An innovation in the subcutaneous island pedicle flap for cutaneous reconstruction.

The aim of this study was to describe an innovation of the transposition pedicle island flap for reconstruction of the medium-sized skin defects in the face, neck and hand. Twenty-seven cases of skin tumours and scars were surgically excised and reconstructed with this island flap. ALL flap survived with primary healing postoperatively. With a follow-up from 1 to 22 months, functionally and cosmetically satisfactory outcomes were achieved. This modification of transposition island flap provides a competitive repair alternative for the treatment of medium-sized skin defects.

[Comparative study on bone marrow megakaryocytes in children with thrombocytopenic purpura, aplastic anemia and myelodysplastic syndrome].

OBJECTIVE: Thrombocytopenic hemorrhage is one of the major appearance in pediatric hemorrhagic diseases, in which, idiopathic thrombocytopenic purpura (ITP) is the most common disease. Thrombocytopenia is the earliest phenomenon or the only one in certain phases of hemorrhagic diseases, such as ITP, aplastic anemia (AA) and myelodysplastic syndrome (MDS). By now, the pathogenesis of thrombocytopenia in different diseases has not been clearly determined. At present, it is very difficult to diagnose these diseases and estimate their prognosis with current clinical data. In this study, morphological characteristics and hematopoiesis function of bone marrow megakaryocyte in pediatric patients with ITP, AA and MDS were observed and the cause and mechanism of different thrombocytopenias were analyzed. METHODS: There were 16 children with ITP, 17 with AA and 16 with MDS in this study. CD41 McAb immunohistochemical technique was used to detect micromegakaryocyte on bone marrow smears. Plasma clot culture and CD41 McAb immunohistochemical technique were used for the MK-colony forming assay. The colony formation rate of colony formation unit-megakaryocyte (CFU-MK) and burst formation unit-megakaryocyte (BFU-MK) were counted. RESULTS: There was no statistical difference on the positive rates of micromegakaryocyte and type I lymphoid small micromegakaryocyte between groups of ITP and control. The number of micromegakaryocyte and the formation rates of CFU-MK in ITP group were significantly higher than those in control group. Among AA patients, the numbers of MK, micromegakaryocyte and the formation rates of CFU-MK, BFU-MK in vitro significantly decreased. There was no significant difference in the positive rate of micromegakaryocyte between groups of MDS and control, but the number of micromegakaryocyte and the positive rate of type I lymphoid micromegakaryocyte were significantly higher than those of control group. There was no statistical difference of the formation rate of CFU-MK between groups of MDS and control. But in 63% childhood patients, the formation rate of CFU-MK decreased, 25% increased, and 13% was normal; BFU-MK formation rate decreased significantly in MDS group. CONCLUSION: Overproliferation of bone MKs may exist in most ITP patients. For obviating the nosogenetic factors, the normal MK releasing platelet could be easily found in the culture system. But the colony formation rate of MK decreased in a few patients with CITP. The abnormality of MK might be one of the reasons for thrombocytopenia in partial patients with ITP. Underproliferation of MKs may exist in AA, but no pathosishemogenesis was found. The dysfunction of early phase MK progenitor and stem cell might be the major reason for AA, but not the abnormality of hematopoietic microenvironment. There may be two kinds of megakaryocyte clones in bone marrow of children with MDS. One may be pathologic and potentially malignant micromegakaryocytes, the other may be the normal megakaryocytic precursors. The increase of pathologic MK resulted in abnormal development and maturation of MK in bone marrow. The change of megakaryopoiesis showed different in ITP, AA or MDS. Using bone marrow smear megakaryocyte counting, small micromegakaryocyte immunohistochemical detecting and the formation rate of bone marrow MK colony assay, the different thrombocytopenia can be diagnosed during the early stage of ITP, AA or MDS.

[Study on the application for near-infrared spectroscopy quantitative analysis and selecting optimum wavelength by the MAXR regression procedure].

This paper introduces the principle and method with which the model about the quantitative analysis of Fourier transformation near infrared (NIR) spectroscopy by MAXR regression procedure can be established. In this way, the authors have selected the wave length information by Matlab language design programming in order to establish the quantitative analysis models with near infrared spectroscopy. Taking sixty-six wheat samples as experiment materials, quantitative analysis models to determine protein content are established with thirty-three samples. The relative coefficient are 0.977 1 and 0.976 5 respectively and the standard error are 0.335 and 0.340 between the predication result of the two models which include respectively two or three wave length information and Kjeldahl's value for the protein content of the another thirty-three wheat samples. When selecting the wave length information, the MAXR regression procedure can establish the optimum regression models which contain 1 or 2...or k wavelength information respectively. MAXR regression procedure is a useful method when selecting the optimum wavelength information because of its shorter computation time, and the method not only can carefully select the essential wavelength information to establish NIR spectroscopy quantitative analysis models of resisting multicollinearity information disturbance, but also to establish the work for selecting optimum wavelength information which can direct to design the special NIR analysis instrument for analyzing specific component in the special samples.

[Applied study on support vector machine (SVM) regression method in quantitative analysis with near-infrared spectroscopy].

This paper introduced the application of support vector machines(SVM) regression method based on statistics studytheory to the quantitative analysis with near-infrared (NIR) spectroscopy. Sixty-six wheat samples were used as experimental materials, and thirty-three of them were used as calibration samples. The protein contents and NIR spectra of the calibration samples were used to build SVM regression models by four different kernel functions. The protein content of the predicting samples are estimated by four different SVM regression models. All of the correlation coefficients between the estimated values by different SVM regression models and the standard chemical values of protein content by Kjeldahl's method are more than 0.97. The average absolute error is less than 0.32. To investigate the predicting effect, it is compared with PLS regression models. The result suggested that the SVM regression, which was built to estimate the protein content of wheat samples, can also be used in the quantitative analysis of real samples by NIR.

[Predicting the chemical composition of intact kernels in maize hybrids by near infrared reflectance spectroscopy].

Intact-kernel samples of normal maize inbred lines and hybrids were collected from field experiments of three locations. Calibration equations were developed by partial least square regression (PLS) of chemical values of near infrared reflectance spectroscopy (NIRS) data and tested through both cross and external validation. In addition, 40 progenies of F1 and F2 generation not included in calibration and validation sets were verified to further evaluate the reliability of three calibration equations. The authors found the coefficients of correlation (r) of 0.98, 0.93 and 0.97 between NIRS predicted and actual protein, starch and oil content in these materials, respectively. However, the greatest relative errors were 2.7% (protein), 2.46% (starch) and 7% (oil). Thus, the accuracy of prediction could be comparable to chemical methods. The feasibility of developing NIRS equations with samples of inbred lines to determine grain quality of hybrids was also examined. The analysis of principal components of spectrum of the inbred lines and hybrids supported a new theory that plant spectrum properties could be heritable.

[A study on micromegakaryocyte in children with idiopathic thrombocytopenic purpura].

OBJECTIVE: Bone marrow megakaryocytes overly proliferate and abnormally develop among patients with idiopathic thrombocytopenic purpura (ITP). Previous studies showed that it resulted from the abnormal immune function of the body. But the changes in megakaryocytes, especially in micromegakaryocytes in this disease are unclear. The present study was designed to explore the growth and development status of megakaryocytes and the significance of changes in micromegakaryocytes in pediatric cases. METHODS: Routine bone marrow smears assay and enzyme labeling for micromegakaryocytes with CD41 monoclonal antibody (McAb) were performed in 46 children with ITP. The level of platelet-associated immunoglobulin (PA-Ig) was measured with ELISA. RESULTS: Among 46 children, 36 had acute ITP (AITP)and 10 chronic ITP (CITP). The number of megakaryocytes increased or was normal in 45 patients, but decreased only in 1 case of CITP. The positive rate of micromegakaryocytes and type I micromegakaryocytes was 98% (45/46) and 35% (16/46), respectively. The positive rate of type I micromegakaryocytes was higher in CITP (50%) cases than that in AITP (31%) cases, but the median of the other three types of micromegakaryocytes in CITP cases (159) was lower than that in the AITP cases (336). There was no relationship between the numbre of all types of megakaryocytes and the level of PA-Ig. CONCLUSION: Majority of patients with ITP showed an increase in micromegakaryocytes, especially in type II, III and IV. The immune disturbance might not be the only reason for ITP. The abnormality of quality of megakaryocytes might be one of the potential causes for thrombocytopenia in some cases of ITP, especially in those of CITP. The appearance and the number of type I micromegakaryocytes might reflect the prognosis of cases of ITP.

[Influence of FT-NIR spectrometer scanning requirements on the math model's precision].

This study is based on the agriculture product near infrared spectra database, which is a foundation database. The database has very important effects on agriculture products quality analysis and agriculture breeding. What the NIR researchers and NIR users care about is how to utilize information of the foundation database fully. To share the NIR resource, unifying the scanning term to get high quality spectra is the first step. This article uses wheat powder as sample to study the influence of different resolution, different He-Ne frequency and sample granularity on the wheat powder protein model. The results show that scanning sample by 4, 8 or 16 cm(-1) resolution has little influence on the wheat powder protein math model. The change in He-Ne frequency has influence to wavenumber accuracy, but when the change is within 1 cm(-1), the influence is indistinctive. For FT-NIR instruments with He-Ne to have better stability, we needn't often adjust the He-Ne wavenumber. Sample granularity has more distinctive influence on the NIR math models.

[Detection of Borna disease virus-p24 specific antibody in the sera of schizophrenic patients of China by means of Western-blot].

BACKGROUND: To investigate whether Borna disease virus (BDV) infection is related to the schizophrenic patients from China. METHODS: A reliable Western-blot method for detection of BDV-p24 antibody was established by adjusting the reaction conditions of BDV-p24 recombinant protein and specific antibodies. The sera of schizophrenic patients and normal controls from Heilongjiang Province were screened for specific BDV-p24 antibody by this method, and the BDV-p24 antibody positive sera were confirmed by the Western-blot method with sera-GST protein absorption. RESULTS: Ten of 116 (8.6%) schizophrenic patients were found to be positive for BDV-p24 specific antibody, while no BDV-p24 specific antibody was found in sera of normal controls. CONCLUSIONS: The results demonstrate that the Borna disease virus infection also exists in China, and the infection is possibly associated with schizophrenia in some way.

[The role of releasing the fibrous bundles across levator muscle in correcting congenital blepharoptosis].

OBJECTIVE: To investigate the role of releasing the fibrous bundles across the levator muscle between the medial canthus and lateral canthsus near the top of tarsus in the correction of the congenital blepharoptosis. METHODS: Twenty-seven patients with 40 eyes of blepharoptosis were undergoing the treatment. It was performed by releasing the fibrous bundles across the levator muscle between the medial canthus and lateral canthsus near the top of tarsus to correct the mild and moderate blepharoptosis. A further procedure can also be added to by folding the levator aponeurosis if necessary. In the severe blepharoptosis, the frontalis aponeurose flap may be applied for the suspension as well during the operation. RESULTS: Of the 40 eyes in 27 cases with mild, moderate and severe blepharoptosis were treated by using this method, with 38 eyes corrected satisfactorily and 2 eyes corrected mostly in the following-ups from 3 months to 1 year. CONCLUSION: The above mentioned technique may be a good, simple and effect method to corret congenital blepharoptosis.

[The rapid analysis of functional components of P. Lobata by near infrared spectrum].

The functional components of P. Lobata were analyzed rapidly by near infrared (NIR) spectral technology. The optimum conditions of mathematics model of three components (total isoflavones, puerarin, daidzin) were studied, including the sample set selection, chemical value analysis, the detection methods and conditions. The analytical results demonstrated that the correlation between the chemical value (true value) of the three components of sample set and the NIR predicated value is 0.975, 0.984, 0.996, respectively. The correlation between the chemical value of the testing sample set and the NIR predicated value is 0.982, 0.975 and 0.981, respectively. It proved that the mathematical model that we established can be used in practice.

[Study on quantitative analysis with near-infrared spectra using latent root regression model].

The latent root regression model with near-infrared spectra of 40 soybean samples was founded for analyzing the content of soybean protein in this study. The contents of protein in another 32 soybean samples were predicted by this model. The predicting results were compared with PLS, which shows that the latent root regression model can practically be used for the quantitative analysis of the biological samples with near-infrared spectra. This method is a new kind of chemometrics calibration method, which is modified from PCR. Because the method takes the role of sample composition into account when extracting the principal component from the NIR spectra of samples, the model has a good result in analyzing samples. Further more, the results showed that it is necessary to take account of the role of sample composition when building quantitative analysis model using NIR spectra.
