Downregulation of human endothelial nitric oxide synthase promoter activity by p38 mitogen-activated protein kinase activation.

Human endothelial nitric oxide synthase (eNOS) plays a crucial role in maintaining blood pressure homeostasis and vascular integrity. eNOS gene expression may be upregulated by a signaling pathway, including PI-3Kgamma--> Jak2--> MEK1 --> ERK1/2--> PP2A. It remains unclear whether other mitogen-activated protein kinase (MAPK) family members, such as JNK, p38 kinase, and ERK5/BMK1, also modulate eNOS gene expression. Our purpose, therefore, is to shed light on the effect of the p38 MAPK signaling pathway on the regulation of eNOS promoter activity. The results showed that a red fluorescent protein reporter gene vector containing the full length of the human eNOS promoter was first successfully constructed, expressing efficiently in ECV304 cells with the characteristics of real time observation. The wild-types of p38alpha, p38beta, p38gamma, and p38delta signal molecules all markedly downregulated promoter activity, which could be reversed by their negative mutants, including p38alpha (AF), p38beta (AF), p38gamma (AF), and p38delta (AF). Promoter activity was also significantly downregulated by MKK6b (E), an active mutant of an upstream kinase of p38 MAPK. The reduction in promoter activity by p38 MAPK could be blocked by treatment with a p38 MAPK specific inhibitor, SB203580. Moreover, the activation of endogenous p38 MAPK induced by lipopolysaccharide resulted in a prominent reduction in promoter activity. These findings strongly suggest that the activation of the p38 MAPK signaling pathway may be implicated in the downregulation of human eNOS promoter activity.

Potentiation of adenosine A1 receptor agonist CPA-induced antinociception by paeoniflorin in mice.

The effect of paeoniflorin (PF), a major constituent isolated from Paeony radix, on N6-Cyclopentyladenosine (CPA), a selective adenosine A1 receptor (A1 receptor) agonist, induced antinociception was examined in mice. In the tail-pressure test, CPA (0.05, 0.1, 0.2 mg/kg, s.c.) could induce antinociception in a dose-dependent manner. PF (5, 10, 20 mg/kg, s.c.) alone failed to exhibit any antinociceptive effect in mice; however, pretreatment of PF (20 mg/kg, s.c.) could significantly enhance CPA-induced antinociception. Additionally, pretreatment of 8-Cyclopentyl-1,3-dipropylxanthine (DPCPX, 0.25 mg/kg, s.c.), a selective A1 receptor antagonist, could antagonize the antinociceptive effect of combining CPA with PF. Furthermore, in the competitive binding experiments, PF did not displace the binding of [3H]-8-Cyclopentyl-1,3-dipropylxanthine ([3H]-DPCPX) but displaced that of [3H]-2-Chloro-N6-cyclopentyladenosine ([3H]-CCPA, a selective A1 receptor agonist) to the membrane preparation of rat cerebral cortex. These results suggested that PF might selectively increase the binding and antinociceptive effect of CPA by binding with A1 receptor.

Maternal antenatal administration of vitamin K1 results in increasing the activities of vitamin K-dependent coagulation factors in umbilical blood and in decreasing the incidence rate of periventricular-intraventricular hemorrhage in premature infants.

AIMS: Infants less than 35 weeks' gestation age are susceptible to periventricular-intraventricular hemorrhage (PIVH). This may be partially attributable to low concentrations of vitamin K-dependent coagulation factors. The purposes of this study were: (1) to determine the umbilical blood activity levels of vitamin K-dependent coagulation factors II, VII, IX and X; (2) to investigate the change in activities of these factors in premature infants' umbilical blood after prenatal administration of vitamin K1 to the mothers; and (3) to study the prophylactic effects on PIVH after maternal antenatal supplemental vitamin K1. METHODS: Pregnant women in preterm labor at less than 35 weeks of gestation were randomly selected to receive antenatal vitamin K1 10 mg per day injection intramuscularly or intravenously for 2-7 days (vitamin K1 group, n = 40), or no such treatment (control group, n = 50). At the same period, cord blood samples were collected from thirty full-term neonates to compare the factor levels with those of premature infants. Intracranial ultrasound was performed by the same sonographer to determine the presence and severity of PIVH. RESULTS: The activities of vitamin K-dependent coagulation factors in umbilical blood in the control group were: factor II 25.64+/-9.49%, factor VII 59.00+/-17.66%, factor IX 24.67+/-8.88%, and factor X 30.16+/-5.02%. In full-term infants, the respective values were: factor II 36.70+/-4.88%, factor VII 64.54+/-10.62%, factor IX 30.18+/-5.69%, and factor X 34.32+/-12.63%. In vitamin K1 group these factors were: factor II 36.35+/-6.88%, factor VII 69.59+/-16.55%, factor IX 25.71+/-10.88%, and factor X 39.26+/-8.02%. The data suggest the absence of vitamin K-dependent coagulation factors in preterm infants, and antenatal supplement of vitamin K1 may increase the cord blood activity of factor II, VII and factor X (P < 0.001). In addition, the overall rates of PIVH in the vitamin K1 group and in controls were 32.4 and 52.0%, respectively (P = 0.036), and the frequency of severe PIVH was 5.0 and 20.0%, respectively (P = 0.038). CONCLUSIONS: Administration of vitamin K1 to pregnant women at less than 35 weeks' gestation age may result in improved coagulation and may reduce the incidence as well as the severity degree of PIVH.

Understanding the neural correlates of visual perception: a survey of our toolbox.

One of the central goals of neuroscience is to elucidate the link between behavior and neuronal activity at both single neuron level and the level of neuronal ensembles. New analytical approaches and techniques constantly emerge that correlate neural activity with perception and behavior with ever finer temporal and spatial scales. In this article I review some of these approaches employed in studies of visual perception in which perceptual reports were required from subjects. I divide these methods into 'correlational' methods and 'perturbation' methods; the former measures normal neural activity and correlates it with behavior, the latter perturbs normal neural activity to cause behavioral alterations. I compare such approaches applied to primates with those applied to human subjects, and discuss how these approaches could be combined and made more powerful in the future.

The infant development, environment, and lifestyle study: effects of prenatal methamphetamine exposure, polydrug exposure, and poverty on intrauterine growth.

OBJECTIVE: Methamphetamine use among pregnant women is an increasing problem in the United States. Effects of methamphetamine use during pregnancy on fetal growth have not been reported in large, prospective studies. We examined the neonatal growth effects of prenatal methamphetamine exposure in the multicenter, longitudinal Infant Development, Environment and Lifestyle study. DESIGN/METHOD: The Infant Development, Environment and Lifestyle study screened 13808 subjects at 4 clinical centers: 1618 were eligible and consented, among which 84 were methamphetamine exposed, and 1534 were unexposed. Those who were methamphetamine exposed were identified by self-report and/or gas chromatography-mass spectrometry confirmation of amphetamine and metabolites in infant meconium. Those who were unexposed denied amphetamine use and had a negative meconium screen. Both groups included prenatal alcohol, tobacco, or marijuana use, but excluded use of opiates, LSD, PCP or cocaine only. Neonatal parameters included birth weight and gestational age in weeks. One-way analysis of variance and linear-regression analyses were conducted on birth weight by exposure. The relationship of methamphetamine exposure and the incidence of small for gestational age was analyzed using multivariate logistic-regression analyses. RESULTS: The methamphetamine exposed group was 3.5 times more likely to be small for gestational age than the unexposed group. Mothers who used tobacco during pregnancy were nearly 2 times more likely to have small-for-gestational-age infants. In addition, less maternal weight gain during pregnancy was more likely to result in a small-for-gestational-age infant. Birthweight in the methamphetamine exposed group was lower than the unexposed group. CONCLUSIONS: These findings suggest that prenatal methamphetamine use is associated with fetal growth restriction after adjusting for covariates. Continued follow-up will determine if these infants are at increased risk for growth abnormalities in the future.

Endomorphins restored the endothelium-dependent relaxation of the rabbit aorta rings exposed to high D-glucose condition via NO-cGMP pathway.

Rings of rabbit aorta that were both incubated in a high concentration of D-glucose and contracted submaximally by phenylephrine showed significantly decreased endothelium-dependent relaxations induced by acetylcholine. The cGMP production of aorta rings was also reduced. Treatment with endomorphins (1-1000 nmol/L) restored acetylcholine-induced relaxations of aorta rings incubated in high glucose concentrations and increased the cGMP synthesis. Moreover, this effect of endomorphins on endothelium was antagonized by naloxone, and the increase in the production of cGMP was also blocked.

[Simultaneous control of microorganism, disinfection by-products and bio-stability by sequential chlorination disinfection].

A new safe chlorination disinfection process, the sequential disinfection process with short-term free chlorine plus chloramines was investigated its efficiency on control of indices of microorganism, disinfection by-products and bio-stability. This disinfection process may take the advantage of free chlorine's quick inactivation of microorganism and chloramines' low DBP yield and long-term residual. Thus, both control of microorganisms and DBPs can be achieved effectively and economically. The pilot test results show that safe chlorination disinfection process has a little better efficiency on total bacteria count, HPC and total coliform count than free chlorination disinfection, which indicate that free chlorine and chloramines might have the synergetic disinfection effect for their different attacking targets. It also had the same inactivation efficiency on polio virus and coliphage f2 as free chlorination in 120 minutes contact time. Sequential chlorination disinfection process could reduce 35.8% - 77.0% yield of trihalomethane and 36.6% - 54.8% yield of haloacetic acids. The poorer quality the water had, the more advantage the safe-chlorination disinfection had than free chlorination disinfection one. Moreover, sequential chlorination had better efficiency on bio-stability control than free chlorination.

[Serum total cholesterol and 10-year cardiovascular risk in a Chinese cohort aged 35-64 years].

OBJECTIVE: To evaluate the association between serum total cholesterol (TC) and the risk of cardiovascular diseases (CVD) in subjects aged 35-64 years. METHODS: This prospective study was performed from 1992 to 2002 in 11 provinces of China and the association between baseline TC level and the development of cardiovascular diseases was analyzed in 30 384 subjects aged 35-64 years using Cox multivariate proportional hazards regression. RESULTS: (1) Compared with the group of TC < 3.64 mmol/L (140 mg/dl), multivariate-adjusted relative risk of ischemic cardiovascular disease (ICVD, including CHD and ischemic stroke) increased continuously with increasing TC level. (2) TC level was positively related to the occurrence of ischemic stroke, but not to hemorrhagic stroke. The risk of CHD increased by 74% (RR = 1.743) and the risk of ischemic stroke increased by 12% (RR = 1.119) in the group with TC > or = 5.72 mmol/L compared with the group with TC < 5.72 mmol/L (220 mg/dl). (3) High serum cholesterol was responsible to 5.9% of ICVD, 11.7% of the acute CHD and 2.9% of the acute stroke. (4) At each TC level, the 10-year risk of ICVD increased in the presence of other risk factors. CONCLUSION: Starting from TC of 3.64 mmol/L, the risk of ICVD increases continuously with increasing TC level. Multiple risk factor intervention should be strengthened to reduce the overall risk of CVD.

[Variant genotyping of CCR2-64I, SDF1-3' A and CCR5Delta32 in HIV-1 infected Chinese long-term nonprogressors].

BACKGROUND: To investigate variant genotyping of CCR2-64I, SDF1-3'A and CCR5Delta32 in HIV-1 infected Chinese Long-term nonprogressors and to study their association with disease progression. METHODS: The genotypes of CCR2-64I, SDF1-3'A and CCR5Delta32 were detected by polymerase chain reaction/restriction fragment length polymorphism (PCR/RFLP) assay in seventeen HIV-1 infected Chinese Long-term nonprogressors (LTNPs) and thirty-nine Chinese typical progressors (TPs). RESULTS: The frequency of CCR2-64I and SDF1-3'A in LTNPs are 50% and 62.5%, higher than those (23.08% and 33.33%) in TPs. Only one heterozygous CCR5 mutant was detected in LTNPs, and no CCR5 mutant in TPs. CONCLUSION: Variant genotyping of CCR2-64ISDF1-3'A and CCR5Delta32 may be protective factors for delaying disease progression in HIV-1 infected Chinese LTNPs.

[Study on the optimal ratio of autoskin and alloskin during mixed microskin grafting].

OBJECTIVE: To investigate the influence of different amount of allogeneic microskin in mixed grafting with certain quantity of autologous microskin on wound healing in rats. Methods Male Wistar rats served as alloskin donor rats. Forty female SD rats with full thickness skin defect were enrolled in the study, and they were randomly divided into four groups, i.e. group I (n=10, with allogeneic microskin graft at area expansion rate of 10:3); group II (n=10, with autologous microskin graft at area expansion rate of 10:1); group III (n=10, with mixed grafting of autologous and allogeneic microskin at area expansion rate of 10:1, respectively); group IV (n=10, with mixed grafting of autologous and allogeneic microskin at area expansion rate of 10:1 and 10:3, respectively). The wound healing rate, wound contraction rate and histological changes were observed at the 2, 3 and 4 post graft weeks (PGW). RESULTS: (1) In group I, there was mainly granulation tissue with some de novo epithelial cells appearing at the wound edge along with the rejection of grafted allogenous skin in the rat wound. In group II, there was still some granulation tissue remaining at 2 PGW due to insufficient amount of microskin. However, the wounds in the mixed grafting group appeared almost totally epithelialized. (2) Various amounts of mononuclear inflammatory cell infiltration and different degrees of angiectasis were observed in the dermal layer after the skin grafting in all groups, especially in group II and IV. There was thickening of the epithelial layer in all groups except group I. (3) The wound healing rate decreased obviously along with the development of rejection in group I at 2 to 4 PGWs. The wound healing rate was (55 +/- 26)% in group II, which was obviously lower than that in group III (88 +/- 6)% and in group IV (76 +/- 10)% at 3 PGWs (P < 0.01). (4) The contraction rate of the wound in group IV (69 +/- 7)% was much higher than that in group I (58 +/- 11)% at 3 PGWs (P < 0.05), and there was no difference among all the other groups. CONCLUSION: Wound healing can be obviously accelerated by mixing some autologous microskin with appropriate amount of alloskin. Moreover, certain amount of autologous microskin (expansion rate 10:1) mixed with the same proportion of allogeneic microskin seems to be more beneficial in promoting wound healing.

[Analysis of promoter sequence and its activity of homologues gene rad25 of eukaryotes from halophilic archaea].

The transcriptional product in Halobacterium halobium R1 similar to eukaryotic gene rad25 was analyzed by RT-PCR. Using bgaH as the reporter genes, the promoter function of eukaryotic rad25-like DNA fragment in halophilic archaea was investigated by promoter probe analysis. The important functional regions, which could influence the promoter activity of rad25-like gene, were identified by deletion analysis of the promoter sequence in Haloferax volcanii. It is found that the DNA fragment of promoter similar to eukaryotic gene rad25 contains the typical characteristic sequence of archaeal promoter. These results indicate that rad25-like gene in Halobacterium halobium R1 is active and may play a role on the NER pathway as the eukaryotic pattern.

[Effects of electroacupuncture on GDNF positive cell immunoreactivity in local dermal tissue of the inflammatory pain focus in the rat of adjuvant arthritis].

OBJECTIVE: To study on the analgesic mechanism of electroacupuncture (EA) in the rat of adjuvant arthritis (AA). METHODS: Forty-eight SD rats were randomly divided into a blank control group (n = 8), an inflammatory group (n = 10), an acupoint EA group (n = 10), a non-acupoint EA group (n = 10) and a contralateral acupoint EA group (n = 10). Complete Freund's adjuvant (CFA) 50 microL were injected into the left malleolus' articular cavity in the rats except the blank control group for preparing single local adjuvant arthritis (AA) model. EA was given every other day to the acupoint EA group, the non-acupoint EA group and the contralateral acupoint EA group. The improving effects of EA at "Huantiao" (GB 30) and "Yanglingquan" (GB 34) on the dorsal flexion pain score and the swell of dorsum of hind paw were investigated, and effects of EA on Glial cell line-derived neurotrophic factor (GDNF) positive cells immunoreactivity in the inflammatory tissue of the AA rats on the 14th day after injection of adjuvant were observed with immunohistochemical technique. RESULTS: Hyperalgesia and local swell, and GDNF in the dermal and subcutaneous tissue around the inflammatory ankle joint in the inflammatory groups were significantly higher than those in the blank control group. EA on the ipsilateral and contralateral acupoints reduced the pain, promoted the recovery of swell, and decreased the positive area percentage and mean optical density of GDNF positive cells in the dermal and the subcutaneous tissues. However, the non-acupoint EA group did not have this action. CONCLUSION: EA can regulate expression of GDNF in local dermal tissue of the inflammatory focus in the AA rat, so as to exert the anti-inflammatory and analgesic effects.

[A new method for EMSA by modifying DIG high prime DNA labeling and detection starter Kit II].

Gel retardation, also named electrophoretic mobility shift assay (EMSA), is a useful tool for identifying protein-DNA interactions. Typically, 32P-labeled DNA probes used in EMSA are sensitive. However, it relies on the handling of hazardous radioisotopes, and is not easily quantified. Recently, some successful cases have been reported using non-radio labelled probes instead of radiolabelled probes in EMSA. The method is rapid, convenient, and safe, but it depends on a very expensive kit. In this study, we offered a new method performing EMSA by modifying DIG High Prime DNA Labeling and Detection Starter Kit II (Rohe). Firstly, the prepared labeled probe was introduced the EcoR I stick in the end of probe for 3'-end labeling, and then was performed the probe labeling and detecting the signals of EMSA with the relatively cheap DIG High Prime DNA Labeling and Detection Starter Kit II Rohe. By adjusting the experiment parameters, the successful result was obtained. The present study provides a successful example and method for modifying DIG High Prime DNA Labeling and Detection Starter Kit II.

[Pilot-plant testing for flue gas desulfurization and dust removal by activated coke].

A developed flue gas desulfurization and dust removal process with a cross-flow moving bed filled with activated coke (AC) was tested on a pilot scale with the gas treatment capacity of 1000 mg x h(-t). The results show that a easy-operating and stable-running pilot-scale testing equipment can be reached, and under the conditions of testing space velocity, the feed gas temperature of 120 degrees C, SO2 concentrations of 3232-6006 mg x m(-3) and dust concentration of 89.3-1599.7 mg x m(-3), the high efficiency of flue gas desulfurization and dust removal is reached with the purity of dust less than 50 mg x m(-3). In the technology, the spent AC was regenerated by heating, and the SO2 concentration in the desorption gas is about 40.1%, which can be efficiently recovered as a sulfur resource. Favorable economy benefit can be reached by using the process.
