Induction of proliferation or hypertrophy of chondrocytes in serum-free culture: the role of insulin-like growth factor-I, insulin, or thyroxine.

In bone forming cartilage in vivo, cells undergo terminal differentiation, whereas most of the cells in normal articular cartilage do not. Chondrocyte hypertrophy can be induced also in vitro by diffusible signals. We have identified growth factors or hormones acting individually on 17-d chick embryo sternal chondrocytes cultured in agarose gels under strictly serum-free conditions. Insulin-like growth factor I or insulin triggered the first steps of chondrocyte maturation, i.e., cell proliferation and increased matrix deposition while the chondrocytic phenotype was maintained. However, cells did not progress to the hypertrophic stage. Proliferation and stimulated collagen production was preceded by a lag period, indicating that synthesis of other components was required before cells became responsive to insulin-like growth factor I or insulin. Very small amounts of FBS exerted effects similar to those of insulin-like growth factor I or insulin. However, FBS could act directly and elicited hypertrophy when constituting greater than 1% of the culture media. Basic FGF has been claimed to be the most potent chondrocyte mitogen, but had negligible effects under serum-free conditions. The same is true for PDGF, a major serum-mitogen. Under the direction of thyroxine, cells did not proliferate but became typical hypertrophic chondrocytes, extensively synthesizing collagen X and alkaline phosphatase.

[Short term effects of carbon disulfide on the intracardial irritation development and transmission in the rat].

Studies were conducted into effects of acute and subacute CS2 application on intracardiac irritability and conduction in rat. Aconitine-induced arrhythmia and the coronary occlusion method were used as pathophysiological models. Under physiological conditions, short-time exposure to CS2 caused deceleration of intracardiac impulse conduction, while under pathophysiological conditions, modified arrhythmia or reduced survival rate was the result.

[Concept in the treatment of pain in patients with cancers].

Pain in cancer patients is shown to arise in different ways, demanding a flexible approach to treatment. The following procedures can be selected from, depending on the origin of the pain. (1) Systemic analgesia with peripheral or central analgesic agents combined with psychotropic preparation; (2) administration of opioids through peridural or intrathecal catheters (for pain responsive to opioids, but side effects considerable); (3) neurolyses, e.g. epigastric tumors, recurrence of rectal carcinoma; (4) cordotomy, e.g. unilateral pain in plexus infiltration; (5) operations (endoprostheses, bridging osteosynthesis in the case of bone metastases with or without fractures); (6) radiotherapy (strontium in the case of multiple bone metastases of some tumors). Other treatment methods, e.g. administration of karsil or cortisone, as adjuvant therapy require further investigation.

[Rate of contamination of intravenous indwelling cannulae with sideport access].

136 cannulae with valved side injection port were introduced intravenously into patients from the general surgery, urology and dermato-surgical service under clinical routine conditions. The cannulae were used from 1 up to 4 days for anaesthesia purposes as well as peri- and postoperative infusion therapy and injection of drugs through the sideport. After use the cannulae were removed following an aseptic procedure and examined microbiologically. 37 cannulae (27.2%) were found contaminated, 25 of these (18.3%) with more than 25 colonies. The infusion line (border of the conus, the interior of the conus and the lumen of the cannula) was 21 times (15.4%), the side injection port 6 times (4.4%) and the intravasal capillary surface 17 times (12.5%) contaminated. Contamination of the cannula or of parts of the cannula did not correlate well with observed signs of inflammation at the puncture site (45 patients = 33%). If, however, the contamination of the cannula is considered to be an infection risk, the addition of a side injection port to such a cannula does not increase the risk of infection. This risk is much greater via the infusion line.

[Studies on the determination of optimal insemination time using electrometric resistance measurements in heifers and young cows after estrus synchronization].

Studies were conducted on how to use under the conditions of industrialised production in the GDR an electrometric method proposed by Soviet scientists to determine oestrus and ovulation date in oestric-synchronised heifers and cows. "Oestrometer-2", an instrument developed in the USSR, was used for 4,818 measurements of electric resistance in 700 oestric-synchronised heifers and 39 oestric-synchronised cows. The typical course of the resistence curve was confirmed by the results. Individual data variation together with unsatisfactory data reliability of the instrument, when applied to measurement at vaginovestibular mucosa, were likely to suggest that the method could not be used with good effect for high-accuracy determination of the ovulation date and therefore, was not useful in helping to save semen pellets after oestric synchronisation.

[The effect of caffeine on the resorption and various central effects of ethanol].

Caffeine given in a dose of 400 mg one hour before 1 g/kg ethanol did not influence the course of blood alcohol levels in male volunteers. Furthermore, caffeine did not improve psychomotoric skills impaired by ethanol. Two cups of coffee ingested 30 min. after ethanol (0.5 g/kg) caused a statistically significant increase of blood ethanol levels one hour afterwards (from 0.49 to 0.61%). This may be due to an accelerated absorption of ethanol caused by the ingestion of warm fluid.

[Attempted suicide in Lübeck 1947-1968, data and tendencies (author's transl)].

Evaluation of 2937 case notes of patients admitted for attempted suicide to the department in Lübeck between 1947 and 1968. This series can be seen as resentative for the Lübeck region. During this period the number of cases has increased about fourfold and has been rising since. Mortality could be reduced to 3% or below. Earlier typical differences between the sexes have since approached each other so that in the end men and women showed the same increase by age during the third decennium. Women were admitted significantly more frequently than men. Drugs, particularly sleeping pills and transquilizers, have greatly increased in suicide attempta. Earlier attempts occurred in 30% of cases. Measured by additional diagnoses, the morbidity of the series lay be 44.8%.
