OBJECTIVE: To investigate the morphological features of the coronary artery visualized in angiography in hypertensive patients. METHODS: A total of 203 patients (including 159 male and 44 female patients aged 29 to 75 years) with suspected coronary artery disease were studied during the period from November 1999 to November 2001. All the patients received clinical examination and selective coronary angiography, and were divided into hypertensive and normotensive groups for comparison. RESULTS: There were 5.7+/-1.2 vascular curves in the hypertensive group, but only 3.1+/-0.8 in normotensive group, showing significant differences between the 2 groups (P<0.01). The bifurcation lesion was found in 34.5% patients in hypertensive group, but in only 23.1% in normotensive group (P<0.01). CONCLUSION: Hypertensive patients have more cardiovascular curves and bifurcation lesions in comparison with normotensive patients.
OBJECTIVE: To evaluate the clinical application of laparoscopic vertical banded gastroplasty (LVBG) for morbid obesity. METHODS: Four patients undergoing LVBG were evaluated by comparison of the preoperative and postoperative morbidities and body weights. RESULTS: Laparoscopic surgery was successfully performed in all the 4 cases without laparotomy. The average operative time was 210 min without intraoperative or postoperative complications. In the 24-month follow-up, the body weight was reduced by over 50% of the excessive weight. CONCLUSION: LVBG is an effective and safe treatment for morbid obesity.
OBJECTIVE: To study the influence of ginkgolic acids on human tumor cells and normal cells. METHODS: Ginkgolic acids (total concentration 90%) was prepared from ginkgo sarcotestas. The inhibitive effect of ginkgolic acids on human tumor cells and normal cells lines was examined by MTT assay. RESULTS: When the concentration was 5.0 micrograms/ml, ginkgolic acids obviously inhibited the growth of tumor cells and didn't influence the normal cells. Inhibitive rate of ginkgolic acids on LTEP-a-2 was 59.1%. High-concentration ginkgolic acids had inhibitive effect on the growth of tumor cells and normal cells. CONCLUSION: Ginkgolic acids had a obvious inhibitive effect on tumor cells in vitro. When the concentration was under 5.0 micrograms/ml, ginkgolic acids didn't influence the growth of normal cells.
OBJECTIVE: In order to explore the effects of lead on the growth and development of cultured hippocampal neural cells and on the expression of Oct-2, the II subtype POU domain protein. METHODS: Experiment cell model was established using primary culture of hippocampal neural cells from SD rat embryos. Target cells were exposed to lead acetate in the different concentrations, i.e. 10(-1), 10(0), 10(1), 10(2), 10(3) micromol/L, while the control group was given the same quantity of the culture medium. The immunohistochemistry method was utilized to detect the expressions of Neurofilament (NF) and Glial Fibrillary Acidic Protein (GFAP), the markers for neuron and astrocyte, respectively, and the expression of Oct-2 as well. RESULTS: The results showed that 10 micromol/L lead acetate treatment caused diminishing of neuronal cell body and the decreases of both axon lengths and inter-cellular connections. In addition, 1 micromol/L lead acetate significantly increased the number of GFAP-positive cells compared with the control group (P < 0.05). By image analysis system, 1 micromol/L lead acetate treatment was found to induce a statistically significant increase of the positive area rate concerning Oct-2 expression in hippocampal neurons and astrocytes, while both positive area rate and integral density of light of Oct-2 expression were found to increase markedly in the groups treated by 10 micromol/L lead acetate (P < 0.01). CONCLUSIONS: Lead acetate treatment may contribute to the inhibitions of both growth and differentiation of hippocampus neurons, and to the stimulation of glial cell hyperplasia simultaneously. In addition, the CNS impairments caused by lead is partly correlated with the enhancement of Oct-2 expression.
AIM: To explore the effects of M-CSF and IL-10 on IL-12 and IL-18 production and expressions of HLA-DR and CD80 by human peripheral blood monocytes. METHODS: Human monocytes were isolated from blood of healthy donors, and cultured with M-CSF and or IL-10 alone or together. Then the monocytes and culture supernatant were collected respectively. IL-12 p40 and IL-18 levels in culture supernatant were detected by ELISA and the expressions of CD80 and HLA-DR on the monocytes were analyzed by FACS. RESULTS: (1)M-CSF induced secretion of IL-18 by the monocytes, while IL-10 inhibited the production of IL-18 and antagonized enhancement of LPS-induced IL-18 production by M-CSF. Both M-CSF and IL-10 inhibited secretion of IL-12 p40 by the monocytes and had a synegistic effect in the inhibition. (2)M-CSF could induce while IL-10 inhibited HLA-DR expression. Moreover, IL-10 had an antagonistic action on HLA-DR expression induced by M-CSF. M-CSF had no influence on CD80 expression, while IL-10 induced CD80 expression. CONCLUSION: M-CSF and IL-10 can regulate IL-12 and IL-18 production and HLA-DR and CD80 expressions by human monocytes, which may affect the activation and differentiation of T cells and T cell-mediated immune response.
OBJECTIVE: To investigate the association between the alleles of human leukocyte antigens (HLA)-DRB1 and essential hypertension (EH) in the Hans living in Yunnan of China. METHODS: The alleles of HLA-DRB1 were typed by polymerase chain reaction-sequence specific primers (PCR-SSP) technique in samples from 83 patients with essential hypertension and 91 healthy controls. All of the subjects, their grandparents and grand-grandparents are Hans living in Yunnan of China. RESULTS: The frequency of DRB1*1501/2 in the EH patients (0.219) was significantly higher than that in the controls(0.060), chi quare=18.331, P<0.01. Relative risk (RR) was 4.46, and etiologic factor (EF) was 0.34. The frequency of DRB1*0901 significantly decreased in the group of patients with EH in comparison to the controls (0.081 versus 0.192) chi-square=8.704 P<0.05. RR was 0.41 and prevention factor(PF) was 0.19. CONCLUSION: HLA-DRB1*1501/2 is associated with the susceptibility to essential hypertension in the Yunnan Hans of China. DRB1*0901 may protect one from the disease.
OBJECTIVE: To examine the changes in the function and expression of multidrug resistance gene (mdr1) and P-gly-coprotein (P-gp) in nasopharyngeal carcinoma (NPC) CNE1 cell following irradiation for determining the sequential order of radiotherapy and chemotherapy in the treatment of NPC. METHODS: The expressions of mdr1 gene and its protein P-gp as well as the function of P-gp efflux were examined in CNE1 cells before and after irradiation exposure by reverse transcriptional polymerase chain reaction (RT-PCR), Western blotting and flow cytometry, respectively. RESULTS: Irradiation of CNE1 cells induced a long-term overexpression of mdr1 gene and P-gp and reduction in intracellular daunorubicin accumulation. CONCLUSION: Irradiation decreases the chemotherapy sensitivity of CNE1 cell, and induction chemotherapy should be therefore performed before radiotherapy in the treatment of advanced NPC.
OBJECTIVE: Explore the effects of lead on the expression of Brn-3a transcription factor and the induction of apoptosis in vitro. METHODS: Experiment cell model was established using primary culture of hippocampus neurons of SD rat embryos. Target cells were exposed to lead acetate with the different concentration, i.e. 0.1, 1, 10, 100, 1000 mumol/L, whilst the control group was given the same quantity of the culture medium. The survival rate was determined 24 h and 48 h after exposure by MTT method. Brn-3a expression and apoptosis induction were investigated by means of immuno-histochemistry and TUNEL methods respectively. RESULTS: 1 mumol/L lead acetate treatment significantly decreased the IOD of Brn-3a positive neurons (P < 0.05) and 10 mumol/L caused a marked decline of the rate of positive area (P < 0.01) compared with the control group. In addition, 1 mumol/L lead acetate was demonstrated to be the minimal concentration to induce apoptosis of cultured hippocampus neurons with a dose response effects found in higher concentrations. CONCLUSION: It is indicated that the impairments of hippocampus caused by lead was at least partly related to the apoptosis induction by lead. Furthermore, decrease of Brn-3a expression followed by lead treatment might be probably one of the mechanisms to the apoptosis induced by lead in hippocampus.
The net photosynthetic rate (Pn) of Dolichomitriopsis diversiformis(Mitt.) was measured under controlled air temperature (Ta), plant water content (PWC) and irradiance (Photosynthetically Active Radiation, PAR) in laboratory. The results showed that Pn was closely related to PAR, Ta and PWC. The response curve of Pn to PAR was a right angle hyperbola, and the parameters were affected by Ta and PWC. The Pn reduction was great when PWC was low and Ta was high, or when PWC was high and Ta was low. The peak Pn (Pmax) was observed underweak light (PAR < 200 mumol.s-1.m-2) and relatively low (50%-80%) PWC. Under higher Ta, Pmax was higher, but decreased when Ta was higher than 25 degrees C. The PWC range within which Pmax occurred increased in accordance with the increase of PAR. When PAR was lower than 200 mumol.s-1.m-2, the Ta range within which Pmax occurred was relatively high (20-25 degrees C), and Pmax increased with the increase of PWC. When PWC was higher than 80%, Pmax decreased with the increase of PWC. The Ta range within which Pmax occurred decreased in accordance with the increase of PAR. Under moderate light (230 < PAR < 270 mumol.s-1.m-2), the sensitivity of Pn response to Ta and PWC was higher than that under higher or lower light. The Pn response to Ta and PWC was saddle-like, and its parameters were altered according to the variation in PAR.
Recently, more and more studies have discovered that some diseases result from gene defect and functional variation of ion channels, which are called ion passage diseases or ion channelopathies. Meanwhile, it has been found that even though many diseases do not fall into the category of the ion passage disease, some links or passages during the disease development are closely related with the malfunction of ion channels, and many drugs can prevent and cure these diseases by acting on ion channels. Therefore, the relationship between physiology/pathophysiology and ion channels is gradually becoming one of the hot topics in the current researches. The recent progress in the researches on the relationship between penile erection and ion channels is briefly reviewed in this article.

OBJECTIVE: To observe the effects of functional inhibition of p-glycoprotein on radiosensitivity of drug-resistant MCF-7/Adr tumor cells. METHODS: Flow cytometry was used to examine the dynamic changes of apoptosis rate and mitochondrial membrane potential (deltapsim) in verapamil-treated and untreated MCF-7/Adr cells after the X-ray exposure. RESULTS: The apoptotic rate of verapamil-treated cells was 25.53%+/-2.85%, 30.43%+/-2.21%, 39.03%+/-2.60% at 6, 12, and 24 h after X-ray exposure, respectively, and was 16.13%+/-1.16%, 21.73%+/-1.31%, and 27.53%+/-2.55% for the untreated cells. The mean fluorescence intensity of verapamil-treated cells was 75.27+/-4.90, 96.47+/-5.59, 57.77+/-2.55, respectively, at the corresponding time points, in comparison with the intensity of 54.17+/-4.05, 62.30+/-5.93, and 39.53+/-2.65 for the untreated cells. The apoptosis rate and deltapsim of the treated cells were significantly higher than those in the untreated cells at each of the specified time point (P<0.01), and in the former cells, the apoptosis rate reached the maximum at 24 h (F=47.870, P=0.000) and the deltapsim at 12 h after X-ray exposure (F=74.485, P=0.000). CONCLUSION: The radiosensitivity of MCF-7/Adr cells is increased after functional inhibition of p-glycoprotein, possibly in relation to the alteration of deltapsim.
OBJECTIVE: To investigate the effect and mechanism of alpha1-adrenoceptor blocker combined with antibiotics in the treatment of chronic prostatitis. METHODS: Eighty patients with chronic prostatitis were divided into two groups, one treated with alpha1-adrenoceptor blocker (Terazosin 2 mg qn) and Levo-ofloxacin (0.2 bid), and the other given Levo-ofloxacin (0.2 bid) alone for 6 weeks. Chronic prostatitis symptom index (CPSI), urodynamic data and prostatic secretion examination were compared before and after treatment. RESULTS: The CPSI score of the treated group decreased from 31.8 +/- 7.4 to 15.5 +/-6.6, while that of the control group decreased from 30.9 +/- 7.1 to 21.4 +/- 6.2. There was significant difference between the two groups (P < 0.05). The maximum flow rates before and after the combined treatment were 16.5 +/- 6.3 ml/s and 20.4 +/- 4.6 ml/s, while those before and after Levo-ofloxacin administration were 16.1 +/-5.8 ml/s and 17.3 +/- 6.8 ml/s. The difference was significant (P < 0.05). The maximum urethral pressure of the combined treatment group decreased from 92.5 +/- 15.3 cm H2O to 72.5 +/- 13.4 cm H2O, while that of the control group decreased from 93.2 +/- 14.8 cm H2O to 91.7 +/- 13.6 cm H2O. CONCLUSION: Alpha1-adrenoceptor blocker can lower the intraurethral pressure, which prevents urine from refluxing to the prostate. Alpha1-adrenoceptor blocker combined with antibiotics is effective for chronic prostatitis.
OBJECTIVE: To investigate the effects of chuanxiongzine on the relaxation of isolated rabbit corpus cavernosum tissues. METHODS: Observations were made on the relaxing effect of chuanxiongzine on phenylephrine (PE)-precontracted corpus cavernosum and cavernosal strips preincubated with L-NAME, and ODQ, using endothelium removal and experimental method of smooth muscle strips. The effect of chuanxiongzine on cAMP and cGMP levels in corpus cavernosum was measured by 125I radioimmunoassay. RESULTS: Chuanxiongzine had concentration-dependent relaxing effect on the cavernosal muscle contraction induced by PE (EC50 1.58 x 10(-4) mol/L). This relaxing effect was partially antagonized by ODQ and not blocked by L-NAME or endothelium removal. In PE-precontracted cavernosal strips, chuanxiongzine induced relaxation accompanied with an increase in cAMP and cGMP levels. CONCLUSION: Chuanxiongzine was effective in relaxing cavernosal muscle precontracted by PE in vitro. And its relaxing effect may be enhanced by increasing cGMP and cAMP levels in the corpus cavernosum.
BACKGROUND: New evidence suggests that the implantable cardiac defibrillator (ICD) may be effective for primary prevention of sudden death. High instrumental cost and the potentially large number of candidates will significantly impact hospital budgets. OBJECTIVE: To review the information relevant to hospital policy decisions on the use of ICDs for primary prevention. METHODS: Modelling based on an evaluation of reported studies and Canadian costs, detailed in a comprehensive review available at . RESULTS: Health outcomes: Two high-quality primary prevention trials suggest a reduction in annual mortality of 2.9% with bounds of probability (PB) of 2.0% to 3.8%, in early years. To undertake 100 implants per year may result in an increasing annual saving of life-years of acceptable quality, stabilizing by 15 years at 110 (PB 72 to 154) each year. Economic impact: The annual costs would stabilize at 15 years at approximately 4.3 million dollars. Cost-effectiveness: The incremental cost-effectiveness from the point of view of the health care system would be approximately 47,000 dollars (PB 35,000 dollars to 70,000 dollars), discounted at 3%. Ethical and legal issues: The decision should not be individual but institutional, using a process that is transparent, consistent and fair. CONCLUSIONS: The ICD can prolong life, with acceptable quality. Restriction of its use would be solely on grounds of expense.
Studies on the dynamics of NH4+, NO3- and TN concentrations in the surface water of paddy field with an independent irrigation system showed that the concentration of NH4+ and TN in surface water was increased to the maximum after 1day of each urea application, and then decreased rapidly with time. The ratio of NH4+/TN, NO3-/TN and (NH4+ + NO3-)/TN was increased firstly, and then decreased. NH4+ and TN could be used as the main indexes for the water pollution of the paddy field, and NO3- used as the assistant one. There existed a positive relation between the concentration of these three kinds of nitrogen and N application amount. Nine days after urea application was the key time to prevent the gross loss of the three kinds of nitrogen in the water surface of the paddy field. The simulation expressions of NH4+ and TN with time factor could be exponential, logarithmic, power or binomial, while NO3- had no appropriate expression. The simulation with factors of time and N application amount showed that the simulation expressions of NH4+ and TN were the same.
