[Producing human lactoferrin by high-density fermentation recombinant Pichia pastoris].

BACKGROUND: To evaluate expression of human lactoferrin gene by high-density fermentation in recombinant Pichia pastoris on the premise of maintaining its biological activities. METHODS: The neutrophil was isolated from human peripheral blood and its total RNA was prepared. Full-length cDNA of human lactoferrin gene was then obtained by RT-PCR, cloned into expression vector pPIC 3.5 K and transformed into Pichia pastoris strain KM71. With two-layer filter method, the transformants with high-productivity of human lactoferrin were screened out into fed-batch high-density fermentation. And later, the physical, chemical and biological activities of fermentation product were detected preliminarily. RESULTS: The strain p3.5-k-7 with better productivity of human lactoferrin was screened out into fed-batch high-density fermentation. The fermentation lasted nearly for nine days, with A-600 of culture once above 260 and the highest productivity of human lactoferrin being 115 mg/L, 7.67 times the amount of that in shake flask cultivation. CONCLUSION: The authors successfully realized high-density fermentation expression of human lactoferrin gene in recombinant Pichia pastoris.

[Immunoglobulin variable region gene rearrangement and hypermutation in paraneoplastic pemphigus associated Castleman's tumor].

OBJECTIVE: We have studied the role of lymphoproliferative tumors in the pathogenesis of autoimmune and the origin of the autoantibodies in paraneoplastic pemphigus (PNP) in recent years. A Castleman's tumor from a patient was identified to produce autoantibody. To identify the relationship between the tumor and pathogenesis of the disease, we analyzed the rearrangement of immunoglobulin variable region gene and its hypermutation in B cells of Castleman's tumor from a patient who was diagnosed of paraneoplastic pemphigus. METHODS: The surface markers of cultured tumor lymphocytes were assessed with immunochemistry staining. After total RNA of the tumor cells were isolated, the mRNA was reversely transcribed into cDNA. V(H) and V(L) genes were cloned and their sequences were analyzed. RESULTS: Immunochemistry staining and flow cytometer analysis showed that the tumor cells were CD20, HLA-DR, smIgM, and smIgG positive. The cloned IgV(H) and IGHV3-9*01 germ-line gene are homologous and so are the Ig V(L) and the IGKV4-1*01 germ-line gene. More nucleotide changes in the V(H) or V(L) occurred in CDRs than those in FRs. CONCLUSION: In this reported case, a clone of specific B-lymphocyte in the Castleman's tumor carrying functional rearranged immunoglobulin heavy and light chain genes was found to have experienced switch recombination and was possible to produce IgG autoantibody.

[The new development on pathogenesis of autoimmune paraneoplastic pemphigus].

Paraneoplastic pemphigus (PNP) is an autoimmune mucocutaneous disease first named by Anhalt, et al in 1990. The disease is characterized by such distinctive clinical symptoms and signs as severe, painful mucosal erosions, polymorphous skin lesions, histopathology hallmarks, and immunological findings. The situation typically presents in patients with lymphoproliferative diseases and primarily malignancies. A main challenge of the study is the relationship between the existence of associated tumors and the autoimmune reaction to the skin. Some researchers suspected that the possible expression of foreign antigens on the tumor can cross react with epidermal antigens inducing the auto-reactive clones of T-lymphocytes. Some speculated that the type of tumors associated with PNP may produce plakin proteins that result in initiation of the immune response. Other reports believed the autoimmune reaction is related to the epitope spreading or to the changing of cytokines. We analyzed 12 PNP patients diagnosed in our department in the past few years. An intensive study to the B cells in the PNP associated with tumors demonstrated that the tumors have structural basis to produce antibody. The similar immunoglobulin heavy chain genes of tumor B cells in 7 patients strongly suggested that the B cell clones were functional and recognized the same antigen epitope. The autoantibodies secreted by the tumor can react against specific plakin proteins in epidermis, lead to the impairment of cell-cell adhesion, and cause the mucocutaneous lesions. The clinical significance of the RESULTS indicates the importances of early finding and total resection of the associated tumors, and the usage of IVIG pre or during operation to prevent Bronchitis Obliterans. The new finding is also important for the study of other antibody mediated autoimmune diseases.

[Study on cytokeratin and CA125 for diagnosing lymph node micrometastases in endometrial cancer].

OBJECTIVE: To study the roles of cytokeratin (CK) and CA(125) in diagnosing lymph node micrometastasis in endometrial cancer. METHODS: The expressions of CK and CA(125) in 50 primary tumors and 298 lymph nodes from 50 patients with endometrial cancers were analysed by immunohistochemical methods. RESULTS: (1) The positive expression rates of CK and CA(125) in primary tumors of cases with endometrial cancer were respectively 100%, and 78%. (2) The expressions of CK and CA(125) in metastatic lymph nodes were both strong with 100% positive rate. In lymph nodes without metastasis, the expressions of CK and CA(125) were both weak, with positive rates of 15% and 12% respectively. (3) Among patients with stage I, II diseases, tumor recurrence rate was significantly higher in patients with positive CK or CA(125) expression in lymph nodes than in patients without CK or CA(125) expression (P < 0.05). (4) Multiple regression analysis revealed that in stages I, II endometrial cancer, CK expression in lymph nodes, and CA(125) expression in lymph nodes together with depth of myometrial invasion were relevant factors with tumor recurrence. CONCLUSIONS: CK expression in lymph nodes without metastasis can predict lymph node micrometastasis and is an independent risk factor for recurrence of disease in stages I, II endometrial cancers. CA(125) expression in lymph nodes without metastasis can also suggest lymph node micrometastasis, but it is not an independent predictive factor for tumor recurrence in stages I, II endometrial cancers.

Real-time three-dimensional echocardiography for quantifying left ventricular mass.

OBJECTIVE: To test the accuracy of real-time three-dimensional echocardiography (RT3DE) imaging system for evaluating left ventricular mass (LVM) in phantom and excised canine heart. METHODS: Ten left ventricular (LV) wall phantoms made of two rubber-bursas, ten excised canine hearts underwent RT3DE and two-dimensional echocardiography (2DE). In RT3DE "full volume" imaging, the myocardial volume was measured using 2, 4, 8, and 16-plane method with the analysis software of RT3DE. Mass was then calculated by multiplying the resulting myocardial volume by specific density of myocardial tissue. In 2DE the masses were measured by area-length method. The true LV wall phantom mass was measured by water displacement and the canine LVM was weighed by anatomy, which served as a reference standard. We compared RT3DE or 2DE with true mass. RESULTS: In LV wall phantoms, RT3DE correlated with true masses strongly (r = 0.813-0.994) and weakly correlated between 2DE and true masses (r = 0.628). In excised canine hearts, there is an excellent correlation between RT3DE and true masses (r = 0.764-0.991), while 2DE value showed a lesser correlation (r = 0.514). There are no difference between RT-3DE and true masses (P > 0.05) but different between 2DE and true masses (P < 0.05). In different planes, there was no difference between 8-plane and 16-plane (P > 0.05) but different between 8-plane and 2, 4-plane (P < 0.05). CONCLUSION: RT3DE can accurately quantify LVM and provide a new tool to evaluate LV function. For LVM by RT3DE, 8-plane measurement method is the best choice for accuracy and convenience.

[Electrochemical behavior of daunorubicin at Co/GC ion implantation modified electrode and the application of the electrode].

AIM: To study the electrochemical behavior of daunorubicin at Co/GC ion implantation modified electrode. METHODS: With Co/GC ion implantation modified electrode as working electrode, daunorubicin was determined by voltammetry in 0.05 mol x L(-1) Na2HPO4-KH2PO4 (pH 6.82) solution. RESULTS: A sensitive reductive peak of daunorubicin was obtained by linear sweep voltammetry. The peak potential was -0.60 V (vs SCE). The peak current was proportional to the concentration of daunorubicin over the range of 2.84 x 10(-8) - 1.42 x 10(-6) mol x L(-1) and 1.42 x 10(-6) - 1.28 x 10(-5) mol x L(-1) with the detection limit of 1.42 x 10(-8) mol x L(-1). The reduction wave was applied to the determination of daunorubicin. The electrochemical behavior and reaction mechanism were studied by linear sweep and cyclic voltammetry. CONCLUSION: The reduction process was quasi-reversible with adsorption characteristics.

Surfactants, polymers and their nanoparticles for personal care applications.

A "touch me not" plant folding up rapidly upon being attacked or microbes depositing on teeth or ocean vessels even under hostile conditions are examples in nature that provide inspiration for developing new classes of personal care release or deposition systems. In this paper, development of such systems based on polymer/surfactant colloid chemistry is explored for achieving transport and release of cosmetic and pharmaceutical molecules at desired rates at desired sites. The successful development of products depends upon understanding and utilizing key interactions among surfactants, polymers and hybrid polymers that are relevant to personal care products. Thus, the absorbed layers or tethers on the particulates can be manipulated for desired dispersion of actives or depositions on substrate under any and all conditions. New hybrid polymers and nanogels have been synthesized for tuning up nanodomains that can extract and deliver at will cosmetics/drugs/toxins by perturbing pH, temperature or ionic strength of the system. Particularly, hydrophobically modified polymers have features of both polymers and surfactants and due to the associative nature of the hydrophobic groups, such polymers can form intramolecular nanodomains for performing carrier functions. Nanogels developed recently include that of polyacrylamide, poly(acrylic acid) and starch nanogels modified for extraction and subsequent slow release of fragrances and overdosed toxic drugs. Binding and release processes were investigated using surface plasmon resonance and fluorescence spectroscopies, powerful techniques for monitoring short term and long term changes.

[Detection of a novel mutation in COL4A5 gene from a Chinese family with X-linked alport syndrome].

By means of PCR and direct sequencing, all 51 exons and their neighbouring intronic sequences of the COL4A5 gene were analyzed to detect mutations in 17 members from a Chinese family with X-linked Alport syndrome(XLAS). At the position 2240 in exon 26, a single-base deletion(2240deIC) is found in all male patients, and a heterozygous deletion is found in all female patients, whereas no mutation is found in normal and 80 control individuals. Meanwhile, the corresponding PCR products of female patients are cloned and sequenced to confirm the results. It is concluded that the 2240deIC mutation is the underlying cause of XLAS in this family, not a polymorphism. Furthermore, this single-base deletion mutation in COL4A5 gene is first reported in X-linked Alport syndrome.

PURPOSE: To compare and evaluate the efficacy, safety and stability of extreme myopia correction with Phakic Posterior Chamber Intraocular Lens (PPCIOL) Keratomileusis (Lasik). and Laser in situ METHODS: This prospective study enrolled 43 cases of extreme myopia. The PPCIOL group was inserted Implantable Contact Lens (ICL) and Lasik group was done excimer laser scanning. The efficacy index and safety index were compared. The rate of regression postoperative was observed and the changes of low contrast vision and Glare vision were statistically studied. RESULT: The mean spherical equivalent refraction of PPCIOL was (-16.77+/-3.37)D(-11.75 approximately -25.75 D)and that of Lasik was (-13.8+/-2.71) D (-9.37 approximately -23.75 D). There was no statistically difference between efficacy index of two groups postoperatively and a significant higher safety index in PPCIOL(P < 0.001) . The rate of regression was 0 in PPCIOL group and 32.56% in Lasik group (P< 0.001). The increase of low contrast vision and Glare vision from the baseline preoperative was more significant in PPCIOL group than which in Lasik group (P < 0.001). Two eyes of 1 PPCIOL case had anterior capsular cataract in 6 months postoperative. No other complication was found in two groups. CONCLUSION: PPCIOL implantation is an efficient method for extreme myopia correction with more safety and stability, and provides good visual quality.

[Diurnal variation of winter wheat water and heat fluxes of a simulation with photosynthesis-evapotranspiration coupled model].

A coupled model of winter wheat photosynthesis-evapotranspiration was established based on SPAC theory. Sensible heat and latent heat fluxes were calculated by two-layer model proposed by Shuttleworth and Wallace, and photosynthesis and evapotranspiration were coupled by the parameterization of canopy resistance. The model was validated with the data measured by eddy covariance method. The results showed that the simulated and observed values were accordant, and the model could simulate the diurnal variation of sensible heat and latent heat fluxes very well. Sensitivity analysis indicated that the sensitive parameters of canopy transpiration were wilting point, stomata conductance, reflectivity of leaves to infrared radiation, and convexity of photosynthesis response to light, while the sensitive parameter of soil evaporation was soil resistance. The model could be used to study the interactions between water and heat fluxes and environmental factors, and to instruct the irrigation scheme in the field.

[A rapid and nondestructive analysis of red wines of different manufacturers].

Alcohol and sugar are two main components in wine. We applied ATR Fourier Transform Infrared (FTIR) spectra and second derivative spectra to analyze the alcoholicity and content of sugar of red wines produced by different manufacturers for the first time. For wines with less sugar, the intensity of absorption peaks assigned to C-O bond at 1120-1000 cm(-1) becomes stronger when the alcoholicity is higher. For wines with more sugar, we can confirm the content of sugar by the intensity of absorption peaks and the number of peaks during the range from 1200 to 830 cm(-1), which includes many the characteristic absorptions of sugar. FTIR spectra not only can show clearly the relationship between the intensity of peaks and content of alcohol and sugar, but also are so rapid and convenient that it can be used to analyse samples directly.

Preliminary study on mouse interleukin-21 application in tumor gene therapy.

Interleukin-21 (IL-21) is a recently characterized T cell-derived cytokine with a significant homology to IL-2, IL-4 and IL-15. To determine whether IL-21 has broad immunoregulatory activity and can stimulate durable antitumour responses, we constructed mouse IL-21 (mIL-21) recombinant plasmid and evaluated its antitumor efficacy. Mouse IL-21 cDNA was amplified from Con A-activated mouse T cells by RT-PCR. Recombinant pcDNA3.1/mIL-21 was constructed and transfected into Sp2/0 cells. Mouse IL-21 expression was analyzed by Western blotting and its activities were detected by 3H-TdR incorporation and MTT assay. The recombinant pcDNA3.1/mIL-21 was injected s.c. into tumor lump. Tumor size, weight, the activities of CTLs, NK cells and LAK cells and serum IFN-gamma level were measured for evaluating mIL-21 mediated antitumor responses. The results indicated that mIL-21 was correctly expressed in Sp2/0 cells and it can improve the proliferation of T cells and B cells, and enhance NK cytotoxic activity in vitro. The activities of CTL and NK cells, and serum IFN-gamma level were significantly improved, furthermore the tumor growth was obviously suppressed in pcDNA3.1/mIL-21 treated mice. However, the LAK activity did not alter significantly. Taken together, this study suggests that the injection with recombinant plasmid containing mIL-21 is a potential approach for tumor gene therapy.

Gender-specific pattern of insulin-like growth factor-binding protein-3 protease activity in mouse thyroid.

There is evidence that the IGF system plays an important role in the growth and function of the thyroid gland. Proteolysis is an important posttranslational process that modulates the affinity of IGF binding proteins (IGFBPs) to IGFs, thus regulating their activity. IGFBP-3 has been shown to be cleaved by members of the kallikrein family, some of which are expressed in human thyroid and are characterized by regulation by steroid hormones. The aim of this study was to determine whether IGFBP-3 protease activity is present in mouse thyroid tissue and to characterize its activity by gender and nutrition. Male and female BALB/c mice, aged 16 wk, were studied in the fasted state, or after 1-h or 4-h refeeding. IGFBP protease activity was present in thyroid tissue and resulted in a decrease in IGFBP-3 affinity for IGF-I. The activity was inhibited by 10 mM ZnCl(2), activated by CaCl(2), and was substantially greater in tissue from male mice compared with that from female animals. These properties and the pattern of effect of a panel of protease inhibitors were consistent with this protease being a member of the tissue kallikrein family. Serum inhibited the proteolytic effect of thyroid extracts. There was no effect of nutrition. In conclusion, the degree of activity of IGFBP-3 protease in mouse thyroid tissue is gender specific and is likely to lead to an increased IGF availability in male mice.

Using denaturing HPLC for SNP discovery and genotyping, and establishing the linkage disequilibrium pattern for the all-trans-retinol dehydrogenase (RDH8) gene.

All-trans-retinol dehydrogenase (RDH8) is a visual cycle enzyme that reduces all-trans-retinal to all-trans-retinol. As part of an on-going effort to map genes involved in complex eye diseases, myopia in particular, using association studies, single nucleotide polymorphisms (SNPs) were identified and linkage disequilibrium (LD) pattern was established within and around the RDH8 gene. We used denaturing high-performance liquid chromatography (DHPLC) to screen SNPs in four DNA pools each consisting of DNA from five individuals and genotyped the identified SNPs in 150 Chinese subjects from Hong Kong. Fifteen SNPs were identified: seven were common with the minor allele frequency >0.05 and ten were novel. Common SNPs were included in LD and haplotype analysis using the ASSOCIATE and EH programmes. Four SNPs in the 3' region exhibited significant LD and formed a haplotype block, while three common SNPs in the 5' region did not exhibit useful LD. The LD pattern around the RDH8 gene suggested that one SNP from the 3'region and two to three SNPs from the 5' region were needed in association studies involving RDH8. Our results demonstrated the efficiency of DHPLC in screening SNPs when coupled with DNA pooling strategy and in genotyping SNPs.

The effects of nocturnal life on endocrine circadian patterns in healthy adults.

We observed the 24-hour patterns of endocrine in medical students who lived either a diurnal life or nocturnal life. Nocturnal life was designed by skipping their breakfast but consuming much (>50% of their daily food intake) in the evening and at night with the sleep from 0130 h to 0830 h the next morning. After 3 weeks in the experimental life, the 24-hour plasma concentrations of melatonin, leptin, glucose and insulin were measured every three hours. Both plasma melatonin and leptin showed peaks at 0300 h in the diurnal lifestyle group, and the night peaks decreased in the nocturnal lifestyle group. The changes in the patterns of melatonin and leptin were highly consistent with that of night-eating syndrome (NES). Plasma glucose increased after all meals in both groups. Its concentration maintained a high level in the nocturnal lifestyle group between midnight and early morning while insulin secretion decreased markedly during this period. Furthermore, the strong association between glucose and insulin in the diurnal lifestyle group after meals was damaged in the nocturnal lifestyle group. It was suggested that nocturnal life leads to the impairment of insulin response to glucose. Taking these results together, nocturnal life is likely to be one of the risk factors to health of modern people, including NES, obesity and diabetes.
