Application of maximum likelihood to direct methods: the probability density function of the triple-phase sums. XI.

The maximum-likelihood method is applied to direct methods to derive a more general probability density function of the triple-phase sums which is capable of predicting negative values. This study also proves that maximization of the origin-free modulus sum function S yields, within the limitations imposed by the assumed approximations, the maximum-likelihood estimates of the phases. It thus represents the formal theoretical justification of the S function that was initially derived from Patterson-function arguments [Rius (1993). Acta Cryst. A49, 406-409].

Synthesis and structure of the bidimensional zeolite ITQ-32 with small and large pores.

A new bidimensional zeolite containing 8R and 12R pores, denoted as ITQ-32, has been synthesized and its structure solved from powder X-ray diffraction data. This zeolite presents a relatively large pore volume (0.16 cm3/g) and pore apertures of 3.5 x 4.3 A and can be prepared as a nearly pure silica zeolite and as aluminosilicate. In the latest case, acidic properties are developed.

NOR-1 is involved in VEGF-induced endothelial cell growth.

Neuron-derived orphan receptor-1 (NOR-1) is a transcription factor over-expressed in human atherosclerotic plaques that is involved in vascular smooth muscle cell proliferation. The aim of this study was to analyze whether NOR-1 plays a role in vascular endothelial growth factor (VEGF) induced endothelial cell growth. VEGF induced an early and transient up-regulation of NOR-1 in human umbilical vein endothelial cells (HUVEC). NOR-1 up-regulation by VEGF is processed through VEGF receptor-2 (VEGFR-2) and involves different signaling pathways including increase in cytosolic Ca(2+), activation of protein kinase C and mitogen-activated protein kinase (MAPK) pathways (both extracellular-signaling regulated kinase [ERK] and p38 MAPK). VEGF induced CREB activation (phosphorylation in Ser(133)). In transfection assays, a dominant-negative of CREB inhibited NOR-1 promoter activity, while mutation of the three CRE sites in the NOR-1 promoter abolished VEGF-induced NOR-1 promoter activity. Antisense oligonucleotides against NOR-1 inhibited VEGF-induced endothelial cell growth (reduced DNA synthesis, and inhibited cell cycle progression and endothelial cell wound repair after mechanical injury). These results indicate that NOR-1 could be a key transcription factor regulating endothelial cell growth induced by VEGF.

Simvastatin inhibits NOR-1 expression induced by hyperlipemia by interfering with CREB activation.

OBJECTIVE: Our aim was to investigate whether neuron-derived orphan receptor-1 (NOR-1), an early gene induced by low density lipoproteins (LDL) in vascular smooth muscle cells (VSMC), is regulated by statins. METHODS: NOR-1 expression was analyzed in human VSMC in culture and in vivo in the aorta of diet-induced hyperlipemic pigs by RT-PCR and real-time PCR. [3H]Thymidine incorporation was used as an index of DNA synthesis. NOR-1 promoter activity was analyzed using a luciferase reporter system. Cyclic AMP response element binding protein (CREB) binding was assessed by EMSA and ELISA and CREB activation (phosphorylation in Ser133) by Western blotting. RESULTS: Simvastatin inhibited NOR-1 expression induced by LDL in VSMC and by hypercholesterolemia in the abdominal aorta of hyperlipemic pigs. The inhibition of the isoprenylation of geranylgeranylated proteins by simvastatin was key in both NOR-1 up-regulation and DNA synthesis induced by LDL. Inhibitors of RhoA (toxin B and exotoxin C3) and ROCK (Y-27632) mimicked the effect of simvastatin on NOR-1. Similarly both simvastatin treatment and cells transfected with a RhoA dominant-negative (RhoAT19N) showed inhibition of LDL-induced NOR-1 promoter activity. These effects were associated to the interference of the activation of CREB, a key transcription factor involved in NOR-1 induction. Finally, simvastatin prevented LDL induction of a reporter construct containing four consensus CRE and inhibited the expression of SMemb (a marker for dedifferentiated VSMC) dependent on CREB. CONCLUSIONS: NOR-1 is a target for simvastatin in the vascular wall. We identified NOR-1 and CREB as key transcription factors mediating the effect of statins on VSMC proliferation through a mechanism dependent on RhoA/ROCK.

Synthesis, characterization, and framework heteroatom localization in ITQ-21.

ITQ-21 has been synthesized in a wide range of compositions. By rationally modifying the synthesis variables and zeolite composition, it is possible to fine-tune the crystallite size from nanocrystals (<80 nm) up to microns and to avoid the competition of other phases such as CIT-5, SSZ-24, or a laminar phase that can also be synthesized with the same organic structure directing agent. By means of XRD and (19)F MAS NMR, Ge and Si have been localized among the different crystallographic positions, and it is shown that Ge preferentially occupies T1 positions at the D4R cages, avoiding formation of Ge-O-Ge pairs. However, at high Ge loadings (Si/Ge = 1.7), a new (19)F MAS NMR signal at -14 ppm has been observed and assigned to the presence of Ge-O-Ge in Ge-rich D4R cages. Energetic configurations obtained by theoretical calculations fully agree with experimental observations, with the following increasing order in energy for Ge substitution: T1 < T2 < Ge-O-Ge in T1 < T3.

Supramolecular self-assembled molecules as organic directing agent for synthesis of zeolites.

Solid materials with uniform micropores, such as zeolites, can act as selective catalysts and adsorbents for molecular mixtures by separating those molecules small enough to enter their pores while leaving the larger molecules behind. Zeolite A is a microporous material with a high void volume. Despite its widespread industrial use in, for example, molecular separations and in detergency, its capability as a petroleum-refining material is limited owing to its poor acid-catalytic activity and hydrothermal stability, and its low hydrophobicity. These characteristics are ultimately a consequence of the low framework Si/Al ratio (normally around one) and the resulting high cationic fraction within the pores and cavities. Researchers have modified the properties of type-A zeolites by increasing the Si/Al compositions up to a ratio of three. Here we describe the synthesis of zeolite A structures exhibiting high Si/Al ratios up to infinity (pure silica). We synthesize these materials, named ITQ-29, using a supramolecular organic structure-directing agent obtained by the self-assembly, through pi-pi type interactions, of two identical organic cationic moieties. The highly hydrophobic pure-silica zeolite A can be used for hydrocarbon separations that avoid oligomerization reactions, whereas materials with high Si/Al ratios give excellent shape-selective cracking additives for increasing propylene yield in fluid catalytic cracking operations. We have also extended the use of our supramolecular structure-directing agents to the synthesis of a range of other zeolites.

Involvement of neuron-derived orphan receptor-1 (NOR-1) in LDL-induced mitogenic stimulus in vascular smooth muscle cells: role of CREB.

OBJECTIVE: Low density lipoproteins (LDLs) modulate the expression of key genes involved in atherogenesis. Recently, we have shown that the transcription factor neuron-derived orphan receptor-1 (NOR-1) is involved in vascular smooth muscle cell (VSMC) proliferation. Our aim was to analyze whether NOR-1 is involved in LDL-induced mitogenic effects in VSMC. METHODS AND RESULTS: LDL induced NOR-1 expression in a time- and dose-dependent manner. Antisense oligonucleotides against NOR-1 inhibit DNA synthesis induced by LDL in VSMCs as efficiently as antisense against the protooncogene c-fos. The upregulation of NOR-1 mRNA levels by LDL involves pertusis-sensitive G protein-coupled receptors, Ca2+ mobilization, protein kinases A (PKA) and C (PKC) activation, and mitogen-activated protein kinase pathways (MAPK) (p44/p42 and p38). LDL promotes cAMP response element binding protein (CREB) activation (phosphorylation in Ser133). In transfection assays a dominant-negative of CREB inhibits NOR-1 promoter activity, while mutation of specific (cAMP response element) CRE sites in the NOR-1 promoter abolishes LDL-induced NOR-1 promoter activity. CONCLUSIONS: In VSMCs, LDL-induced mitogenesis involves NOR-1 upregulation through a CREB-dependent mechanism. CREB could play a role in the modulation by LDL of key genes (containing CRE sites) involved in atherogenesis.

Insertion reactions of [M(SR)3(PMe2Ph)2] with CS2 (M = Ru, Os; R = C6F4H-4, C6F5). X-ray structures of [Ru(S2CSC6F4H-4)2(PMe2Ph)2], trans-thiolates [M(SR)2(S2CSR)(PMe2Ph)2] (M = Ru; R = C6F5 and M = Os; R = C6F4H-4), and trans-thiolate-phosphine [Os(SC6F5)2(S2CSC6F5)(PMe2Ph)2].

Reactions of [M(SR)(3)(PMe(2)Ph)(2)] (M = Ru, Os; R = C(6)F(4)H-4, C(6)F(5)) with CS(2) in acetone afford [Ru(S(2)CSR)(2)(PMe(2)Ph)(2)] (R = C(6)F(4)H-4, 1; C(6)F(5), 3) and trans-thiolates [Ru(SR)(2)(S(2)CSR)(PMe(2)Ph)(2)] (R = C(6)F(4)H-4, 2; C(6)F(5), 4) or the isomers trans-thiolates [Os(SR)(2)(S(2)CSR)(PMe(2)Ph)(2)] (R = C(6)F(4)H-4, 5; C(6)F(5), 7) and trans-thiolate-phosphine [Os(SR)(2)(S(2)CSR)(PMe(2)Ph)(2)] (R = C(6)F(4)H-4, 6; C(6)F(5), 8) through processes involving CS(2) insertion into M-SR bonds. The ruthenium(III) complexes [Ru(SR)(3)(PMe(2)Ph)(2)] react with CS(2) to give the diamagnetic thiolate-thioxanthato ruthenium(II) and the paramagnetic ruthenium(III) complexes while osmium(III) complexes [Os(SR)(3)(PMe(2)Ph)(2)] react to give the paramagnetic thiolate-thioxanthato osmium(III) isomers. The single-crystal X-ray diffraction studies of 1, 4, 5, and 8 show distorted octahedral structures. (31)P [(1)H] and (19)F NMR studies show that the solution structures of 1 and 3 are consistent with the solid-state structure of 1.

Synthesis of a new zeolite structure ITQ-24, with intersecting 10- and 12-membered ring pores.

A new 10- and 12-membered ring zeolite, named ITQ-24, has been synthesized, and its structure has been solved. It has been found that this zeolite structure is topologically identical to that proposed for the hypothetical polymorph C of the SSZ-33/SSZ-26/CIT-1 family. This new zeolite has been achieved by using a rational approach of introducing Ge in the framework that has a directing effect toward zeolite structures with double-four-membered rings as secondary building units. Notoriously, active catalytic centers, such as Ti and Al, have been incorporated into this new zeolite, demonstrating that it is catalytically active for alkylation of aromatics.

Neuron-derived orphan receptor-1 (NOR-1) modulates vascular smooth muscle cell proliferation.

Vascular smooth muscle cells (VSMCs) migration and proliferation play a key role in the pathophysiology of cardiovascular disease. However, the transcription factors that regulate VSMC activation are not completely characterized. By a mRNA-differential display approach, we have identified neuron-derived orphan receptor-1 (NOR-1), a transcription factor within the NGFI-B subfamily of nuclear receptors, as a immediate-early gene in VSMCs. Two NOR-1 isoforms (alpha and beta) were identified and cloned from serum-induced porcine VSMC that shared high homology with the human isoforms. Northern blot analysis revealed a strong and transient (1 to 6 hours) upregulation of NOR-1 in both porcine and human coronary SMCs by growth factors (serum, platelet-derived growth factor-BB, and epidermal growth factor) and alpha-thrombin but not by cytokines. NOR-1 upregulation is processed through G protein-coupled receptors and tyrosine kinase receptors, and involves Ca2+ mobilization, protein kinase C activation, and the mitogen-activated protein kinase pathway. This induction was closely dependent of the cAMP response elements present in NOR-1 promoter as transfection assays indicate. Human coronary atherosclerotic lesions overexpress NOR-1, and balloon angioplasty transiently induces NOR-1 in porcine coronary arteries with a pattern similar to that observed in VSMCs in culture. Antisense oligonucleotides against NOR-1 inhibited human coronary SMC proliferation (reduced de novo DNA synthesis, cell cycle progression, and VSMC wound repair) as efficiently as antisense against the protooncogene c-fos. These results show that NOR-1 modulates VSMC proliferation, and suggest that this transcription factor may play a role in both spontaneous and accelerated atherosclerosis.

A zeolite with interconnected 8-, 10- and 12-ring pores and its unique catalytic selectivity.

One of the major goals in catalysis is to improve the capabilities of microporous materials. This can be accomplished by synthesizing new zeolites with controlled pore architectures. In particular, zeolites containing channels of different sizes in the same structure are desirable. Here, we report a zeolite containing fully interconnected 8-, 10- and 12-membered-ring pores, synthesized by combining the structure-directing effect of the organic 1,5-bis-(methylpyrrolidinium)-pentane and the framework isomorphic substitution of germanium for silicon. Analysis of the crystal structure determined by direct methods from synchrotron powder diffraction data reveals 16 independent tetrahedrally coordinated atoms. This thermally and hydrothermally stable zeolite structure, when synthesized in its acidic form by incorporating aluminium in the framework, presents unique catalytic shape-selectivity effects derived from its particular pore topology.

A large-cavity zeolite with wide pore windows and potential as an oil refining catalyst.

Crude oil is an important feedstock for the petrochemical industry and the dominant energy source driving the world economy, but known oil reserves will cover demand for no more than 50 years at the current rate of consumption. This situation calls for more efficient strategies for converting crude oil into fuel and petrochemical products. At present, more than 40% of oil conversion is achieved using catalysts based on faujasite; this zeolite requires extensive post-synthesis treatment to produce an ultrastable form, and has a large cavity accessible through four 0.74-nm-wide windows and thus limits the access of oil molecules to the catalytically active sites. The use of zeolites with better accessibility to their active sites should result in improved catalyst efficiency. To date, two zeolites with effective pore diameters exceeding that of faujasite have been reported, but their one-dimensional pore topology excludes use in oil refining. Similarly, zeolites with large pores and a three-dimensional pore topology have been reported, but in all these materials the pore openings are smaller than in faujasite. Here we report the synthesis of ITQ-21, a zeolite with a three-dimensional pore network containing 1.18-nm-wide cavities, each of which is accessible through six circular and 0.74-nm-wide windows. As expected for a zeolite with this structure, ITQ-21 exhibits high catalytic activity and selectivity for valuable products in preliminary oil refining tests.

A new interpretation and practical aspects of the direct-methods modulus sum function. VIII.

Since the first publication of the direct-methods modulus sum function [Rius (1993). Acta Cryst. A49, 406-409], the application of this function to a variety of situations has been shown in a series of seven subsequent papers. In this way, much experience about this function and its practical use has been gained. It is thought by the authors that it is now the right moment to publish a more complete study of this function which also considers most of this practical knowledge. The first part of the study relates, thanks to a new interpretation, this function to other existing phase-refinement functions, while the second shows, with the help of test calculations on a selection of crystal structures, the behaviour of the function for two different control parameters. In this study, the principal interest is focused on the function itself and not on the optimization procedure which is based on a conventional sequential tangent formula refinement. The results obtained are quite satisfactory and seem to indicate that, when combined with more sophisticated optimization algorithms, the application field of this function could be extended to larger structures than those used for the test calculations.
