new Ir(III) complexes can be used in many interesting applications such as sensors and luminescent devices.

Nanostructuring of a polymeric substrate with well-defined nanometer-scale topography and tailored surface wettability.

This study demonstrates a simple and highly reproducible method for fabricating well-defined nanostructured polymeric surfaces with aligned nanoembosses or nanofibers of controllable aspect ratios, showing remarkable structural similarity with interesting natural biostructures such as the wing surface of Cicada orni and the leaf surface of Lotus. Our studies on the present biomimetic surfaces revealed that the wetting property of the nanostructured surface of a given chemical composition could be systematically controlled by rendering nanometer-scale roughness. The nanofabricating method we developed can be readily extended to other thermoplastic polymeric materials (e.g., light-emitting polymers, conducting polymers, block copolymers, liquid crystalline polymers), and it could be applied to developing a new generation of optical and electronic devices.

Innate BALB/c enteric epithelial responses to Trichinella spiralis: inducible expression of a novel goblet cell lectin, intelectin-2, and its natural deletion in C57BL/10 mice.

Infection of mice with the nematode parasite Trichinella spiralis induces changes in the proteome of the jejunal epithelium, including substantial up-regulation of a novel variant of interlectin. In this study we sequence this novel lectin, termed intelectin-2, and compare expression levels during T. spiralis infection of resistant (BALB/c) with susceptible (C57BL/10) mouse strains. Intelectin-2 was cloned and sequenced from BALB/c mRNA extracted on day 14 of infection, and was found to have 91% amino acid identity with intelectin (within our study termed intelectin-1). Intelectin-2 transcripts were up-regulated early (day 3) during infection with T. spiralis in BALB/c mice, suggesting an innate response, and levels remained high through to day 14 (time of parasite rejection). Immunohistochemistry of jejunal sections with a rabbit polyclonal Ab to Xenopus laevis 35-kDa cortical granule lectin (XL35; 68% identity with intelectin-2) followed a similar pattern, with intense labeling of goblet and Paneth cells at day 14. However, intelectin-2 transcripts and protein were absent, and immunohistochemistry negative when C57BL/10 mice were infected with T. spiralis. Genomic PCR and Southern blotting confirmed that the intelectin-2 gene is absent from the C57BL/10 genome. The presence of intelectin-2 in resistant BALB/c mice, its absence from the susceptible C57BL/10 strain and the kinetics of its up-regulation during T. spiralis infection suggest that this novel lectin may serve a protective role in the innate immune response to parasite infection.

Novel synthesis of organic nanowires and their optical properties.

Aligned nanowires of organic luminescent material were prepared by introducing the organic luminants into nanochannels of variable size in an anodic aluminum oxide (AAO) membrane, and the emission spectra from these nanowire arrays exhibited novel size-dependent luminescent properties.
