[Cloning, expression and subcellular localization of a novel human gene-RNF122].

OBJECTIVE: To clone a functionally unknown gene RNF122 and analyze its expression pattern and subcellular localization. METHODS: PCR was used to clone the novel gene-RNF122 from the mixed human tissue cDNA library. Bioinformatics analysis was used to identify structure characteristics of the gene. Northern blot was used to analyze its expression in normal tissues; RT-PCR was employed for its expression in cell lines and tumor tissues. By co-focal microscope, we identified its subcellular localization with organelle markers. RESULTS: A novel human gene-RNF122 was cloned, which was proved to have been expressed in several normal and tumor tissues and many cell lines, and localized in ER and Golgi apparatus. CONCLUSION: RNF122 is a novel gene which encodes a protein that has a classic RING domain. It is widely expressed in several tissues and cell lines. The encoded protein is localized in ER and Golgi apparatus, which indicates that it may play a role in the process of protein degradation.

Characterization of human dopamine responsive protein DRG-1 that binds to p75NTR-associated cell death executor NADE.

Expression of human dopamine responsive gene-1 (DRG-1) is up-regulated in response to treatment of dopamine in the rat astrocytes. However, its functions are not clear up to now. In the presented studies, DRG-1 was identified to be a conserved gene in the vertebrate and expressed abundantly in human testis, brain and skeletal muscle. DRG-1 was shown to interact with human p75NTR-associated cell death executor (NADE) in vivo and in vitro, and the interaction occurred in cytoplasm. The regions required for the interaction were subsequently mapped to the N-terminal of DRG-1 and the C-terminal of NADE. Furthermore, MTT assay showed that stable expression of DRG-1 in 293 cells could promote cell proliferation, and this promotion was suppressed by overexpression of NADE. In flow cytometry cell cycle analysis, overexpression of DRG-1 in 293 or PC12 cells increased the population of cells in the S phase with a concomitant decrease in G0/G1 population. These findings suggest that DRG-1 may contribute to the dopamine-induced cell growth, which is negatively regulated by NADE.

Leucine zipper-like domain is required for tumor suppressor ING2-mediated nucleotide excision repair and apoptosis.

The plant homodomain (PHD) of ING2 was shown to regulate p53-dependent apoptosis through phosphoinositides signaling. However, the role of a predicted leucine zipper-like (LZL) motif in N-terminus of ING2 is unclear. Here, we show that LZL motif is critical for the proper functions of ING2 in DNA repair, apoptosis and chromatin remodeling after UV irradiation. Deletion of LZL domain also abrogated the association between ING2 and p53, but not between ING2 and p300, suggesting that ING2 modulates p53-dependent chromatin remodeling, apoptosis and DNA repair by functioning as a scaffold protein to mediate the interaction between p53 and p300.

Dynamic changes of sandy land in northwest of Beijing, China.

The area northwest of Beijing is one of the most important regions where many organizations invest and pay most attention. The environmental problems in this region affect not only Beijing but also the surrounding area. Based on observation of the characteristics of the change in sandy land, this study classified four types of dynamic change of sandy land, including extended sandy land, the reversely changed sandy land, the potential sandy land and no change in sandy land. Then the process and the trend of changes in sandy land and their environmental impact on the area northwest of Beijing were analyzed. The results show that the area of sandy land has increased in this region in the period of 1991 to 2002. Change between sandy land and grassland was the dominant change. It is found that the monitoring zones of Hunshandake sandy land and north of Yin Shan are regions with high ratio of extended sandy land, and are connected with widespread potential change of sandy land. This implies that these two regions have a high probability of increase in sandy land in the future. On the other hand, in the monitoring zone of Horqin sandy land and Ba Shang Plateau and its surrounding area, desertification had been controlled and the area of sandy land is expected to decrease. This indicates that the direction of the sandstorm to Beijing is expected to gradually move to the northwest. Furthermore, the decreases in sandy land and the reversing change from arable land to grassland and forests in the study region will affect the land quality and atmosphere. And the logistic multiple regression (LMR) model was employed to better understand the complexity and processes of increases in sandy land. This model predicts that there is a high probability of increases in sandy land in north of Siziwang Banner, Zhengxiangbai Banner and Zhenglan Banner. Finally, suggestions to the ecological construction of the study area have been proposed.

Mechanisms for human cytomegalovirus-induced cytoplasmic p53 sequestration in endothelial cells.

Human cytomegalovirus (HCMV) infection results in endothelial dysfunction, typically known as dysregulated apoptosis, and aberrant expression and sub-cellular localization of p53, a tumor suppressor that accumulates at the late stage of infection. In this study, we examined three hypotheses that could be responsible for HCMV-induced cytoplasmic p53 accumulation at the later stage of infection: hyperactive nuclear export, cytoplasmic p53 tethering and delayed p53 degradation. Leptomycin B treatment, a nuclear export inhibitor, was unable to reduce cytoplasmic p53, thereby eliminating the hyperactive nuclear export mechanism. The findings that nascent p53 still entered nuclei after the nuclear export inhibition indicated that cytoplasmic tethering may play a minor role. Cytoplasmic p53 was still observed after the translation activities were blocked by cycloheximide. There was more than an eight-fold increase in the cytoplasmic p53 half-life with abnormal p53 ubiquitination. Taken together, these results suggest that delayed degradation could be responsible for the cytoplasmic p53 accumulation. The general slow-down of the proteasomal activity and the dysregulated p53 ubiquitination process at the later stage of infection could contribute to the reduced cytoplasmic p53 degradation and might be relevant to dysregulated endothelial apoptosis. The HCMV-induced changes in p53 dynamics could contribute to endothelial dysfunction.

Efficacy of one-day quadruple therapy for H pylori infection in Chinese patients.

AIM: To compare the efficacies of one-day quadruple therapy and seven-day triple therapy in Chinese patients. METHODS: Sixty consecutive patients with nonulcer dyspepsia and confirmed H pylori infection were randomized to receive either omeprazole 40 mg, amoxycillin 1 g, and furazolidone 100 mg, all twice a day for 7 d or omeprazole 20 mg (at breakfast and dinner), amoxicillin 1 g, furazolidone 200 mg, and colloidal bismuth subcitrate 220 mg four times for only one day. H pylori status was determined before and at least 5 weeks after therapy by endoscopy with antral and corpus biopsies for rapid urease test and histology. RESULTS: H pylori eradication was successful in 66.67% (20/30) patients in the 7-d group and in 36.67% (11/30) patients in the 1-d group (P = 0.037). Side effects were induced by the treatment in 13.3% (4/30) patients of each group, but these were all self-limiting, short-lasting, and did not require any specific treatment. CONCLUSION: The one-day quadruple therapy is less effective than the one-week regimen in curing H pylori infection in Chinese patients.

The DNA sequence, annotation and analysis of human chromosome 3.

After the completion of a draft human genome sequence, the International Human Genome Sequencing Consortium has proceeded to finish and annotate each of the 24 chromosomes comprising the human genome. Here we describe the sequencing and analysis of human chromosome 3, one of the largest human chromosomes. Chromosome 3 comprises just four contigs, one of which currently represents the longest unbroken stretch of finished DNA sequence known so far. The chromosome is remarkable in having the lowest rate of segmental duplication in the genome. It also includes a chemokine receptor gene cluster as well as numerous loci involved in multiple human cancers such as the gene encoding FHIT, which contains the most common constitutive fragile site in the genome, FRA3B. Using genomic sequence from chimpanzee and rhesus macaque, we were able to characterize the breakpoints defining a large pericentric inversion that occurred some time after the split of Homininae from Ponginae, and propose an evolutionary history of the inversion.

HCMV infection attenuates hydrogen peroxide induced endothelial apoptosis-- involvement of ERK pathway.

Human cytomegalovirus (HCMV) exerts anti-apoptotic effect during early stage of infection, which provides HCMV time for propagation. We investigated pathways mediating the resistance to H(2)O(2)-induced cell death - a self-defense mechanism to remove infected cells. We found that human aortic endothelial cells (HAECs) infected with VHL/E strain of HCMV during first 3 days were resistant to H(2)O(2) (0-2 mM) induced apoptosis. This anti-apoptotic effect may be mediated by the upregulation of Bcl-2, an anti-apoptotic protein through the activation pro-survival pathway extracellular signal regulated kinase (ERK). Through this mechanism, HCMV is able to propagate and causes endothelial dysfunction, hence vascular disease.

[Neuronal activities in the ventrolateral thalamus and basal ganglia in relation to Parkinson's disease].

OBJECTIVE: To investigate the neuronal activities in the ventrolateral thalamus (VL), internal globus pallidus (GPi), and subthalamic nucleus (STN) in relation to parkinsonian symptoms. METHODS: Twenty-seven patients with Parkinson's disease, 17 males and 10 females, aged 59 +/- 8, received stereotactic surgical treatment on the VL (n = 10), GPi (n = 10), or STN (n = 7) respectively. Microelectrode recording in the three nuclei and electromyography (EMG) on the limbs contralateral to the surgical side were performed intraoperatively. All patients were evaluated with the unified Parkinson's disease rating scale (UPDRS) in "off" medication state pre-and post-operatively. Single unit analysis and cross-correlation analysis were carried out to explore the relationship among the neuronal activities and limb EMG. One-way ANOVA was performed to study the numbers of neuron in these 3 nuclei in different discharge patterns and the improvement of the 3 main symptoms of Parkinson's disease. RESULTS: Three hundred and sixty-one neurons were obtained from 27 microrecording trajectories (146 for VL, 139 for GPi, and 76 for STN). Three patterns of neuronal activities were identified: tremor-related activities ("tremor cells", n = 159); tonic neuronal activities (n = 134), and irregular neuronal activities (n = 68). The rhythm of the tremor-related activities was correlated with the frequency of limb tremor in VL (r(2) = 0.8, P < 0.01), GPi (r(2) = 0.7, P < 0.01), and STN (r(2) = 0.5, P < 0.01). The average number of cells in each tract of VL, GPi, and STN were 9.0, 4.9, and 3.0 for tremor-related activities (ANOVA, P < 0.05); 4.2, 6.2 and 4.3 for tonic neuronal activities (ANOVA, P > 0.05) and 1.4, 2.8 and 3.7 for irregular neuronal activities (ANOVA, P < 0.05). The UPDRS revealed that VL had the highest effect on tremor improvement (91.7%) as compared to GPi (84.8%) and STN (62.7%), whereas STN and GPi had significant effects on bradykinesia improvement (70.8% and 62.3% respectively) as compared to VL (36.9%). However there was no significant difference in the improvement of rigidity among VL, GPi, and STN (56.2%, 71.3%, and 68.8% respectively, all P > 0.05). CONCLUSION: Different patterns of neuronal activities in VL, GPi and STN are likely to associate with parkinsonian symptoms. The results are important not only for the target selection, but also for understanding of neurophysiological basis underlying Parkinson's disease.

Phosphonylation mechanisms of sarin and acetylcholinesterase: a model DFT study.

Potential energy surfaces for the phosphonylation of sarin and acetylcholinesterase (AChE) have been theoretically studied at the B3LYP/6-311G(d,p) level of theory. The obtained results show that the phosphonylation process involves a two-step addition-elimination mechanism, with the first step (addition process) being the rate-determining step, while by comparison, the ensuing steps are very rapid. Stable trigonal bipyramidal intermediates are formed in the studied pathways. It is also revealed that the catalytic triad of acetylcholinesterase plays the catalytic role in the reaction by speeding up the phosphonylation process, as it does in the acylation reaction of ACh and AChE. The effect of aqueous solvation was accounted for via the polarizable continuum model. It is concluded that the enzymatic reaction here is influenced strongly by the solvent environment.

[Protective and curative effects of prophylactic administration of pulmonary surfactant on neonatal respiratory distress syndrome].

OBJECTIVE: To evaluate the protective and curative effects of prophylactic administration of pulmonary surfactant (PS) on neonatal respiratory distress syndrome (NRDS). METHODS: One hundred neonates aged 0.5 h after birth, with the gestational age < 32 w, birth weight < 1500 g, and number of gastric, stable microbubble < or = 7/mm(2) by gastric stable microbubble test (SMT), but without clinical or radiological manifestations of RDS at the admission, were randomly divided into 2 equal groups: prophylactic group (PG), receiving curosurf, a product of PS, immediately after admission; and non-prophylactic group (N-PG), receiving curosurf only after development of RDS. RESULTS: One hour after the administration of PS, the PaO(2), a/APO(2), pH, and PaO(2) of the PG group were 86.2 mm Hg +/- 8.1 mm Hg, 0.30 +/- 0.04, 7.38 +/- 0.06, and 178 +/- 37, all significantly higher than those of the non-PG group (all P < 0.01), and the PaCO(2) of the PG group was 37.3 mm Hg +/- 9.8 mm Hg, significantly lower than that of the non-PG group (53.6 mm Hg +/- 11.1 mm Hg, P < 0.01). In comparison with the level before the administration of PS (0.75 +/- 0.06), the level of FiO(2) of the 47 pediatric patients receiving mechanical ventilation decreased after the administration of curosurf time-dependently, e.g., was 0.50 +/- 0.09, 0.34 +/- 0.06, and 0.25 +/- 0.07 8, 48, and 96 hours after the administration. In comparison with the level before the administration of curosurf 9.0 +/- 1.0 cm H2O, the level of mean airway pressure (MAP) decreased time-dependently after the administration, e.g., were 7.5 +/- 0.8 and 6.0 +/- 0.3 48 and 96 hours after the administration (all P < 0.01). Compared with that before the administration of curosurf (3.02 +/- 0.2), the X-ray chest score decreased time-dependently after the administration of curosurf, e.g., were 1.89 +/- 0.34, 1.82 +/- 0.33, and 1.17 +/- 0.42 6, 12, and 72 hours after the administration (all P < 0.01). The RDS rate of the PG group was 30%, significantly lower than that of the non-PG group (P < 0.01). The severe case rate of the PG group was 20%, significantly lower than that of the N-PG group (53%, P = 0.01). The mortally of the PG group was 0, significantly lower than that of the non-PG group (P < 0.05). The total times of supplemental oxygen administration, assisted ventilation and hospitalization of the 47 patients with RDS in the PG group were significantly shortened compared with the RDS patients in the N-PG group [(3.6 +/- 1.7) d vs. (5.9 +/- 3.6) d, P < 0.05; (8.6 +/- 5.5 d vs. (14.1 +/- 6.2) d, P < 0.01; and (20.5 +/- 10.0) d vs. (32.8 +/- 17.8) d, P < 0.05). CONCLUSION: Prophylactic administration of PS to the preterm neonates with high risk of RDS effectively decreases the incidence of RDS, development of severe cases and mortality, shorten the disease course, the duration of supplemental oxygen administration and assisted ventilation, thus decreasing the potential morbidity associated with long-term oxygen supplement and assisted ventilation.

DNA strand breaks induced by near-zero-electronvolt electron attachment to pyrimidine nucleotides.

To elucidate the mechanism of DNA strand breaks by low-energy electrons (LEE), theoretical investigations of the LEE attachment-induced C(5')-O(5') sigma bond breaking of pyrimidine nucleotides (5'-dCMPH and 5'-dTMPH) were performed by using the B3LYP/DZP++ approach. The results indicate that the pyrimidine nucleotides are able to capture electrons characterized by near-0-eV energy to form electronically stable radical anions in both the gas phase and aqueous solution. The mechanism of the LEE-induced single-strand bond breaking in DNA might involve the attachment of an electron to the bases of DNA and the formation of base-centered radical anions in the first step. Subsequently, these radical anions undergo either C-O or glycosidic bond breaking, yielding neutral ribose radical fragments and the corresponding phosphoric anions or base anions. The C-O bond cleavage is expected to dominate because of its low activation energy. In aqueous solutions, the significant increases in the electron affinities of pyrimidine nucleotides ensure the formation of electronically more stable radical anions of the nucleotides. The low activation energy barriers for the C(5')-O(5') bond breaking predicted in this work are relevant when the counterions are close enough to the phosphate moiety of DNA.

The distribution and neurochemistry of the parathyroid hormone 2 receptor in the rat hypothalamus.

This study reports the distribution of parathyroid hormone 2 receptor (PTH2R)-immunoreactive fibers in the hypothalamus using fluorescent amplification immunocytochemistry. The pattern of immunolabeling is strikingly similar to that of tuberoinfundibular peptide of 39 residues (TIP39), a peptide recently purified from bovine hypothalamus and proposed to be a ligand of the PTH2R based on pharmacological data. To investigate the anatomical basis of suggestions that TIP39 affects the secretion of several hypophysiotropic hormones we performed double-labeling studies and found that only somatostatin fibers contain PTH2R in the median eminence, which suggests that somatostatin release could be directly regulated via the PTH2R. However, several hypothalamic nuclei projecting to the median eminence contain a high density of both TIP39 and PTH2R fibers and terminals. We report here, that the PTH2R terminals also contain vesicular glutamate transporter-2, and suggest that TIP39 terminals are ideally positioned to modulate glutamatergic influences on hypophysiotropic neurons.

Anticancer activity of litchi fruit pericarp extract against human breast cancer in vitro and in vivo.

Litchi fruit pericarp (LFP) extract contains significant amounts of polyphenolic compounds and exhibits powerful antioxidative activity against fat oxidation in vitro. The purpose of this study is to confirm the anticancer activity of LFP extract on human breast cancer in vitro and in vivo, and to elucidate the mechanism of its activity. Human breast cancer cells were tested in vitro for cytotoxicity, colony formation inhibition, BrdU incorporation, and gene expression profiling after treatment with LFP extract. Seven nude mice bearing human breast infiltrating duct carcinoma orthotopically were tested for its anticancer activity and expression of caspase-3 in vivo by oral administration of 0.3% (0.3 mg/ml) of LFP water-soluble crude ethanolic extract (CEE) for 10 weeks. LFP extract demonstrated a dose- and time-dependent inhibitory effect on cell growth (IC(50) = 80 microg/ml), and it significantly inhibited colony formation and BrdU incorporation of human breast cancer cells. Oligonucleotide microarray analysis identified 41(1.22%) up-regulated and 129 (3.84%) down-regulated genes after LFP water-soluble CEE treatment; the predominantly up-regulated genes were involved in various biological functions including cell cycle regulation and cell proliferation, apoptosis, signal transduction and transcriptional regulation, and extracellular matrix/adhesion molecules; and down-regulated genes were mainly associated with adhesion, invasion, and malignancy of cancer cells. A 40.70% tumor mass volume reduction and significant increase of casepase-3 protein expression were observed in vivo experiment. The findings in this study suggested that LFP extract might have potential anticancer activity on both ER positive and negative breast cancers, which could be attributed, in part, to its DNA damage effect, proliferating inhibition and apoptosis induction of cancer cells through up-regulation and down-regulation of multiple genes involved in cell cycle regulation and cell proliferation, apoptosis, signal transduction and transcriptional regulation, motility and invasiveness of cancer cells; ADP-ribosyltransferase (NAD+; poly (ADP-ribose) polymerase)-like 1 (ADPRTL1), Cytochrome P450, subfamily I (CYP1A1) and Hyaluronan-mediated motility receptor (HMMR) might be the main molecular targets at which LFP water-soluble CEE acted.

Polymeric self-assembly into micelles and hollow spheres with multiscale cavities driven by inclusion complexation.

The inclusion complexation between beta-CD (beta-cyclodextrin) and adamantyl group (ADA) is used as a driving force in constructing polymeric micelles. The micelles composed of a hydrophobic core of PtBA-ADA and a hydrophilic shell of PGMA-CD show unique properties due to the presence of beta-CDs on the micellar surface. The micelles can be converted to hollow spheres of PGMA-CD networks. The hollow spheres possess a central hole in the size of submicrometers and many cavities of beta-CDs of 0.7 nm on the surface.
