Epi-Allele elicits compensatory expression of the non-targeted allele and prevents haploinsufficiency in dominant genetic diseases.

Epigenetic regulation may underlie asymmetric allelic expression of many genes during development and disease pathogenesis. Allele-specific epigenetic modification could provide an efficient therapy for dominant genetic diseases due to heterozygous mutations. We developed an allele-specific epigenetic editing method ("Epi-Allele") for silencing pathogenic alleles and found surprisingly elevated expression of the non-targeted alleles, leaving total gene expression unchanged. Genome-wide screening revealed that such compensated allelic expression represents a common phenomenon, suggesting that the Epi-Allele approach could avoid the haploinsufficiency induced by current allele-specific silencing therapies. This notion was validated by allele-specific epigenetic remodeling of Myh6 and MYH7 genes in ameliorating cardiac phenotypes in a hypertrophic cardiomyopathy (HCM) mouse model and HCM patient iPSC-derived cardiomyocytes, respectively. Thus, Epi-Allele offers an allele-specific haploinsufficiency-free therapeutic approach for treating dominant genetic diseases.

Full-length single-cell spatial transcriptomics reveals spatial and cell-type-specific transcript isoforms in the primate brain.

The primate brain exhibits complex RNA alternative splicing heterogeneity crucial for functional complexity, yet systematic spatial isoform characterization has been lacking. We developed Fullscope-seq, a full-length single-molecule large field-of-view spatial transcriptomics sequencing method at single-cell resolution, based on programmed concatenation cDNA for multiple long-read sequencing platforms. Applying Fullscope-seq to the macaque brain, we uncovered thousands of genes exhibiting differential transcript usage (DTU) across cortical layers, cell types and brain regions. Fullscope-seq resolved hundreds of major isoform switches across distinct brain regions and identified DTUs between superficial and deep cortical layers. Cortical layer-specific DTUs showed cell-composition dependence, whereas regional DTUs were regulated according to both cellular composition and spatial contexts. These isoform variations showed substantial enrichment for neuropsychiatric disorder-associated genes and were conserved across platforms and species. Our study establishes a scalable framework for spatial isoform analysis and provides a resource for understanding transcriptomic diversity in complex tissues.

Theoretical study of one-, two-, and three-photon absorption properties for a series of Y-shaped molecules.

In this paper, the equilibrium geometries, one-, two-, and three-photon absorption properties, and the transition nature of a series of Y-shaped molecules which possess an imidazole-thiazole core have been theoretically studied by using the parametrization model 3 and Zerner's intermediate neglect of differential overlap methods. Our calculated results have confirmed the experimental findings that the investigated molecules are all promising multiphoton absorption materials and both the two-photon absorption and the three-photon absorption cross sections are seriatim increscent along with the increase of the electron-donor strength. The nonlinear optical phenomenon originates from the intramolecular charge transfer within the pi-conjugated system. The calculated results indicate that the heterocyclic core increases the two- and three-photon absorption cross sections due to its pi-excessive nature. So it can provide more free electrons to enlarge the charge transfer within the molecule system. In addition, the design of Y shape and the sulfonyl-based electron-accepting group play a part in the enhancement of multiphoton absorption. It is notable that molecules with heterocyclic core will provide favorable condition for multiphoton absorption applications.

[A study on the expression of myeloid cell leukemia 1 proteins and survivin and their relation with B cell apoptosis in non-Hodgkin's lymphoma].

OBJECTIVE: To explore the expression of myeloid cell leukemia 1 (MCL-1) proteins and survivin and its correlation with cell apoptosis as well as with the development and progression of B cell non-Hodgkin's lymphoma (B-NHL). METHODS: TdT-mediated dUTP nick end labeling and immunohistochemistry were used to study cell apoptosis and expression of MCL-1 proteins and survivin proteins in 43 patients with B-NHL and 10 with reactive hyperplasia (RH) lymphoid tissue. RESULTS: The positive rate of MCL-1 proteins and survivin proteins was 58.1% (25/43) and 69.8% (30/43) respectively. The expression of MCL-1 proteins was not detected in RH lymphoid tissue, but that of survivin was detected in 10.0% (1/10). The expression of MCL-1 proteins in aggressive B-NHL was higher than that in indolent B-NHL (70.0 % vs 30.8 %, P < 0.05). The expression of survivin in aggressive B-NHL was also higher than that in indolent B-NHL (80.0% vs 46.2%, P < 0.05). Apoptotic index (AI) was not correlated positively with the expression of MCL-1, but correlated positively with the expression of survivin (r = 0.429, P < 0.01). MCL-1 and survivin were correlated positively in B-NHL (r = 0.598, P < 0.001). CONCLUSIONS: MCL-1 proteins as family member of BCL-2 have no influence on apoptosis but survivin may participate in the regulation mechanism of B-NHL apoptosis. It is indicated that the two proteins with a close relationship may take part in the development and progression of B-NHL.

[Simultaneous determination of twelve glucocorticoids residues in milk by ultra performance liquid chromatography-electrospray tandem mass spectrometry].

A comprehensive analytical method based on ultra performance liquid chromatography-electrospray tandem mass spectrometry (UPLC-MS/MS) with negative mode has been developed for the simultaneous determination of twelve glucocorticoids residues in milk. The multi-reaction monitoring mode was employed for the determination. Milk samples were extracted by sonication in a methanol/acetate buffer (pH 5.20) solution, and then defatted with n-hexane. Sample concentration and purification were performed using Oasis HLB, Sep-pak silica and Sep-pak amino-propyl solid phase extraction cartridges. The separation was performed on a Waters ACQUITY UPLC BEH C18 column (100 mm x 1.0 mm i. d., 1.7 microm) with gradient elution using methanol and water (containing 0.1% formic acid) at a flow rate of 0.1 mL/min. Identification of the glucocorticoids was done using retention times and the distribution of diagnostic ion pairs. Quantification of the glucocorticoids was based on the peak areas of the parent ion and a fragment ion with a higher signal. The limits of detection (LOD) of the method were from 0.02 to 0.38 microg/kg and the limits of quantification (LOQ) ranged from 0.07 to 1.27 microg/kg. Average recoveries for the twelve glucocorticoids (spiked at the levels of 2 and 0.4 microg/kg) ranged from 69.3% to 94.3%, with relative standard deviations between 3.5% and 16.7%. Routine tests showed that the method is fast, sensitive and specific for the determination of glucocorticoids residues in milk.

[Analysis of nonylphenol, octylphenol and bisphenol A in animal tissues by liquid chromatography-tandem mass spectrometry with accelerated solvent extraction].

A comprehensive analytical method based on liquid chromatography-electrospray ionization tandem mass spectrometry with negative ionization mode has been developed for measuring alkylphenols (AP) and bisphenol A (BPA) in animal tissues. Samples of animal tissues were extracted by accelerated solvent extraction with dichloromethane. Sample concentration and purification were performed using an OASIS NH2 solid extraction cartridge. The effects of mobile phase and additives on ionization were assessed. The recoveries for the compounds ranged from 88% to 101% and the relative standard deviations were below 15%. The detection limits of the method under multiple-reaction monitoring mode were 0.3, 0.05 and 0.1 microg/kg for BPA, nonylphenol (NP) and octylphenol (OP), respectively. The contents of NP ranging from 0.49 to 55.98 microg/kg were found in 27 real samples of animal tissues from Beijing market. The results indicate that the endocrine disrupting nonylphenol is ubiquitous in food of animal origin. Nonylphenol was found in all fish samples with concentration levels ranging from 9. 13 to 55.98 microg/kg.

[A clinical study on delayed encephalopathy after carbon monoxide poisoning].

OBJECTIVE: To investigate clinical and imaging characteristics of delayed encephalopathy after carbon monoxide poisoning(DEACMP). METHODS: A retrospective and comparative analysis of neurological manifestations, course, periods of false-cure, prognosis and neuroimaging findings were conducted in 198 patients with DEACMP. RESULTS: Of these patients, 60.1% obtained an initial recovery within 30 d, 23.7% within 31-60 d and 14.6% over 60 d. Total clinical rate was 32.8%, and effectiveness rate 70.1%. According to imaging findings, 15.2% patients were found to have bilateral lesions of basal ganglion, and 70.0% with lesions of subcortical white matter, 12.6% with lesions of two types above mentioned and 2.1% with no lesions. The intervals between onset of illness and initial recovery were (44.6 +/- 10.1), (38.2 +/- 11.9), (61.3 +/- 17.0) d, and (7.5 +/- 2.4) d respectively. Imaging findings can demonstrate the severity of brain injury, but not necessarily parallel to the improvement of illness, for which SPECT proves more useful. CONCLUSIONS: DEACMP is involved in ischemic brain injury and a self-limited illness.
