Development of gene vectors for pinpoint targeting to human hepatocytes by cationically modified polymer complexes.

We developed a vector that might enable gene therapy of metabolic liver disease or hepatoma. Here we demonstrate the use of cationically modified biocompatible phospholipid polymer conjugated with hepatitis B surface (HBs) antigen for the specific transfer of genes into human hepatocytes. Poly(2-methacryloyloxyethyl phosphorylcholine (MPC)- co-N,N-dimethylaminoethyl methacrylate (DMAEMA)-co- p-nitrophenylcarbonyloxyethyl methacrylate(NPMA))(polyMDN) was prepared as a frame of vector. The specific expression of sFlt-1 or GFP by polyMDN conjugated with HBs containing plasmid (plasmid/polyMDN-HBs), polyMDN containing plasmid (plasmid/polyMDN), plasmid only and PBS were assessed in tumor cells (HepG2 or WiDr) in vitro and in vivo. The histological findings, organ weight changes, and degree of liver dysfunction were examined in the mice administered by several reagents. The sFlt-1 and GFP expression was observed only in the HepG2 cells transfected with sFlt-1 or GFP/polyMDN-HBs. None of the side effects mentioned above was observed. In conclusion, these results suggest that polyMDN-HBs is a human hepatocyte-specific gene delivery vector that might not have serious side effects.

Several types of bilayer smectic liquid crystals with ferroelectric and antiferroelectric properties in binary mixture of dimeric compounds.

The mesomorphic behavior and phase structure were examined in the mixture of two kinds of dimeric compounds, alpha,omega-bis(4-alkoxyanilinebenzylidene-4'-carbonyloxy)pentane (mOAM5AMOm), by optical microscopy, X-ray diffraction, polarization switching, and second-harmonic generation measurements. One compound is 4OAM5AMO4 with a short terminal alkyl chain that forms a single-layer smectic phase (SmCAs) with a random mixing of spacer and tail groups. Another compound is 16OAM5AMO16 with a long terminal alkyl chain that forms a chiral, anticlinic, and antiferroelectric bilayer phase (SmCAb) with the bent molecules tilted to the bilayer. By mixing these two compounds, the SmCAs phase of 4OAM5AMO4 is easily destabilized, leading to the wide content region of the bilayer phases. In the bilayer regime, three other smectic phases are newly induced. Two of them are antiferroelectric and ferroelectric phases in which the molecules lie perpendicularly with respect to the layer. The other shows no polar response to an external electric field and behaves like a smectic A. The new appearance of these bilayer phases is discussed as a mixing effect of long and short tail groups.

Self-assembled lamellar nanostructures of wholly aromatic rod-rod-type block molecules.

Wholly aromatic rod-rod type di- and triblock molecules, oligo(ether sulfone)-b-oligo(ether ketone)s (OES-OEK), were synthesized to study a solid-state self-assembled nanostructure. The OES and OEK segments in the block molecules form segregated crystalline domains. The energy-filtering transmission electron microscopy images revealed that the di- and triblock OES-OEK co-oligomers formed lamellar nanostructures with a periodicity of approximately 9 and 13 nm, respectively. Interpretation of the odd-even behavior for the emergence of ferroelectricity and antiferroelectricity in bent-core mesogens.

Simple theoretical interpretation has been made on the previously reported odd-even behavior of the emergence of ferroelectricity and antiferroelectricity in homologous series of bent-core mesogens with the same chiral end chains (S,S) ; compounds with even and odd carbon numbers (including oxygen) exhibit ferroelectric and antiferroelectric B2 phases, respectively. The odd-even behavior was confirmed in newly synthesized racemic compounds with chiral end chains of R and S forms (R,S) together with (R,R) and (S,S) forms. According to our theoretical interpretation, ferroelectricity should be more stable in (R,S) compounds than in (S,S) compounds. Actually, the transition temperature from the isotropic phase to the Sm-CP phase was higher and the temperature range of the B2 phase was broader in the (R,S) compound than in the (S,S) compound.

Dual mode bioreactions on polymer nanoparticles covered with phosphorylcholine group.

We investigated the preparation of polymer nanoparticles covered with phosphorylcholine (PC) groups and the immobilization of proteins in order to observe dual mode bioreactions on the nanoparticles. For the surface modification on the nanoparticles, a water-soluble amphiphilic phospholipid polymer with PC groups as a hydrophilic moiety was synthesized. In this polymer, an active ester group, which can immobilize proteins, was introduced. Using the phospholipid polymer as a solubilizer, poly(L-lactic acid) nanoparticles were prepared from its methylene chloride solution in an aqueous medium by the solvent evaporation method. The diameter of the nanoparticles was ca. 200 nm and the surface was covered with the PC groups and active ester groups. Proteins could immobilize on the nanoparticles under mild conditions by the reaction between the active ester group and amino group in the proteins. Both an antibody and enzyme were immobilized on the nanoparticles and bioreactions such as the antigen/antibody reaction and enzymatic reaction were observed. When an antigen was added to the suspension of the nanoparticles, aggregation of the nanoparticles occurred and then they precipitated. Also, the enzymatic reaction proceeded well when the enzyme substrate was added to the suspension. Based on these results, we provided polymer nanoparticles functionalized with both the antibody and enzyme, and the dual mode bioreactions could occur. We concluded that the novel polymer nanoparticles could be used for nano-/micro-scaled diagnostic and medical treatment systems.

Low-birefringent, chiral banana phase below calamitic nematic and/or smectic C phases in oxadiazole derivatives.

Bent-shaped molecules based on the oxadiazole central core with various side wings and terminal chain groups have been synthesized, and their liquid-crystalline behavior was investigated by optical microscopic, X-ray, and electrooptic measurements. These molecules exhibit liquid-crystal polymorphism including both the calamitic and banana phases. Such a characteristic polymorphism is attributable to the larger bend angle of the oxadiazole core compared to that of the resorcinol core used in conventional banana molecules. Only one type of banana phase, designated as the Bx phase, is formed. It appears upon cooling from the nematic and smectic liquid crystals and exhibits chiral domains with a very weak birefringence (apparently optically isotropic). By applying an electric field, the Bx phase is altered to a high-birefringence B2 phase with a homochiral SmC(A)P(A) structure that exhibits an antiferroelectric response. From detailed analyses of the optical texture and X-ray patterns through the transformation from well-oriented calamitic phases, the Bx phase was found to exhibit a helical structure, which arises as a frustration from the ground-state B2 phase in such a manner that the blocks of B2 layers are twisted with respect to each other in a direction parallel to the layer plane similarly to the twisted grain boundary (TGB) phase.

Oral delivery of diclofenac sodium using a novel solid-in-oil suspension.

The present work reports on a new pharmaceutical formulation for oral delivery of diclofenac sodium (DFNa), a non-steroidal anti-inflammatory drug (NSAID). Although DFNa itself is water-soluble at neutral pH, it was readily suspended in soybean oil via complex formation with an edible lipophilic surfactant and a matrix protein. The resulting solid-in-oil (S/O) suspension containing stably encapsulated DFNa in an oil phase markedly reduced the risks for gastrointestinal ulcers upon oral administration even at the LD(50) level in rats (ca. 50 mg/kg DFNa). In addition, plasma concentration of DFNa upon administration of an S/O suspension was comparable with that of the aqueous counterpart at the same DFNa dose. These results indicate the potential use of S/O suspensions as novel oil-based pharmaceutical formulations for oral delivery of water-soluble drugs without causing severe mucitis.

Distinct layered structure with density modulation in solid phase formed from B2 phase of banana molecules.

A unique two-dimensional (2D) long-range structure has been observed in a low-temperature phase X1 for a banana molecule having bromine atom substituted on the central core using synchrotron radiation (SR) x-ray scattering measurements. The X1 phase is formed from the B2 phase with the SmC(A)P(A) structure upon cooling and then shows multiple reflections around the first layer line, which are interpreted as a peculiar frustrated structure with long-range layer modulation order. Furthermore, the observation of a well-defined (100) reflection with a spacing of 171A means that there is the electron density modulation along the layers. By coupling these reflections, a 2D lattice with a=173 A, c=53.4A, and beta=81.1 degrees, is determined where the a axis is parallel to the original layer of the phase. This unique structure with modulation can be interpreted as an undulated layer structure and suggested to be the result from deformation with polarization splay defects periodically occurring along the layer. The angle, beta=81.1 degrees, between a and c axes indicates that the position of splay defects in one layer is staggered from that in the neighboring layer. In other words, the splay defect lines run in a direction tilted by roughly 80 degrees with respect to the a axis.

Asymmetrically functional surface properties on biocompatible phospholipid polymer membrane for bioartificial kidney.

To obtain a bioartificial kidney composed of a porous polymer membrane and renal cells, a polysulfone (PSf) membrane (PSM) blended with 2-methacryloyloxyethyl phosphorylcholine (MPC) polymer was prepared. The PSM flat membrane with a porous structure could be prepared from the polymer blend containing 1 wt % of the MPC polymer in PSf by the phase inversion technique in a dry-wet process. Asymmetrical surface properties were observed on both sides of the membrane surfaces. That is, the sponge layer formed at the substrate-contacting surface of the membrane had 10-20 microm pores, but the pores in the micrometer range could not be observed for a skin layer formed at the air-contacting surface of the membrane. At the sponge layer surface, the MPC unit composition was 7 times larger than that at the skin layer surface. The amount of proteins adsorbed on the surface corresponded to the MPC unit composition. On the skin layer, a small amount of adsorbed proteins and platelet adhesion could be suppressed compared with those on the sponge layer. However, the skin layer had a moderate protein adsorption, so it showed a sufficient cytocompatibility to enable renal tubule epithelial cells to adhere and proliferate in the membrane. Thus, it functioned well as a renal tubule. Therefore, because of both its hemocompatibility and cytocompatibility, we could conclude that the PSM membrane is useful for as a renal tubule device for a bioartificial kidney.

Sequential enzymatic reactions and stability of biomolecules immobilized onto phospholipid polymer nanoparticles.

Polymer nanoparticles for sequential enzymatic reactions were prepared by combining a phospholipid polymer shell with a polystyrene core. The active ester groups for the bioconjugation and phospholipid polar groups were incorporated into the phospholipid polymer backbone using a novel active ester monomer and 2-methacryloyloxyethyl phosphorylcholine. For the sequential enzymatic reactions, acetylcholinesterase, choline oxidase, and horseradish peroxidase-labeled IgG were immobilized onto the nanoparticles. As substrates, acetylcholine chloride, choline chloride, and tetramethylbenzidine were added to the nanoparticle suspension, the acetylcholine chloride was converted to choline chloride, the choline chloride was oxidized by choline oxidase, and hydrogen peroxide was then formed as an enzymatic degradation product. The hydrogen peroxide was used for the next enzymatic reaction (oxidized by peroxidase) with tetramethylbenzidine. The sequential enzymatic reactions on the nanoparticles via degradation products (hydrogen peroxide) were significantly higher than that of the enzyme mixture. This result indicated that the diffusion pathway of the enzymatic products and the localization of the immobilized enzyme were important for these reactions. These nanoparticles were capable of facilitating sequential enzymatic reactions.

Novel biomineralization for hydrogels: electrophoresis approach accelerates hydroxyapatite formation in hydrogels.

As a first step toward hydroxyapatite (HAp) formation in agarose hydrogels, we have tailored the internal chemistry using an electrophoresis approach. HAp was formed using aqueous solutions of calcium chloride and disodium hydrophosphate, which were set in a conventional agarose electrophoresis apparatus. Calcium and phosphate ions provided cations and anions, respectively, and were shown to migrate into the agarose hydrogel toward the corresponding electrode side. HAp was formed after colliding with each ion. The time needed to reach complete HAp formation was 30 min, and 130 ng of HAp was formed in 1 mg of agarose hydrogel when the equilibrium swelling state was reached. The electrophoresis approach accelerated the HAp formation, and the linear velocity of 1 mm/min was shown to be roughly 15 times larger than that of simple diffusion (0.06 mm/min).

Biocompatible and biodegradable phosphorylcholine ionomers with reduced protein adsorption and cell adhesion.

In this paper a recently developed biodegradable phosphorylcholine ionomer (PC ionomer) was evaluated in different biological environments with a focus on the adsorption of proteins (fibrinogen) and the adhesion of cells. Our results have shown that the polar phosphoryl choline (PC) group may be enriched at the surface of cast films with an added hydrophilic environment. X-ray photoelectron spectroscopy confirmed the surface depletion of PC groups in dry conditions, as nitrogen and phosphorous atoms were found in the bulk of the material but not at the outermost surface layer. The surface enrichment leads to a strongly hydrophilic surface that prevents the adsorption of proteins and reduces the adhesion of cells. The non-functional and hydrophobic reference poly(trimethylene carbonate) (PTMC) adsorbs both proteins and cells, thus the wetting and low adhesion behavior of the PC ionomer can be attributed to the introduced PC functionality. Since the in vivo acceptance of biomaterials is determined by their ability to withstand protein adsorption the PC ionomer described in this paper is highly interesting for a number of in vivo applications in which the adsorption of proteins may be critical, for example, blood contact events.

Relation between dielectric and low-frequency Raman spectra of hydrogen-bond liquids.

We analyzed the complex dielectric and Raman spectra of hydrogen-bond liquids in the microwave to terahertz frequency range. As for water and methanol, the high-frequency component of the dielectric spectrum, i.e., the small deviation from the principal Debye relaxation, clearly corresponds to the Raman spectrum. This indicates that the cooperative relaxation, accompanied by huge polarization fluctuation, is virtually not Raman active, whereas the faster processes reflect common microscopic dynamics. For ethylene glycol, the shape of the Raman spectrum also resembles that of the high-frequency deviation of the dielectric spectrum, but, additionally, a weak manifestation of the cooperative relaxation arising from quadrupolar conformers is detected.

High functional hollow fiber membrane modified with phospholipid polymers for a liver assist bioreactor.

For practical application of a liver assist system with a tissue-conjugated hollow fiber membrane (HFM) bioreactor used in an extracorporeal therapy, it would require a highly sophisticated HFM which has both hemocompatibility on one side and cytocompatibility on the other side. In this study, we present a cellulose acetate (CA) HFM modified with 2-methacryloyloxyethyl phosphorylcholine (MPC) copolymers (PMB30 (MPC-co-n-butyl methacrylate) and PMA30 (MPC-co-methacrylic acid) for preparing a novel liver assist HFM bioreactor. A CA/PMB-PMA30 HFM modified asymmetrically on the inner and outer surface with the PMB30 and PMA30 was prepared successfully. Analysis with an X-ray photoelectron spectroscope showed that the intensity of the phosphorus atom attributed to the MPC units on the outer surface of the modified HFM was stronger than that of the inner surface. The PMA30 was immobilized on the outer surface of the CA/PMB30 blend HFM by a chemical condensation reaction. The CA/PMB-PMA30 HFM showed good water and solute permeability in comparison with the CA HFM. The morphologies of the adherent hepatocytes were round in shape in comparison with the cells that adhered on CA HFM. Furthermore, hepatocytes cultured on the inner surface of the CA/PMB-PMA30 HFM showed higher functional expression in terms of urea synthesis and albumin synthesis than that of the CA HFM.

Odd-even behavior of ferroelectricity and antiferroelectricity in two homologous series of bent-core mesogens.

Two chiral bent-core mesogens Pn-O-PIMB(n - 2)* (n = 9 and 10) and their oxygen analogues Pn-O-PIMB(n - 2)*-(n - 4)O (n = 8, 9, and 10) with omega-[(S)-amyloxy]alkoxy terminal groups were prepared, and their phase structures were investigated by means of electro-optic, polarization reversal current and second harmonic generation measurements in order to clarify the effect of the interlayer steric interaction on the emergence of polar orderings. The odd-even behavior for the alternative appearance of ferroelectricity and antiferroelectricity was observed in two homologous series; the bent-core mesogens P10-O-PIMB8*, P8-O-PIMB6*-4O, and P10-O-PIMB8*-6O in addition to the previously reported P6-O-PIMB4* and P8-O-PIMB6*, where the length of chains n is even, exhibited ferroelectric phases. On the contrary, the mesogens P7-O-PIMB5*, P9-O-PIMB7*, and P9-O-PIMB7*-5O, where n is odd, showed antiferroelectric phases. It is obvious that the interlayer steric interaction plays a major role for the emergence of a variety of phase structures.

Terahertz spectroscopy in smectic phases of a bent-core molecule.

The dynamics of a bent-core molecule in two smectic phases is studied using terahertz (THz) spectroscopy, which enables us to observe some rapid molecular motions and segmental motions reflecting intermolecular interactions. The isotropic liquid, liquid crystal and crystal phases of the bent-core liquid crystal P-8-PIMB are clearly distinguished by THz spectra. Unexpectedly, the transmittance in the B3 phase, which has been assigned to either a highly ordered smectic or crystal phase from results of x-ray diffraction analysis, is almost the same as that in the switchable liquid-crystalline B2 phase. This suggests that the molecular dynamics in the B3 phase is liquid-crystal-like on this time scale.
