Synthesis of novel chemical probes for the study of tanshinone binding proteins.

Novel diazirine or biotin-labeled tanshinone probes were synthesized and evaluated for TRAP inhibitory activity against RANKL-induced osteoclastogenesis in RAW264.7 cells. We found that diazirine-labeled derivatives (18 and 20) are potent inhibitors of RANKL-induced osteoclastogenesis. IC50 values were 18.02 and 15.00 microM, respectively. These probes will be useful reagents for investigating tanshinone-proteins interactions.

Collagen mimetic peptide-conjugated photopolymerizable PEG hydrogel.

Collagen mimetic peptide (CMP) with a specific amino acid sequence, -(Pro-Hyp-Gly)(x)-, forms a triple helix conformation that resembles the native protein structure of natural collagens. CMP previously has been shown to associate with type I collagen molecules and fibers via a strand invasion process. We hypothesized that when poly(ethylene glycol) (PEG) hydrogel, a non-adhesive tissue engineering scaffold, is conjugated with CMP, it may retain cell-secreted collagens and also form physical crosslinks that can be manipulated by cells. A photopolymerizable CMP derivative was synthesized and copolymerized with poly(ethylene oxide) diacrylate to create a novel PEG hydrogel. In a model retention experiment, diffusional loss of type I collagen that was added to the hydrogel was limited. Chondrocytes were encapsulated in the hydrogel to examine its use as a tissue engineering scaffold. After 2 weeks, the biochemical analysis of the CMP-conjugated PEG gel revealed an 87% increase in glycosaminoglycan content and a 103% increase in collagen content compared to that of control PEG hydrogels. The histology and immunohistochemistry analyses also showed increased staining of extracellular matrix. These results indicate that the CMP enhances the tissue production of cells encapsulated in the PEG hydrogel by providing cell-manipulated crosslinks and collagen binding sites that simulate natural extracellular matrix.

Human risk assessment for heavy metals and as contamination in the abandoned metal mine areas, Korea.

Cleanup goals for the contaminated sites are established on the basis of risk assessments and rely on the estimated toxicity of the chemicals of concern (COC). Toxicity estimates are based on bioavailability causing risk of adverse health effects on humans. In this study, bioavailability of As, Cu, Pb and Zn in soil was determined by SBET (Simple Bioavailability Extraction Test), and chemical analysis for groundwater and stream water collected from the abandoned mine areas (Dukeum, Dongil, Dongjung, Myungbong and Songchun mine areas) was conducted. High values of cancer risk for As (1.16x10(-5)) were detected through soil ingestion pathways in the Songchun mine area and assessed through water exposure pathways in the all mines except Dukeum. The hazard index value for As in the Songchun mine area (3.625) exceeded 1.0. The results indicated that the ingestion of As-contaminated soil and water by local inhabitants can pose a potential health threat in these mine areas.

A case of Staphylococcus aureus endocarditis after ear piercing in a patient with normal cardiac valve and a questionnaire survey on adverse events of body piercing in college students of Korea.

Body piercing in young adults is on the rise in western countries and also in Korea. It is usually practised by non-medical persons, and therefore frequently entails local adverse events. Serious complications, such as septic arthritis or infective endocarditis, are occasionally reported, but infective endocarditis in patients with normal cardiac valve is rare. We experienced a case of acute S. aureus endocarditis after ear piercing. Thereafter we studied the status and complications of body piercing in college students of Korea based on a questionnaire survey, and revealed that the rate of ear piercing is high (96.5%) and 61.3% of the pierced persons experience local complications.

Human risk assessment of As, Cd, Cu and Zn in the abandoned metal mine site.

The cancer risk and the non-cancer hazard index for inhabitants exposed to As, Cd, Cu and Zn in the soils and stream waters of the abandoned Songcheon Au-Ag mine area were evaluated. Mean concentrations of As, Cd, Cu, Pb and Zn in agricultural soils were 230, 2.5, 120, 160, and 164 mg kg(-1), respectively. Mean concentrations of As, Cd and Zn of the water in the stream where drinking water was drawn was 246 microg L(-1), 161 microg L(-1) and 3899 microg L(-1), respectively. These levels are significantly higher than the permissible levels for drinking water quality recommended by Korea and WHO. The resulting human health risks to farmers who inhabited the surrounding areas due to drinking water were summarized as follows: (1) the non-cancer health hazard indices showed that the toxic risk due to As and Cd in drinking water were 10 and 4 times, respectively, greater than those induced by the safe average daily dosages of the respective chemicals. (2) the cancer risk of As for exposed individuals through the drinking water pathway was 5 in 1000, exceeded the acceptable risk of 1 in 10,000 set for regulatory purposes.

Baseline study on essential and trace elements in polished rice from South Korea.

In 2000, 63 (polished) white rice samples were collected in eight administrative areas all over South Korea and analyzed for 16 elements by inductively coupled plasma atomic emission spectrometry (ICP-AES). Potassium had the highest content, next to Mg, Ca, Si, Zn, Na, Al and Fe. Most of the samples contained worldwide average concentrations of essential and trace elements in rice grains reported by various researches. For inter-area differences in those elements in the rice, the statistical analysis showed no significant differences (p > 0.05) among the eight administrative areas, suggesting that inter-area differences were not substantial in most cases. Thus, the present data can be used as national background levels of elements in rice produced in South Korea. Using the published data on daily consumption of rice in South Korea, it was possible to estimate the daily intake of As, Cd, Cu, Pb and Zn via rice. The results showed that a regular consumption of rice produced in Korea plays an important role in accumulation of essential and trace elements in Korean, especially for farm-households consuming relatively large amounts of rice.

Change of enamel after Er:YAG and CO2 laser irradiation and fluoride treatment.

OBJECTIVE: Our aim was to compare acquired acid resistance in dental enamel after Er:YAG and CO(2) laser irradiation in vitro with additional fluoride treatment. BACKGROUND DATA: The application of lasers in dental hard tissue can impact dental constituents and fluoride with complex interactions. METHODS: Bovine enamels were divided into four groups according to their surface condition: unlased, Er:YAG laser-ablated, fluoridetreated after Er:YAG laser ablation, and CO(2) laser-irradiated after Er:YAG laser and fluoride treatment. Calcium distribution in enamel after pH-cycling process and crystallographical change after laser treatment were evaluated. RESULTS: The crystallinity of enamel was much improved after Er:YAG laser ablation. The CO(2) laser irradiation in the fluoride-treated laser enamel formed alpha-TCP and fluorapatite. The change of calcium distribution in enamel was least in the CO(2) laser-irradiated specimens; the fluoride-treated specimen after Er:YAG laser ablation was next. Additional fluoride treatment both after Er:YAG and before CO(2) laser irradiation improved the acid resistance of enamel. CONCLUSION: Both external treatment of fluoride and irradiation of CO(2) laser after Er:YAG laser ablation greatly reduced calcium loss in enamel during the pH cycling process.

Regional outbreak of mumps due to genotype H in Korea in 1999.

Serological and virological studies were carried out of a mumps outbreak which occurred in one region, Yoeju County, Southeast of Seoul in Korea from September to December, 1999. Sera from 736 children at 8-13 years of age of patients with mumps and healthy children were tested for mumps-specific antibodies by enzyme immunoassay. The overall IgM positive rate was 7.6% (56/736), compared with 69.8% (514/736) for IgG. Of the 49 children with both IgG and IgM, 32 were also confirmed by both clinical and serological diagnosis. IgM antibodies were detected even in the samples collected up to 3 months after the onset of symptoms. Although 436 children had been vaccinated before the outbreak, 27 (6.2%) were found to be IgM positive, particularly 6 (4.4%) of 136 were positive serologically despite a second-dose vaccinees. Sequence analysis of the small hydrophobic (SH) gene of 4 mumps viruses isolated from 42 saliva specimens revealed that these were related to the genotype H, but distinguishable from European strains. This is the first study on the outbreak due to mumps virus genotype H and provides information to assess the understanding of recent outbreaks of mumps in Korea.

Genetic and antigenic characterization of measles viruses that circulated in Korea during the 2000-2001 epidemic.

Despite the marked reduction in the incidence of measles in Korea by the introduction of measles vaccine, a large measles epidemic occurred during 2000-2001. During the epidemic, more than 55,000 measles cases were reported and at least 7 children were dead. In this study, we analyzed the genetic and antigenic properties of 15 measles viruses that isolated during the epidemic. Sequence analyses of entire hemagglutinin (H) and nucleoprotein (N) genes of the viruses indicated that all Korean isolates had a high degree of homology (>99.8%) when compared with each other. They differed from other wild-type viruses by as much as 6.8% in the H gene and 6.5% in the N gene at the nucleotide level. The deduced amino acid variability was up to 6.4% for the H protein and up to 6.5% for the N protein. Phylogenetic analysis of nucleotide sequences and deduced amino acid sequences of the H and N genes revealed that all Korean viruses were grouped into the genotype H1. This strongly demonstrated that single genotype of measles virus has been circulated in Korea during the 2000-2001 epidemic. Plaque reduction neutralizing antibody titers against vaccine strains, Edmonston and Schwarz, and recently isolated Korean strains were measured using sera from vaccinees and recently infected children. Although sera of recently infected children demonstrated higher neutralizing antibody titers against wild-type strains than against vaccine strains, both sera neutralized both strains and the reciprocal geometric mean titers (GMTs) were not significantly different against both strains.

Molecular characterization of two genotypes of mumps virus circulated in Korea during 1998-2001.

Sequence analyses of the entire small hydrophobic (SH) and hemagglutinin-neuraminidase (HN) genes of mumps viruses circulated in Korea from 1998 to 2001 showed that these isolates were grouped into two genotypes, H and I. While genotype I was predominant throughout the country during this period, genotype H was found in the restricted region, 1999. The nucleotide and deduced amino acid sequences of Korean isolates showed the type-specific changes including the signature motif at positions 28-30 in the SH gene and the neutralizing epitopes in the HN gene. Particularly, Asian strains including Korean isolates and European strains differed from 2.3 to 3.8% at the nucleotide sequence level in the SH gene although they belonged to the same genotype H. Furthermore, none of Korean isolates were genetically related to the vaccine strains used in Korea. The results provide important information to understand the epidemiology of mumps infection and to facilitate the development of more efficient vaccine program in Korea.

Detection and typing of respiratory adenoviruses in a single-tube multiplex polymerase chain reaction.

A multiplex polymerase chain reaction (PCR) assay that is capable of detecting and typing six serotypes of respiratory adenovirus (Ad) was developed, using multiple sets of type-specific primers. The detection of each different serotype depended on distinguishing different numbers and sizes of amplification products on agarose gels following PCR. The multiplex PCR was tested with 26 clinical Ad isolates and other respiratory viruses including influenza viruses, parainfluenza viruses, and respiratory syncytial viruses as well as respiratory bacterial pathogens such as Chlamydia pneumoniae, Streptococcus pneumoniae, Haemophilus influenzae, and Mycoplasma pneumoniae. The multiplex PCR for the detection and typing of Ads gave an excellent correlation with the results by conventional typing with type-specific antisera. This assay may serve as a rapid means of confirming Ad with simultaneous serotype identification of the isolates. It will also have relevance as an adjunctive tool to conventional serotyping for diagnostic and epidemiological purposes.

A simple technique for correcting the hump on a deviated nose.

The traditional technique for correcting the hump on a deviated nose frequently results in redeviation. The main cause of redeviation is incomplete correction of the septal deviation, but complete septal correction is very difficult. The authors have therefore adopted a new technique to reduce the possibility of redeviation. The new technique is simple. It no longer requires complete septal correction and gives good results.
