[Interactions of ovarian carcinoma cells and human peritoneal mesothelial cells involved in matrix metalloproteinases expressions of ovarian carcinoma cells].

OBJECTIVE: To investigate the interactions of ovarian carcinoma cells and human peritoneal mesothelial cells (HPMC) involved in matrix metalloproteinases (MMP) expressions of ovarian carcinoma cells. METHODS: The conditioned medium (CM) of ovarian carcinoma cell SKOV3 was tested by enzyme-linked immunosorbent assay (ELISA) for transforming growth factor beta1 (TGF-beta1). The impact of SKOV3-CM in the presence or absence of TGF-beta1 neutralizing antibody on fibronectin (Fn) gene expression of HPMC was studied by RT-PCR. HPMC were pretreated with serum-free medium, SKOV3-CM, SKOV3-CM + TGF-beta1 neutralizing antibody, and SKOV3-CM + IgG, then the supernatant was collected as HPMC-CM(1), HPMC-CM(2), HPMC-CM(3) and HPMC-CM(4). SKOV3 were incubated with different HPMC-CM, HPMC-CM(1) + antibody against Fn or HPMC-CM(1) + IgG. MMP-2 and MMP-9 gene mRNA expressions and protein expressions of SKOV3 were detected by RT-PCR and ELISA respectively. RESULTS: TGF-beta1 in SKOV3-CM was (236 +/- 22) ng/L. Fn gene mRNA expressions of HPMC before and after stimulation by SKOV3-CM were 1.328 +/- 0.025 and 2.643 +/- 0.051, and the latter was higher than the former (P < 0.05). Fn gene mRNA expressions of HPMC stimulated by SKOV3-CM + TGF-beta1 neutralizing antibody was 1.897 +/- 0.035, which was less than that of SKOV3-CM group (P < 0.01). Before SKOV3 were incubated with HPMC-CM, MMP-9 protein and mRNA expressions were (14.5 +/- 1.6) microg/L and 1.50 +/- 0.04, but protein and mRNA expressions of MMP-2 were scarcely detected. When SKOV3 were incubated with HPMC-CM(1), mRNA expressions of MMP-2 and MMP-9 were 0.226 +/- 0.012 and 2.66 +/- 0.07, protein expressions were (15.0 +/- 0.8) and (37.2 +/- 3.5) microg/L, which were all higher than those of SKOV3 without treatment of HPMC-CM (P < 0.01). Following incubation of SKOV3 with HPMC-CM(1) + antibody against Fn, mRNA expressions of MMP-2 and MMP-9 were 0.138 +/- 0.007 and 1.82 +/- 0.06, protein expressions were (8.8 +/- 0.7) and (25.8 +/- 2.5) microg/L, which decreased compared with those of HPMC-CM(1) group (P < 0.01). Following incubation of SKOV3 with HPMC-CM(2), mRNA expressions of MMP-2 and MMP-9 were 0.467 +/- 0.018 and 4.28 +/- 0.09, protein expressions were (39.3 +/- 3.6) and (62.0 +/- 5.3) microg/L, which were higher than those of HPMC-CM(1) group (P < 0.01). Following incubation of SKOV3 with HPMC-CM(3), gene expressions of MMP-2 and MMP-9 were 0.331 +/- 0.015 and 3.52 +/- 0.08, protein expressions were (27.6 +/- 1.9) and (50.0 +/- 4.1) microg/L, which decreased compared with HPMC-CM(2) group (P < 0.05), but were still higher than those of HPMC-CM(1) group (P < 0.05). CONCLUSIONS: Ovarian carcinoma cells activate HPMC through TGF-beta1, which induces higher expressions of MMP of ovarian carcinoma cells. Up-regulating Fn expression of HPMC may be one of action mechanisms of ovarian tumor cells. Fn derived from HPMC stimulates MMP-2 and MMP-9 expressions of ovarian carcinoma cells at gene and protein levels.

Adsorption of Pb, Cd to Fe, Mn oxides in natural freshwater surface coatings developed in different seasons.

Metal oxides( Fe, Mn oxides) in natural surface coatings(biofilms and associated minerals) are believed to play a significant role in the fate and transport of trace metal in aquatic environments. Seasonal variation of Fe, Mn oxides and organic materials in surface coatings, which were developed periodically on glass slides in Nanhu Lake, Jilin Province, China over the time frame of three seasons, was investigated in order to understand the influence of metal oxides on Pb and Cd adsorption to heterogeneous surface coating materials (biofilm). Pb and Cd adsorption was measured under controlled laboratory conditions(mineral salts solution with defined speciation, ionic strength 0.05 mol/L, 25 degrees C and pH 6.0). The classical Langmuir adsorption isotherm was applied to estimate equilibrium coefficients of Pb and Cd adsorption to the surface coatings. In general, components in the surface coatings varied greatly with seasons altering and obtained higher concentrations in summer while the content of iron oxides always exceeded that of manganese oxides. Correlation analyses between the maximum adsorption of Pb and Cd and components in the surface coatings developed periodically indicated that Pb phase association with Mn oxides and Cd phase association with Fe oxides as well as Mn oxides were statistically significant. Effect of Mn oxides on Cd adsorption was confirmed in view of its higher content in the surface coatings. The importance of ferromanganese oxides for Pb and Cd adsorption to the natural surface coatings developed in different seasons was evidenced.

Lead adsorption capacities of different components in natural surface coatings.

Pb adsorption capacities of Fe oxide, Mn oxide and organic materials in natural surface coatings( biofilms and associated minerals) collected in three lakes, two ponds and a river in Jilin Province, China and Cayuga Lake in US were studied. A novel extraction technique was employed to remove one or more component(s) from the surface coatings. Pb adsorption to surface coatings before and after extraction was performed to determine the adsorptive properties of the extracted component(s). The statistical analysis of observed Pb adsorption was carried out using nonlinear least squares fitting(NLSF) to estimate the Pb adsorption capacity of each component of surface coatings. For each body of water, the estimated Pb adsorption capacity of Mn oxide(mol Pb/mol Mn) was significantly higher than that of Fe oxide(mol Pb/ mol Fe). The value of estimated adsorption capacities of organic materials with the unit mol Pb per kg COD was similar to or less than that of Fe oxides with the unit mol Pb per mol Fe. Comparison of components of surface coatings in different waters showed that the estimated Pb adsorption capacities of components in surface coatings developed in different natural waters were different, especially for Mn oxides.

[A novel mutation of NPHS2 identified in a Chinese family with steroid-resistant nephrotic syndrome].

OBJECTIVE: Autosomal recessive steroid-resistant nephrotic syndrome (SRNS) is a subgroup of familial nephrotic syndrome. A causative gene has been identified, that is NPHS2, in chromosome 1q25-31, which encodes podocin. This study aimed to detect NPHS2 mutation in a Chinese family with SRNS. METHODS: Renal biopsy was performed on the proband and her sibling for routine histologic and immunohistochemical investigation and electron microscopic examination. The expressions of podocin, nephrin, alpha-actinin and WT1 in glomeruli of the proband were detected by indirect immunofluorescence. Peripheral blood samples were collected for genetic analysis from the proband and her parents, and 53 adults with normal urinalysis. Genomic DNA was isolated from peripheral blood leucocytes. Eight exons of NPHS2 were amplified by polymerase chain reaction. Mutational analysis was performed using denaturing high-performance liquid chromatography (DHPLC) and DNA fragments with aberrant elution profiles of both strands revealed by DHPLC were re-amplified and sequenced directly. RESULTS: The histologic findings on kidney biopsies were focal segmental glomerulosclerosis. In controls, the distribution of staining with P35, rabbit against a human podocin recombinant protein (amino acids 135 - 383 = all the C-terminal part of the protein downstream the transmembrane domain), and P21, rabbit against a human podocin recombinant protein (amino acids 15 - 89 = all the N-terminal part of the protein upstream the transmembrane domain) showed a linear pattern along glomerular capillary walls on glomeruli, and the fluorescent intensity of the staining with P35 was intensely positive. The fluorescent intensity of the staining with P21 was positive. In the proband, the distribution of the staining with P35 showed uneven and nonlinear, and the fluorescent intensity of the staining with P35 was weakly positive. The staining with P21 was negative. The area, location, distribution and fluorescent intensity of the staining with nephrin, alpha-actinin and WT1 on glomeruli of the proband were the same as those in the controls. The DHPLC elution profiles of exon 4 of NPHS2 from the proband and her parent were aberrant. The chromatograms by sequencing detected in the exon 4 of NPHS2 showed a composite heterozygous mutation of both 467_468insT and 503G > A in the proband, a heterozygous mutation of 503G > A in her father, and a heterozygous mutation of 467_468insT in her mother, respectively. CONCLUSION: The study demonstrated for the first time a novel mutation, 503G > A, of NPHS2 in Chinese kindred with autosomal recessive SRNS. A significantly decreased or negative expression was also revealed in glomeruli of the proband stained with two kinds of anti-podocin antibodies.

[Antineutrophil cytoplasmic autoantibody positive vasculitis induced by propylthiouracil: a case report].

OBJECTIVE: Propylthiouracil (PTU) as a drug used during the treatment of hyperthyroidism could induce antineutrophil cytoplasmic autoantibody-positive vasculitis. Here the author reported a childhood case of antineutrophil cytoplasmic autoantibody-positive vasculitis induced by PTU, which is rarely described. METHODS: The diagnosis was made according to the symptoms, signs, serum markers and renal biopsy, and the relevant literature was reviewed. RESULTS: The 12-year-old girl presented with gross hematuria, proteinuria, renal function damage [Ccr 52.46 ml/(min. 1.73 m(2))], positive antineutrophil cytoplasmic autoantibody (ANCA-MPO) (MPO ELISA 140%) and a vasculitis lesion in the renal biopsy sample. She had been treated with PTU for 5 years because of Graves disease. After the diagnosis, the PTU was withdrawn, and prednisone (40 mg/d) and cyclophosphamide (25 mg, Bid) were applied. Three weeks after the therapy with prednisone and cyclophosphamide the gross hematuria disappeared. Three months after the treatment the renal function returned to normal [Ccr 124 mg/(min.1.73 m(2))], and the titer of ANCA-MPO decreased from 140% to 57%. CONCLUSION: PTU may induce antineutrophil cytoplasmic autoantibody positive vasculitis. A right diagnosis and treatment can improve its prognosis of the disease.

[Clinical and pathological characteristics of focal segmental glomerulosclerosis in children].

OBJECTIVE: To investigate the clinical and pathological characteristics of focal segmental glomerulosclerosis (FSGS) in children. METHODS: The data of 38 children, aged from one and half to 15 years, 25 boys and 13 girls, with primary FSGS were studied retrospectively. RESULTS: Majority of the cases in this study were school-aged children. The average age of initial onset was 8.9 +/- 3.68 years. The ratio of boys to girls was 1.92. The clinical manifestation included isolated proteinuria in 3 cases, proteinuria and hematuria in 1 and nephrotic syndrome in 34 (simple type in 16 and nephritic type in 18). Of 38 cases, 24 (63%) presented with hematuria, 11 (29%) with hypertension and 7 (18%) with decreased creatinine clearance. The pathologic classification included perihilar variant in 17 cases, peripheral variant in 14 and tip variant in 7. The predominant clinical feature of children with tip variant was simple type of nephrotic syndrome (86%). Microscopic hematuria was not common (29%). Blood pressure and renal function were normal. The children with diffuse mesangial hypercellularity superimposed on changes of FSGS (in 21 of 38 cases) were more likely to have hematuria (76%) and less simple nephrotic syndrome (30%). The initial treatment response to prednisone in 34 cases with nephrotic syndrome showed sensitive in 12 cases, resistant in 21 and unknown in 1. Transition from sensitive to resistant occurred in six of 12 children. Three of 4 cases with non-nephrotic syndrome showed no response and the remaining one had unknown response. It was found that 44% of children who received cyclophosphamide and 83% of children who received pulse methylprednisolone and pulse cyclophosphamide or cyclosporin A in addition to oral steroids had complete or partial remission. Correlation analysis showed that the level of proteinuria after treatment was correlated directly with renal tubulointerstitial lesion and renal function (Pr = 0.48, P < 0.05; Pr = 0.45, P < 0.05). CONCLUSION: FSGS was common in school-aged children. The predominant presenting feature was nephrotic syndrome. Hematuria was common. Hypertension and renal insufficiency were less frequently seen. The renal biopsy showed multiple variants. Pulse methylprednisolone and pulse cyclophosphamide or cyclosporin A treatments showed relatively good response.

[Maternal plasma leptin levels and their relationship to insulin and glucose in pregnant women with gestational diabetes mellitus and gestational impaired glucose tolerance].

OBJECTIVE: To investigate the changes in leptin levels and the relationship between the substance and insulin and glucose in pregnant women with gestational diabetes mellitus (GDM) and gestational impaired glucose tolerance (GIGT). METHODS: Radio immunoassay was used to measure the leptin levels at fasting and 3 h after oral glucose load (50 g) in 36 patients with GDM and GIGT, and also in 24 normal pregnant women. In each pregnant women, fasting serum insulin levels and glycosylated haemoglobin values were measured by electrochemiluminescent immunoassays and pressure liquid chromatography (LPLC) respectively, glucose levels 1 h after the administration of glucose by glucose oxidase method. RESULTS: The serum leptin levels in pregnant women with GDM and GIGT and normal pregnant women were (14.9 +/- 4.3) micro g/L and (10.0 +/- 1.8) micro g/L, respectively, suggesting the serum leptin levels were significantly higher in pregnant women with GDM and GIGT than in normal controls. The levels of fasting serum insulin, glycosylated haemoglobin values and plasma glucose levels obtained 1h after oral administration of 50 g glucose were (12.9 +/- 4.3) mU/L, (6.1 +/- 1.1)%, (11.0 +/- 1.4) mmol/L, respectively, whereas the corresponding values in normal controls were (8.6 +/- 3.2) mU/L, (4.5 +/- 1.0)%, (7.8 +/- 1.2) mmol/L. A positive correlation was found between maternal serum leptin levels and fasting serum insulin levels in pregnant women with GDM and GIGT (r = 0.835, P < 0.01). A positive correlation was also found between maternal leptin concentrations and glycosylated haemoglobin values, as well as between leptin levels and plasma glucose levels obtained 1 h after the administration of 50g glucose in women with GDM and GIGT (r = 0.758, P < 0.01; r = 0.561, P < 0.01). CONCLUSIONS: Leptin levels are elevated in pregnant women with GDM and GIGT compared with healthy pregnant women, a positive and significant correlation was found between the maternal leptin levels and fasting insulin levels, as well as between the maternal leptin levels and plasma glucose levels obtained 1 h after the administration of 50 g glucose in women with GDM and GIGT.
