[Effect of theophylline on the thyroid function in healthy subjects: comparison between the short- and long-term medication].

Effect of theophylline on the thyroid function was studied in 9 healthy subjects. Theophylline (200mg) was taken two times (AM8:00, PM10:00) and blood was collected two times (AM9:00, PM4:00) a day for five days. Five of these 9 subjects took same dose two times (AM8:00, PM10:00) a day and blood was collected once (AM9:00) a week for four weeks. In short-term medication, serum free thyroxine (FT4) increased significantly (1.19 +/- 0.19 ng/dl, before medication) in 4th day AM (1.29 +/- 0.14 ng/dl, p < 0.05), in 4th day PM (1.30 +/- 0.12 ng/dl, p < 0.05) and in 5th day AM (1.30 +/- 0.16 ng/dl, p < 0.05). Serum free triiodothyronine (FT3) increased significantly (3.9 +/- 0.36 pg/ml, before medication) in 4th day AM (4.3 +/- 0.66 pg/ml, p < 0.05) and in 5th day AM (4.3 +/- 0.61 pg/ml, p < 0.05). Serum thyroid stimulating activity (TSA), which was measured as an increase in cAMP in FRTL-5 cells, increased significantly (116.5 +/- 49.5%, before medication) in 1st day PM (152.3 +/- 60.0%, p < 0.05), in 2nd day PM (192.6 +/- 86.7%, p < 0.05), in 3rd day PM (208.5 +/- 94.1%, p < 0.05) and in 4th day AM (183.5 +/- 80.1%, p < 0.05). In contrast, serum FT4, FT3, TSH, and TSA did not increase significantly in long-term medication.(ABSTRACT TRUNCATED AT 250 WORDS)

Effect of hyperdynamic circulatory support on hepatic hemodynamics, oxygen supply and demand after massive hepatectomy.

This study was designed to clarify the effect of hyperdynamic circulatory support with dobutamine or dopamine after massive hepatectomy on hepatic hemodynamics, oxygen supply and demand, and lactate uptake. Mongrel dogs were allocated three groups: 70% hepatectomy group; a group that received dobutamine after 70% hepatectomy; and a group that received dopamine after 70% hepatectomy. Hepatic blood flow and oxygen delivery to the liver decreased after 70% hepatectomy. Although oxygen consumption in the liver also decreased, oxygen consumption per liver weight increased after massive hepatectomy; an increase of oxygen uptake ratio and a decrease of lactate uptake in the liver were observed. When dobutamine or dopamine were administered after 70% hepatectomy, hepatic blood flow increased, followed by normalization of the balance of oxygen supply and demand in the liver; lactate uptake subsequently improved in the liver. The data support the conclusion that hepatic oxygen consumption affected by dopamine and dobutamine is associated with an increase in the extraction of lactate. Hyperdynamic hepatic circulatory support was advantageous to hemodynamics and metabolism in the residual liver after massive hepatectomy.

[Experimental study of hepatic hemodynamics and hepatic oxygen consumption after hepatectomy].

This study was undertaken in order to clarify the hepatic hemodynamics and hepatic oxygen consumption in the remaining liver after 30% and 70% hepatectomy using adult mongrel dogs and moreover was investigated the improving effect of drugs (dobutamine (DOB), dopamine (DA), dibutyryl cyclic AMP (DBcAMP) and SS-094) after hepatectomy. The hepatic blood flow after hepatectomy decreased depending on the percentage of the liver resected but the hepatic blood flow per unit liver weight increased. The hepatic oxygen consumption also decreased but the hepatic oxygen consumption per unit liver weight increased, particularly after massive hepatectomy, resulting in the elevation of hepatic oxygen extraction ratio. And the uptake of lactate per unit liver weight decreased after massive hepatectomy. The administration of DOB, DA, DBcAMP and SS-094 after massive hepatectomy increased the hepatic arterial flow and portal venous flow and as a result the hepatic oxygen delivery was increased. The improvement of hepatic hemodynamics resulted in normalizing of the relation between hepatic oxygen demand and supply, and improvement of the uptake of lactate in the remaining liver. Therefore it was considered that those drugs favored the remaining hepatic hemodynamics and metabolisms after massive hepatectomy.

Radioiodination with 125I and reductive methylation with tritium of a vegetalizing inducer protein. Specific radio-activities and effect on biological activity.

An inducer protein, the vegetalizing factor, which causes amphibian ectoderm to differentiate into endoderm was labelled with carrier-free 125I (2000 Ci/m-Atom) to specific activities of 17-32 micronCi/microgram protein. The partial loss of the biological inducing activity is not caused by the iodination of the factor but is due to the oxidation and reduction steps in the iodination procedure. By reductive methylation with formaldehyde and [3H]NaBH4 under mild conditions the factor was labelled to specific activities of 0.8-2.3 micronCi/microgram protein depending on the specific activity of the [3H]NaBH4 (5-15 Ci/mmol). Under these conditions up to 25% of the amino groups of the protein were methylated without a reduction of the biological activity of the factor. The inducing activity of the protein was however considerably reduced, when under more stringent conditions 60-70% of the amino groups were methylated. It was shown by NMR spectroscopy that about one third of protein amino groups is dimethylated under these conditions. The factor could be labelled with N-(propionyloxy)succinimide without a loss of the biological activity, when very few amino groups were substituted. The experiments show that the factor can be labelled to high specific activity without loss of its inducing activity by reductive methylation, which is therefore the most suitable method.

Template active chromatin structures: degradation by deoxyribonuclease I.

Chicken embryos were pulse-labelled in vivo with [3H]uridine (10 min), the chromatin isolated and treated with DNAse I. The residual chromatin was separated from the degradation products by centrifugation. The nascent pulse-labelled RNA is completely recovered in the residual chromatin even after prolonged incubation with DNAase I, whereas the DNA is completely degraded to 80 base polynucleotide fragments and smaller fragments.
