Preservation of ultrastructure in phosphatidylcholine vesicles by tannic acid and OsO4.

To improve preservation and visualization of membrane structure in artificial phosphatidylcholine vesicles (PC liposomes), the following three fixation formulas including tannic acid (TA) and osmium (OsO4) steps were tested: (1) TA-OsO4, (2) OsO4-TA, and (3) OsO4-TA-OsO4. The TA-OsO4 method, known to stabilize PC against conventional dehydration, was inadequate for characterizing liposome structure because it induced a serious artifact that the liposomes were aggregated and fused. On the other hand, the OsO4-TA method did not cause the artifact but only a dull EM image was obtained for the membrane structure. The best results with egg PC liposomes have been obtained by the OsO4-TA-OsO4 method which gives improved preservation and enhanced contrast of membrane structure.

Programmed Escherichia coli cell lysis by expression of cloned T4 phage lysis genes.

Self-disruptive Escherichia coli that produces foreign target protein was developed. E. coli was co-transformed with two vector plasmids, a target gene expression vector and a lysis gene expression vector. The lytic protein was produced after the expression of the target gene, resulting in simplification of the cell disruption process. In this study, the expression of cloned T4 phage gene e or t was used for the disruption of E. coli that produced beta-glucuronidase (GUS) as a model target protein. The expression of gene e did not lead to prompt cell disruption but weakened the cell wall. Resuspension with deionized water facilitated cell lysis, and GUS activity was observed in the resuspended liquid. Expression of gene e at mid logarithmic growth phase was the optimal induction period for GUS production and release. On the other hand, the expression of gene t induced immediate cell lysis, and intracellular GUS was released to the culture medium. Maximum GUS production was obtained when gene t was induced at late logarithmic growth phase.

Electrical and morphological changes of human erythrocytes under high hydrostatic pressure followed by dielectric spectroscopy.

The electrical and morphological properties of human erythrocytes under high hydrostatic pressure up to 500 MPa have been studied by dielectric spectroscopy. The pressure-induced changes in the dielectric behavior of erythrocyte suspensions indicate that hydrostatic pressure causes the change in cell shape from discoidal to spherical and hemolysis at 200-300 MPa, the formation of buds and spicular processes followed by vesiculation at 300-400 MPa, and the increase in the membrane capacitance at 400-500 MPa.

Effects of hypercapnia on cochlear and cerebral blood flow in rabbits.

Cochlear blood flow (CoBF) and cerebral blood flow (CBF) were measured using laser Doppler flowmetry and the microsphere method during CO2 and O2 gas mixture inhalation in rabbits. Both methods revealed that CBF was increased by CO2 inhalation. CoBF measured by laser Doppler measurement decreased during CO2 inhalation while that measured by the microsphere method showed a slight increase, but lateral wall blood flow in the cochlea measured by the microsphere method showed no significant change. Our results obtained with laser Doppler flowmetry were not inconsistent with those reported in humans. It is considered that the rabbit, in which thick bone surrounds the cochlea, is a good model for the experimental evaluation of the laser Doppler technique used in the clinical measurement of CoBF.

Overview of clinical studies of childhood acute lymphoblastic leukemia for more than ten years by the Japanese Children's Cancer and Leukemia Study Group.

Since 1981, the Children's Cancer and Leukemia Study Group (CCLSG) has developed a series of protocols for treatment of acute lymphoblastic leukemia (ALL) in childhood. In the first randomized controlled study of the 811 protocol (1981-1983) a comparison of conventional daily 6-mercaptopurine and methotrexate with a pulsed regimen of the two drugs was performed. The superiority of the pulsed regimen was shown. In the next 841 protocol (1984-1987) a comparison of two drugs and three drugs during induction therapy was conducted. The three-drug regimen resulted in a significantly higher event-free survival (EFS) rate. In the 874 protocol (1987-1990) two regimens with or without cranial irradiation were randomly compared, and there was no significant difference between the two regimens for the standard-risk group. To further improve the EFS rate a risk group-directed protocol 911 was conducted starting in January 1991. Life-table analysis of serial CCLSG protocols revealed that the outcome of overall ALL has gradually improved with an increase of the EFS rate; 41.4% +/- 3.6% at 14 years for the 811 protocol, 51.3% +/- 3.5% at 11 years for the 841 protocol, 56.7% +/- 3.1% at 8 years for the 874 protocol, and 78.2% +/- 3.1% at 4 years for the more recent 911 protocol.
