[Effects of bone morphogenetic protein and transforming growth fractor-beta on biomechanical property for fracture healing in rabbit ulna].

OBJECTIVE: To investigate the effects of exogenous bone morphogenetic protein (BMP) and transforming growth factor-beta (TGF-beta) on biomechanical property for ulna of fracture healing. METHODS: Thirty-six adult rabbits were made the model of right ulnar fracture and treated locally with TGF-beta/PLA, BMP/PLA, TGF-beta + BMP/PLA or PLA (as control group). Fracture healing was evaluated by measurement of the mechanical parameters and geometric parameters. RESULTS: As compared with control group, the geometric parameters, the bending broken load, the ultimate bending strength, the bending elastic modulus, the ultimate flexural strength, the flexural elastic modulus, the ultimate compressing strength, the compressing elastic modulus, and the ultimate tensile strength for ulna of fracture healing increased significantly in the treatment groups(P < 0.01). These parameters were higher in TGF-beta + BMP/PLA group than in TGF-beta/PLA group or in BMP/PLA group and in TGF-beta/PLA group than in BMP/PLA group(P < 0.05). There was no significant difference in bone density between the treatment groups and control group. CONCLUSION: Local application of exogenous TGF-beta and BMP can increase the callus formation and enhance biomechanical strength of bone after fracture healing. A combination of TGF-beta and BMP has synergetic effect in enhancing fracture healing.

[Analysis of severe acute respiratory syndrome in Beijing].

OBJECTIVE: To study the clinical, laboratory, and radiologic features of 34 cases of severe acute respiratory syndrome (SARS) in Beijing. METHODS: All patients were admitted to the isolation wards. Their demographic, clinical, laboratory, and radiologic characteristics were analyzed. Univariate and multivariate analyses were performed. RESULTS: Eight patients came from a family, and 15 patients were medical staff. The mean age of patients was (33.4 +/- 13.4) years. The latent period varied from 2 to 14 days (median 4 days). The most common symptoms were fever (100%), palpitation (91.7%), myalgia (79.2%), headache (70.8%), diarrhea (73.9%) and cough (58.3%). The mean leucocyte count was (4.6 +/- 1.4) x 10(9)/L, and the mean lymphocyte ratio was 0.27 +/- 0.11. 68.4% of the patients had lymphopenia (absolute lymphocyte count < 1.3 x 10(9)/L). Other common findings included elevated levels of serum alanine aminotransferase, lactate dehydrogenase and erythrocyte sedimentation (76.2%, 28.6% and 47.8%, respectively), and decreased levels of serum iron and albumin (63.2% and 47.8%, respectively). Thirty-two cases had abnormal chest radiographs. In 2 cases in whom typical lung opacities could not be found on the initial plain chest radiographs, thoracic CT proved to be useful. Postmortem examination of 1 patient revealed marked edema with foci of hemorrhage and hyaline membrane formation in the lungs, hemorrhage necrosis and a obvious decline of cells in lymph glands. In a multivariate analysis (Stata 7.0), the independent predictor of an adverse outcome was advanced age (odds ratio per decade of life, 1.6; 95% CI, 1.08 to 2.63; P = 0.007). CONCLUSIONS: Fever, lymphopenia, low serum iron and chest radiograph are helpful to diagnose SARS early; age is the independent predictor of an outcome.

[Effects of the novel gene, LAPTM4B, highly expression in hepatocellular carcinoma on cell proliferation and tumorigenesis of NIH3T3 cells].

OBJECTIVE: To study the biological effects of the novel gene LAPTM4B high expressed in hepatocellular carcinoma on cell proliferation and tumorigenesis of NIH3T3 cells. METHODS: Eukaryotic expression plasmid pcDNA3-TM4B was constructed and transfected into mouse NIH3T3 cell line using Lipofectamin-2000 mediating gene transfer technique. Monoclonal transfected cells with high expression of LAPTM4B gene were identified and selected by RT-PCR, Northern blot and Western blot method. Cell surface morphology was detected by scanning electronic microscopy (SEM). The cell attachment/spreading on various matrices was examined. The cell growth curves were measured by acid phosphatase method. Cell cycle was detected by flow cytometry (FCM). The expression level of Cyclin E protein was examined using Western blot. RESULTS: The cell surface of the LAPTM4B transfected cells showed abundant tube/finger-like microvilli, which were distinguished significantly from the MOCK cells. The cell attachment/spreading of transfected cells increased on fibronectin, matrigel and laminin. The growth rate of transfected cells was faster than that of the MOCK cells. FCM analysis indicated that the amount of the transfected cells in S phase was increased significantly. Cyclin E protein expression of transfected cells was much higher than that of the MOCK cell. The serum-dependence of transfected cells was decreased. The fibrosarcoma was formed at a tumorigenic rate of 50% in NIH mice inoculated with LAPTM4B transfected cells. CONCLUSION: LAPTM4B gene promotes the cell proliferation by involving in the regulation of cell cycle control and causes tumorigenesis of NIH3T3 cells, indicating that it plays important roles in tumorigenesis.

[Correlation of NDRG1 gene with liver tissue differentiation and hepatocarcinogenesis].

OBJECTIVE: To detect the expression profile of NDRG1 gene in different tissues and cell lines and explore the relationship of NDRG1 with liver tissue differentiation and hepatocarcinogenesis. METHODS: The expression profiles of NDRG1 in hepatocellular carcinoma (HCC) tissues, paired noncancerous liver (PNL) tissues, adult normal liver (NL) tissues, baby mouse liver tissues and fetal liver tissues in different developmental stages and cultured cell lines were performed using RT-PCR and Northern Blot analysis. RESULTS: Expression of NDRG1 was significantly up-regulated in HCC tissues compared to that of NL tissues and PNL tissues, however, the expressions of NDRG1 in NL and PNL tissues showed no evident difference. In ten cell lines, the highest expression was in the 293T human kidney cell line, the next one in liver cell L02, and the lowest one in the undifferentiated HLE cell line. Expression of NDRG1 in liver tissues enhanced with the development of the baby mouse and human fetus. CONCLUSION: NDRG1 is probably related to the liver cell differentiation and is highly expressed in the specific stage of the differentiation. However, it could not be recognized as a marker of differentiation. The high expression of NDRG1 in HCC indicates that HCC is not just distributed to a simple de-differentiation. Much more study is necessary in understanding the comprehensive relationship of the disorder proliferation and differentiation of HCC and the related genes.

The experimental and clinical study on the effect of curcumin on cell cycle proteins and regulating proteins of apoptosis in acute myelogenous leukemia.

To investigate whether the Bcl-2 gene family is involved in modulating mechanism of apoptosis and change of cell cycle protein induced by curcumin in acute myeloid leukemia HL-60 cell line and primary acute myelogenous leukemic cells, the Bcl-2 family member Mcl-1, Bax and Bak and cell cycle proteins including P27kipl, P21wafl, cyclin D3 and pRbp- were selected and their expression detected by SABC immuno-histochemical stain method. The attitude of sub-G1 peak in DNA histogram was determined by FCM. The TUNEL positive cell percentage was identified by terminal deoxynucleotidyl transferase (TdT)-mediated Biotin dUNP end labeling technique. It was found that when HL-60 cells were treated with 25 mumol/L curcumin for 24 h, the expression level of Mcl-1 was down-regulated, but that of Bax and Bak up-regulated time-dependently. There was significant difference in the expression level of Mcl-1, Bax and Bak between the curcumin-treated groups and control group (P < 0.05-0.01). At the same time, curcumin had no effect on progress of cell cycle in primaty acute myelogenous leukemia at newly diagnosis, but could increase the peak of Sub-G1 (P < 0.05), and down-regulate the expression of Mcl-1 and up-regulate the expression of Bax and Bak with the difference being statistically significant. The expression of P27kipl, P21wafl and pRbp- were elevated and that of cyclin D3 decreased in the presence of curcumin. These findings suggested that the Bcl-2 gene family indeed participated in the regulatory process of apoptosis induced by curcumin in HL-60 cells and AML cells. Curcumin can induce apoptosis of primary acute myelogenous leukemic cells and disturb cell cycle progression of HL-60 cells. The mechanism appeared to be mediated by perturbing G0/G1 phases checkpoints which associated with up-regulation of P27kipl, P21wafl and pRbp- expression, and down-regulation of cyclin D3.

Impaired mixed lymphocyte reaction in intrahepatic cholestasis of pregnancy.

OBJECTIVE: To clarify the alteration of maternal-fetal mixed lymphocyte reaction (MLR) in intrahepatic cholestasis of pregnancy (ICP). METHODS: Twenty-two patients with intrahepatic cholestasis of pregnancy and 21 normal pregnant women were recruited into the present study. Of patients with ICP, 5 also experienced pregnancy-induced hypertension (PIH). Maternal-fetal mixed lymphocyte reaction was conducted by using a morphologic transformation method, and the transformation rates of lymphocytes were compared either between ICP patients with and without pregnancy-induced hypertension, or between control and ICP group. RESULTS: In the ICP group, the transformation rates of lymphocytes were not significantly different between pregnancies with and without pregnancy-induced hypertension (p > 0.5). Compared with control, the transformation rate of lymphocytes in intrahepatic cholestasis of pregnancy decreased significantly (p < 0.001). CONCLUSION: Suppressed mixed lymphocyte reaction was observed in intrahepatic cholestasis of pregnancy and its mechanism behind and role in the disease need to be further explored.

Dysfunction of mitochondria in human skeletal muscle in type 2 diabetes.

Skeletal muscle is strongly dependent on oxidative phosphorylation for energy production. Because the insulin resistance of skeletal muscle in type 2 diabetes and obesity entails dysregulation of the oxidation of both carbohydrate and lipid fuels, the current study was undertaken to examine the potential contribution of perturbation of mitochondrial function. Vastus lateralis muscle was obtained by percutaneous biopsy during fasting conditions from lean (n = 10) and obese (n = 10) nondiabetic volunteers and from volunteers with type 2 diabetes (n = 10). The activity of rotenone-sensitive NADH:O(2) oxidoreductase, reflecting the overall activity of the respiratory chain, was measured in a mitochondrial fraction by a novel method based on providing access for NADH to intact mitochondria via alamethicin, a channel-forming antibiotic. Creatine kinase and citrate synthase activities were measured as markers of myocyte and mitochondria content, respectively. Activity of rotenone-sensitive NADH:O(2) oxidoreductase was normalized to creatine kinase activity, as was citrate synthase activity. NADH:O(2) oxidoreductase activity was lowest in type 2 diabetic subjects and highest in the lean volunteers (lean 0.95 +/- 0.17, obese 0.76 +/- 0.30, type 2 diabetes 0.56 +/- 0.14 units/mU creatine kinase; P < 0.005). Also, citrate synthase activity was reduced in type 2 diabetic patients (lean 3.10 +/- 0.74, obese 3.24 +/- 0.82, type 2 diabetes 2.48 +/- 0.47 units/mU creatine kinase; P < 0.005). As measured by electron microscopy, skeletal muscle mitochondria were smaller in type 2 diabetic and obese subjects than in muscle from lean volunteers (P < 0.01). We conclude that there is an impaired bioenergetic capacity of skeletal muscle mitochondria in type 2 diabetes, with some impairment also present in obesity.

[Study on clinical significance of fetal posterior fossa fluid].

OBJECTIVE: To discuss the clinical significance of accumulated fluid in fetal posterior fossa. METHODS: Prenatal ultrasonography examination was performed on 5 400 woman at more than 20 weeks gestation, 110 women with fetus accumulated fluid in the posterior fossa more than 5mm were included in this study. The changes of accumulated fluid in fetal posterior fossa and the associated intracranial and extracranial anomalies were observed regularly every 2 or 3 weeks until delivery. The infants were also followed up. RESULTS: The incidence of the fetal posterior fossa fluid was 2.0%. Generally, the accumulated fluid in fetal posterior fossa was diagnosed at first time at 22 approximately 41 gestation weeks, median was (31 +/- 4) weeks. Most of them were be found between 29 approximately 32 weeks (42 cases, 38.2%), and the maximum amount of accumulated fluid in fetal posterior fossa was also in 29 approximately 32 gestation weeks (39 cases, 35.5%). The amount of accumulated fluid was from 6 mm to 26 mm, mean (11 +/- 3) mm, mostly between 10 approximately 14 mm (79 cases, 71.8%). The incidence of defected infants was 4.0%, 7.6% and 83.3% respectively, when the posterior fossa fluid was less than 10 mm, 10 approximately 14 mm and more than 15 mm. CONCLUSION: Most of cases could be diagnosed between 29 approximately 32 weeks gestation. The more fluid in the posterior fossa found, the more defected fetal or infant would be observed. In cases of more than 10 mm, especially more than 15 mm, anomales should be observed carefully.

[Maternal-fetal mixed lymphocyte culture in intrahepatic cholestasis of pregnancy].

OBJECTIVE: To determine the maternal-fetal histocompatibility in intrahepatic cholestasis of pregnancy (ICP) by means of a morphologic method for mixed lymphocyte culture. METHODS: Twenty-two pregnant women with ICP and 21 normal pregnant women were enrolled in the present study. Mixed lymphocyte culture was conducted using lymphocytes obtained from maternal peripheral blood and cord blood of their fetuses. The transformational rates of lymphocytes were calculated and compared between normal and ICP-complicated pregnancies. RESULTS: The transformational rate in ICP was (24 +/- 5)%, while that in normal pregnant women was (36 +/- 9)%, the difference was significant between two groups (P < 0.001). The presence of pregnancy-induced hypertension did not affect the transformational rates in ICP group (P > 0.05). The transformational rate of lymphocyte in ICP did not correlate with cholylglycine level in serum significantly (P > 0.05). CONCLUSION: Decreased maternal-fetal lymphocyte reaction is demonstrated in ICP and it may play an etiological role.

[Influence of P-glycoprotein expression on chemotherapeutic response of metastatic breast carcinoma].

BACKGROUND & OBJECTIVES: The clinical study showed that the P-glycoprotein(P-gp) expression was closely associated with the chemotherapeutic effect, response rate, prognosis, and survival time. Until now, few clinical papers have reported about metastatic sites and its response to chemotherapy with P-gp expression in metastatic breast carcinoma. The current study was designed to investigate the role of P-gp expression and its clinical value of chemotherapy for the patients with different metastatic sites of this carcinoma. METHODS: P-gp expression in 46 postoperative patients with metastatic breast carcinoma was detected by SABC immunohistochemical method. 43 cases treated with combination regimen: Cyclophosphamide 600 mg/m2 i.v. on day 1, Pirarubicin 60 mg/m2 i.v. on day 1, 5-Fluorouracil 600 mg/m2 i.v. on days 1 and 8. Repeat the cycle every 3 weeks for at least 2 cycles. The correlation between P-gp expression and chemotherapeutic response was analyzed. RESULTS: 1) P-gp expression positive rate was 56.5%, the P-gp expression in the patients with lung or liver viscera metastasis was higher than that in skin or lymph node metastasis (P = 0.049). 2) The overall response rate was 58.1% in 43 patients. The response rate of the P-gp negative group was higher than the P-gp positive group(P < 0.01). 3) The response rate in the patients with skin and lymph node metastasis was higher than the patients with lung and liver metastasis (P < 0.05). 4) The postoperative patients who had received CAF(cyclophosphamide + adriamycin + 5-fluorouracil) or CMF(cyclophosphamide + methotrexate + 5-fluorouracil) regimen adjuvant chemotherapy previously, the response rate of metastatic diseases to chemotherapy had no significant difference (P > 0.05). CONCLUSIONS: P-gp expression may be considered as an index for evaluating multidrug resistance, guiding drug use, and judging prognosis of the patients with metastatic breast carcinoma.

Thermochemical characteristics of 2,4-dichlorophenol degrading strain Pseudomonas GT241-1 of growth metabolism by microcalorimetry.

By using LKB-2277 Bioactivity Monitor, ampoule method, the heat output of the growth metabolism of a 2.4-dichlorophenol degrading bacteria strain, Pseudomonas strain GT241-1, has been determined at 30 degrees C. From the thermogenic curves, it can be established that thermokinetic equation of their growth metabolism is Pt = Pt = 0 exp(k(m) t), dP/dt = k(m)P1, with the order of growth metabolism n = 1. The experimental results indicate that the relationship between the metabolic power (P) and the cell concentration (C), and relationship between the metabolic power of each cell (P0) and the cell concentration can be characterized by the following thermal equation: C = a + kP, InC = a' + k'P0 or dC/dP0 = KC1. The order of the P0-C equation n is also 1. These results are very significant in environmental sciences, biology and thermochemistry.
