Screening, optimization and artificial recombination of dsRNA fragments for RNAi-mediated pest resistance in Apolygus lucorum.

RNA interference (RNAi) is an eco-friendly strategy for pest management, with double-stranded RNA (dsRNA) as the core functional component. In this study, three RNAi target genes (Ubx, wupA and Dpp) with strong lethal effects on Apolygus lucorum were screened via microinjection. The 7-day cumulative mortalities were 56.67 ± 3.33% for dsUbx, 94.44 ± 1.11% for dswupA and 92.22 ± 1.11% for dsDpp. We optimized dsRNA sequences by removing conserved sequences in non-target organisms based on homology alignment and off-target risk analysis. The optimized fragments dswupA-OTE and dsDpp-OTE still exhibited high insecticidal activity, with 7-day cumulative mortalities of 77.78 ± 2.94% and 70.00 ± 1.93%, respectively. We also evaluated the effects of dsRNA length and target sites on RNAi efficiency and screened potent short dsRNA fragments. Novel artificially recombinant dsRNAs were constructed by assembling effective short fragments from different genes, which retained strong insecticidal activity despite shorter sequence length. This study verifies the feasibility of multi-target recombinant dsRNA for pest control and provides a theoretical basis for developing multi-gene RNAi technologies against A. lucorum.

Biodegradable heparin-loaded microspheres: carrier molecular composition and microsphere structure.

Microspheres of amphiphilic triblock polymers PLLA-PEG-PLLA were investigated as carriers for heparin delivery. Two series of PLLA-PEG-PLLA triblock were synthesized and prepared into microspheres with heparin loaded. The microspheres were hollow and the surface morphology varied from smooth to porous. The pore size increased with increasing PEG content. The microsphere size distribution showed that higher PEG content increased the average microsphere size. The release rate of heparin was closely related to the surface morphology of the microspheres. DSC spectra showed that both cold crystalline temperature (Tc) and crystalline melting temperature (Tm) of heparin-loaded microspheres were related to the copolymer composition and the Tc was lower than those of corresponding pure microspheres. A new method for exposing the animal's heart with maintenance of natural breathing.

In the experimental study on the electrophysiology and functions of heart in the animal, the thorax should be opened to expose the heart, at the same time natural breathing is maintained instead of artificial breathing. The key procedure of this new method is to avoid injuring the pleural cavity, so pneumothorax can be prevented. By means of this new method some subject studies have been finished.

[The effect of anti-ICAM-1/LAF-1 McAb on the cytotoxicity of tumor infiltrating lymphocytes from oral squamous cell carcinoma].

OBJECTIVE: To investigate the effect of anti-ICAm-1/LAF-1 McAb on the cytotoxicity of tumor infiltrating lymphocytes from oral squamous cell carcinoma. METHODS: TIL were isolated from fresh tumor tissues in 15 patients with oral squamous cell carcinoma and cultured in vitro. The expression of ICAM-1/LAF-1 in fresh and activated TIL was examined by FACS. The cytotoxicity of TIL against auto tumor cells and the level of TNF-alpha produced by TIL were compared before and after interrupted with anti-ICAM-1/LAF-1 McAb. RESULTS: The expression of ICAM-1 and LAF-1 in the activated TIL significantly increased comparing with fresh TIL (P < 0.05). Interrupted by anti-ICAM-1 McAb, the cytotoxicity of TIL against auto tumor cell was not significantly changed. Interrupted by anti-ICAM-1 and anti-LAF-1 McAb, the level of TNF-alpha produced by TIL showed no differences comparing with the control (P < 0.05), but the cytotoxicity of TIL against auto tumor cells significantly decreased (P < 0.05). CONCLUSION: The effect of anti-LAF-1 McAb may result from steric interference of antigen-nonspecific adhesion process.

Surface modification of poly (D,L-lactic acid) with chitosan and its effects on the culture of osteoblasts in vitro.

Chitosan is a good biodegradable natural polymer, widely used in biomedical fields. In this study, chitosan was used to modify the surface of poly (D,L-lactic acid) (PDLLA) in order to enhance its cell affinity. The properties of a modified PDLLA surface and control were investigated by contact angle and electron spectroscopy for chemical analysis (ESCA), which indicated the changes in surface energy and chemical structure. Scanning electron microscopy (SEM) observation displayed differences in surface morphology between the chitosan-modified film and the control. These data reflected that PDLLA films could be modified with chitosan and in turn may affect the biocompatibility of the modified films. Therefore, adhesion and growth of osteoblasts on modified PDLLA films as well as control were studied. Cell morphologies on the films were examined by SEM and cell viability was evaluated using an MTT assay; the differentiated cell function was assessed by measuring alkaline phosphatase (ALP) activity. The ALP activity of modified PDLLA films was significantly higher than that found on the control (p < 0.01). The proliferation of osteoblasts on modified films was also found to be higher than that on the control (p < 0.05), suggesting that chitosan could be used to modify PDLLA and then enhance its cell biocompatibility.
