[Effects of infrasound on ultrastructure of testis cell in mice].

OBJECTIVES: To investigate the effects of infrasound on ultrastructure of testis in mouse. METHODS: Twelve male BALB/C mice were randomly divided into three groups according to exposed duration on 1, 7 and 14 day. The mice were separately exposed to infrasound environment under 8 Hz/90 dB, 8 Hz/130 dB, 16 Hz/90 dB, 16 Hz/130 dB 2 hours per day. There was another control group which had three mice were separated into module with no infrasound. All the mice were killed on schedule. Then all the sections of testis were observed under electronic microscope. The alterations of structure and the chromatin were observed. RESULTS: Some acute alteration in one day group was found in testis cell, such as cellular denaturation and necrosis, intercellular edema, mitochondria swelling, liposome hyperplasia. When the infrasound was up to 8 Hz/130 dB, the damage showed seriously. In 7 and 14 day group, the acute alteration was gradually decreased. A plenty of abnormal sperm were found. And other alteration was chromatin condense. The effect of variational frequency was important in ultrastructure. CONCLUSIONS: The infrasound markedly effected to testicular cell morphology and secreting function. Infrasound will lead to the alteration of procreation in mouse.

[A clinical study of haploidentical and G-CSF primed bone marrow transplantation by using CD25 for aGVHD prophylaxis].

To explore the feasibility of using CD25 monoclonal antibody (Basiliximab) in T-cell undepleted allo-BMT with graft from haplotype-matched related donor for acute GVHD prophylaxis. Twenty-eight patients with leukemia received allo-BMT from HLA two or three loci mismatched related donors. The donors were given G-CSF (Lenograstim) 250 micro g/d for 7 doses prior to marrow harvest. CSA, MTX, ATG and mycophenolate mofetil (MMF) were combined for GVHD prophylaxis. ATG 5 mg/(kg.d) was infused from day 4 to day 1 before transplantation and MMF was administered from day 7. In the study group, the patients received additional CD25 monoclonal antibody for aGVHD prophylaxis. CD25 20 mg each by 30 min intravenous infusion on 2 hours before transplantation and day 4 after transplantation was administered while no application of CD25 in the controls. The outcomes of transplantation were compared between the stud y and control groups. The results showed that the median number of CD34(+) cell in graft was 5.9 x 10(6)/kg in the control group and 7.9 x 10(6)/kg in the study group. The median number of CD3(+) cell was 48 x 10(6)/kg and 52 x 10(6)/kg respectively (P > 0.05). All patients showed 100% donor-typed hematopoietic cells after transplantation by cytogenetic evidence. Five out of fifteen patients in the control group experienced II - IV acute GVHD. While none of thirteen in the study group developed the II - IV acute GVHD. However, none in both groups developed extensive cGVHD. The median follow-up duration was 8 (3-15) months in the study group and 26 (15-36) months in control. In the study group, one patient died from transplant related mortality (CMV infection); no one relapsed; and 12/13 patients survived in disease-free situation within the period of follow-up. In the control group, six patients died from transplant related mortality (3 GVHD, 2 infection and 1 relapsed) and 9/15 patients survived in disease-free situation. The one-year probabilities of disease-free survival (DFS) in two groups were significantly different (P < 0.05). It is concluded that the transplant from haploidentical donors used CD25 antibody is effective and feasible in preventing acute severe GVHD and improving DFS. The major histocompatibility barrier in the haploidentical related allo-BMT could be crossed by donors primed with G-CSF and GVHD prophylaxis with CD25 antibody in the hosts.

[Correlation of bone marrow stromal function in vitro with efficacy of anisodamine therapy for aplastic anemia].

To observe normal bone marrow stromal function for supporting hematopoiesis and realize the correlation of bone marrow stromal function in patients with aplastic anemia (AA) and anisodamine therapy, normal human marrow CFU-GM was assayed on bone marrow stromal layer (SL) formed on week 3 culture, and the CFU-GM without SL as a control (100%). Results showed that the production of CFU-GM on the normal SL was significantly higher than that of the control. The production of CFU-GM on the SL of 4 AA patients, who responded to anisodamine, was < 80% of the control, and when 2 of them were reexamined for stromal function after treatment, the number of CFU-GM on SL rose up to 168.8% and 249.2% from 38.7% and 39.7% of the control respectively. While in the rest 6 patients, the number of CFU-GM was > 80% of the control, only one patient was improved by anisodamine therapy. So, normal bone marrow stromal layer can obviously support the growth of granulocyte/macrophage progenitor cells. Anisodamine therapy could be an effective agents for aplastic anemia with stromal dysfunction

[The relationship between apoptosis of the cardiac myocytes and myocardial dysfunction in severely scalded rats].

OBJECTIVE: To investigate relationship between apoptosis of the cardiac myocytes and myocardial dysfunction in severely scalded rats. METHODS: Wistar rats inflicted by 40% TBSA III degree scalding were employed as the model. The myocardial tissue was obtained from the left ventricle at different postburn time points. Apoptosis was determined by the determination of myocardial tissue Caspase-3 activity and TUNEL staining (terminal deoxynucleotidyl transferase (TdT)-mediated dUTP-biotin nick end labeling). The LVSP (left ventricular systolic pressure), LVEDP (left ventricular end-diastolic pressure) and +dp/dtmax (the rate of the rise of left ventricular pressure) and -dp/dtmax (the rate of the fall of left ventricular pressure) were all monitored by four-channel physiological recorder. In addition, myocardial activities of MPO and SOD were detected. RESULTS: The TUNEL staining of rat myocardial cells was positive at 6 postburn hour (PBH) and reached top level at 12 PBH. The change in Caspase-3 activity was earlier than that of apoptotic morphology and reached peak values at 3 and 6 PBHs. The left ventricular function (systolic and diastolic function) was significantly impaired after the scalding and dropped to the lowest levels at 12 PBHs. The increase in myocardial tissue MPO activity was accompanied by the decrease in SOD activity. CONCLUSION: Myocardial cellular apoptosis was one of the reasons of postburn myocardial injury in scalded rats. Caspase protease cascade pathway might be involved in the process of apoptosis, which suggested that the initiation of apoptosis was closely related to the infiltration of neutrophils and to the release of large amounts of oxygen free radicals in myocardial tissue.

[Effect of jianpi wenshen decoction on serum gastrin, plasma motilin and somatostatin in patients of diabetic diarrhea].

OBJECTIVE: To observe the effect of Jianpi Wenshen Decoction (JWD) on serum gastrin, plasma motilin and somatostatin in patients of diabetic diarrhea (DD). METHODS: Patients with DD were randomly divided into two groups, the JWD group and the control group treated with Loperamide (LPA). Besides, a normal control group was set up. Changes of serum gastrin, plasma motilin and somatostatin were observed. RESULTS: Before treatment, the levels of gastrin and motilin in both groups were higher and somatostatin lower than those in the normal control group. After 1 month treatment, levels of the three indices were restored in both group approaching the normal range with insignificance as compared with those in the normal control group (P > 0.05). Level of plasma motilin and serum gastrin showed an increasing trend along with the therapeutic effect elevation, while level of somatostatin showed a decreasing trend. CONCLUSION: JWD could promote the recovery of the impaired function of vegetative nerve system in DD patients. At the same time, serum gastrin, plasma motilin and somatostatin may be taken as the indexes for evaluating the efficacy in treating DD.

Cathepsin L is essential for embryogenesis and development of Caenorhabditis elegans.

Cysteine proteases play critical biological roles in both intracellular and extracellular processes. We characterized Ce-cpl-1, a Caenorhabditis elegans cathepsin L-like cysteine protease. RNA interference with Ce-cpl-1 activity resulted in embryonic lethality and a transient delayed growth of larvae to egg producing adults, suggesting an essential role for cpl-1 during embryogenesis, and most likely during post-embryonic development. Cpl-1 gene (Ce-cpl-1:lacZ) is widely expressed in the intestine and hypodermal cells of transgenic worms, while the fusion protein (Ce-CPL-1::GFP) was expressed in the hypodermis, pharynx, and gonad. The CPL-1 native protein accumulates in early to late stage embryos and becomes highly concentrated in gut cells during late embryonic development. CPL-1 is also present near the periphery of the eggshell as well as in the cuticle of larval stages suggesting that it may function not only in embryogenesis but also in further development of the worm. Although the precise role of Ce-CPL-1 during embryogenesis is not yet clear it could be involved in the processing of nutrients responsible for synthesis and/or in the degradation of eggshell. Moreover, an increase in the cpl-1 mRNA is seen in the intermolt period approximately 4 h prior to each molt. During this process Ce-CPL-1 may act as a proteolytic enzyme in the processing/degradation of cuticular or other proteins. Similar localization of a related cathepsin L in the filarial nematode Onchocerca volvulus, eggshell and cuticle, suggests that some of the Ce-CPL-1 function during development may be conserved in other parasitic nematodes.

Purification and Characterization of a Platelet Agglutinating Inhibiting Protein (Agkisacutacin) from Agkistrodon acutus Venom.

A platelet agglutinating inhibiting protein (agkisacutacin) was isolated from the venom of Agkistrodon acutus by DEAE Sepharose Fast Flow and size exclusion chromatography. The purified product was a 29 kD protein composed of two disulfide bond-linked polypeptide chains of molecular weight of 14 kD, 15 kD, respectively. It completely inhibited ristocetin-induced platelet agglutination with an IC(50) value of 18.5 mg/L. It also inhibited thrombin-induced platelet aggregation with an IC(50) value of 1.22 g/L, but it did not affect the platelet aggregation induced by collagen and ADP. No fibrinolytic activity, phospholipase A(2) activity, anticoagulant activity, haemorrhagic activity or lethal activity were detected in agkisacutacin. Therefore this protein may offer considerable therapeutic potential in treatment of platelet-rich thrombosis.

Molecular Cloning and Sequence Analysis of cDNA Encoding Acutolysin C, a Hemorrhagic Metalloproteinase, from Agkistrodon acutus.

A full-length cDNA of 1 650 bp was amplified from the snake venom gland cDNA library of Agkistrodon acutus. Analysis of the nucleotide sequence indicated that the amplified cDNA contained a complete open reading frame encoding 417 amino acid residues including signal peptide sequence, zymogen sequence and proteinase domain. The zymogen sequence contained CGVT motif that was highly conserved in almost all venom metalloproteinases. The metalloproteinase domain contained a conserved signature zinc-binding motif HEXXHXXGXXH in the catalytic region and the CIM turn. It shares high similarity with the sequence of acutolysin C deduced from crystallographic data, and with other class P-I snake venom hemorrhagic toxins.

Development of a new mini-invasive tumour hyperthermia probe using high-temperature water vapour.

A new mini-invasive hyperthermia probe using high-temperature water vapour for deep regional tumour treatment was developed in this paper. The vacuum insulation mechanism was incorporated into the probe to avoid heating damage to the normal tissues around the edge of the insertion path. To better understand the heat transfer behaviour in living tissues due to operation of the probe, theoretical models based on the Pennes' equation were established and two closed form analytical solutions under constant flux or temperature heating at the tip of probe were obtained. Parametric studies were performed to investigate the influence of various parameters on the temperature response of tissues heated by the probe. Further, several simulating experiments on the actual heating performance of the probe fabricated in this paper were conducted on the in vitro biological materials (fresh pork) and phantom gel. It was demonstrated that the probe can cause a high enough temperature over the treatment area to thermally destroy the tumour tissue in due time, while the temperature over the surrounding healthy tissues can be kept below a safe threshold value. This mini-invasive heating probe may have significant applications in future clinical tumour hyperthermia.

Attachment status in children prenatally exposed to cocaine and other substances.

Attachment status of children exposed in utero to cocaine, opiates, and other substances was examined at 18 months (n=860) and 36 months (n=732) corrected age. Children exposed to cocaine and opiates had slightly lower rates of attachment security (but not disorganization), and their insecurity was skewed toward ambivalent, rather than avoidant, strategies. Continued postnatal alcohol use was associated with higher rates of insecurity and disorganization at 18, but not 36, months of age. Stability of attachment across the 18-month period was barely above chance expectation. Attachment status at 18 months was associated with child temperament and caregiver-child interaction; at 36 months, attachment was associated with child temperament, child behavior problems, and caregivers' parenting self-esteem.

Use of mailed urine specimens in diagnosing urothelial carcinoma by cytology and DNA image analysis.

OBJECTIVE: To evaluate the usefulness of urine specimens collected via a mailer system and analyzed by cytology and DNA ploidy for the detection of urothelial carcinoma (UC). STUDY DESIGN: We retrospectively reviewed the diagnoses of 91 mailed urine specimens received from 72 patients, 67% of whom had a history of UC. The specimens were fixed in an equal volume of 50% ethanol solution before being mailed. The cytologic findings were interpreted in conjunction with DNA ploidy image analysis. We compared these initial diagnoses with those of follow-up examinations, including biopsies, cystoscopic findings and urinary cytology/DNA ploidy analyses. In addition, to examine the quality of the mailed samples, 3 cytopathologists performed a blinded assessment of cytologic slides of 20 mailed and 17 fresh urinary samples for bacterial overgrowth, urothelial degeneration, and presence of proteinaceous material and crystals. RESULTS: Follow-up was available for 68 of the 91 mailed specimens. The sensitivity for detecting UC using mailed urine specimens that were analyzed by both cytology and DNA ploidy was 61%, while specificity was 92%. The levels of bacterial overgrowth and urothelial degeneration in the mailed specimens were not significantly greater than in the fresh specimens (p>0.05). The levels of proteinaceous material and crystals were significantly higher in the mailed specimens (p<0.05). CONCLUSION: The results of combined cytology and DNA ploidy image analysis by using mailed urine samples were comparable to those of fresh urine specimens for the detection of UC reported in previous publications. The increase in crystals and proteinaceous material did not impede diagnostic interpretation. The mailing system is a reliable and convenient method of monitoring and triaging patients with UC or related symptoms.

Thyroid transcription factor-1 immunocytochemical staining of pleural fluid cytocentrifuge preparations for detection of small cell lung carcinoma.

OBJECTIVE: To assess whether thyroid transcription factor-1 (TTF-1) immunostaining of cytocentrifuge slides previously stained by the Papanicolaou technique is feasible and useful in diagnosing small cell lung carcinoma (SCLC) in pleural fluid. STUDY DESIGN: Eleven Papanicoalou-stained cytocentrifuge preparations of pleural fluid from 11 patients with SCLC were reviewed. The initial cytologic diagnoses were 5 positive, 1 suspicious and 5 negativefor SCLC. After transferring the cytologic material to a positively charged slide, immunostaining was performed using mouse monoclonal antibody against TTF-1. RESULTS: The neoplastic cellularity varied among the single suspicious and 5 positive cases. The case with a suspicious diagnosis had scant tumor cells. All 6 cases showed nuclear stainingfor TTF-1 in most of the tumor cells. The other 5 cases with a negative cytologic diagnosis were negativefor TTF-1. CONCLUSION: The results of this study indicate that immunostaining for TTF-1 can be successfully applied to previously Pap-stained cytocentrifuge preparations to assist in the diagnosis of SCLC in pleural fluid.

Addition of cryoprotective agent into biological tissue through minimally invasive injection.

In this article, a minimally invasive injection method to enhance the addition of cryoprotective agent into the target biological tissues or organs to be cryopreserved was studied. Compared with the traditional treatment through immersion and permeation, direct injection could be much more effective and faster. To illustrate the benefit of this approach, a mathematical model for the addition of cryoprotective agent was established and experiments were performed to test the differences given by the two methods. Both the theoretical and the experimental results demonstrate that the minimally invasive injection method for enhancing the addition of cryoprotective agent is a promising substitution for the commonly used immersing method.
