Combinatorial method for the study of new co-fluorescence enhancement system.

The new co-fluorescence enhancement system was found in polymer matrix and studied with a combinatorial approach based on Re(DBM)3phen (Re3+: Tb3+, La3+, Gd3+, Y3+; dbm: dibenzoylmethane; phen: phenanthroline) and Sm3+ complex co-doped poly(methyl methacrylate)(PMMA). We have presents a new methodology for the rapid optimization of the luminescence conditions of thin-film sample in arrays of microwells. Two libraries were generated in order to study the effect of the species and content of enhancing ions on the luminescence enhancing efficiency, respectively. At the optimal content of 20 wt.% Sm(DBM)3phen, the maximum sensitization efficiency of Tb(DBM)3phen is about nine times. The intramolecular and intermolecular energy transfer processes in rare-earth complex-doped PMMA are discussed. The energy transfer processes make the co-fluorescence effect come true. Although, the methodology described here was implemented for optimization of co-fluorescence conditions, it is can be further implemented for a variety of applications in which optimization of parameters can be studied in situ using spectroscopic tool.

[Single nucleotide polymorphism analysis in sow mu opioid receptor gene].

Mu opioid receptor (MOR) is a member of G protein-coupled receptor family, distributed in the pain transduction region in the brain and related to emotion and behaviour. This study was designed to investigate the Single Nucleotide Polymorphism (SNP) of Mu opioid receptor gene in various breeds,including duroc, landrace and Yorkshire. 5' UTR (untranslate region), coding region and 3' UTR of Mu opioid receptor gene were amplified by eight pairs of primers, and the Single Nucleotide Polymorphism (SNP) were detected by SSCP. Five polymorphisms were found (Genebank Accession number: AF521309). The results of chi(2) test showed that the frequencies of genotypes in different breeds were significantly different (P<0.01). The frequencies of mutation genotypes in Yorkshire were significantly higher than Duroc and Landrace. According to the above results, we can speculate the difference of the frequencies of genotypes may be the results of long term choice pressure.

[Single nucleotide polymorphism in exon partial sequence of sow kappa opioid receptor gene].

The possibility of the Kappa opioid receptor (KOR) gene as the candidate gene affecting the stereotyped behavior traits in sows was discussed in the current study. Based on the exon partial sequence of sow KOR gene, three pairs of primers were designed to analyze single nucleotide polymorphisms to develop PCR-SSCP method in Landrace, Yorkshire and Duroc breeds. One polymorphism, found in PCR product amplified with F1/R1, was caused by a single nucleotide mutation of C to T at the position of mRNA 105 compared with the sequence published in GenBank, which was a silence mutation. The results of chi2 test showed that the frequencies of genotypes in different breeds were different significantly (P<0.01). The least square analysis between three genotypes and the stereotyped behavioral traits showed that individuals with BB genotype had significantly higher (P<0.01) still standing behaviors than those with AA and AB genotype, but no difference were found in other behavioral traits. According to the above results, we may conclude that SNP of KOR gene has effect on the still standing behavioral trait of sows.

[Study on function of decoction for invigorating the kidney and improving blood circulation on rabbits blood stasis model].

OBJECTIVE: To evaluate the effect of decoction for invigorating the kidney and improving blood circulation to thrombosis on rabbits blood stasis model. METHOD: Thirty rabbits were randomly divided into normal group, model group, heavy dose group, slight dose group and xue shuan ning group. Tissue-type plasminogen activator (t-PA), plasminogen activator inhibitor (PAI), fibrinogen (Fbg) and D-dimer (DD) were investigated after those rabbits had been treated. One was selected randomly from each group to observe pathological changes. RESULT: There was significant difference in t-PA, PAI, Fbg and DD between normal group and other groups (P < 0.01). Among groups of heavy dose, slight dose, xue shuan ning and model, the statistical differences were significant, as well as among groups of heavy dose, slight dose and xue shuan ning (P < 0.05). However, there was no statistical difference between heavy dose group and slight dose group (P > 0.05). The pathological changes in model group were most serious, and those in xue shuan ning were less serious. There were slight pathological change in heavy dose group and light dose group. CONCLUSION: Models were made successfully. Heavy dose group and slight dose group have stronger effect on thrombosis than xue shuan ning group.

[Single nucleotide polymorphism analysis in sow mu opioid receptor gene exon III].

Stereotypies are the most common abnormal behaviours in sows. Stereotypies are repeated, relatively invariable sequences of movements which have no obvious purpose. Mu opioid receptor is a member of G protein-coupled receptor family, distributed in the pain transduction region in the brain and related emotion and behavior regions where influence the animal neural reaction and behavior. The possibility of the MOR gene as the candidate gene to affect the stereotyped behavior traits in sows was discussed in this study. The primer of sow MOR gene exon III partial sequence was designed to analyze single nucleotide polymorphisms by PCR -SSCP in Landrace, Yorkshire and Duroc breeds. Two polymorphisms were found, which was caused by a single nucleotide mutation of C to T and C to A at the positions of 1169 and 1226, respectively compared with the sequence in GenBank, but it was the silence mutation. The results of chi 2 test showed that the frequencies of genotypes resulted in different breeds were significantly different (P < 0.01). The least square analysis between the nucleotide acid mutant induced three genotypes and the stereotyped behavioral traits in Yorkshire showed that individuals with BB genotype have significant higher (P < 0.01) sham-chewing behavior than those with AA genotype, but not for bar-biting and standing. According to the above results, we can putatively draw the conclusion that MOR gene is probably the major gene affecting the sham-chewing behavioral traits or linked to the major gene.

[The study of yi-shen-huo-xue fang's effects on expression of GMP-140 and cleaning out the oxygenic free radicle on rabbits blood stasis model].

OBJECTIVE: To evaluate the effects of Yi-Shen-Huo-Xue Fang on expression of GMP-140 and cleaning out the oxygenic free radicle on rabbits blood stasis model. METHOD: Thirty rabbits were divided randomly into five groups as the normal group, model group, large dose of "Yi-Shen-Huo-Xue Fang" group, small dose of "Yi-Shen-Huo-Xue Fang" group and "Xue-Shuan-Xin-Mai-Ning" group. After being treated respectively, granule membrane protein 140(GMP-140), erythrocyte sueroxide dismutase (E-SOD), erythrocyte lipid peroxide(E-LPO), plasma lipid peroxide(P-LPO) were checked up. RESULT: The GMP-140, E-SOD, E-LPO, P-LPO in normal control were compared with those in model groups, With the difference(P < 0.01), model control group was compared with large dose group and small dose group (P < 0.01), with "Xue-Shuan-Xin-Mai-Ning" group(P < 0.05), large dose group was compared with "Xue-Shuan-Xin-Mai-Ning" group(P < 0.05), and large dose group were compared with small dose group (P > 0.05). CONCLUSION: The model was made successfully. Large dose group, small dose group and "xue-shuan-xin-mai-ning" group can inhibit expression of GMP-140, enhence SOD activity and decrease LPO content on blood stasis rabbit model. Large dose group and small dose group have stronger effect than "xue-shuan-xin-mai-ning" group.

Combinatorial study of cofluorescence of rare earth organic complexes doped in the poly(methyl methacrylate) matrix.

The luminescence enhancement effect of different kinds and contents of rare earth complexe (RE(DBM)3Phen, RE = Dy, La, Gd, Sm, Y; DBM = dibenzoylmethane; Phen = 1,10-phenanthroline) sensitized Eu(DBM)3Phen doped in poly(methyl methacrylate) (PMMA) matrix was investigated using the combinatorial method. The efficiency of the luminescence enhancement increases with a decrease in the weight percentage of the Eu(DBM)3Phen and an increase in the molecular weight of the PMMA in the systems. Among these sensitization ion complexes, La(DBM)3Phen shows the highest sensitization efficiency. At the optimal content of 5 wt % Eu(DBM)3Phen and 350,000 g/mol weight average molecular weight (Mw) of PMMA, the maximum sensitization efficiency of La(DBM)3Phen is approximately 20 times. We believe that the PMMA with high molecular weight enwraps the rare earth complexes and keeps the donors and acceptors close, which results in the effective intermolecular energy transfer and, consequently, the high sensitization efficiency.

Parallel solution combustion synthesis for combinatorial materials studies.

A parallel solution combustion synthesis technique was developed for combinatorial materials studies. The vigorous combustion reactions were successfully limited in the microreactors by using a substrate-net-mask microreactor system and the lowest adoptable furnace temperature. Using this technique, a luminescent materials library of Y3Al5O12/Tb(chi) was synthesized with the aid of an ink-jet delivery system. Structure and luminescence characterizations were implemented using X-ray diffraction and UV/X-ray spectroscopies, respectively. The results show that this technique is reliable and applicable to combinatorial study of powder materials with high synthesis temperature.
